        Production and Characterization of Enzymes Involved in Chitin Catabolic Cascade from a Bacterial Strain Isolated from Soil

        The present study was aimed to isolate the bacteria producing chitinolytic enzymes including chitin deacetylase (CDA) from soil origin. About 24 bacteria were screened for their chitinolytic enzymes producing ability on the basis of colloidal chitin. Amongst one of the most potent isolates designated to JI-02 was further selected as the most prominent chitinolytic and CDAlike enzyme (CDA-X) producer based on the ability of deacetylation of an artificial substrate pNP-acetamilide. The maximum production of CDA-X in crude enzyme from JI-02 was observed in the presence of 1% colloidal chitin at 37°C and pH 6.4 after 120 h of incubation. Potent enzyme activity of CDA-X was determined toward pNP-acetamilide, demonstrating that optimal pH, temperature and ionic strength of the CDA-X were assessed to be 7.0, 52°C and 125 mM, respectively. Unexpectedly, Nacetylglucosamine (GlcNAc) was not fully converted by the action of CDA-X to glucosamine (GlcN) under the established conditions. Further study on enzyme activity toward chitin-oligosaccharides consisting of multi-N-acetylglucosamine (GlcNAc)n, n=2-5 may be necessary to elucidate the mode of action which requires the minimum size of (GlcNAc)n. However, our data suggest that CDA-X can convert chitin to chitosan with the maximum yield of approximately 0.08g/L during the fermentation of the strain in the presence of chitin. Furthermore, we found that crude enzyme showed the significant chitosanolytic activity. Taken together, our data suggest that the strain JI-02 may be a potential candidate as a particular strain for better understating of chitin catabolic pathway.

        This paper is the research to suggest for the long-term preservative media through analyzing the types of the recording media. To do so, firstly, the general properties of the recording media will be analyzed and the suggestion for the legal ground about the recording media will be done. Secondly, we divided the recording media into analogue media and digital media, and examined the types and the properties with the analysis of their merits and demerits. As the representative recording media for the research, the analogue media is classified as paper and micro film, and the digital media is divided into magnetic disk, optical disk, magneto-optical disk, and flash memory. And thirdly, based on the results of the analysis of the analogue media and the digital media, we presented the possible way for the development of the long-term preservation of the analogue media in the future. And we searched for the best possible long-term preservative media among the four digital media; magnetic disk, optical disk, magneto-optical disk, and flash memory. The development of the recording media must be constantly proceeded for the records which are produced in the past and the records that will be produced in the future. And not to be turned over the radical changes of the time, these researches for the long-term preservative media should be positively done.

      • 토양환경지도 자료를 이용한 콩밭 토양의 화학성 비교

        It is important to interpret the soil chemical properties in order to effectively manage the farmland. This study was conducted to investigate the soil chemical properties of upland soybean field according to the soil series at 32 upland fields located in the parts of Gangwon-do. Soil sampling sites were selected by using the National Statistics DB and KSIS DB that is a soil environment information system provided by the Rural Development Administration. Soil samples were collected from 0~15 cm of top soil before seeding or transplanting, air-dried, passed through a 2 mm sieve, and analyzed for soil pH, EC, organic matter, available phosphorus (P2O5), exchangeable potassium, exchangeable calcium and exchangeable magnesium. The average chemical properties have exceeded the each nutrient optimum range level of RDA, except the organic matter in experimental soil 2015. And despite application of conventional fertilization, each soil series nutrient contents were change respectively. The results of this study suggest that conventional fertilization should be applied based on the analysis about the amount of accumulated nutrient contents in the soil.

