        The Global Histone Modification Patterns of Osteosarcoma

        Background: Epigenetic alteration may affect a patient’s prognosis by altering the development and progression of the tumor. Some recent reports have identified a correlation between histone modification and patient outcome. However, no studies have been conducted on global histone modification in osteosarcomas. Methods: We investigated histone modification in 54 cases of osteosarcoma by performing immunohistochemical staining. The immunohistochemical expres­sion of four histone modification markers, acetylated H4 lysine 12 (H4K12Ac), acetylated H3 ly­sine 18, trimethylated H3 lysine 27, and dimethylated H3 lysine 4 were evaluated. Results: High H4K12Ac expression was correlated with patient age (p=0.011). However, the other histone mod­ification markers showed no correlation with any of the clinicopathological data such as survival, tumor grade, tumor site, metastasis, age, or gender. Conclusions: Our study showed that all four histone modification markers are expressed in osteosarcoma (median expression rate, 40 to 60%). However, we did not find a correlation with the clinicopathological factors except for age. Further study to evaluate the reason for the association between H4K12Ac and patient age is needed.

        Correlation Between Cyclooxygenase-2 Expression and HuR Cytoplasmic Translocation of Breast Cancer

        Background : Embryonic lethal abnormal vision (ELAV)-like protein HuR is known to stabilize mRNA through binding AU-rich elements in the 3’-untranslated region. Recent studies show that HuR expression is associated with the expression of several genes including cyclooxygenase- 2 (COX-2). HuR exists predominantly in the nucleus, but cytoplasmic translocation of HuR is thought to be more important for its activity. COX-2 is a well-known enzyme that promotes tumor growth. Methods : To evaluate the correlation of HuR and COX-2 expression, we analyzed expression of HuR and COX-2 in 91 cases of breast cancer using immunohistochemistry. Results : Nuclear and cytoplasmic expression of HuR was seen in 76 (83.5%) and 19 (20.9%) of 91 cases respectively. COX-2 immunoreactivity was seen in 54 (59.4%) cases. Cytoplasmic HuR expression showed significant correlation with COX-2 expression (p=0.001). Nuclear HuR showed no correlation with COX-2 expression or other clinicopathological parameters. COX-2 expression is significantly associated with tumor grade (p=0.028). COX-2 (p=0.092) and cytoplasmic (p=0.569) and nuclear HuR (p=0.247) expression showed no correlation with survival. Conclusions : These results suggest that cytoplasmic HuR expression is associated with COX-2 expression in breast cancer and cytoplasmic location of HuR might contribute to the stabilization of COX-2 mRNA.         Associations between the Expression of Mucins (MUC1, MUC2, MUC5AC, and MUC6) and Clinicopathologic Parameters of Human Breast Ductal Carcinomas

        Purpose: Mucins are members of the glycoprotein family expressed in benign and malignant epithelial cells. The aim of this study is to evaluate the relationships between the expression of mucins in breast ductal carcinoma and clinicopathologic parameters. Methods: We constructed tumor microarrays based on 240 cases of invasive ductal carcinoma and 40 cases of ductal carcinoma in situ (DCIS) using formalin fixed, paraffin embedded tissues. We examined the expressions of MUC1, MUC2, MUC 5AC, and MUC6 by immunohistochemistry. Results: MUC1 demonstrated cytoplasmic, membranous, apical, and combinative expressions. Other mucins demonstrated cytoplasmic expression. In invasive ductal carcinoma, MUC1, MUC2, MUC5AC, and MUC6 were expressed in 93.6%, 6.2%, 4.8%, and 12.4% of cases, respectively; these rates were slightly, but not significantly, higher than observed in cases of DCIS. MUC1 expression was associated with estrogen receptor (ER) expression and negative MUC1 expression was associated with triple negativity. MUC6 expression was correlated with higher histologic grade, lymphatic invasion, lymph node metastasis, and HER2 positivity. No associations with any other clinicopathologic parameters were observed. Conclusion: Most invasive ductal carcinomas of the breast express MUC1, and this expression is associated with ER expression. MUC6 expression is correlated with some clinicopathologic parameters that are indicators of poor prognosis. To evaluate the role of MUC6 as a potential biomarker, further studies are warranted.

