Genetic effect of <i>CCR3</i> and <i>IL5RA</i> gene polymorphisms on eosinophilia in asthmatic patients

<P><B>Background</B></P><P>Eosinophilic infiltration and peripheral blood eosinophilia in asthma require the cooperation of eosinophil-specific cytokines and chemokines and their receptors.</P><P><B>Objective</B></P><P>We investigated the association of polymorphisms in <I>CCR3</I> and <I>IL5RA</I> with asthma susceptibility or peripheral blood eosinophilia and the effects of the polymorphisms on receptor expression.</P><P><B>Methods</B></P><P>Polymorphisms in <I>CCR3</I> and <I>IL5RA</I> were identified and genotyped in 576 asthmatic patients and 180 healthy control subjects. CCR3 and IL-5 receptor α (IL-5Rα) protein expression on eosinophils was measured by means of flow cytometry.</P><P><B>Results</B></P><P>Although polymorphisms in <I>CCR3</I> were not associated with asthma susceptibility, the <I>CCR3</I> haplotype <I>ht2</I> showed a negative gene dose effect on the eosinophil count (<I>P</I> = .003–.009). <I>IL5RA c.−5091G>A</I> was weakly associated with eosinophil count. The effects of <I>ht2</I> were greater when paired with <I>IL5RA c.−5091A</I> (<I>P</I> = .001–.002). CCR3 protein expression was higher on eosinophils of asthmatic patients without <I>ht2</I> than in those with <I>ht2</I>. Asthmatic patients with the <I>IL5RA c.−5091A</I> allele showed higher IL-5Rα expression than those who were homozygous for the G allele.</P><P><B>Conclusion</B></P><P>The genetic association between <I>CCR3</I> polymorphisms and the number of circulating eosinophils was revealed as a novel finding. These associations were more pronounced when the <I>CCR3</I> polymorphisms were paired with polymorphisms in <I>IL5RA</I>. The protein expression levels of CCR3 and IL-5Rα on peripheral blood eosinophils are associated with the polymorphisms on their own genes.</P><P><B>Clinical implications</B></P><P>The identification of single nucleotide polymorphisms and haplotypes of <I>CCR3</I> and <I>IL5RA</I> might be useful in developing markers for intermediate phenotypes of eosinophil number and in designing strategies to control diseases related to hypereosinophilia.</P>

Suppressive Effects of Anthocyanins on Athma-specific T-helper 2 Cytokine Expression and β-Hexosaminidase Release in IgE-Antigen Complex-Stimulated RBL-2H3 Cells

Anthocyanins belong to a group of flavonoid compounds and are well known for their various health beneficial effects including antioxidative activities. Among them, the major anthocyanins isolated from the seed coat of black soybean (Glycine max L.) were previously characterized as glycosides having glucopyranose. Asthma is an allergic disease that is strongly associated with various immune cells including basophils and mast cells. Eosinophils, basophils and mast cells play important roles in asthma through the release of inflammatory mediators such as athma-specific T-helper (Th)2 cytokines and subsequent amplification of asthma symptoms by their degranulation. Rat basophilic leukemia RBL-2H3 cells was the most common in vitro models used for evaluating asthmatic reactions. We examined the effects of anthocyanin from the seed coat of black soybean (Glycine max L.) on the antigen-stimulated degranulation and Th2 cytokines production in RBL-2H3. Cell degranulation was evaluated by detecting the release of β -hexosaminidase. The β-hexosaminidase release and Th2 cytokine production in RBL-2H3 cells upon stimulation with IgE-antigen complex was much higher those that in untreated control cells. Anthocyanins significantly suppressed the IgE-antigen complex-induced degranulation of RBL-2H3 and inhibited IgE-antigen complex-mediated interleukin (IL)-4, IL-13 and TNF-α production in RBL-2H3 cells. These findings suggest that anthocyanins from the seed coat of black soybean (Glycine max L.) effectively inhibit asthmatic reactions and may have beneficial effects against allergic asthma.

Recently the two studies - the Heart and Estrogen/Progestin Replacement Study (HERS) and the Women's Health Initiative (WHI) - have provided us new and shocking knowledge that questions long-standing clinical practice and prescribing. Thereafter on October 6, 2002, NAMS (the North American Menopause Society) convened an Advisory Panel to develop clinical recommendations regarding the use of postmenopausal hormone therapy. Despite the proven benefits of HRT (hormone replacement therapy), beliefs about side effects and concerns about safety hinder acceptance of HRT among patients. So we present some updated and revised guidelines on HRT and their alternatives for the physicians and patients who have interests in HRT regimen. It seems clear that one of the essential requirements in HRT followed from this critical data of risk increases is to individualize HRT, choosing ingredients, doses and applications more adequate for the special condition of the woman.