        중학교 가정과 '인간발달과 가족관계' 영역의 교육자료 (CD-Rom) 개발 및 현장 적용 연구 : 실천적 추론 수업을 중심으로

        The purpose of this study is to develop lesson plans. teaching guide, and teaching materials applying practical reasoning teaching model in the area of 'human development and family relationship' in middle school home economics. The practical reasoning teaching model consists of five factors: Desired results, Awareness of context. Alternative approaches, Consequences of action, and Action. This study based on practical reasoning has following process: Curriculum development, Developing lesson plan. teaching material, and teaching guide. Experimental teaching and evaluation, Feedback, Production of CD-Rom. Teaching guide includes lesson Plan. workbook, multimedia materials and teaching resources, Especially teaching guide in CD-Rom can be used effectively in the actual teaching. In the classroom, this teaching model accomplished active and interesting Participation of teachers and students. It is proposed that practical reasoning teaching model should be applied to other areas of home economics. In addition, various teaching materials based on Practical reasoning need to be developed.

        단측 미세혈관문합에서 종축절개 또는 타원형절제에 의한 동맥구형성이 문합부 개통성에 미치는 영향에 관한 비교연구

        The fashion of vascular anastomosis, end-to-end or end-to-side (E-to-S), are depended upon surgeon' s preference or surgical situations. In E-to-S anastomosis two different methods of arteriotomy are applicable but it has been suggested the type, either elliptical or slit arteriotomy, play a different role in the flow hemodynamics. We thought that the difference is more considerable in microvascular surgery. This study examines the effects of elliptical versus slit arteriotomy on morphologic vessel patency. Twenty male Sprague-Dawley rats were divided into two groups of ten rats each. Both carotid arteries were selected as a experimental model. During the procedures the rat brain was tolerable to ischemia and all animals were survived after operations. The morphologic analysis of anastomosis site was through resin-casting method with scanning electromicroscopic examination. The results showed notable difference between two groups in three-dimensional morphology at two-month of operation. This difference may affect the flow hemodynamics and long-term vessel patency. In microvascular anastomosis, the mortpologic difference of the elliptical arteriotomy is worse than slit arteriotomy which compromises the vessel circumference.

        GTP cyclohydrolase I (EC 3.5.4.16) catalyzes the conversion of GTP to D-erythro-7,8-dihydroneopterin triphosphates and formic acid. and thus performs the first committing step in the biosynthesis of the pteridine moiety of folk acid in microorganisms and of tetrahydrobiopterin in higher animals. GTP cyclohydrolase I isolated from Streptomyces tubercidicus was inhibited by DL-6-methyl-5,6,7,8-tetrahydropterin and xanthopterin, and was not inhibited by tetrahydrofolic acid, pterin, pterin-6-carboxylic acid, biopterin, neopterin, ribofravin. These results suggest that bacterial GTP cyclohydrolase I may be regulated by its metaboilic end product.

