        Heme oxygenase-1 (HO-1)/carbon monoxide (CO) axis suppresses RANKL-induced osteoclastic differentiation by inhibiting redox-sensitive NF-κB activation

        Heme oxygenase (HO-1) catalyzes heme to carbon monoxide (CO), biliverdin/bilirubin, and iron and is known to prevent the pathogenesis of several human diseases. We assessed the beneficial effect of heme degradation products on osteoclastogenesis induced by receptor activator of NF-κB ligand (RANKL). Treatment of RAW264.7 cells with CORM-2 (a CO donor) and bilirubin, but not with iron, decreased RANKLinduced osteoclastogenesis, with CORM-2 having a more potent anti-osteogenic effect. CORM-2 also inhibited RANKLinduced osteoclastogenesis and osteoclastic resorption activity in marrow-derived macrophages. Treatment with hemin, a HO-1 inducer, strongly inhibited RANKL-induced osteoclastogenesis in wild-type macrophages, but was ineffective in HO-1<sup>+/- </sup>cells. CORM-2 reduced RANKL-induced NFATc1 expression by inhibiting IKK-dependent NF-κB activation and reactive oxygen species production. These results suggest that CO potently inhibits RANKL-induced osteoclastogenesis by inhibiting redox-sensitive NF-κB-mediated NFATc1 expression. Our findings indicate that HO-1/CO can act as an antiresorption agent and reduce bone loss by blocking osteoclast differentiation. [BMB Reports 2017; 50(2): 103-108]

        A new logistics concept is needed through the sharing information between suppliers and consumers, which maximizes the customers service and its flexibility by changing functional-oriented to process-oriented. However in the real world, it is known that it takes long time to build vehicle scheduling in the process of transporting the amount of orders from the logistics center to the vendors due to the realistic constraints. This paper presents a design for each process enabling delivery planning system for the improvement logistics plant services. In addition, an interactive automatic delivery planning system is implemented utilizing the proposed algorithm.

      • 비만의 원인과 치료에 대한 동서의학적 고찰

        The purpose of this study was to investigate efficient treatment in obesity. In order to help clinical application in obesity treatment, this study was performed to investigate the cavse and treatment in obesity. The main finding of this study were an follows. 1. In the cause of oriental medicine in obesity, that were moisture(濕), phlegm(痰), gore(瘀), the productive heat(積熱), overeating, supernutrition, underaction, heredity and habitude, psychologic factor(stress) etc. 2. In the western medicine, the cause of obesity were genetic cause, endocrine disorder, hypothalamic extraodinary and secondary effect of drug. But simple obesity is the greater part. Also there were psychologic and environmental causes. 3. Ear acupuncture of wide application were Naebunbi, Sinmun, Taejang ijo˘m. Pyejo˘m, Bijo˘m etc. 4. Body acupuncture of application were Naejo˘ng, Sanggo˘ho˘, Pungnyung, Kokchi, Samo˘mgyo, Umno˘ngcho˘n etc. 5. In oriental medical treatment of obesity, Bangpungtongs ngsan(防風通聖散), Daesihotang(大柴胡湯), Seungkitang(承氣湯), Bangkihwangkitang(防己黃??湯), Richulsaryeungtang(二朮四??湯), Taeksatang(澤瀟湯), Opieum(五皮飮), Gongyundan(控涎丹) etc were wide applicated. 6. The western medical treatment in obesity encourage the dietary cure, exercise cure and behavior adjustment than medical therapy or operation. In treatment of obesity the dietary cure and Herb-drug therapy, ear acupuncture and body acupuncture are effective. But weight maintenance after treating is more important. After all, the continuous dietary cure, behavior adjusting cure and exercise cure are necessary.

        Antioxidant and hepatoprotective effects of Korean ginseng extract GS-KG9 in a D-galactosamine-induced liver damage animal model

        BACKGROUND/OBJECTIVES: This study was designed to investigate the improvement effect of white ginseng extract (GS-KG9) on D-galactosamine (Ga1N)-induced oxidative stress and liver injury. SUBJECTS/METHODS: Sixty Sprague-Dawley rats were divided into 6 groups. Rats were orally administrated with GS-KG9 (300, 500, or 700 mg/kg) or silymarin (25 mg/kg) for 2 weeks. The rats of the GS-KG9- and silymarin-treated groups and a control group were then intraperitoneally injected Ga1N at a concentration of 650 mg/kg for 4 days. To investigate the protective effect of GS-KG9 against GalN-induced liver injury, blood liver function indicators, anti-oxidative stress indicators, and histopathological features were analyzed. RESULTS: Serum biochemical analysis indicated that GS-KG9 ameliorated the elevation of aspartate aminotransferase (AST), alanine aminotransferase (ALT), alkaline phosphatase (ALP), and lactate dehydrogenase (LDH) in GalN-treated rats. The hepatoprotective effects of GS-KG9 involved enhancing components of the hepatic antioxidant defense system, including glutathione, glutathione peroxidase (GPx), superoxide dismutase (SOD), and catalase (CAT). In addition, GS-KG9 treatment inhibited reactive oxygen species (ROS) production induced by GalN treatment in hepatocytes and significantly increased the expression levels of nuclear factor erythroid-2-related factor 2 (Nrf2) and heme oxygenase-1 (HO-1) proteins, which are antioxidant proteins. In particular, by histological analyses bases on hematoxylin and eosin, Masson"s trichrome, α-smooth muscle actin, and transforming growth factor-β1 staining, we determined that the administration of 500 mg/kg GS-KG9 inhibited hepatic inflammation and fibrosis due to the excessive accumulation of collagen. CONCLUSIONS: These findings demonstrate that GS-KG9 improves GalN-induced liver inflammation, necrosis, and fibrosis by attenuating oxidative stress. Therefore, GS-KG9 may be considered a useful candidate in the development of a natural preventive agent against liver injury.

