Effect of Hericium Gold® on Hepatic Dysfunction and Alcoholic Hangover

The improving efficacies of Hericium Gold® containing fermented extract of Hericium erinaceum on acute hepatic dysfunction and alcoholic hangover were investigated via the effects on carbon tetrachloride (CCl4)-induced hepatitis and ethanol overdosage, respectively. Oral administration with Hericium Gold®, especially at 100 mg/kg, attenuated the delayed excretion and blood clearance of bromosulphalein induced by CCl4 (1 ml/kg). Hericium Gold® also reduced the increase in blood alanine transaminase (ALT) and aspartate transaminase (AST) activities following CCl4 exposure. Although Hericium Gold® did not reduce the peak blood concentration of ethanol which was observed 1 hr after oral administration of alcohol (3 ml/kg), it enhanced the blood clearance at 100 mg/kg. Taken together, the results suggest that Hericium Gold® possesses hepatoprotective activity against exposure to CCl4 or alcohol, and that could be a candidate for the improvement of chemical-induced or alcoholic liver disorders.

Misaengtang (MST), a formula of Korean herbal medicines, has been used as a weight-controlling recipe. We have investigated two experiment of body weight regulation by MST in rats. i) The anti-obesity effect of MTS on a high fat diet-induced obesity, male Sprague-Dawley rats were fed with a high-fat diet containing 30% in the absence or presence of MST (0.3, 1 or 3%) or a reference orlistat (0.05%) for 6 weeks. ii) weight-decrease effect of MST on normal diet, same animal were fed with a normal diet in the absence or presence of MST (3%) for 6 weeks. And the body weights, daily feed and water consumptions, organ weights, fat weights serum biochemistry were measured. In both experiments, MST and orlistat did not affect the body weight gain. But orlistat significantly increased the feed and water consumptions, leading to low-feed efficiency, and orlistat markedly reduced abdominal, paratesticular and perirenal fat weights, although increased the kidney weights. In MST, low dose (0.3%) of MST decreased the perirenal fat and increased the kidney weights in rats fed HFD, and MST 3% decreased the abdominal fat weights in rats fed normal diet. In addition, Orlistat caused changes in parameters of hepatotoxicity (AST and glucose), nephrotoxicity (BUN and B/C ratio) and lipid metabolism (HDL and triglycerides). In comparison, MST decreased AST, ALP and ALT, the hepatotoxicity markers, and somewhat improved the hepatic fatty degeneration. Taken together, it is suggested that MST does not exert anti-obesity activity as well as remarkably direct effects, but MST may be potentially benefit for dietary cure and exercise-cure of obesity.
BTT0811의 in vitro 3T3 Neutral Red Uptake 시험법을 이용한 광독성 평가

Phototoxicity is a toxicity reaction induced by UV after applying the chemical to part or entire body. BTT0811 which has co-developed by Biotoxtech Co., Ltd., Korea and Northland, China is recombinant material of thymosin β4. We performed to evaluate the phototoxicity of BTT0811 using in vitro 3T3 neutral red uptake phototoxicity test in OECD guideline. Before evaluating the phototoxicity of this study, we selected two substances for the controls. One of the positive controls was chlorpromazine, already known as a phototoxicity substance, and the other was L-histidine, which has no phototoxicity. Then, we evaluated the phototoxicity of BTT0811 compared with those substances. In results of study, photo irritation factor (PIF) and mean photo effect (MPE) was measured for evaluating phototoxicity. PIF and MPE value of chlorpromazine was more than 6 and 0.15, respectively. Chlorpromazine had phototoxicity resulting from PIF and MPE. On the other hand, L-histidine was measured no PIF value and less than 0.1 of MPE. There was no PIF value of BTT0811 and MPE value of BTT0811 was less than 0.1. Based on the results of this study, BTT0811 did not show any evidence of phototoxicity in BALB/c 3T3 clone A31 cells under the conditions of this study.
