      • Poster Session:PS 0234 ; Gastroenterology : Effect of Mir-34a in Regulating Steatosis by Targeting PPARa Expression in NAFLD

        Background: The association between altered expression of miR-34a and pathophysiological features of nonalcoholic fatty liver disease (NAFLD) and whether there is a connection between susceptibility to NAFLD has not been completely clarifi ed. Methods: The vitro model was established by culturing L02 cells with a high concentration of free fatty acid (FFA) and the vivo model was established by feeding C57BL/6 mice with HFD. To determine the effects of miR-34a, cultured L02 cells transfected with miR-34a inhibitor and C57BL/6 mice injected with miR-34a inhibitor through vein tail were analysed for the level of PPARaand the metabolic sensor AMPK. In functional experiments, TG content and steatosis degree were measured by TG assay kit, HE and Oil Red O staining. Results: miR-34a expression is signifi cantly upregulated in steatosis-induced hepatocytes and in liver tissues of HFD fed mice. The upregulation of miR-34a resulted in the downregulation of hepatic peroxisome proliferator-activated receptor-a(PPARa), the direct target of miR-34a. Moreover, the action of miRNA-34a on PPAR-a depends on the presence of a single miRNA-34a binding site. Silencing miR-34a led to an initial increase the level of PPARaand the targets of PPARa, including CPT1, CPT2, SLC27A4, SLC27A1 and ACBD3. Activation of the central metabolic sensor AMPK was also increased. In functional experiments, miR-34a inihibitor suppressed cell and mice liver TG content and improve steatosis degree. Furthermore, inhibition of PPARa expression aggravated hepatocellular steatosis in vitro models. Conclusions: NAFLD is associated with altered hepatic miR-34a expression. Decreased expression of miR-34a potentially contributes to altered lipid metabolism implicated in the pathogenesis of NAFLD. These results suggest that regulating it`s target PPARa by down-regulation of miR-34a levels may be a therapeutic strategy against NAFLD.

        Background: The mechanisms of pathogenesis underlying nonalcoholic fatty liver disease (NAFLD) remain unclear at present and the depth study of HADHA in the development of NAFLD has never been investigated. Methods: The NAFLD cell model was established by treating L02 cells with free fatty acid (FFA) overload. Small interfering RNA (siRNA) was used to knock down HADHA level. The expression of HADHA and key enzymes associated with fatty acid beta- oxidation in L02 cells were determined by q-PCR. Key protein associated in energy metabolism, endoplasmic reticulum(ER) stress and infl ammatory were determined by western blotting. ATP, hydroperoxide (H2O2), catalase (CAT) and mitochondrial membrane potential (MMP) were measured. The prediction of HADHA upstream regulation of miRNA was carried out and luciferase reporter assays were implemented to validate the prediction. Results: After culturing L02 cells by FFA for 48h, we detect the increased protein level of HADHA. HADHA knockdown in L02 cells resulted in an increased of lipid accumulation and downregulation of gene expression involved in fatty acid beta-oxidation, including PPARa, ACOX1, CPT2, EHHADH, ECHS1, HADHB and HADH. Additionally, administering HADHA siRNA exhibited improvement of oxidative stress, embodied in decreased level of H2O2 and MDA, meanwhile, increased levels of ATP, CAT and MMP. Furthermore, HADHA knockdown demonstrated weakened AMPK pathway, activation of MAPK and MKK3 pathway, and improve ER stress by downregulation of C/EBPa and C/EBPß. Moreover, HADHA was regulated directly by upstream gene of miR-124. Conclusions: Our results show that HADHA may plays a role of double-edged sword in hepatic steatosis and cell injury in NAFLD, and provide a new insight into the pathogenic mechanisms of NAFLD, may becoming a potential new therapeutic target for NAFLD.

      • RAMS evaluation for a steel-truss arch high-speed railway bridge based on SHM system

        The evaluation theory of reliability, availability, maintainability and safety (RAMS) as a mature theory of state evaluation in the railway engineering, can be well used to the evaluation, management, and maintenance of complicated structure like the long-span bridge structures on the high-speed railway. Taking a typical steel-truss arch bridge on the Beijing-Shanghai high-speed railway, the Nanjing Dashengguan Yangtze River Bridge, this paper developed a new method of state evaluation for the existing steel-truss arch high-speed railway bridge. The evaluation framework of serving state for the bridge structure is presented based on the RAMS theory. According to the failure-risk, safety/availability, maintenance of bridge members, the state evaluation method of each monitoring item is presented. The weights of the performance items and the monitoring items in all evaluation levels are obtained using the analytic hierarchy process. Finally, the comprehensive serving state of bridge structure is hierarchical evaluated.

        S100A4 Gene is Crucial for Methionine-Choline-Deficient Diet-Induced Non-Alcoholic Fatty Liver Disease in Mice