        알부민이 첨가된 시험관내 약역학 감염모델을 이용한 폐렴알균 치료에서 Ceftriaxone 일일 1회 요법

        배경 : Penicillin 내성 폐렴알균이 증가하는 지역에서 대안 중의 하나로 사용되는 ceftriaxone (CTR)의 적절한 용법, 용량을 파악하는 것이 중요하다. 본 연구는 임상에서 분리된 폐렴알균을 대상으로 시험관내 약역학 감염모델을 이용하여 알부민이 첨가된 경우와 첨가되지 않은 경우 CTR의 일일 1회 요법의 효과를 비교해 보고자 하였다. 재료 및 방법 : Penicillin 감수성(SM24), 중간내성(SM47), 내성(SM60)주를 대상으로 2-구획 시험관내 약역학 감염모델을 적용하였다. CTR 주입은 2 g씩 24시간마다로 사람의 약동학을 모의하였다. 알부민은 모델에서 4g/ dL가 유지되도록 하였다. 살균효과는 0, 2, 4, 6, 24, 30, 48시간째 집락수 변화로 측정하였다. 결과 : 모든 균주에서 알부민이 첨가되지 않은 경우 6시간 이내에 살균력이 관찰되었다. 최저억제농도 및 알부민 첨가유무에 관계없이 3주 모두 24시간째 살균력이 관찰되었고, 48시간째는 모두 측정한계 이하의 집락수로 감소하였다. 전 실험과정에서 CTR에 대한 내성주는 출현하지 않았다. 결론 : CTR의 일일 1회 요법은 알부민이 첨가되었을 때, 감수성, 중간내성, 내성주 모두에서 살균력 발현이 지연되었으나 최종효과는 알부민이 없을 때와 다르지 않았다. 앞으로 알부민 결합효과와 관련된 기초적인 추가 연구가 필요할 것으로 생각된다. Background : During the era of increasing penicillin resistant Streptococcus pneumoniae, it is important to have knowledge about adequate dosage and dosing interval of ceftriaxone (CTR). We examined efficacies of once-daily CTR and compared results in an in vitro pharmacodynamic infection model (IVPDIM) supplemented with albumin and those without albumin. Methods : Using three clinically isolated S. pneumoniae that were susceptible (SM24), intermediate (SM47) and resistant (SM60) against CTR, we utilized a two-compartment IVPDIM. CTR 2 g was administered intravenously every 24 h. Human albumin was added with concentration of 4 g/dL. Samples were removed at multiple time points over a 48-h period to determine the colony counts. Results : In SM24 and SM60, bactericidal effects were observed within 6 hours in groups without albumin. The number of colonies during 1st 6 hours were more decreased in albumin-free groups than in albumin-supplemented groups (P<0.05). In SM47, similar results were found during 1st 6 hours (P=0.03). But, regrowth was observed in albumin supplemented group at 30 h. Irrespective of results of minimal inhibitory concentrations and albumin supplementation, bactericidal effects were shown at 24 h in all 3 strains. All groups were decreased below the detection limit at 48 h. Development of resistance was not detected throughout the entire study period in either strain. Conclusions : Although extents of killing in albumin supplemented broth of once-daily CTR dosing were delayed in all 3 strains compared with those of albumin free broth, final efficacies were not different between the two groups.

        Because many centers jointly select the subject, multi-center clinical study can promptly recruit various patient groups into the research, thus being advantageous in terms of generalization. However, prospective clinical study has difficulties in implementation, because it takes a lot of efforts to trace a large scale of subjects. The purpose of this study is to develop a web database system suitable for multi-center prospective clinical study. This study has presented three basic strategies-accurate data input, easy data tracing, and real-time data verification-to supplement the demerits of prospective clinical study. The developed system has been applied in the clinical study proceeded by the study group for Respiratory failure which is affiliated to The Korean Academy of Tuberculosis and Respiratory Diseases's Association for Research on Respiratory Failure and has been verified of its performance.

        중학교 가정교과의 열린교육 활성화를 위한 수업 모듈 및 웹 기반 교육자료 개발

        This study is about open and secondary level Home Economics Education which is applicable to field and which includes the characteristics, purpose and problems of each open education. secondary education and home economics education. Based on the ideology of open education, this study will identify the meaning of openness in terms of Home Economics Education reconstructing meaning. purpose, characteristics, curriculum paradigm and curriculum. The instructional model and module which use web, will be Presented as the results of the reconstruction works. The open instructional module in Home Economics Education, which will be operated on the web, is consisted of 1)setting the orientation of curriculum development, 2)instructional model development. 3)rebuilding the Home Economics Education curriculum 4)development of instructional modules and web-based materials and 5)construction of the web-site . Six instructional module begin with critical/emancipatory curriculum Paradigm in open Home Economics Education. use module as a unit of class management instead of 1 class. facilitates nest-type integration which is applied from open education keeping the nature of Home Economics Education. and focus on practical problem and problem solving through reconceptualization with dialectical perspective on individual, family and society. The practical problem solving model has four steps : Identifying practical problems. Understanding the contexts of Problem, Searching for solutions and examining its consequences . Action and evaluation . The web-site has to be developed based on the six instructional module includes those issues : self-identification for reinforcing autonomy and self-esteem. living with famity. living with friends, autonomous living. healthy living, and understanding sexuality.
