Samples of agricultural soils possibly contaminated by polychlorinated biphenyls (PCBs) were collected from Anseong, Korea. The agricultural areas chosen have possibly been contaminated by nearby factories, either directly or via atmospheric deposition, and require monitoring. The concentrations of 29 out of 209 PCB congeners were determined at five sites using high-resolution gas chromatography/high-resolution mass spectrometry. Total concentrations of the 29 PCB congeners in soil samples obtained from each study site were 106.65, 149.15, 222.67, 166.15, and $118.28pg\;g^{-1}$ dry weight, respectively, with pentaCBs and hexaCBs giving the highest concentrations of the congeners studied. The total toxic equivalent (TEQ) for 12 PCBs from each site was 0.05, 0.11, 0.08, 0.05, and $0.04ng\;kg^{-1}$ dry weight, respectively. These values were mainly due to high TEQ values for PCB 126 (3,3',4,4',5-pentachlorobiphenyl) from each site (at 0.05, 0.10, 0.07, 0.04, and $0.03ng\;kg^{-1}$ dry weight, respectively). The TEQ values are much lower than the action level of 20 ng WHO-TEQ $kg^{-1}$ recommended by the World Health Organization, but PCB concentrations in this area should be monitored nonetheless, since these man-made compounds bioaccumulate, thus threatening the ecosystem and human health.

Cytochrome P450 1A2 (CYP1A2) is a member of the cytochrome P450 superfamily enzymes in mammals and plays a major role in metabolizing endogenous hormones in the liver. In recent days, CYP1A2 expression has been found in not only the liver but also other tissues including the pancreas and lung. However, little information is available regarding the expression of CYP1A2 in the ovary, in spite of the facts that the ovarian follicle growth and atresia are tightly associated with controls of endocrine hormonal networks. Therefore, the expression of CYP1A2 in the ovaries of prepubertal and pubertal rats was investigated to assess its expression pattern and puberty-related alteration. It was demonstrated that the expression level of CYP1A2 was significantly (p < 0.01) higher in the pubertal ovaries than prepubertal counterparts. At the ovarian follicle level in both groups, whereas CYP1A2 expression was less detectable in the primordial, primary and secondary follicles, the strongly positive expression of CYP1A2 was localized in the granulosa cell layers in the antral and pre-ovulatory follicles. However, the ratio of CYP1A2-positive ovarian follicle was significantly (p < 0.01) higher in the ovary of pubertal group (73.1 ± 3.1%) than prepubertal one (41.0 ± 10.5%). During the Immunofluorescence, expression of CYP1A2 was mainly localized in Fas-positive follicles, indicating the atretic follicles. In conclusion, these results suggested that CYP1A2 expression was mainly localized at the atretic follicular cells and affected by the onset of puberty. Further study is still necessary but we hypothesize that CYP1A2 expresses in the atretic follicles to metabolize residue of the reproductive hormones. These findings may have important implications for the fields of reproductive biology of animals.

Under the stressed condition, a complex feedback mechanism for stress is activated to maintain homeostasis of the body and secretes several stress hormones. But these stress hormones impair synthesis and secretion of the reproductive hormones, followed by suppression of ovarian function. Cytochrome P450 1A2 (CYP1A2) plays a major role in metabolizing exogenous substances and endogenous hormones, and its expression is recently identified at not only the liver but also several organs with respect to the pancreas, lung and ovary. Although the expression of CYP1A2 can be also affected by several factors, understanding for the changed pattern of the ovarian CYP1A2 expression upon stress induction is still limited. Therefore, CYP1A2 expression in the ovaries from immobilization stress-induced rats were assessed in the present study. The stress-induced rats in the present study exhibited the physiological changes in terms of increased stress hormone level and decreased body weight gains. Under immunohistological observation, the ovarian CYP1A2 expression in both control and the stressed ovary was localized in the antral to pre-ovulatory follicles. However, its expression level was significantly (p < 0.01) higher in the stress-induced group than control group. In addition, stress-induced group presented more abundant CYP1A2-positive follicles (%) than control group. Since expression of the ovarian CYP1A2 was highly related with follicle atresia, increased expression of CYP1A2 in the stressed ovary might be associated with changes of the ovarian follicular dynamics due to stress induction. We hope that these findings have important implications in the fields of the reproductive biology.