      • 데이타베이스 시스템을 이용한 웹 자원 관리 엔진의 설계 및 구현

        In this paper, we propose a web resource management engine based on general DBMSs. The web resources management engine manages the meta information describing the characteristics of resourves in a target web site. The engine extracts the meta information from HTML documents in the site and stores it into a predefined database called virtual schema. We use a general-purpose DBMS for maintaining the virtual schema. For design of the virtual schema, we model each element of HTML documents as a DBMS relation, and each attribute of an element as an attribute of its corresponding DBMS relation. We also model the link information among HTML documents as another relation. In order to merge the web and database having the virtual schema, we employ some additional tags in HTML documents, which were originally proposed in the INFORMIX WebDataBlade. Our web resource management engine has the following good features: (1) it supports dynamic changes of the web resources in a site by issuing update queries on the virtual schema, (2) it provides such searches that are based on structural characteristics of HTML documents since the virtual schema keeps the necessary information, (3) it provides the standard SQL interface for users to simply search and update the web resources, and finally, (4) it is portable to any DBMSs by just extending the gateway between the web and database since it only uses the common facilities supported by general-purpose DBMSs.

      • 대장균내에서 발현된 가용성 IL-6의 정제 및 Alanine-Scanning Mutagenesis에 의한 IL-6 Mutant의 제조

        Interleukin-6(IL-6) is a multifunctional cytokine that acts on various target cells to induce a diversity of biological responses. Studies were performed on the purification of soluble and functional IL-6 expressed in the periplasm of Escherichia coli. The coding region of the IL-6 gene was fused to the pe1B secretion signal sequence and the expression was under the control of T7 promotor. Upon cell induction under 26 C, recombinant IL-6 was overexpressed in the periplasmic soluble fraction. The periplasmic soluble extract was already 50% pure and was further purified to homogeneity by gel filtration and Mono-Q column chromatography. About 1.5 mg pure protein was obtained from 11 cultured cells. The recombinant IL-6 had a specific activity_ of 1 x 108 U/mg in B9 bioassay, which is eqivalent to that of the natural IL-6. To understand the structure-functional relationship of IL-6, twenty two alanine substitutions of IL-6 have been generated in helix B and surrounding loop regions (amino acid residues 51-109). Most of the mutant proteins were soluble, but those having alanine substitution at Asp7l, G1u93, Phe 94, G1u95, Tyr97, and Arg104 showed little expression or the reduced yield, suggesting that these residues may involve in the folding process or the stability of IL-6 tertiary structure.

        Tag식 개방계 장치를 이용한 알콜류의 인화점 및 연소점 측정

        The flash point is one of the most important combustible properties used to determine the potential for the fire and explosion hazards of industrial material and the fire point is the temperature of the flammable liquid at which there will be flaming combustion, sustained 5 seconds in response to the pilot flame. In this study, the flash point and fire point were measured to present raw data of the flammable risk assessment for alcohols, using Tag open-cup apparatus(ASTM D 1310-86). The measured values were compared with the calculated values based on 0.78 times stoichiometric concentration. The values calculated by the proposed equations were in good agreement with the measured values.

        SC 합성 기둥-철골 보 접합부의 내진성능에 관한 실험적 연구

        Steel-concrete composite column is a recently developed composite system, in which the two opposite flanges of H-shape section are connected by welded links, and vacant space enclosed by the flanges, web and links is filled with concrete. The purposes of this paper are to suggest efficient moment-connection details for SC composite column system and to verify their seismic performance. Two types of SC composite column-steel beam connections were proposed; one for strong axis of column and the other for weak axis of column. Cyclic loading tests for two full-scaled specimens were performed according to the standard loading history of SAC(2000). Moment-connection detail for strong axis of column showed sufficient seismic performance required for SMRF(special moment resisting frame). However, connection detail for weak axis of column exhibited inferior seismic performance than that for strong axis of column, which satisfied the requirements for OMRF(ordinary moment resisting frame). Additional studies are needed to increase elastic stiffness and seismic performance of connection detail for weak axis of SC composite column.
