        人蔘의 配合에 따른 抗酸化作用에 關한 硏究 (1) : 人蔘과 配合하는 數種 韓藥制의 抗酸化 作用에 對한 比較 硏究 Study on comparative anti - oxidative effects of some herbal medicine frequently combined with Ginseng Radix

        This study was carried out in order to investigate the anti-oxidative effects of main tonic herb medicine, Ginseng Radix, Rehmammiae Radix Preparat, Astragali Radix, Poria, Aconiti Iateralis Preparata Radix, Gecko and Bupleuri Radix. These herbs were measured on inhibitory activity against lipid peroxidation of linoleic acid mixture and rat liver and hydrogen-giving-temper effect by DPPH. The study showed the following valuable results: 1. In inhibitory activity of water extract against lipid peroxidation of linoleic acid mixture, Gecko showed the highest anti-oxidant activity among the some herbal medicine and Rehmammiae Radix Preparat showed the lowest anti-oxidant activity when compared with the examined samples. 2. In inhibitory activity of water extract against lipid peroxidation of rat liver, Gecko showed the highest anti-oxidant activity among the some herbal medicine and Rehmammiae Radix Preparat showed the lowest anti-oxidant activity. 3. In hydrogen-giving-temper effect of water extract by DPPH, Gecko showed the highest anti-oxidant activity. 4. In the linoleic acid mixture, Gecko showed the highest anti-oxidant activity in Butanol fraction. 5. In the lipid peroxidation of rat liver, Gecko showed the highest anti-oxidant activity in Butanol fraction. 6. In hydrogen-giving-temper effect of water extract by DPPH, Gecko showed the highest anti-oxidant activity in Butanol fraction. From the above results, it was concluded as follow: All the seven herb medicine including Ginseng Radix showed the anti-oxidant activity. Especially, Gecko showed the highest anti-oxidantactivity among them.

