        What Are the Differences in Psychological Outcomes Between People Who Posted COVID-19-Related Content on Social Media and Those Who Did Not?

        Objective During the lockdown of cities and home quarantine, media became the only way for people to conveniently get coronavirus disease-2019 (COVID-19)-related information. And media engagement was closely related to psychological outcomes. But fewer researchers took COVID-19-related posting behaviors into consideration. Therefore, the present study aimed at examining the differences in psychological outcomes between people who posted COVID-19-related content on social media and those who did not. Methods The present study included 917 participants (304 males, 613 females) who had answered the questionnaires of media engagement, positive affect, negative affect, depression, anxiety, stress, satisfaction with life, death anxiety, and meaning in life. Results Results of t-tests showed that the Post group had lower levels of negative affect, anxiety, stress, and death anxiety than the Not Post (Npost) group. Network comparison tests indicated that the Npost group’s network and the Post group’s network differed in global strength, two edge-weights, and node centrality indices. Conclusion The results indicated that more attention should be paid to people who did not post any COVID-19-related content, especially when they have higher levels of stress and depression to prevent comorbidities. And for people who posted content, more attention should be paid when they have a higher level of negative affect.

      • Equivocal Association of RAD51 Polymorphisms with Risk of Esophageal Squamous Cell Carcinoma in a Chinese Population

        Aim: To study the contribution of genetic variation in RAD51 to risk of esophageal squamous cell carcinoma (ESCC). Methods: Three single nucleotide polymorphisms (SNPs) in RAD51 (rs1801320, rs4144242 and rs4417527) were genotyped in 316 ESCC patients and 316 healthy controls in Anyang area of China using PCR-RFLP (polymerase chain reaction-restriction fragment length polymorphism). Demographic variables between cases and controls were statistically compared by T test and Chi-square test. Hardy-Weinberg equilibrium was evaluated by the Chi-square test. Odds ratios (ORs) and 95% confidence intervals (CIs) were calculated to measure any association with ESCC. Haplotype frequencies were estimated by Phase 2.1. Result: The genotype frequencies of rs1801320, rs4144242 and rs4417527 in patients with ESCC demonstrated no significant differences from those in control group (P>0.05). When the haplotypes of these three SNPs were constructed and their relationships with ESCC risk investigated, however, CGG was observed to increase the risk (P=0.020, OR=2.289). Conclusions: There was no association between the three SNPs of RAD51 and ESCC susceptibility in our Chinese population. However, the CGG haplotype might be a risk factor.

        Genotoxicity of Zizyphi Spinosi Semen in Bacterial Reverse Mutation (Ames) Test, Chromosomal Aberration and Micronucleus Test in Mice

        Zizyphi spinosi semen (Z. spinosi) has been used in traditional Chinese medicine for the treatment of rheumatoid arthritis and wounds. However, toxicity in high doses was often observed due to the presence of alkaloids. This study was conducted to investigate the potential genotoxicity of Z. spinosi in vitro and in vivo. This was examined by the Bacterial reverse mutation (Ames) test using Salmonella typhimurium TA98, TA100, TA1535, TA1537 and Escherichia coli WP2uvrA, Chromosomal aberration was investigated using Chinese hamster lung cells and the micronucleus test using mice. Z. Spinosi did not induce mutagenicity in the Ames test, and it did not produce chromosomal aberration in Chinese hamster lung cells with and without metabolic activation, nor in the micronucleated polychromatic erythrocytes in the bone marrow cells in mice. Based on these results, it is concluded that Z. spinosi does not have mutagenic potential under the conditions examined in each study.

      • Lack of Mutagenicity Potential of Periploca sepium Bge. in Bacterial Reverse Mutation (Ames) Test, Chromosomal Aberration and Micronucleus Test in Mice

        Objectives: The root barks of Periploca sepium Bge. (P. sepium) has been used in traditional Chinese medicine for healing wounds and treating rheumatoid arthritis. However, toxicity in high-doses was often diagnosed by the presence of many glycosides. The potential mutagenicity of P. sepium was investigated both in vitro and in vivo. Methods: This was examined by the bacterial reverse mutation (Ames) test using Escherichia coli WP2uvrA and Salmonella typhimurium strains, such as TA98, TA100, TA1535, and TA1537. Chromosomal aberrations were investigated using Chinese hamster lung cells, and the micronucleus test using mice. Results: P. sepium did not induce mutagenicity in the bacterial test or chromosomal aberrations in Chinese hamster lung cells, although metabolic activation and micronucleated polychromatic erythrocytes were seen in the mice bone marrow cells. Conclusions: Considering these results, it is suggested that P. sepium does not have mutagenic potential under the conditions examined in each study.

