        고온가열처리에 의한 바다방석고둥 ( Omphalius pfeifferi capenteri ) 의 식품성분 변화

        바다방석고둥을 원료로 한 신제품 개발에 관한 일련의 기초 연구로 저장성 부여를 위하여 시도하는 고온가열처리 조건 (F_0 5분, F_0 10분, F_0 15분 및 F_0 20분)에 따른 바다방석고둥의 성분변화에 대하여 살펴보았다. 바다방석고둥 통조림은 살균을 5분 이상 처리하는 경우 저장 중 팽창관이 발생하지 않았고, 또한 생균수가 검출되지 않아 저장성이 있었다. 고온가열처리 정도가 증가할수록 바다방석고둥의 일반성분 중 수분함량은 감소하였고, 조단백질 함량은 증가하였으며, 조지방 및 조회분 함량은 거의 변화가 없었다. 또한 바다방석 통조림은 고온가열처리 정도가 증가할수록 이취에 관여하는 휘발성염기질소 함량, pH와 색조에 관여하는 갈변도는 증가하였고, 영양에 관여하는 구성아미노산, 무기질과 맛에 관여하는 유리아미노산, TMAO, total creatinine 및 수율 둥은 감소하였다. 이들을 색조, 조직감 및 맛에 대하여 관능검사한 결과 자숙 처리한 시료에 비하여 모든 항목에서 F_0 10분까지는 차이가 없었으나, 그 이상 가열처리한 경우 품질이 저하하였다고 인지되었다. 이상의 결과로 미루어 보아 바다방석고둥을 고온가열처리하여 신제품을 개발하고자 하는 경우 F_0 값이 10분 이상이면 바람직하지 않은 것으로 판단되었다. The top shell, Omphalius pfeifferi capenteri meat vacuum-packed in can (diameter× height, 74.1 ㎜ × 50.7 ㎜) were heated at 115℃ up to F_0 values of 5 min, 10 min, 15 min and 20 min, and the changes in food components were studied. After 14 days storage at 37℃ and 55℃, no growth of microorganism and panelling were recognized from the canned meats which were sterlized at 115℃ with F_0 value of 5 min and over. In the case of proximate composition of the canned meats, the moisture content decreased with the increase of F_0 value, while crude protein increased. The increase of volatile basic nitrogen content, pH and degree of browning and the decrease of mineral, total amino acid, free amino add, trimethylamine oxide, total creatinine contents and yields were observed during thermal processing. In sensory evaluation on color, texture and taste in the canned meats, no significant difference was observed among a boiled sample and the canned meats heated at F_0 value of 10 min and below. But, in the canned meats heated at F_0 value of over 15 min, its sensory scores decreased with the increase of F_0 value. From these results, the reasonable F_0 value for preparation of the heat-treated top shell meats was in the range of 5∼10 min.

        Accuracy of cardiac output measurements during off-pump coronary artery bypass grafting: according to the vessel anastomosis sites

        Background: During beating heart surgery, the accuracy of cardiac output (CO) measurement techniques may be influenced by several factors. This study was conducted to analyze the clinical agreement among stat CO mode (SCO), continuous CO mode (CCO), arterial pressure waveform-based CO estimation (APCO), and transesophageal Doppler ultrasound technique (UCCO) according to the vessel anastomosis sites. Methods: This study was prospectively performed in 25 patients who would be undergoing elective OPCAB. Hemodynamic variables were recorded at the following time points: during left anterior descending (LAD) anastomosis at 1 min and 5 min; during obtuse marginal (OM) anastomosis at 1 min and 5 min: and during right coronary artery (RCA) anastomosis at 1 min and 5 min. The variables measured including the SCO, CCO, APCO, and UCCO. Results: CO measurement techniques showed different correlations according to vessel anastomosis site. However, the percent error observed was higher than the value of 30% postulated by the criteria of Critchley and Critchley during all study periods for all CO measurement techniques. Conclusions: In the beating heart procedure, SCO, CCO and APCO showed different correlations according to the vessel anastomosis sites and did not agree with UCCO. CO values from the various measurement techniques should be interpreted with caution during OPCAB.         PTC/NTC Behaviors of Nanostructured Carbon Black-filled HDPE Polymer Composites