복강경하 질식 자궁적출술 30예의 임상적 고찰 : A Review of 30 Cases

Hysterectomy is the most common gynecologic operation. Traditionally, hysterectomy was performed either through an abdominal approach or through a vaginal approach. Recently, laparoscopic hysterectomy is quite often tried. To evaluate the effectiveness and value of LA VH(Laparoscopy Assisted Vaginal Hysterectomy), and to evaluate the possibility whether to rep;ace abdominal hysterectomy to LAVH, we analyzed our clinical cases. This clinical study is a retrospective review of 30 cases of LAVH from May 2002 to February 2003 at the department of Obstetrics and Gynecology, Chung- Ju hospital, Konkuk University. We evaluated age, parity, previous abdominal operations, indications of hysterectomy, combined operation, operating time, hemoglobin change, weight of uterus, hospital stay and postoperativeThe average operating time was 146 ± 42 minutes, the uterine weight was 239 ± 126 grams, and the mean postoperative hemoglobin loss was 1.7 ± 1.4 g/dl. The most common pathologic finding was myoma uteri(37%), and the second most pathologic finding was adenomyosis(20%) and myoma uteri with adenomyosis (20%). Serious complications were not appeared. LAVH provides a shorter hospital stay, quicker recovery, and the acceptable complication rate except incerased operating time than abdominal hysterectomy. This study suggests that LAVH appears more useful than TAH(Total Abdominal Hysterectomy) and could replace abdominal hysterectomy.

Artificial neural network has offered distributed processing power, error correcting capability and structural simplicity of the basic computing element. Basically, neural nets are self-adaptive systems that try to configure themselves to store new information. The Error Backpropagation Algorithm is one of the most widely-used learning algorithms for multi-layered neural networks, but it has a serious drawback that it requires a lengthy learning time, even for a moderate number of input patterns. In this paper, we proposed a specific definition of generalization concept and comparatively analyzed methods which enhance the generalization property and techniques which reduce the learning time, based on this definition.

This paper deals with a PID neural networks control technique that efficiently control, its elevation angle and azimuth. The Helicopter system is taken as the control system in the current paper, and a mathematical model is derived to design the multivariable controller. The coefficients of the controller are automatically adjusted by the back- propagation algorithm of the neural network. The simulation results using MATLAB verified it's usefulness.

Ventricular septal defect remains an infrequent but devastating complication of acute MI. Approximately 1∼3% of cases of acute myocardial infarction are complicated by ventricular septal rupture in the prethrombolytic era and the incidence has decreased to 0.2% with reperfusion therapy. This mechanical complication usually occurs within the first 10 to 14 days when necrotic tissue is most abundant and the collateral coronary circulation is not well developed. This lesion is generally associated with complete coronary obstruction rather than severe stenosis. Rupture of the ventricular septum is a severe mechanical complication of acute myocardial infarction, usually resulting in death unless surgical repair is performed. The bad prognosis of this event within the first 2 weeks indicates the need for early surgical rapair. This complication is more frequent after the first acute myocardial infarction in the elderly and secondary to a transmural myocardial infarction. We report two cases of ventricular septal rupture after acute myocardial infarction.

Structure and Immune Modulating Activity of a Polysaccharide Purified from Mulberry Fruits Pericarp

In the present study, we investigated structure and immune modulating activity of a polysaccharide (tentatively named JS-MP-1), previously purified from Mulberry fruits (Morus bombycis Koidz) pericarp. Based on HPAEC-PAD analysis, major monosaccharides of JS-MP-1 were shown to be galactose (37.6% in mole percentage), arabinose (36.3%) and rhamnose (18.4%), respectively. EA analysis did not show the presence of sulfur (0.07%). Biochemical analysis showed neutral sugars (35.25%), uronic acid (11.25%), sulfate (0.16%) and proteins (0.3%). FT-IR spectra of JS-MP1 confirmed the presence of carboxyl groups (uronic acid – α-GalAp in RG1) and COC, CO and CC stretching vibrations at 948 – 1150 cm-1 corresponding to polysaccharides. Peak at 894 cm-1 indicates the presence of β-Galp units. 1H-NMR spectrum (in D2O) confirmed the presence of α-1,2-Rhap, α-Araf, β -Galp and α-1,4-GalAp units. The most intense HMQS signals were observed for terminal α-Araf. The presence of methylester groups in the residues of α -1,4-GalAp was confirmed by the HMQS peak at C 54.3/H 4.01 ppm (OCH3). Taken collectively, the JS-MP-1 is probably a rhamnogalacturonan type 1 (RG1), which contains: (a) partially methyl esterified α-1,4-D-GalAp and α -1,2-L-Rhap units in backbone, and (b) α-L-Araf and β-Galp are bound to Rha as terminal units or more complex side chains (arabinan, galactan and/or arabinogalactan). The ELISA assay showed that JS-MP-1 remarkably stimulates the release of the proinflammatory cytokines, TNF-α and IL-6, in a dose-dependent manner, from the RAW 264.7 cells. RT-PCR analysis also showed a significant induction of the iNOS and COX-2 gene expression. These results suggest that JS-MP-1 isolated from the Mulberry fruits has immune modulating activities.
