        호흡기 상피세포에서 MUC5AC와 MUC5B 발현에 대한 Betulinic Acid의 효과

        Background and Objectives MUC5AC and MUC5B are representative secretory mucin genes in the human airway, whose expressions are increased by a variety of inflammatory mediators. Betulinic acid, a naturally occurring pentacyclic triterpenoid, is known to have an anti-inflammatory property. However, the effects of betulinic acid on mucin secretion of airway epithelial cells still have not been reported. Therefore, in this study, the effect of betulinic acid on inflammatory mediators-induced MUC5AC and MUC5B expressions was investigated in human airway epithelial cells. Materials and Method In the mucin-producing human NCI-H292 airway epithelial cells, the effects of betulinic acid on interleukin-1β (IL-1β)-, lipopolysaccharide (LPS)-, and phorbol myristate acetate (PMA)-induced MUC5AC and MUC5B expressions were analyzed by reverse transcription-polymerase chain reaction and enzyme-linked immunosorbent assay. Results Betulinic acid attenuated IL-1β-, LPS-, and PMA-induced MUC5B mRNA and glycoprotein expression in NCI-H292 cells. On the other hand, betulinic acid did not attenuate IL-1β-, and LPS-, but induced PMA-induced MUC5AC mRNA and glycoprotein expressions in NCI-H292 cells. Conclusion These results suggest that betulinic acid attenuates IL-1β-, LPS-, and PMA-induced MUC5B expression in the airway epithelial cells.         나노세공 분자체를 이용한 천연가스 연료로부터 황 화합물의 선택적 흡착

        천연가스로부터 유기 황 화합물인 TIIT 와 TBM의 제거를 위한 적합한 흡착제의 선정이 수행되었다. 황 화합물에 대한 포화 흡착량은 Na-Y, Na-ZSM-5, Na,K-ET(A)S-10, Na-모더나이트, Na,K-클리놉틸올라이트, Ti/MCM-41, Ti/SBA-15이 포함되어진 나노세공 물질 및 무정형 티타노실리케이트에 대하여 펄스 흡착 방법에 의해 측정되었다. 측정되어진 물질들 중 Na-Y 와 Na,K-ET(A)S-10 제올라이트에서 THT 와 TBM에 높은 흡착 용량을 보였다. Na,K-ET(A)S-10에서의 THT에 대한 포화 흡착량은 효율적인 흡착제로 잘 알려진 Na-Y 제올라이트와 비슷하였다. Na,K-ET(A)S-10에서의 THT 와 TBM의 흡착량과 흡착능은 Na,K-ET(A)S-10의 결정성이 좋을수록 증가하였다. 모사되어진 천연가스를 이용한 파과 형가로부터 THT 와 TBM 사이의 경쟁적 흡착에 관한 연구에서 Na,K-ET(A)S-10은 THT에 선택적으로 흡착하는 것으로 나타났다. Na,K-ET(A)S-10의 THT에 대한 파과 용량은 1.19 m㏖/g 이었다. 이러한 결과들은 Na,K-ETS-10 과 Na,K-ETAS-10의 높은 흡착 성능이 제올라이트 구조의 넓은 기공 성질과 유기 황 화합물과의 강한 정전기적 상호작용을 지닌 제올라이트 구조 속의 높게 교환되어진 양이온에 기인하는 것으로 보인진다. The selection of a suitable adsorbent for removing organic sulfur compounds tetrahydrothiophene (THT) and t-butylrnercaptan (TBM) from natural gas has been carried out. The saturation adsorption capacity for the sulfur compounds were determined by pulse adsorption method for a group of nanoporous materials, including Na-Y, Na-ZSM-5, Na,K-ET(A)S-10, Na-Mordenite, Na,K-Clinoptilolite, Ti/MCM-41, Ti/SBA-15 and amorphous titanosilicates. Among the materials tested, Na-Y and Na,K-ET(A)S-10 zeolites showed high adsorptive capacities for THT and TBM. The saturation capacity for THT on Na,K-ETS-10 was comparable with that on Na-Y zeolite, which is well known as an effective adsorbent. The capacity and adsorptivity for THT and TBM on Na,K-ETAS-10 were improved by an increase in crystallinity of Na,K-ETAS-10. An investigation of the competitive adsorption between THT and TBM from the breakthrough test using a simulated natural gas indicates that Na,K-ETS-10 selectively adsorbs THT. The breakthrough capacity for THT on Na,K-ETS-10 was 1.19 m㏖/g. The results show that the high adsorption performance of Na.K-ETS-10 and Na,K-ETAS-10 is due to the highly exchanged cations in the zeolitic structure which exhibit the strong electrostatic interactions with organic sulfur compounds and their wide pore nature.

        Insect Fauna of Adjacent Areas of DMZ in Korea

        Insect fauna in adjacent areas of Demilitarized Zone (DMZ) in Korea was surveyed seasonally in2001∼2003. The survey area was divided into 3 regions (eastern mountain, middle inland, and western coastal sieving, beating, brushing and suction were used depending on the environmental and military conditions. Total 361 genera and 437 species of 116 families belonging to 14 orders were identified. Among these, 46 species were new to insect fauna of DMZ areas. Species richness was the highest in the eastern mountain region. Numbers of habitat-common and -specific species were 96 (22%) and 195 (47.2%), respectively. The insect species community similarity was highest (0.64) between eastern mountain region and western coastal region. Insect orders showing high species richness were Coleoptera (38.9%), Lepidoptera (19.2%), Orthoptera (9.4%), and future conservation in DMZ areas.