        In this study, the effects of carbon black (CB) content and anodic oxidation treatment with AgNO3 on positive temperature coefficient (PTC) behavior of CB/HDPE nanocomposites were investigated. Also, the addition of elastomer as a toughing agent was studied. The 20~50 wt% of CB, 0~5 wtt% of elastomer, and 1 wt% of AgNO3-filled HDPE nanocomposites were prepared using the internal mixer in 60 rpm at 160˚C and the compression-molded at 180˚C for 10 min. As a result, the room temperature resistivity and PTC intensity of the composites were dependent, to a large extent, on the content of CB, addition of elastomer, and surface chemical properties that were controlled in the relative arrangements of the carbon black aggregates in a polymeric matrix. Moreover, the composites with relatively low room temperature resistivity and suitable PTC intensity could be achieved by treatment of AgNO3. Consequently, it was noted that PTC effect was due to the deagglomeration or the breakage of the conductive networks caused by thermal expansion or crystalline melting of the polymeric matrix.

      • 고지방식이가 운동시 흰쥐의 글리코겐 이용에 미치는 영향

        Carbohydrate and fat ingestion during exercise are essential nutrient, but a number of opinion is different from their contents of food. In this point of view, to study how high-fat dietary contents affects glycogen utilization, 48 female Sprague-Dawley rats were trained on the treadmill trained for 4wk with consuming two high-fat diet and one high-carbohydrate diet. All animals were fed 79%high-fat diet(n=16, 79F), 40% high-fat diet(n=16, 40F), 70% high-carbohydrate diet(n=16, 70C). After the final day of training, one-half of rats in each the three groups were run to exhaustion (E), whereas the remaining rats served as rested controls (R). All animals were trained on a motorized rodent treadmill. The animals ran for 30min/d at 28m/min(0% grade), analysis items were glycogen, body weight and %Fat were measured. All data were examined by means of one-way ANOVA and significance was set at the p<0.05. The Duncan method was used for a post hoc test. Body weight were similar among the 79F, 40F and 70C groups. %Fat was significantly greater in animals that consumed the 79F group than in those on the 70C group (P<0.05), even though similar body weight each other. Generally liver and muscle glycogen concentration had more in 79FR, on the other hand, had less in 79FE compared with the others. The mean overall glycogen utilization rate was depressed in muscle and liver after animals were on the 79% F diet. In summary, the present study has suggested that consumption of 79% F diet during endurance training can lead to an increase in exercise performance in female rats. Some of the consequences training on F diet are an increase in body fat deposition and slower utilization of glycogen during exercise. These data indicate that rats exposed to a high-fat diet are capable of prolonging exercise in spite of limited glycogen stores. This improved capacity for exercise appears to be partially the result of muscular adaptations to the diet, which apparently increase the ability to oxidize fat and concomitantly spare glycogen.

        Differential Manifestation of Ocular Phenotypes in TALEN-mediated p19arf Knockout FVB/N and C57BL/6J mouse lines

        Background p19arf, primarily known as a tumor suppressor, has also been reported to play an essential role in normal development of mouse eyes. Consistently, lack of p19arf has been associated with ocular defects, but the mixed background of the knockout (KO) mouse strain used raised a concern on the accuracy of the phenotypes observed in association with the targeted gene due to genetic heterogeneity. Object We carried out a study to investigate into the efect of genetic background on the manifestation of p19arf KO associated phenotypes. Methods We characterized the phenotypes of novel p19arf KO mouse lines generated in FVB/N and C57BL/6J using a transcription activator-like efector nuclease (TALEN) system in comparison to the reported phenotypes of three other p19arf-defcient mouse lines generated using homologous recombination. Results Ninety-fve percent of FVB/N-p19arf KO mice showed ocular opacity from week 4 after birth which worsened rapidly until week 6, while such abnormality was absent in C57BL/6J-p19arf KO mice up to the age of 26 weeks. Histopathological analysis revealed retrolental masses and dysplasia in the retinal layer in FVB/N-p19arf KO mice from week 4. Besides these, both strains developed normally from birth to week 26 without increased tumorigenesis except for a subcutaneous tumor found in a C57BL/6J-p19arf KO mouse. Conclusion Our fndings demonstrated surprisingly variable manifestation of p19arf-linked phenotypes between FVB/N and C57BL/6J mice, and furthermore between our mouse lines and the established lines, indicating a critical impact of genetic background on functional study of genes using gene targeting strategies in mice.