        Purpose: To explore the influence of S100 calcium binding protein A4 (S100A4) knockout (KO) on methionine-choline-deficient(MCD) diet-induced non-alcoholic fatty liver disease (NAFLD) in mice. Materials and Methods: S100A4 KO mice (n=20) and their wild-type (WT) counterparts (n=20) were randomly divided into KO/MCD, Ko/methionine-choline-sufficient (MCS), WT/MCD, and WT/MCS groups. After 8 weeks of feeding, blood lipid and liverfunction-related indexes were measured. HE, Oil Red O, and Masson stainings were used to observe the changes of liver histopathology. Additionally, expressions of S100A4 and proinflammatory and profibrogenic cytokines were detected by qRT-PCR andWestern blot, while hepatocyte apoptosis was revealed by TUNEL staining. Results: Serum levels of aminotransferase, aspartate aminotransferase, triglyceride, and total cholesterol in mice were increasedafter 8-week MCD feeding, and hepatocytes performed varying balloon-like changes with increased inflammatory cell infiltrationand collagen fibers; however, these effects were improved in mice of KO/MCD group. Meanwhile, total NAFLD activity scoresand fibrosis were lower compared to WT+MCD group. Compared to WT/MCS group, S100A4 expression in liver tissue of WT/MCD group was enhanced. The expression of proinflammatory (TNF-α, IL-1β, IL-6) and profibrogenic cytokines (TGF-β1, COL1A1,α-SMA) in MCD-induced NAFLD mice were increased, as well as apoptotic index (AI). For MCD group, the expressions ofproinflammatory and profibrogenic cytokines and AI in KO mice were lower than those of WT mice. Conclusion: S100A4 was detected to be upregulated in NAFLD, while S100A4 KO alleviated liver fibrosis and inflammation, inaddition to inhibiting hepatocyte apoptosis.

        An available technology of special microprocessor communication (SMC) in a local telephone network is put forward in this paper. This techology has been used in a long distant computer communication system successfully. The system of SMC may not use common computer and MODEM, thereby the communication cost would be decreased largely. The system is suitable for ordinary users and will accelerate the step of computer communication in China.

        Phylogenetic Diversity and Antifungal Activity of Endophytic Fungi Associated with Tephrosia purpurea

        Sixty-one endophytic fungus strains with different colony morphologies were isolated from the leaves, stems and roots of Tephrosia purpurea with colonization rates of 66.95%, 37.50%, and 26.92%, respectively. Based on internal transcribed spacer sequence analysis, 61 isolates were classified into 16 genera belonging to 3 classes under the phylum Ascomycota. Of the 61 isolates, 6 (9.84%) exhibited antifungal activity against one or more indicator plant pathogenic fungi according to the dual culture test. Isolate TPL25 had the broadest antifungal spectrum of activity, and isolate TPL35 was active against 5 plant pathogenic fungi. Furthermore, culture filtrates of TPL25 and TPL35 exhibited greater than 80% growth inhibition against Sclerotinia sclerotiorum. We conclude that the endophytic fungal strains TPL25 and TPL35 are promising sources of bioactive compounds.

        Age-stage, two-sex life tables of the maple poison moth (Lymantria nebulosa Wileman) at different ambient temperatures

        The maple poison moth (MPM), Lymantria nebulosa (Wileman), is a Lepidopteran defoliator and one of the most important pests of Liquidambar formosana Hance. The objective of this study was to determine the effects of temperature on the population growth and reproduction of this species. MPM were raised in artificial climate chambers in a laboratory at 21 ± 1°C, 24 ± 1°C, 27 ± 1°C, 30 ± 1°C, 33 ± 1°C and 36 ± 1°C, under a relative humidity (RH) of 70% ± 10% and a 14 L:10D photoperiod, and age-stage, two-sex life tables complied for insects at each temperature. The developmental duration, adult preoviposition period and overall preoviposition period decreased with temperature in a time-dependent manner. The growth curves of the different temperature groups overlapped considerably. The highest larval survival rate (80%) was recorded at 30 ± 1°C. At 36°C only a few eggs hatched and the first instar could not complete the first molt. We consequently regard 36°C as the lethal temperature threshold for this species. Life expectancy decreased with age under all six temperatures. The endogenous growth rate was significantly lower at 21 ± 1°C than at the other temperatures, and was highest in 30 ± 1°C group. The net growth rate of the 30 ± 1°C group was also significantly higher than others. Reproductive parameters were highest at 27 ± 1°C and 30 ± 1°C. Although environmental factors absent from the laboratory environment may limit the generality of our results, they nonetheless provide a theoretical basis for developing integrated control methods for MPM in the wild.

        Hepatitis B Virus Core Protein Mediates the Upregulation of C5α Receptor 1 via NF-κB Pathway to Facilitate the Growth and Migration of Hepatoma Cells

        Purpose C5α receptor 1 (C5ΑR1) is associated with the development of various human cancers. However, whether it is involved in the development of hepatitis B virus (HBV)–related hepatocellular carcinoma (HCC) is poorly understood. We explored the expression, biological role, and associated mechanisms of C5AR1 in HBV-related hepatoma cells. Materials and Methods The expression of C5ΑR1 mediated by HBV and HBV core protein (HBc) was detected in hepatoma cells. The function of nuclear factor кB (NF-κB) pathway in HBc-induced C5AR1 expression was assessed. The roles of C5ΑR1 in the activation of intracellular signal pathways, the upregulation of inflammatory cytokines, and the growth and migration of hepatoma cells mediated by HBc, were investigated. The effect of C5α in the development of HCC mediated by C5AR1 was also measured. Results C5ΑR1 expression was increased in HBV-positive hepatoma cells. Dependent on HBc, HBV enhanced the expression of C5ΑR1 at the mRNA and protein levels. Besides, HBc could promote C5ΑR1 expression via the NF-κB pathway. Based on the C5ΑR1, HBc facilitated the activation of JNK and ERK pathways and the expression and secretion of interleukin-6 in hepatoma cells. Furthermore, C5ΑR1 was responsible for enhancing the growth and migration of hepatoma cells mediated by HBc. Except these, C5α could promote the malignant development of HBc-positive HCC via C5AR1. Conclusion We provide new insight into the mechanisms of hepatocarcinogenesis mediated by HBc. C5ΑR1 has a significant role in the functional abnormality of hepatoma cells mediated by HBc, and might be utilized as a potential therapeutic target for HBV-related HCC.