        아미노 결합 정지상 HPLC 컬럼을 이용한 마 껍질의 allantoin 함량 분석

        마에는 다양한 기능성 성분이 함유되어 있어서 식용 및 약용으로 다양하게 이용되고 있다. 특히 allantoin은 대표적인 마의 2차 대사산물로서 의약품 및 기능성 화장품 제조에도 활용되는 고부가가치 산물이다. 본 연구에서는 아미노 결합 정지상 HPLC 컬럼을 이용하여 마의 속과 껍질의 allantoin 함량을 분석하였다. Allantoin 표준용액의 검량선의 결정계수(R<sup>2</sup>)값은 0.9999로 높은 직진성을 보였으며, 검출한계(LOD)와 정량한계(LOQ)는 각각 0.0229 mg/mL 및 0.0694 mg/mL로 설정되었다. 마 속과 껍질의 건조중량 기준 allantoin 함량은 각각 3.09±0.02 mg/g, 3.91±0.11 mg/g으로 속보다 껍질에 더 많은 allantoin이 함유되어있음을 확인하였다. 따라서 이러한 결과를 토대로 향후 농업부산물인 마껍질이 새로운 고부가가치 기능성 소재로 활용될 수 있음을 확인하였다. Yam (Dioscorea japonica) is widely utilized as food and a pharmaceutical ingredient as it contains a variety of valuable constituents. Allantoin is one of the bioactive components in yam that is used in pharmaceutical and cosmetic industry. This study was conducted to analyze and compare the allantoin content of yam flesh and peel by HPLC analytical method using an amino bonded-phase column to make up for the limitations of the previous HPLC analytical methods. The allantoin contents of yam flesh and peel were 3.09±0.025 and 3.91±0.11 mg/g (dry weight), respectively. The results of this study indicated that yam peel has higher allantoin content than yam flesh, and that the discarded yam peel could be used as a source for high value-added functional materials.

        Background and Objectives Naringenin and delphinidin are types of anthocyanidin, which are flavonoids and thus have anti-inflammatory property. Moderate consumption of natural dietary naringenin and delphinidin is believed to do anti-inflammatory action, but the action mechanism is unclear. Therefore, this study aimed to investigate the effects of naringenin and delphinidin on interleukin-1β (IL-1β)- and lipopolysaccharide (LPS)-induced MUC5AC and MUC5B expressions in airway epithelial cells. Materials and Method In NCI-H292 cells and cultured nasal polyp epithelial cells, the effects of naringenin and delphinidin on IL-1β- and LPS-induced MUC5AC and MUC5B expressions were analyzed by real-time polymerase chain reaction and enzyme-linked immunosorbent assay. Results Delphinidin attenuated IL-1β- and LPS-induced MUC5AC and MUC5B mRNA and glycoprotein expression in a dose-dependent pattern in NCI-H292 cells and in cultured nasal polyp epithelial cells. Naringenin partially attenuated IL-1β- and LPS-induced MUC5AC and MUC5B mRNA and glycoprotein expression at a high dose. Conclusion These results suggest that delphinidin attenuates MUC5AC and MUC5B expressions in the airway epithelial cells. Between anthocyanidin and delphinidin, delphinidin exhibits greater potential as an ideal therapeutic agent for the control of mucus-hypersecretion in the treatment of airway inflammatory diseases.

        인간 호흡기 상피세포에서 MUC5B 발현에 대한 Udenafil의 효과

        Background and Objectives Mucus hypersecretion in the airway may lead to increased frequency and duration of infection, declined lung function, and increased morbidity and mortality in inflammatory respiratory diseases. Udenafil, a phosphodiesterase (PDE) 5 inhibitor, is an oral medication for erectile dysfunction. Recent studies show that PDE5 inhibitor has various anti-inflammatory properties. However, the effect of udenafil on mucus secretion in human airway epithelial cells is unclear. Therefore, the effect and brief signaling pathway of udenafil on MUC5B expression were investigated in human airway epithelial cells. Materials and Method We analyzed the effects and brief signaling pathway of udenafil on the lipopolysaccharide (LPS) induced MUC5B expression in mucin-producing NCI-H292 epithelial cells using reverse transcription-polymerase chain reaction, enzyme-linked immunosorbent assay, and immunoblot analysis. Results Udenafil attenuated the LPS-induced MUC5B mRNA expressions and glycoprotein production in NCI-H292 epithelial cells. It also attenuated LPS-induced toll like receptor 4 (TLR4) mRNA expression and phosphorylation of extracellular regulated kinase1/2 (ERK1/2) and p38 in NCI-H292 epithelial cells. Conclusion These results suggest that udenafil attenuates the LPS induced MUC5B expression via TLR4, ERK1/2 and p38 mitogen activated protein kinase (MAPK) pathway in human airway epithelial cells, and that it could be a novel therapeutic agent for controlling chronic airway disease.
