        Acteoside and Acyl-Migrated Acteoside, Compounds in Chinese Kudingcha Tea, Inhibit α-Amylase In Vitro

        Acteoside, the predominant polyphenol of small-leaved kudingcha, the Chinese tea, has various biological activities. In this study, we examined the acyl migration of acteoside to isoacteoside with high-temperature treatment of acteoside. The inhibitory effects of acyl-migrated acteoside and acteoside on α-amylase were investigated, as were their binding interaction with α-amylase. The binding of acteoside and isoacteoside to α-amylase was investigated by using the fluorescence spectra assay, circular dichroism, and protein–ligand docking studies. Acteoside was more effective than preheated acteoside and isoacteoside in inhibiting α-amylase activity. Acteoside and isoacteoside binding to α-amylase may induce conformational changes to α-amylase, and the binding site of acteoside and isoacteoside being near the active site pocket of α-amylase may explain the decreased activity of α-amylase. The different affinities and binding sites of acteoside and isoacteoside for α-amylase resulted in different inhibition rates, which may be due to structural differences between acteoside and isoacteoside.

        Precisely tailoring pore structure in sunflower plate-derived N, O co-doped carbons for high-performance supercapacitors

        The controllable adjustment of pore structure is of great significance for the high capacitive performanceof porous carbon materials. In this research, a versatile K2CO3 activation approach is proposed to adjustand control the porosity of sunflower plate-derived hierarchical porous carbon materials (HPC-x) by controllingthe K2CO3/carbons mass ratio. Notably, the variety of pore types, including micropores, mesoporesand macropores, can be detected at different K2CO3/carbons mass ratios (0 4). Impressively,the specific surface area of optimal HPC-2 material is 2526 m2/g. Compare with the other samples(HPC-1, HPC-3 or HPC-4), the HPC-2 possess superior capacitance activity (369.4 F/g at 0.5 A/g). Ontop of that, the energy density of HPC-2 assembled supercapacitors in 1 M Na2SO4 and 6 M KOH solutionscan even reach 32.6 and 7.3 Wh/kg, respectively.

        Bagasse as functional fillers to improve and control biodegradability of soy oil-based rigid polyurethane foams

        Using agricultural wastes (bagasse) and polyol derived from soy oil as raw materials to develop biodegradable rigid polyurethane foams (RPUFs) is beneficial to reduce the dependence on petroleum resources and promote the sustainable socioeconomic and environmental development. This study focuses on bagasse as a functional filler to improve and control the mechanical properties and biodegradability of the RPUFs. With the increasing levels of bagasse content, the density and mechanical properties of RPUFs increase. The biodegradation of RPUFs has a greater enhancement compared to the foam without bagasse, and all bio-foams have excellent thermal insulation properties. All changes in foam performance are due to the fact that the active hydroxyl groups on bagasse react with excess isocyanate to form urethane, which affects the structure of RPUFs. In this study, scientific design and molecular regulation theory were applied to improve the utilization value of bagasse and develop high-performance biodegradable RPUFs for thermal insulation materials

        Solution-based synthesis of PEDOT:PSS films with electrical conductivity over 6300 S/cm

        Poly(3,4-ethylenedioxythiophene) mixed with poly (styrenesulfonate) (PEDOT:PSS) is considered as oneof the most valuable conductive polymers due to its high conductivity, transparency and mechanicalflexibility. Many experiments have proved that H2SO4 post-treatment is an effective way to enhance theelectrical conductivity of PEDOT:PSSfilms. Here we reported a method for the fabrication of PEDOT:PSSfilms with conductivity as high as 6323.9 364.5 S/cm, based on a two-step H2SO4 post-treatment. Thisvery high conductivity is, as far as we know, the highest reported value for PEDOT:PSSfilms by solutionpreparation. The removal of PSS and redox reaction are the keys to enhance the conductivity of PEDOT:PSSfilms.