        This study was conducted to optimize the processing of high quality surimi gel from unmarketable cultured bastard halibut Paralichthys olivaceus. According to endogenous enzyme activity and processing optimization, high quality surimi gel from unmarketable cultured bastard halibut was prepared by mixing 3.0% (w/w) salt, 2.4% (w/w) starch, 5.0% (w/w) egg white and 4.8% (w/w) ice water in a Stephan mixer, set at 5˚C for 24 h, followed by boiling for 30 min, and finally cooling for 30 min. The strength of the surimi gel from unmarketable cultured bastard halibut prepared by the above processing method was 1,257 g×cm, which was 33% higher than that of a commercial surimi gel from Alaska pollock, grade SA.

        Controlled post-sulfurization process for higher efficiency nontoxic solution-deposited CuIn<sub>0.7</sub>Ga<sub>0.3</sub>Se<sub>2</sub> absorber thin films with graded bandgaps

        <P><B>Abstract</B></P> <P>A practical way of adjusting post-sulfurization process with variable durations is proposed to improve the cell-conversion efficiency of non-toxic solution driven CuIn<SUB>0.7</SUB>Ga<SUB>0.3</SUB>Se<SUB>2</SUB> absorber thin films. The degree of bandgap grading was controllable by changing the sulfurization time from 2 min to 30 min due to the dissimilar distribution of sulfur across the absorber layer. Deeper intensified distributions of Ga and S were responsible for enhancements in the open-circuit voltage and cell-conversion efficiency. A competitive efficiency of ∼8.81% for the 10 min-sulfurized CIGSSe cell was achieved with other promising parameters, i.e., short-circuit current density of 33.17 mA/cm<SUP>2</SUP>, open-circuit voltage of 0.496 V, and fill factor of 53.5%, resulting from the improved carrier collection and reduced recombination by the bandgap grading. This improved efficiency corresponds to a ∼15.6% enhancement compared to the unsulfurized sample.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Controlled post-sulfurization process enhances cell efficiency of CuIn<SUB>0.7</SUB>Ga<SUB>0.3</SUB>Se<SUB>2</SUB>. </LI> <LI> Sulfur penetrates easily toward the back contact with concentration gradient. </LI> <LI> A graded bandgap structure is responsible for the improved charge collection. </LI> <LI> The achieved cell efficiency of ∼8.81% corresponds to the ∼15.6% enhancement. </LI> <LI> Actual bandgap variations are projected on the basis of elemental distributions. </LI> </UL> </P>

        Effect of Jaeumkanghwatang (JEKHT), a Polyherbal Formula on the Pharmacokinetics Profiles of Tamoxifen in Male SD Rats (2) - Oral Combination Treatment of Tamoxifen 50 mg/kg with JEKHT 100 mg/kg on JEKHT 6-day Repeated Pretreated Rats with 8-day Repeated

        Objectives : The effects of Jaeumkanghwatang (JEKHT) co-administration on the pharmacokinetics of tamoxifen were observed after oral combination treatment of tamoxifen 50 mg/kg with JEKHT 100 mg/kg on JEKHT 6-day repeated oral pretreated rats with 8-day repeated co-administration to confirm the effects of JEKHT co-administration on the pharmacokinetics of tamoxifen. Methods : Six days after pretreatment of JEKHT 100 mg/kg, tamoxifen 50 mg/kg was co-administered with JEKHT 100 mg/kg, once a day for 8 days within 5 min. The blood were collected at 30 min before administration, 30 min, 1, 2, 3, 4, 6, 8 and 24 hrs after end of first and last 8th tamoxifen treatment, and plasma concentrations of tamoxifen were analyzed using LC-MS/MS methods. PK parameters of tamoxifen ($T_{max}$, $C_{max}$, AUC, $t_{1/2}$ and $MRT_{inf}$) were analysis as compared with tamoxifen single administered. Results : Six-day repeated oral pretreatment of JEKHT and 8-day repeated oral co-administration of tamoxifen within 5 min did not influenced on the plasma concentrations and pharmacokinetic parameters of tamoxifen, oral bioavailability, as compared with tamoxifen single treated rats, except for some negligible effects. Conclusions : It is concluded that JEKHT did not influenced on the plasma concentrations and pharmacokinetic parameters, the oral bioavailability of tamoxifen. Therefore, it is considered that co-administration of JEKHT and tamoxifen will be provide an effective novel treatment regimen on the comprehensive and integrative medicine for breast cancer patients, if they showed favorable synergic effects on the pharmacodynamics or reduce the tamoxifen treatment related toxicity and side effects in future studies.