        화장품 소재로서 고마리 추출물의 생리활성

        Purpose: The purpose of this study was to investigate the potential of Persicaria thunbergii (P. thunbergii) extracts as physiologically active cosmetic ingredients. Methods: To elucidate the anti-oxidative, anti-inflammatory, and anti-microbial properties of P. thunbergii , its ethanol and hot-water extracts were examined for 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity, nitric oxide (NO) production, protective effects against oxidative stress in HaCaT cells, anti-inflammatory activity, anti-microbial activity, and inhibition of β-hexosaminidase expression, an allergy factor. Results: The anti-oxidative activity of the two P.thunbergii extracts was compared, and the anti-oxidative activity of the ethanol extract was found to be superior to that of the hot-water extract. The polyphenol contents of P. thunbergii ethanol and hot-water extracts were 132.00 and 89.46 μg/ mL, respectively. And total flavonoid contents of P. thunbergii ethanol and hot-water extracts were 115.27, 84.94 μg/mL, respectively. The DPPH radical scavenging activity was found to be 79.80% for the ethanol extract and 70.33% for the hot-water extract at a concentration of 500 μg/mL. No significant cytotoxicity was observed in HaCaT, RAW 264.7 and RBL-2H3 cells. The protective effect of the extracts on HaCaT cell against oxidative stress induced by hydrogen peroxide (H 2 O 2 ) was confirmed by 23% for ethanol extract and 18% for hot water extract. The anti-inflammatory activity of the extracts was examined in RAW 264.7 cells, and nitric oxide (NO) production was suppressed in a concentration-dependent manner. Both ethanol and hotwater extracts also inhibited the degranulation of immune cells in a concentration-dependent manner, as assessed by the secretion of β-hexosaminidase. In addition, the concentration dependent anti-microbial activities of the extracts were demonstrated in several bacterial strains, such as those of Staphylococcus aureus (S. aureus), Staphylococcus epidermidis (S. epidermidis), and Propionibacterium acnes (P. acnes). Conclusion: Based on the findings from this study, P. thunbergii extracts could be used as functional cosmetic ingredients that possess anti-oxidative, anti-inflammatory, and anti-microbial properties. In order to determine their applicability, further research should be systematically conducted to verify their efficacy through clinical studies in animals and humans. 목적: 고마리 추출물에 대한 생리활성 및 그에 따른 화장품 소재로서의 응용 가능성을 알아보고자 하였다. 방법: 고마리 에탄올및 열수 추출물의 항산화, 항염, 항균 효능을 확인하기 위하여 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical 소거능, nitric oxide (NO) 생성량 측정, 산화적 스트레스에 대한 HaCaT 세포 보호 효과, 항염, 항균, 알러지인자인 β-hexosaminidase 발현량 억제를 측정하였다. 결과: 고마리 추출물의 항산화 효능을 비교한 결과, 에탄올 추출물이 열수 추출물보다 우수하였으며, 고마리 에탄올, 열수 추출물의 폴리페놀 함량은 각각 132.00, 89.46 μg/mL이고, 총 플라보노이드 함량은 각각 115.27, 84.94 μg/mL으로 확인되 었다. 그리고 DPPH radical 소거능 측정결과, 고마리 에탄올, 열수 추출물의 농도가 500 μg/mL 일 때 각각 79.80, 70.33%로 확인 되었으며, 세포 독성 실험결과, HaCaT, RAW 264.7, RBL-2H3 세포에서 유의한 세포독성은 나타나지 않았다. Hydrogen peroxide (H 2 O 2 )에 의해 유발되는 산화적 스트레스에 대한 HaCaT 세포 보호 효과 확인결과, 고마리 에탄올, 열수 추출물은 23%, 18%로 세포 보호 효과가 확인되었다. RAW 264.7 세포에 대한 고마리 추출물의 항염 효과를 확인한 결과, 농도 의존적으로 nitric oxide (NO) 생성이 억제되었다. β-Hexosaminidase의 분비를 측정한 결과, 고마리 에탄올과 열수 추출물은 농도 의존적으로 면역세포의 탈과 립을 억제하였다. 고마리 추출물의 항균효능을 측정한 결과, Staphylococcus aureus (S. aureus), Staphylococcus epidermidis (S. epidermidis), Propionibacterium acnes (P. acnes)에서도 농도 의존적으로 항균 활성이 확인되었다. 결론: 이상의 연구결과를 통해 고마리 추출물은 항산화, 항염증 및 항균 효과를 가지는 기능성 화장품 소재로서의 효능이 확인되었다. 앞으로 기능성 화장품 소재 로서의 활용 가능성을 판단하기 위하여 임상적으로 동물, 인체실험을 통한 고마리 추출물의 효능 검증 연구가 체계적으로 진행되어야 할 것이다.

        오덕환(五德丸)과 사신환(四神丸)의 항산화 및 항염증 효과

        Objectives : The aim of this study was verification of the anti-oxidative and anti-inflammatory effects of Odukhwan(ODH) and Sasinhwan(SSH) in mouse macrophage, RAW264.7 cells. Methods : To investigate the anti-oxidative effect and scavenging activities of DPPH radical, superoxide anions, nitric oxide and peroxynitrite were measured. Cytotoxic activity of extract of ODH and SSH on RAW264.7 cells was measured using MTS assay. To proof the reductive activity of intracellular oxidation, DCFH-DA assay was performed. The nitric oxide(NO) production was measured and pro-inflammatory cytokines and $PGE_2$ were measured by ELISA kit. The levels of inducible nitric oxide synthase(iNOS), cyclooxygenase-2(COX-2) and nuclear NF-${\kappa}B$ p65 expression were detected by western blot. Results : After those analyses, we bring to a conclusion as follows. Both herbal formulations scavenged DPPH radical and nitric oxide. But ODH had no scavenging activity of superoxide anions and SSH had low scavenging activity in peroxynitrite. And the results indicated that ODH and SSH inhibited the LPS-induced NO, $PGE_2$ production and iNOS, COX-2 expression accompanied by an attenuation of IL-$1{\beta}$ and IL-6 production in RAW264.7 cells. They also have suppression effects of LPS-induced NF-${\kappa}B$ activation. Conclusions : ODH and SSH have anti-oxidative and anti-inflammatory effects and they may be a part of database for development of new anti-oxidative and anti-inflammatory drugs.

