        Distortional buckling of a CFS channel section with and without stiffened flanges

        Cold-formed steel (CFS) sections are commonly applied to modern engineering structures, such as roof truss, purlin and industrial goods rack. This study proposes an analytical model to investigate the distortional buckling behavior of CFS-lipped channel sections considering two load scenarios (i.e., axial compression and pure bending). The formulae and analytical solution for calculating the distortional buckling critical stress of CFS channel sections are derived on the basis of the total potential energy principle. The proposed model is extended to the channel section columns and beams with a stiffened flange. CUFSM and generalized beam theory (GBT) are used to conduct numerous channel section columns and beams to validate the proposed method. Results obtained from the proposed model are compared with those calculated using GBT and/or finite-strip code CUFSM. These numerical results are consistent with the model calculations for channel section with and without stiffeners.

        A hemoglobin-based oxygen-carrying biomimetic nanosystem for enhanced chemo-phototherapy and hypoxia alleviation of hepatocellular carcinoma

        Tumor microenvironment is characterized by low pH, high reactive oxygen species and hypoxia, whichprovides a suitable environment for cancer growth. The hypoxia not only elevates tumor angiogenesisand metastasis, but also is responsible for the development of treatment resistance, which graduallybecomes a significant impediment for cancer therapy. Therefore, we developed a biomimetic nanosystemcontaining hemoglobin extracted from red blood cells, chemotherapy drug sorafenib, sensitizer ursolicacid and photosensitizer indocyanine green for enhanced chemo-photo combination therapy of hepatocellularcarcinoma, which could not only enhance the chemotherapy effect of sorafenib bowing to thesensitizing effect of ursolic acid, but also achieved synergetic phototherapy in virtue of indocyaninegreen. Besides, the nanoparticles could effectively delivery exogenous oxygen to tumor site and amelioratethe tumor hypoxic environment with the assistance of hemoglobin. The dual-sensitization drugdelivery system was expected to effectively reduce the resistance of traditional treatment methodsagainst tumor hypoxia, providing a novel prospect for the synergistic hepatocellular carcinomatreatment.

        Structures, biomimetic synthesis, and anti-SARS-CoV-2 activity of two pairs of enantiomeric phenylpropanoid-conjugated protoberberine alkaloids from the rhizomes of Corydalis decumbens

        (±)-Decumicorine A ( 1 ) and (±)- epi -decumicorine A ( 2 ), two pairs of enantiomeric isoquinoline alkaloids featuring a novel phenylpropanoid-conjugated protoberberine skeleton, were isolated and purifi ed from the rhizomes of Corydalis decumbens . The separation of (±)- 1 and (±)- 2 was achieved by chiral HPLC to produce four optically pure enantiomers. The structures and absolute confi gurations of compounds (−)- 1 , (+)- 1 , (−)- 2 , and (+)- 2 were elucidated by spectroscopic analysis, ECD calculations, and X-ray crystallographic analyses. The two racemates were generated from a Diels-Alder [4 + 2] cycloaddition between jatrorrhizine and ferulic acid in the proposed biosynthetic pathways, which were fully verifi ed by a biomimetic synthesis. Moreover, compound (+)- 1 exhibited an antiviral entry eff ect on SARS-CoV-2 pseudovirus by blocking spike binding to the ACE2 receptor on HEK-293T-ACE2 h host cells.

        Comparative Study of Bioactive Constituents in Crude and Processed Glycyrrhizae radix and Their Respective Metabolic Profiles in Gastrointestinal Tract In Vitro by HPLC-DAD and HPLC-ESI/MS Analyses

        Two HPLC methods with diode array detection (HPLC-DAD) and electrospray ionization-mass spectrometry (HPLC-ESI/MS), respectively, were developed to investigate the differences of chemical constituents and their metabolism in gastrointestinal tract in vitro between two decoctions of crude and processed Glycyrrhizae radix. Total of eleven constituents (liquiritin apioside, liquiritin, licuraside, isoliquiritin, ononin, glycyrrhizin, liquiritigenin-7,4'-diglucoside, licorice saponin A3, 22β-acetoxylglycyrrhizic acid, licorice saponin G2, and yunganoside E2) were identified in the two decoctions, whereas lower contents of these constituents were usually found in the decoction of processed G. radix. Furthermore, these constituents were metabolized into their respective aglycons in human intestinal bacteria juice, and the metabolism ratios were all higher in processed G. radix decoction. No change was found in artificial gastric or intestinal juice. This study revealed that the processing can alter the contents of main constituents in crude G. radix and their metabolism in gastrointestinal tract, in which intestinal bacteria play an important role in the metabolism of licorice constituents.