Piroxicam-$\beta$-cyclodextrin은 piroxicam을 $\beta$-cyclodextrin으로 포접시킨 비스테로이드성 항염증약물이다. 이러한 포접형 약물은 위장관에서의 흡수속도가 증가하는 것으로 외국자료에서 보고되고 있으며 이는 이 약물의 위장관 내성에 보다 나은 영향을 끼칠 수 있음을 시사하고 있다. 본 연구는 정상성인 한국인을 대상으로 randomized, crossover design에 의해 piroxicam-$\beta-cyclodextrin(Brexin^{(R)})$과 piroxicam 확산정$(Feldene^{(R)})$ 흡수속도를 비교하고자 하였다. 건강한 성인 8명의 피험자를 2군으로 나누어 시험약 또는 대조약을 각각 20 mg씩 20일의 휴약 기간을 두고 이중 맹검으로 교차 투여하였다. 시험약 또는 대조약의 투여 후 24시간 동안 일정 간격으로 채혈하여 HPLC 방법으로 혈장 내 piroxicam 농도를 측정하였다. $AUC_{0-24}\;({\mu}g/mL)$는 piroxicam-$\beta$-cyclodextrin군에서 $56.1\pm4.9$, piroxicam군에서 $57.3\pm5.6$으로 통계적인 유의성이 없었으나, 투여 후 0.5시간에서의 혈중농도는 piroxicam-$\beta$-cyclodextrin군 $2.9\pm0.4\;{\mu}g/mL$, piroxicam군 $1.6\pm0.3\;{\mu}g/mL$으로 통계적인 유의성을 보였다(p<0.05). 또한 최고혈중농도는 piroxicam-$\beta$-cyclodextrin$(4.3\pm0.5\;{\mu} g/mL)\;piroxicam(3.5\pm0.3\l{\mu}g/mL)$,으로 유의성이 있었으며(p<0.05), 흡수 속도상수는 piroxicam-$\beta$-cyclodextrin$(3.00\pm0.49\;h^{-1}), \;piroxicam(1.80\pm0.21\;h^{-1})$이었다(p<0.1). 이상의 결과에서, piroxicam-$\beta$-cyclodextrin정은 piroxicam 확산정과 비교하여 흡수되는 정도는 서로 비슷하지만 흡수 초기의 혈장농도 및 흡수속도상수에서 보다 빠른 약동학적 특성을 나타내었다.

N/A Aeromonas hydrophila is a facultatively anaerobic, gram-negative rod which has been often regarded as an opportunistic pathogen in hosts with compromised defense mechanisms. A. hydrophila can produce septicemia with skin lesions such as necrosis, edema, and blisters. Therefore, it is necessary to investigate what kinds of exocellular ?oxic or pathogenic factors are produced by A. hydrophila for elucidation of pathogenesis and effective treatment. This study was designed to determine the extracellular enzymes produced by two strains of A. hydrophila which were isolated from the blood and wound of patients with septicemia and osteomyelitis, respectively. Histamine release from rat peritoneal mast cells evoked by A. hydrophila culture supernatant was assayed. The morphologica changes of rat peritoneal mast cells treated with A. hydrophila culture supernatant were observed by electron microscopy. A. hydrophila produced elastease, protease DNase, hemolysin(alpha and beta), lipase, lecithinase, and gelatinase but it did not produce coagulase, hyaluron dase, chondroitin sulfatase or mucinase. The culture supernatant fluid of A. hydrophila induced aistamine release from rat peritoneal mast cells. Transmission electron microscopic studies of mast cells treated with the culture supernatant revealed microvesicles of various size in the cytoplasm and around the cell membrane. These findings suggest that A. hydrophila culture supernatant induces histamine release from rat peritoneal mast cells by a process that may involve a resicular transport mechanism, namely, piecemeal secretion.

N/A Functional cerebral impairments have been verified objectively by brain SPECT and q-EEG (quantitative electroencephalography). Microcerebral circulatory defects without anatomical changes cannot be detected by the brain CT or MRI. Brain SPECT using Tc-99m-HMPAO (Hexamethyl propyleneamine oxime) as a key radioisotope may be accepted as the useful method for identifying functional cerebral impairments. We studied 25 patients with mild head trauma to define whether the SPECT was helpful in detecting cerebral impairment. Results were as follows: The SPECT was positive in 23 patients out of 25, q-EEG positive in 16 patients and brain CT was positive in 3 cases. SPECT and q-EEG were more sensitive than CT. SPECT would be more useful method than brain CT to investigate cerebral function after head injury.