        추출 방법에 따른 감길탕가미방(甘桔湯加味方)의 항염증(抗炎症), 진통(鎭痛), 해열(解熱), 항산화(抗酸化) 및 Histamine 투과 억제효과

        Objectives : Modified Gamgil-tang is a prescription commonly used for respiratory diseases. This thesis was carried out to check the treatment effects and diversity of drug formulation by comparing extraction method of ethanol and water of modified Gamgil-tang. Methods : All experiments were carried out with water and 50% ethanol extraction for comparison. In vivo experiment, hyaluronidase inhibitory effects and trypsin inhibitory effects were tested to measure the anti-inflammatory effects activity. Scavenging effects of DPPH free radical, xanthine oxidase inhibitory effects and inhibition on TBA-RS formation were experimented to measure anti-oxidative effects. With the in vivo experiment, ICR group mice and SD group rats were used as experimental animals. An anti-inflammatory effects experiment were carried out to measure the action on carrageenin-induced hind paw edema: analgesic effects were measured using writhing syndrome induced by 0.7% acetic acid in mice: antipyretic effect was measured using endotoxin, and inhibitory effects of increase vascular permeability induced by 0.5% histamine were measured. Results : For extraction of glycyrrhizin contents, ethanol extract was extracted 2 times of that of water extract. Anti-inflammatory effects showed high in ethanol extract. Anti-oxidative effects measured high in ethanol extract. No significant result was found in inhibition on TBA-RS formation. Analgesic effects were found to be similar in water and ethanol extract. Antipyretic effects were found to be stronger in water extract. Inhibitory effects of increase vascular permeability induced by 0.5% histamine showed stronger in ethanol extract. Conclusion : By measuring anti-inflammatory effects, analgesic effects, antipyretic effects, anti-oxidative effects, and histamine permeation inhibition effects both in water extract and ethanol extract after adding agents such as Mentha Herba, Gardenias Fructus, and propolis to existing Gamgil-Tang, ethanol extract was found to be more effective in anti-inflammatory effects, analgesic effects, anti-oxidative effects, and histamine permeation inhibition effects. The converse was found for antipyretic effect.

        찔레뿌리 초임계 추출물의 항균, 항산화 및 항염증 활성

        The roots of Rosa multiflora Thunberg have been used in traditional oriental medicine as remedies for rheumatic arthralgia and scabies. In this study, the anti-fungal, anti-oxidant, and anti-inflammatory activities of a supercritical extract of Rosa multiflora root were investigated in vitro. To investigate the anti-oxidant and anti-inflammatory effects of the supercritical extract, 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid (ABTS) radical scavenging activity, and the inhibition of nitric oxide production in lipopolysaccharide (LPS)-stimulated RAW 264.7 cells were examined, respectively. In addition, the anti-fungal activities of the extract were assessed. The results showed a concentration-dependent, increase in ABTS radical scavenging activity. The supercritical fluid extracts of Rosa multiflora root exhibited low toxicity to RAW 264.7 cells at 100 µg/mL the highest concentration tested. Cells stimulated with LPS produced more nitric oxide than normal control cells; however, cells treated with the supercritical fluid extract decreased this production in a concentration-dependent manner. Finally, the supercritical fluid extracts showed significant anti-fungal activity. These results suggest that extracts of the roots of Rosa multiflora might be used to develop potent anti-fungal, anti-oxidant, and anti-inflammatory agents, and may be useful as ingredients for related new functional cosmetic materials.

        Anti-oxidant, Anti-inflammatory, and Anti-obesity Effects of Ethanol Extracts from Ulmus divididiana var. japonica