        Natural History of Untreated Coronary Total Occlusions Revealed with Follow-Up Semi-Automated Quantitative Coronary CT Angiography: The Morphological Characteristics of Initial CT Predict Occlusion Shortening

        Objective: To investigate the morphological changes of coronary chronic total occlusion (CTO) as determined by coronary computed tomography angiography (CCTA) follow-up using semi-automated quantitative analysis. Materials and Methods: Thirty patients with 31 CTO lesions confirmed by invasive coronary angiography and baseline/follow-up CCTA were retrospectively included. CTOs were quantitatively analyzed by a semi-automated coronary plaque analysis software (Coronary Plaque Analysis, version 2.0, Siemens) after manually determining the lesion border. Recanalized lumen was defined as the linear-like enhanced opacity traversing the non-opacified occluded segment. Other parameters, such as total occlusion length, total occlusion volume, volume with low attenuation component (< 30 Hounsfield unit [HU]), volume with middle to high attenuation component (30−190 HU) as well as the calcification volume, were also recorded. Results: Recanalized lumen was found within 48.4% (15/31) occlusions on the follow-up CCTA, compared to 45.2% (14/31) occlusions on the baseline CCTA. Eleven of 14 lesions (78.6%) with CT-visible recanalized lumen within CTOs had a shorter occlusion length on follow-up compared to only 3 of 17 lesions (17.6%) without CT-visible recanalized lumen (odds ratio, 17.1, p < 0.001). The percentage of low attenuation component of occlusions was smaller on follow-up CCTA compared to baseline value (18.1 ± 20.1% vs. 22.6 ± 19.6%, p = 0.033). Conclusion: Coronary computed tomography angiography enables non-invasive characterization of natural progression of untreated CTO lesions. Recanalized lumen within CTOs observed at baseline CCTA was associated with shortening of occlusion length on follow-up. Compared to their earlier stage, occlusions of later stage were presented with higher density of non-calcified components.

      • Expression Characteristics of Proteins of the Insulin-like Growth Factor Axis in Non-small Cell Lung Cancer Patients with Preexisting Type 2 Diabetes Mellitus

        Background: Preexisting type 2 diabetes mellitus (T2DM) affects the prognosis and mortality of patients with some cancers. Insulin like growth factor (IGF) and insulin receptor (IR) signaling axes play important roles in both cancer and diabetes development. We aimed to explore the expression characteristics of proteins in IGF/IR axis in non-small cell lung cancer (NSCLC) cases with preexisting T2DM. Methods: Fifty-five NSCLC patients with preexisting T2DM were retrospectively included and matched by 55 NSCLC without diabetes at a 1:1 ratio. The expression of proteins in IGF/IR axis was detected by immunohistochemical staining. Clinicopathological data were collected to analyze their relationship with the protein expression. Results: Both IGF 1 receptor (IGF-1R) and insulin receptor substrate 2 (IRS-2) showed higher expression in the NSCLC with T2DM group, compared with those without T2DM. The high expression of IGF-1R and IRS-2 were found to be negatively associated with lymph node metastases and T staging in the T2DM group, respectively, and IRS-2 expression was also found more in the subgroup whose T2DM duration was more than 4 years. No difference was detected in the expression of IRS-1, IGF-1, IGF-2, IGFBP3, IR and mTOR between groups with or without T2DM. Conclusion: Our study found higher expression of IGF-1R and IRS-2 proteins in NSCLC patients with preexisting T2DM, and that there was an association with early stage NSCLC, which suggested that IGF signaling may play an important early event in development of NSCLC associated with diabetes.

        Characteristic profiles of biofilm, enterotoxins and virulence of Staphylococcus aureus isolates from dairy cows in Xinjiang Province, China

        As an important zoonotic pathogen, Staphylococcus aureus (S. aureus) infection has led to serious mastitis and endometritis in infected dairy cows. HereIn this study, a total of 164 strains of S. aureus were isolated from dairy cows in Xinjiang Province, China, and subjected to assays to determine of drug susceptibility and biofilm (BF) formation ability. The eEnterotoxin-related genes were detected, and then the transcription levels of genes related to BF formation were also determined by using reverse transcription-quantitative polymerase chain reactionPCR. Moreover, the pathogenicity of these isolates with different BF formation abilities was determined via by measuring their hemolysis activity, half lethal dose (LD50) and organ bacterial load, respectively. The results showed that 83.4 % of S. aureus isolates could form BF. Among them, 40.8 % of the strains had weak BF formation ability, and most strains with a strong BF formation ability were ica gene carriers. The S. aureus isolates displayed multidrug resistance and their drug resistance was positively correlated with their BF formation ability. Moreover, 96.3 % of the S. aureus isolates carried enterotoxin genes. Among them, the detection rates of the novel enterotoxin genes were higher than thoseat of conventional enterotoxin genes. Furthermore, the isolates with a strong BF formation ability had higher LD50 but lower hemolysis ability and organ bacterial load than thoseat of the isolates with weak or no BF ability. However, those isolates without BF ability caused produced more severe pathological changes than thoseat of the isolates with strong BF formation ability. These findings suggested that higher frequencies of BF ability and presence of novel enterotoxin genes were are important characteristics of S. aureus isolates from dairy cows in Xinjiang Province, China, and such isolateswhich may posed potential threats to food safety.

