        Bioassay-guided Isolation of a Novel Protein with Antitumor Activity from Trachyrhamphus serratus (Syngnathidae)

        A novel protein with antitumor activity, Hailongin, was purified from the aqueous extract of the whole body of Trachyrhamphus serratus, which is commonly used in traditional Chinese medicine, by bioassay-guided fractionation. Hailongin exhibited strong inhibition of proliferation of the tested human cell lines, such as A549, HeLa, LoVo and CCRF-CEM. The IC_50 values of Hailongin ranged from 5.4 to 25.7 μg/mL. An in vivo study showed that the growth of implanted S-180 solid tumors in mice was significantly inhibited by Hailongin treatment, while the immunological function of the tumor-bearing mice was enhanced. The molecular weight and the isoelectric point of Hailongin were 57.074 kDa (by MALDI-TOF-MS) and 6.2 (by isoelectric focusing-polyacrylamide gel electrophoresis), respectively. Seventeen amino acids were identified in Hailongin. The acidic amino acids accounted for the majority of Hailongin’s amino acid composition. The N-terminal amino acid sequence of Hailongin was determined to be IVPYSHNAGNKGLTQMR and showed no significant homology with known proteins.

        Effects of Ohmic Area Etching on Buffer Breakdown Voltage of AlGaN/GaN HEMT

        This study is on how ohmic area etching affects the buffer breakdown voltage of AlGaN/GaN HEMT. The surface morphology of the ohmic metal can be improved by whole etching on the ohmic area. The buffer breakdown voltages of the samples with whole etching on the ohmic area were improved by the suppression of the metal spikes formed under the ohmic contact regions during high-temperature annealing. The samples with selective etching on the ohmic area were investigated for comparison. In addition, the buffer leakage currents were measured on the different radii of the wafer, and the uniformity of the buffer leakage currents on the wafer were investigated by PL mapping measurement.

        Leaky-Wave Antennas Based on Noncutoff Substrate Integrated Waveguide Supporting Beam Scanning From Backward to Forward

        <P>In this paper, we propose an approach to realize substrate integrated waveguide (SIW)-based leaky-wave antennas (LWAs) supporting continuous beam scanning from backward to forward above the cutoff frequency. First, through phase delay analysis, it was found that SIWs with straight transverse slots support backward and forward radiation of the -1-order mode with an open-stopband (OSB) in between. Subsequently, by introducing additional longitudinal slots as parallel components, the OSB can be suppressed, leading to continuous beam scanning at least from -40 degrees through broadside to 35 degrees. The proposed method only requires a planar structure and obtains less dispersive beam scanning compared with a composite right/left-handed (CRLH) LWA. Both simulations and measurements verify the intended beam scanning operation while verifying the underlying theory.</P>

        Systematic polymorphism analysis of the CYP2C9 gene in Chinese Han and Tibetan populations

        Study about polymorphism of the CYP2C9 was not reported in the Chinese Tibetan population and there was no comparison of genetic polymorphism pattern of CYP2C9 between Chinese Han and Tibetan populations. Here we screened the genetic polymorphisms of functional regions of the CYP2C9in 100 unrelated healthy Chinese Han and Tibetan volunteers,respectively, using direct sequencing. A total of 20 variants were detected and there were different distribution of allelic and genotype frequencies, linkage disequilibrium patterns,haplotype structures and htSNPs between the two populations. CYP2C9*3 is a major functional variant of CYP2C9 in the two populations and *11 allele was only detected in Tibetan population. The determined genetic information of CYP2C9in Chinese Han and Tibetan populations might serve as a baseline for larger studies on determining metabolic phenotypes of CYP2C9 substrate drugs and also provide important data for the advance of personalized medicine in Chinese Han and Tibetan populations.

        Fire-Induced Failure of Long-Term Service and Fresh Concrete Beams Subjected to Fire

        In engineering, reinforced concrete beams are subjected to natural erosion and the working stress state. However, previous studies regarding structural fire have focused on new concrete beams, and most results are obtained from no-stress-state tests. Hence, research that considers the natural erosion and working stress states of beam members in fire is lacking. In this study, fire experiments were performed to investigate the differences between long-term service concrete beams and new elements under loads. A technique using ARAMIS three-dimensional (3D) optical strain measurement was adopted; it provides increased sample throughput and 3D measurements that indicate the change in strain more intuitively. Results show that the moisture content of concrete determines the spalling degree of concrete under the influence of working stress. With the increase of concrete moisture content, the range of temperature of heated and backfire surface increases and the spalling phenomenon is more obvious, especially when the moisture content exceeds 2.5%. In addition, the long-term service specimens are affected by fire more easily, thereby accelerating the destruction process owing to the degree of carbonation.