        Impact of Initial Moisture Content on the Shrinkage-Swelling Behavior of Heishan Bentonite

        Bentonite is generally used as buffer or backfill material in geological repositories and its mechanical behavior is important to affect surrounding structures. In this paper, cyclic shrinkage-swelling tests are performed on Heishan bentonite taking into account the effect of initial moisture content. Results reveal that the shrinkage-swelling behavior of Heishan bentonite relies on initial moisture content and an obvious water retention property is obtainedwith the increases of initial moisture content. To investigate the accurate determination of aforementioned behavior of studied bentonite during drying and wetting cycle, the captured image of digital camera and the binary image processing technique are employed in this study. A simplified analytical model is proposed to simulate the shrinkage-swelling behavior at different initial moisture contents and the model parameters are calibrated by experimental results of Heishan bentonite. Based on the model performance, the model simulation agrees well with the tests and the proposed approach is able to predict the behavior of Heishan bentonite.

        NF-κB decoy potentiates the effects of radiation on vascular smooth muscle cells by enhancing apoptosis

        NF-κB promotes cell survival gainst external stress such as radiation. We examined whether NF-κB decoy transfection enhances the antiproliferative effects of radiation on vascular sm ooth muscle cells (VSMCs) in vitro. The irradiation induced activation or nuclear translocation of NF-κB p65 in VSMCs was confirmed by mmunofluorescence. NF-κB decoy transfection resulted in inhibition of the radiation-induced NF-κB activation in VSMCs and the subsequent reduction of transcription and translocation of ICAM, iNOS, and TNF-α, downstream molecules under the control of NF-κB. By using MTT assay, NF-κB decoy augmented the antiproliferative effects of radiation, where the effect of low dose radiation (2 and 8-Gy) of the cells transfected with NF-κB decoy was equivalent to the high dose (16-Gy) irradiated non-transfected cells at 48 h after irradiation: 1.06 ± 0.16, 1.11 ± 0.22, 1.20 ± 0.25, respectively. The decrease in proliferation and survival of the radiation treated cells by flow cytometry analysis showed that NF-κB inhibition did not show any additive effects on the cell cycle of the irradiated VSMCs, while apoptosis was significantly increased after NF-κB decoy transfection in the irradiated VSMCs (apoptosis fraction: 13.33 ± 2.08% vs. 26.29 ± 7.43%, for radiation only vs. radiation+NF-κ B decoy transfection, P < 0.05). In addition, at 48 h, NF-κB decoy transfection dose dependently (10 µM vs. 20 µM) inhibited proliferation of 16Gy-irradiated VSMCs, and showed greater antiproliferative efficacy than 100 µM sulfasalazine, a specific NF-κB inhibitor. These results indicate that NF-κB inhibition reduces proliferation and survival of irradiated VSMCs, likely by increased apoptosis rather than additive cell cycle arrest and suggest the possibility of adjunctive gene therapy using NF-κB decoy to improve efficacy and to decrease the adverse effects of intracoronary radiation therapy.

      • MiRNA Synergistic Network Construction and Enrichment Analysis for Common Target Genes in Small-cell Lung Cancer

        Background: Small-cell lung cancer (also known as SCLC) is an aggressive form and untreated patients generally die within about 3 months. To obtain further insight into mechanism underlying malignancy with this cancer, an miRNA synergistic regulatory network was constructed and analyzed in the present study. Method: A miRNA microarray dataset was downloaded from the NCBI GEO database (GSE27435). A total of 546 miRNAs were identified to be expressed in SCLC cells. Then a miRNA synergistic network was constructed, and the included miRNAs mapped to the network. Topology analysis was also performed to analyze the properties of the synergistic network. Consequently, we could identified constitutive modules. Further, common target genes of each module were identified with CFinder. Finally, enrichment analysis was performed for target genes. Results: In this study, a miRNA synergistic network with 464 miRNAs and 2981 edges was constructed. According to the topology analysis, the topological properties between the networks constructed by LC related miRNAs and LC unrelated miRNAs were significantly different. Moreover, a module cilque0 could be identified in our network using CFinder. The module included three miRNAs (hsa-let-7c, hsa-let-7b and hsa-let-7d). In addition, several genes were found which were predicted to be common targets of cilque0. The enrichment analysis demonstrated that these target genes were enriched in MAPK signaling pathways. Conclusions: Although limitations exist in the current data, the results uncovered here are important for understanding the key roles of miRNAs in SCLC. However, further validation is required since our results were based on microarray data derived from a small sample size.

        Social Capital in Non-profit Organizations - A Case Study in Japan -

        This empirical research focuses on the staff engaged in NPOs based on the aggressiveness and degree of fulfillment of NPO activities and it is aimed to verify the relationship with social capital, using the survey results on the non-profit organization S.S.F. in Saga Prefecture in Japan. The correlation analysis between "Fulfillment and Aggressiveness" and "Social Capital", "Sample characteristics" and "Social Capital", "Fulfillment and Aggressiveness" and "Sample characteristics" are conducted, and conclude the following results. (1) People who have high confidence in society have a keen sense of fulfillment obtained by the activities of NPOs. (2) People working at elevated position in the non-profit organization and with a lot of working hours are not active with friends, but have aggressive exchanges with colleagues. (3) People being in an elevated position in the NPO is associated with frequent cooperation with an external organization and detailed information in region. Overall, findings show that the role of staff in NPOs is important to further activate the acvtivities of NPOs, so deepening the ties between staff and organizations, and building good trusting relationships will make NPO acvtivities more efficient.