        Influence of Ligustrum lucidum and Schisandra chinensis Fruits on Antioxidative Metabolism and Immunological Parameters of Layer Chicks

        The experiment was conducted to evaluate the effects of Ligustrum lucidum (LL) and Schisandra chinensis (SC) on the growth, antioxidative metabolism and immunity of laying strain male chicks. The results showed that diets supplemented with 1% of either LL or SC had no effects on the growth performance of chicks compared with the control. Furthermore, both LL and SC significantly reduced malondialdehyde (MDA) concentration of serum and heart of chicks (p<0.05). In addition, superoxide dismutase (SOD) activity of serum of the birds was significantly elevated by supplementation with SC (p<0.05). Glutathione reductase (GR) activity of heart and serum of the birds was significantly elevated by supplementation with LL or SC (p<0.05). LL supplementation significantly elevated antibody values against Newcastle Disease virus (NDV)(p<0.05) and lymphoblastogenesis (p<0.05) of the birds. The results suggest that diets supplemented with 1% of either LL or SC may improve immune function and antioxidant status of chicks.

        Tissue distribution of marbofloxacin in pigs after a single intramuscular injection

        Tissue distribution of marbofloxacin was studied in pigs after a single intramuscular injection at 2.5 mg/kg body weight. Samples of plasma, muscle, liver, kidney, heart, lung, and muscle at the injection site were randomly collected from five pigs at 2, 6, 10, 24, 48, 72, and 96 h after administration. Marbofloxacin concentrations were determined by using high-performance liquid chromatography with ultraviolet detection and were subjected to non-compartmental analysis to obtain kinetic parameters. The elimination half-life (t1/2lz) of marbofloxacin at the injection site was 22.12 h, while those in kidney, plasma, liver, lung, heart, and muscle were 16.75, 21.48, 21.84, 24.00, 24.45, and 28.91 h, respectively. Areas under the concentration-time curve from 0 h to ∞ (AUC0–∞s) were calculated to be 31.17 hㆍmgㆍmL−1 for plasma and 32.97, 33.92, 34.78, 37.58, 42.02, and 98.80 hㆍmgㆍg−1 for heart, muscle, lung, liver, kidney, and injection site, respectively. The peak concentration (Cmax) of marbofloxacin was 1.62 µg/mL in plasma and 1.71, 1.74, 1.86, 1.93, 2.45, and 7.64 µg/g in heart, lung, muscle, kidney, liver, and injection site, respectively. The results show that marbofloxacin was fast absorbed, extensively distributed, and slowly eliminated from pigs after a single intramuscular administration.

        Study of upfront surgery versus neoadjuvant chemotherapy followed by interval debulking surgery for patients with stage IIIC and IV ovarian cancer, SGOG SUNNY (SOC-2) trial concept

        Background: Two randomized phase III trials (EORTC55971 and CHORUS) showed similarprogression-free and overall survival in primary or interval debulking surgery in ovariancancer, however both studies had limitations with lower rate of complete resection and lack ofsurgical qualifications for participating centers. There is no consensus on whether neoadjuvantchemotherapy followed by interval debulking surgery (NACT-IDS) could be a preferred approachin the management of advanced epithelial ovarian cancer (EOC) in the clinical practice. Methods: The Asian SUNNY study is an open-label, multicenter, randomized controlled,phase III trial to compare the effect of primary debulking surgery (PDS) to NACT-IDS instages IIIC and IV EOC, fallopian tube cancer (FTC) or primary peritoneal carcinoma (PPC). The hypothesis is that PDS enhances the survivorship when compared with NACT-IDS inadvanced ovarian cancer. The primary objective is to clarify the role of PDS and NACT-IDS inthe treatment of advanced ovarian cancer. Surgical quality assures include at least 50% of nogross residual (NGR) in PDS group in all centers and participating centers should be nationalcancer centers or designed ovarian cancer section or those with the experience participatingsurgical trials of ovarian cancer. Any participating center should be monitored evaluatingthe proportions of NGR by a training set. The aim of the surgery in both arms is maximalcytoreduction. Tumor burden of the disease is evaluated by diagnostic laparoscopy orpositron emission tomography/computed tomography scan. Patients assigned to PDS groupwill undergo upfront maximal cytoreductive surgery within 3 weeks after biopsy, followed by6 cycles of standard adjuvant chemotherapy. Patients assigned to NACT group will undergo 3cycles of NACT-IDS, and subsequently 3 cycles of adjuvant chemotherapy. The maximal timeinterval between IDS and the initiation of adjuvant chemotherapy is 8 weeks. Major inclusioncriteria are pathologic confirmed stage IIIC and IV EOC, FTC or PPC; ECOG performancestatus of 0 to 2; ASA score of 1 to 2. Major exclusion criteria are non-epithelial tumors as wellas borderline tumors; low-grade carcinoma; mucinous ovarian cancer. The sample size is 456subjects. Primary endpoint is overall survival.         SET8 suppression mediates high glucose-induced vascular endothelial inflammation via the upregulation of PTEN