        Heavy mineral analysis to identify sediment provenance in the Dan River drainage, China

        Identifying the provenance of sediments carried by modern rivers can provide information on earth surface processes and help us to interpret geological history more clearly. The types and distribution patterns of surface sediments deposited in river systems are determined mainly by the fluvial flux and weathering of the proximal source rocks. In this paper, analysis of the heavy mineral content of 99 surface sediment samples was used to identify the source of the sediments carried by the Dan River in China. The objectives of this study are to: (1) determine the characteristics and distribution of the sediments, and (2) identify and classify the various depositional areas according to their differing source areas. The Dan River is divided into the lower, middle, and upper reaches to investigate the spatial distribution of heavy minerals. The main minerals detected were amphiboles, pyroxene, epidote, garnet, tourmaline, zircon, apatite, rutile, sphene, anatase, pyrite, and ilmenite. Generally, ilmenite (0.7% to 69.1%), amphibole (0% to 93.6%), epidote (0% to 53.3%), garnet (0% to 36.7%) and pyroxene (0% to 42.0%) were the most frequent detrital heavy minerals in the studied river sections. The Dan River and its tributaries are characterized with respect to their heavy mineral assemblages and heavy mineral indexes. Six depositional areas are distinguished and classified as Depositional area A (the Laoguan, Qi, and Xian rivers), Depositional area B (the Qingyou and Wuguan rivers), Depositional area C (the Laojun River), Depositional area D (the upper reaches of the Dan River), Depositional area E (the Yinhua River), and Depositional area F (the Tao River). The provenance study indicated that the sediments were derived mainly from the proximal exposed rocks. Sediments in the middle reaches of the Dan River were derived from Depositional areas B, C, and E, whereas sediments in the lower reaches were derived mainly from the local rocks along the river. Similar heavy mineral assemblages were obtained from Danjiangkou Reservoir and Depositional area A, especially for the Laoguan River, indicating the significant influence of Depositional area A on the reservoir.

        Semi-active control on long-span reticulated steel structures using MR dampers under multi-dimensional earthquake excitations

        This paper focuses on the vibration control of long-span reticulated steel structures under multi-dimensional earthquake excitation. The control system and strategy are constructed based on Magneto-Rheological (MR) dampers. The LQR and Hrovat controlling algorithm is adopted to determine optimal MR damping force, while the modified Bingham model (MBM) and inverse neural network (INN) is proposed to solve the real-time controlling current. Three typical long-span reticulated structural systems are detailedly analyzed, including the double-layer cylindrical reticulated shell, single-layer spherical reticulated shell, and cable suspended arch-truss structure. Results show that the proposed control strategy can reduce the displacement and acceleration effectively for three typical structural systems. The displacement control effect under the earthquake excitation with different PGA is similar, while for the cable suspended arch-truss, the acceleration control effect increase distinctly with the earthquake excitation intensity. Moreover, for the cable suspended arch-truss, the strand stress variation can also be effectively reduced by the MR dampers, which is very important for this kind of structure to ensure that the cable would not be destroyed or relaxed.

        LncRNA TMPO-AS1 promotes esophageal squamous cell carcinoma progression by forming biomolecular condensates with FUS and p300 to regulate TMPO transcription

        Esophageal squamous cell carcinoma (ESCC) is one of the most life- and health-threatening malignant diseases worldwide, especially in China. Long noncoding RNAs (lncRNAs) have emerged as important regulators of tumorigenesis and tumor progression. However, the roles and mechanisms of lncRNAs in ESCC require further exploration. Here, in combination with a small interfering RNA (siRNA) library targeting specific lncRNAs, we performed MTS and Transwell assays to screen functional lncRNAs that were overexpressed in ESCC. TMPO-AS1 expression was significantly upregulated in ESCC tumor samples, with higher TMPOAS1 expression positively correlated with shorter overall survival times. In vitro and in vivo functional experiments revealed that TMPO-AS1 promotes the proliferation and metastasis of ESCC cells. Mechanistically, TMPO-AS1 bound to fused in sarcoma (FUS) and recruited p300 to the TMPO promoter, forming biomolecular condensates in situ to activate TMPO transcription in cis by increasing the acetylation of histone H3 lysine 27 (H3K27ac). Targeting TMPO-AS1 led to impaired ESCC tumor growth in a patient-derived xenograft (PDX) model. We found that TMPO-AS1 is required for cell proliferation and metastasis in ESCC by promoting the expression of TMPO, and both TMPO-AS1 and TMPO might be potential biomarkers and therapeutic targets in ESCC.