        In this study, the anti-oxidant, anti-inflammatory, and anti-obesity activities of an ethanol extracts of Ulmus divididiana var. japonica (UDE) were investigated in vitro and in vivo. UDE anti-oxidant activity was evaluated with an Electron Spin Resonance (ESR) spectrometer, which measured 1, 1-diphenyl-2-picryl-hydrazyl (DPPH) radical scavenging activity. Cell viabilities were estimated using 3-[4,5-Dimethylthiazol-2-yl]-2,5-diphenyl-tetrazolium bromide (MTT) assay. LPS-stimulated BV-2 microglia were used to study the production of nitric oxide (NO). Cells stimulated with LPS produce more NO than normal control cells. However, cells treated with the UDE decreased this production in a concentration dependent manner (100, 250, 500, 1,000 μg/mL). Also, we investigated the anti-obese activity of UDE in SD rats. The SD rats were randomly divided into five groups: 10% low fat diet (N), 45% high fat diet (H), 45% high fat diet + garcinia extracts 200 mg/kg/day (HG200), high fat diet + UDE 200 mg/kg/day (HU200), high fat diet + UDE 400 mg/kg/day (HU400). UDE was found to lower whole body and abdominal and epididymal adipose tissue weights and lowered plasma levels of triglyceride (TG), compared to those in H group. Histological analyses of the liver and fat tissues of rat treated with UDE revealed significantly decreased number of lipid droplets and decreased size of adipocytes compared to the H group. These results suggest that UDE might be used to develop potent anti-oxidant, anti-inflammatory, and anti-obesity agents, and may be useful as ingredients for related new functional raw materials.

        Endlicheria anomala (Nees) Mez 추출물의 항산화, 항염증 및 미백 활성

        본 연구에서는 Endlicheria anomala (Nees) Mez 메탄올 추출물(EAME)의 항산화, 항염증 및 미백 생리활성을 in vitro assay 및 cell culture model system을 이용하여 분석 하였다. EAME의 항산화능을 분석한 결과 DPPH, H2O2로 유도한 ROS, LPS로 유도한 NO 등 다양한 산화적 스트레스원을 효과적으로 소거하였다. 대표적인 항산화 효소들로 천연물에 의한 항산화능 활성에 의해 주로 발현이 유도되는 세효소인 HO-1, TrxR1, NQO1 및 그 전사 인자인 Nrf2의 단백질 발현에 미치는 영향을 분석한 결과 시료 처리 농도의 증가에 따라 세 효소 및 Nrf2의 발현이 유의적으로 증가됨을 보였다. 또한 EAME는 in vitro DOPA oxidation을 강하게 저해하여 tyrosinase inhibitor로서 작용할 가능성을 시사 하였고 이에 B16F10 melanocyte를 이용하여 미백 효능을 분석한 결과 유의적인 melanin 생성억제능 및 tyrosinase 효소 활성 억제능을 보였으며 이는 tyrosinase, TRP-1, TRP-2 등 melanin 생성의 핵심 작용 효소들의 단백질 발현 저해를 통해 일어나는 것으로 나타났다. 이러한 결과를 통해 EAME가 높은 항산화능과 항염증 활성, 그리고 미백 활성을 보유함을 처음으로 밝혔으며 향후 기능성 식품 및 피부 미용 소재로서유용하게 활용될 수 있을 것으로 판단된다. In this study, the anti-oxidative, anti-inflammatory, anti-melanogenic activities of Endlicheria anomala (Nees) Mez methanol extract (EAME) were evaluated by use of in vitro assays and cell culture model systems. The results revealed that EAME scavenges various radicals such as 1,1-diphenyl-2-picryl hydrazyl hydrogen peroxide induced reactive oxygen species, and lipopolysaccharide induced nitric oxide. Furthermore, EAME induced the expression of anti-oxidative enzymes such as heme oxygenase 1, thioredoxin reductase 1, NAD(P)H dehydrogenase 1, and their upstream transcription factor, nuclear factor-E2- related factor 2. Moreover, EAME inhibited in vitro DOPA oxidation and 3-isobutyl-1-methylxanthine induced melanogenesis in B16F10 cells. Its anti-melanogenic activity will have originated from the inhibition of tyrosinase enzyme activity and melanogenesis related protein expression. Taken together, these results provide the important new insight that E. anomala possesses various biological activities such as anti-oxidative, anti-inflammatory, and anti-melanogenic. Therefore, it might be utilized as a promising material in the fields of nutraceuticals and cosmetics.