        Hyperglycemia-mediated endothelial inflammation participates in the pathogenesis of cardiovascular complications in subjects with diabetes. Previous studies reported that phosphatase and tensin homolog deleted on chromosome ten (PTEN) and SET8 participate in high glucose-mediated endothelial inflammation. In this study, we hypothesize that SET8 regulates PTEN expression, thus contributing to high glucose-mediated vascular endothelial inflammation. Our data indicated that plasma soluble intercellular adhesion molecule-1 (sICAM-1) and endothelial selectin (e-selectin) were increased in patients with diabetes and diabetic rats. PTEN expression was augmented in the peripheral blood mononuclear cells of patients with diabetes and in the aortic tissues of diabetic rats. Our in vitro study indicated that high glucose increased monocyte/endothelial adhesion, endothelial adhesion molecule expression and p65 phosphorylation in human umbilical vein endothelial cells (HUVECs). Moreover, high glucose led to endothelial inflammation via upregulation of PTEN. Furthermore, high glucose inhibited SET8 expression and histone H4 lysine 20 methylation (H4K20me1), a downstream target of SET8. SET8 overexpression reversed the effects of high-glucose treatment. shSET8-mediated endothelial inflammation was counteracted by siPTEN. Furthermore, SET8 was found to interact with FOXO1. siFOXO1 attenuated high glucose-mediated endothelial inflammation. FOXO1 overexpression-mediated endothelial inflammation was counteracted by siPTEN. H4K20me1 and FOXO1 were enriched in the PTEN promoter region. shSET8 increased PTEN promoter activity and augmented the positive effect of FOXO1 overexpression on PTEN promoter activity. Our in vivo study indicated that SET8 was downregulated and FOXO1 was upregulated in the peripheral blood mononuclear cells of patients with diabetes and the aortic tissues of diabetic rats. In conclusion, SET8 interacted with FOXO1 to modulate PTEN expression in vascular endothelial cells, thus contributing to hyperglycemia-mediated endothelial inflammation.

        Effects of feeding different levels of dietary corn silage on growth performance, rumen fermentation and bacterial community of post-weaning dairy calves

        Objective: The objective of this study was to evaluate the growth performance, rumen fermentation parameters and bacterial community of post-weaning dairy calves in response to five diets varying in corn silage (CS) inclusion.Methods: A total of forty Holstein weaned bull calves (80±3 days of age;128.2±5.03 kg at study initiation) were randomized into five groups (8 calves/group) with each receiving one of five dietary treatments offered as total mixed ration in a 123-d feeding study. Dietary treatments were control diet (CON; 0% CS dry matter [DM]); Treatment 1 (T1; 27.2% CS DM); Treatment 2 (T2; 46.5% CS DM); Treatment 3 (T3; 54.8% CS DM); and Treatment 4 (T4; 67.2% CS DM) with all diets balanced for similar protein and energy concentration.Results: Results showed that calves offered CS had greater average daily gain, body length and chest depth growth, meanwhile altered rumen fermentation indicated by decreased rumen acetate concentrations. Principal coordinate analysis showed the rumen bacterial community structure was affected by varying CS inclusion diets. <i>Bacteroidetes</i> and <i>Firmicutes</i> were the predominant bacterial phyla in the calf rumens across all treatments. At the genus level, the abundance of <i>Bacteroidales_RF16_group</i> was increased, whereas <i>Unclassified_ Lachnospiraceae</i> was decreased for calves fed CS. Furthermore, Spearman’s correlation test between the rumen bacteria and rumen fermentation parameters indicated that <i>Bacteroidales_RF16_group</i> and <i>Unclassified Lachnospiraceae</i> were positively correlated with propionate and acetate, respectively.Conclusion: The results of the current study suggested that diet CS inclusion was beneficial for post-weaning dairy calf growth, with 27.2% to 46.5% CS of diet DM recommended to achieve improved growth performance. <i>Bacteroidales_RF16_group</i> and <i>Unclassified Lachnospiraceae</i> play an important role in the rumen fermentation pattern for post-weaning calves fed CS.