      • 중성염을 첨가하여 알칼리 가수분해한 PET 직물의 물성

        Poly(ethylene terephthalate) (PET) was hydrolzyed with aqueous solutions of many salts in alkali metal hydroxides under various conditions. Several physical properties were investigated and the photographs of scanning eletron microscope of the ethched surface of the hydrolysed PET were taken. Some conclusions obtained from the experimental results were summarized as follows. 1. No significant change was observed with different salts used. The physical properties of the hydrolysed PET were independent of the difference in kind of salts used. The tensile strength and the handle value were decreased with the increase in the weight less. The density and the degree of crystallinity of hydrolysed PET were decreased initially, after passing the minimum value, then increased to about the degree of the crystallinity of untreated PET. Added salts with greater positive effects on the hydrolysis etched fiber surface more severely, and the severeness was nearly agreed with the order of the acceleration effect of salts added. 2. It was thought that salts in aqueous NaOH solution accelearating the hydrolysis of PET would contribute to the conservation of energy and NaOH in the weight loss finishing process of PET fabric without further purchasing or equpping other facilities.

      • 골 견인술에 의한 경추골절 탈구환자의 치료에 대한 임상적 연구

        For the patient with fracture dislocation of the cervical spine, skeletal traction method is simple, safe and effective way to decompress and protect the spinal cord reestablishing the normal alignment of the spine. Skeletal traction is indicated as the initial treatment for the reduction of cervical fracture dislocations, and when properly applied, this method eliminate the necessity of laminectomy, spinal fusion, and the use of plaster supports in almost all cases of injury to this part of the spine. The results were satisfactory in most of our patients and the incidence of com-plication was negligiable or minor. The technique for correct application is briefly described.

        Seven kinds of neutral salts with different cations and anions, LiCl, NaCl, KCl, CsCl, NaF, NaBr, and Nal, were added to the dye bath to accurately understand the effect of electrolytes on the reactive dyeing rate of silk with C.I. Reactive Black 5. The cations of salts added lowered the negative surface potential of the silk, improving equilibrium adsorption and the accessibility of the dyestuff to the fiber geatly and speeding up the dyeing rate in the order of Li^+>Na^+>K^+>Cs^+. The sodium cation lowered the negative surface potential of the silk and increased the dye-uptake on the fabric. However, because of the discrepancy in the anions' inhibition power form sodium lowering the surface negative potential the amount of the dye on the silk fiber was different from each other in the order of F> Cl >Br > I. For cations, the activation energy(E_a) of the dyeing was in the order of Li^+>Na^+>K^+>Cs^+but the activation free energy(ΔG^*), or the real energy barrier for the reaction, was in the order of Li^+>Na^+>K^+>Cs^+ because the degree of the contribution of E_a to the activation entropy(ΔS^*) was Li^+>Na^+>K^+>Cs^+. it was found form this result that LiCl had the strongest lowering effect on the negative surface potential of silk. The decrease in ΔS^* should be attributed to the loosely bonded activated complex of dyestufffs, cations and fiber molecules at transition state. For anions, the activation energy(E_a) of the dyeing was in the order of F>Cl >Br>I but the dye-uptake on the fabric and the activation free energy(ΔG^*) were in the order of F<Cl<Br<I because the strength of the interaction of the anions with sodium cations was the same as the order of the latter. In other words F exerted the weakest eletrostatic force on Na^+ force on Na^+ and competed with the dyestuff anions least of all. It was clarified from the Bro¨nsted equation that electrolytes had the ionic strength effect and the specific salt effect on the reactive dyeing.