        유기 게르마늄 농축수로 재배한 농산물의 항염 및 함암효과

        The study was conducted to identify the anti-inflammatory and anti-cancer effects in sprouts of mouse-eyed bean (Rhynchosia nulubilis), ginseng (Panax ginseng), perilla (Perilla frutescens), broccoli (Brassica oleracea var. italica), and lettuce (Lactuca sativa) grown with organic germanium concentrate. Western blot analysis was performed to assess the anti-inflammatory activity of the extract. All extracts exhibited noticeable anti-oxidant activity, indicating a significant correlation between the germanium content and anti-oxidant activity (p<0.05). In particular, rat-eyed bean sprouts with the highest germanium content showed significant anti-inflammatory activity (p<0.05) by significantly inhibiting the expression of the inflammatory complexes, NLRP3, cytokines IL-1β and caspase-1. Ginseng and broccoli sprouts showed strong anti-cancer properties and had high anti-oxidant effects (p<0.001). Germanium-concentrated water allows the mass production of agricultural products containing high concentrations of organic germanium. Agricultural produce grown with germanium concentrate add organic germanium to various physiological active ingredients, increasing the anti-oxidant and anti-cancer effects. These results strongly suggest that agricultural products containing high germanium concentrations can be used as novel health supplements to improve health.

        청아지황탕(靑蛾地黃湯)의 RAW264.7 Cell에서 항산화 및 항염증 효능 연구

        Objectives This study was designed to investigate whether the Cheongajihwang-Tang (CT) has an inhibitory effect association with oxidation or inflammation in RAW264.7 cells. Methods Cytotoxic activity of CT extract on RAW264.7 cells was evaluated by using 3-[4, 5-dimethylthiazol-2-yl]-2, 5-diphenyltetrazolium bromide (MTT) solution. Nitric oxide production was measured using Griess reagent system. The total phenolic contents and 1,1-Diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity was measured to evaluate the anti-oxidative effects of CT. Dichlorofluorescin diacetate (DCFH-DA) has been used as a substrate for measuring intracellular oxidant production. Results Cheongajihwang-Tang does not impair the cell viability in tested concentration. CT showed anti-oxidative effects in vitro by decreasing electron donating ability, and also showed anti-inflammatory effects suppressing NO and ROS expression in LPS induced RAW264.7 activation. CT inhibited the generation of intracellular ROS production as dose dependant manner. Conclusions CT has anti-oxidative effects and anti-inflammatory activities. These results indicate that CT extract has an anti-inflammatory activities via anti-oxidative effects. (J Korean Med Rehabil 2016;26(3):51-58)

        Comparison of anti-oxidant activities of seventy herbs that have been used in Korean traditional medicine

        Many herbs have been used as therapeutics in Korean traditional medicine. In view of their clinical indications, anti-oxidant activity may contribute to their pharmacological effects. However, anti-oxidant information on these plants has not been available. In this study, seventy herbs which have been used in Korean traditional medicine were selected and screened for anti-oxidant activity using their water extracts. The anti-oxidant activity was assessed by their ability to inhibit three oxidation reactions; luminol/Fenton reagent, 2, 7-dichlorodihydrofluorescein (DCHF)/Fenton reagent and DCHF/peroxynitrite. In each assay, 70 herbs were divided into two groups; anti-oxidant group which inhibited the respective oxidation reaction and was majority (about 60 herbs), and pro-oxidant group which enhanced the oxidation reaction but was minority (more or less 10 herbs). When the herbs were listed in the order of their anti-oxidant strength, the orders obtained from each assay were found to be quite similar. The upper top rankers (more or less 10 herbs) in each assay showed strong activity compared to the others. The uppermost rankers in each assay were Rubus coreanus Miquel/ Rubus schizostylus (覆盆子), Schisandra chinensis Baillon/ Schizandra chinensis (五味子) and Terminalia chebula Retzius/ Terminalia chebula (訶子). Of the pro-oxidant herbs, about 4-5 herbs were strongly pro-oxidant, which enhanced the control oxidation reactions to 150-300%. But the meaning of this observation is not known since few of them in one assay were also anti-oxidant in other assays. The results obtained in the present study may serve as information for understanding pharmacological effects of these herbs and developing new drugs from them.