        G Protein–Coupled Receptor 30 Mediates the Anticancer Effects Induced by Eicosapentaenoic Acid in Ovarian Cancer Cells

        Purpose: While numerous epidemiological studies have indicated that omega-3 polyunsaturated fatty acids have anticancer properties in various cancers, the effects and mechanisms of eicosapentaenoic acid (EPA) in ovarian cancer cell growth are poorly understood. Materials and Methods: ES2 ovarian clear cell carcinoma cells and SKOV3 adenocarcinoma cells were treated with palmitic acid or EPA, followed by flow cytometry and cell counting to measure apoptosis and proliferation, respectively. A modified protein lipid overlay assay was used to further verify whether EPA was a ligand of G protein–coupled receptor 30 (GPR30) in ES2 cells. The levels of apoptosis-related genes, phosphorylated AKT, and phosphorylated ERK1/2 were detected to explore the underlying mechanism. Finally, inhibitory effect of EPA on tumor growth via GPR30 was determined in vitro and in vivo. Results: EPA suppressed ES2 ovarian clear cell carcinoma cells growth via GPR30, a novel EPA receptor, by inducing apoptosis. As a ligand of GPR30, EPA activated the GPR30-cAMP– protein kinase A signaling pathway. When GPR30 was suppressed by siRNA or its inhibitor G15, the antiproliferative action of EPA was impaired. Furthermore, EPA inhibited tumor growth by blocking the activation of AKT and ERK. In the mouse xenograft model, EPA decreased tumor volume and weight through GPR30 by blocking tumor cell proliferation. Conclusion: These results confirm that EPA is a tumor suppressor in human ovarian clear cell carcinoma cells and functions through a novel fatty acid receptor, GPR30, indicating a mechanistic linkage between omega-3 fatty acids and cancers.

        Selection and evaluation of reference genes for quantitative gene expression analysis in broomcorn millet (Panicum miliaceum L.)

        The appropriate reference genes are crucial for normalization of the target gene expression in qRT-PCR analysis. Broomcorn millet (Panicum miliaceum L.) is one of the most important crops in drought areas worldwide, while the systematical investigation and evaluation of reference genes has not been investigated in this species up to now. Here, 9 commonly used reference genes were selected to detect their expressional stability in different tissues and under different stresses in broomcorn millet. ΔC t , BestKeeper, NormFinder and GeNorm approaches were used to evaluate the potentiality of these candidate genes as the reference gene in broomcorn millet. Taken together, results found that 18S and GAPDH were the suitable reference genes for gene expression normalization in different tissues and under stress treatment in broomcorn millet. This was the first study to investigate the reference genes for qRT-PCR analysis in broomcorn millet, which will facilitate the gene expression studies and also accelerate revealing the molecular mechanism of well-adapted extreme climatic conditions.

        Corticotropin-releasing Factor Changes the Phenotype and Function of Dendritic Cells in Mouse Mesenteric Lymph Nodes

        Background/Aims Dendritic cells (DCs) are a significant contributor to the pathology of numerous chronic inflammatory autoimmune disorders; however, the effects of Corticotropin-releasing factor (CRF) on intestinal DCs are poorly understood. In this study, we investigated the role of CRF in alterations of intestinal dendritic cell phenotype and function. Methods Mouse mesenteric lymph node dendritic cells (MLNDCs) were obtained using magnetic bead sorting. Surface expression of CRF receptor type 1 (CRF-R1) and CRF-R2 was determined by double-labeling immunofluorescence and quantitative polymerase chain reaction (qPCR) and MLNDCs were subsequently exposed to CRF in the presence or absence of CRF-R1 and CRF-R2 antagonists. Expression of surface molecules (MHC-I and MHC-II) and co-stimulatory molecules (CD80 and CD86) was determined by flow cytometric and western blot analyses, and the T cell stimulatory capacity of MLNDCs was evaluated by mixed lymphocyte reaction. Results Immunofluorescent staining and quatitative polymerase chain reaction indicated that both the CRF receptors (CRF-R1 and CRF-2) are expressed on the surface of MLNDCs. Exposure to CRF increased the expression of MHC-II on MLNDCs as well as their capacity to stimulate T cell proliferation. MLNDCs treated with CRF-R1 antagonist exhibited a phenotype characterized by a less activated state and reduced surface expression of MHC-II, and consequently showed reduced capacity to stimulate T cells. In contrast, treatment of MLNDCs with CRF-R2 antagonist yielded an opposite result. Conclusions CRF can alter the phenotype and function of intestinal DCs through direct action on CRF-R1 and CRF-R2, and activation of the CRF-R1 and CRF-R2 pathways yields opposing outcomes.(J Neurogastroenterol Motil 2015;21:571-580)

        Synthesis of Carbon-Coated LiFePO4 Cathode Material by One-Step Microwave-Assisted Pyrolysis of Ionic Liquid Process

        A one-step microwave-assisted pyrolysis of ionic liquid process is developed to synthesize carbon-coated LiFePO4 by using the ionic liquid [BMIm]N(CN)2 as a carbon source. A thin layer of uniform and nanoscaled carbon film is formed on the LiFePO4 particles surface with a strong interface. The electrochemical properties of LiFePO4 are improved due to the carbon coating which increases the electronic conductivity of LiFePO4. The results indicate that the microwaveassisted pyrolysis of ionic liquid process is a rapid and efficient method to prepare carbon-coated LiFePO4 material and other carbon-coated electrode materials.
