        Application Research on Mechanical Strength and Durability of Porous Basalt Concrete

        Porous basalt aggregate is commonly used in roadbed engineering, but its application in concrete has rarely been studied. This paper studies the application of porous basalt in concrete. Porous basalt aggregate is assessed for its effects on mechanical strength and durability of prepared C50 concrete; because it has a hole structure, porous basalt aggregate is known for its porosity, and porous basalt aggregates can be made full of water through changing the content of saturated basalt; after full-water condition is achieved in porous basalt aggregate mixture of C50 concrete, we discuss its mechanical properties and durability. The effects of C50 concrete prepared with basalt aggregate on the compressive strength, water absorption, and electric flux of concrete specimens of different ages were studied through experiments, and the effects of different replacement rates of saturated porous basalt aggregate on the properties of concrete were also studied. The results show that porous basalt aggregate can be prepared as C50 concrete. For early saturated porous basalt aggregate concrete, its compressive strength decreases with the increase of the replacement rate of saturated aggregate; this occurs up to concrete curing at 28 d, when the replacement rate of saturated basalt aggregate is greater than or equal to 40 %. The compressive strength of concrete increases with the increase of the replacement rate of saturated aggregate. The 28 d electric flux decreases with the increase of the replacement rate of saturated aggregate, indicating that saturated porous basalt aggregate can improve the chloride ion permeability resistance of concrete in later stages.

        Polymorphism, Expression of Natural Resistance-associated Macrophage Protein 1 Encoding Gene (NRAMP1) and Its Association with Immune Traits in Pigs

        Natural resistance-associated macrophage protein 1 encoding gene (NRAMP1) plays an important role in immune response against intracellular pathogens. To evaluate the effects of NRAMP1 gene on immune capacity in pigs, tissue expression of NRAMP1 mRNA was observed by real time quantitative polymerase chain reaction (PCR), and the results revealed NRAMP1 expressed widely in nine tissues. One single nucleotide polymorphism (SNP) (ENSSSCG00000025058: g.130 C>T) in exon1 and one SNP (ENSSSCG00000025058: g.657 A>G) in intron1 region of porcine NRAMP1 gene were demonstrated by DNA sequencing and PCR-RFLP analysis. A further analysis of SNP genotypes associated with immune traits including contain of white blood cell (WBC), granulocyte, lymphocyte, monocyte (MO), rate of cytotoxin in monocyte (MC) and $CD4^-CD8^+$ T lymphocyte subpopulations in blood was carried out in four pig populations including Large White and three Chinese indigenous breeds (Wannan Black, Huai pig and Wei pig). The results showed that the SNP (ENSSSCG00000025058: g.130 C>T) was significantly associated with level of WBC % (p = 0.031), MO% (p = 0.024), MC% (p = 0.013) and $CD4^-CD8^+$ T lymphocyte (p = 0.023). The other SNP (ENSSSCG00000025058: g.657 A>G) was significantly associated with the level of MO% (p = 0.012), MC% (p = 0.019) and $CD4^-CD8^+$ T lymphocyte (p = 0.037). These results indicate that the NRAMP1 gene can be regarded as a molecular marker for genetic selection of disease susceptibility in pig breeding.

        Relationship between porcine miR-20a and its putative target low-density lipoprotein receptor based on dual luciferase reporter gene assays

        Objective: Mutations in low-density lipoprotein receptor (LDLR), which encodes a critical protein for cholesterol homeostasis and lipid metabolism in mammals, are involved in cardiometabolic diseases, such as familial hypercholesterolemia in pigs. Whereas microRNAs (miRNAs) can control LDLR regulation, their involvement in circulating cholesterol and lipid levels with respect to cardiometabolic diseases in pigs is unclear. We aimed to identify and analyze LDLR as a potential target gene of SSC-miR-20a. Methods: Bioinformatic analysis predicted that porcine LDLR is a target of SSC-miR-20a. Wild-type and mutant LDLR 3′-untranslated region (UTR) fragments were generated by polymerase chain reaction (PCR) and cloned into the pGL3-Control vector to construct pGL3 Control LDLR wild-3′-UTR and pGL3 Control LDLR mutant-3′-UTR recombinant plasmids, respectively. An miR-20a expression plasmid was constructed by inserting the porcine pre-miR-20a-coding sequence between the HindIII and BamHI sites in pMR-mCherry, and constructs were confirmed by sequencing. HEK293T cells were co-transfected with the miR-20a expression or pMR-mCherry control plasmids and constructs harboring the corresponding 3′-UTR, and relative luciferase activity was determined. The relative expression levels of miR-20a and LDLR mRNA and their correlation in terms of expression levels in porcine liver tissue were analyzed using reverse-transcription quantitative PCR. Results: Gel electrophoresis and sequencing showed that target gene fragments were successfully cloned, and the three recombinant vectors were successfully constructed. Compared to pMR-mCherry, the miR-20a expression vector significantly inhibited wild-type LDLR-3′-UTR-driven (p<0.01), but not mutant LDLR-3′-UTR-driven (p>0.05), luciferase reporter activity. Further, miR-20a and LDLR were expressed at relatively high levels in porcine liver tissues. Pearson correlation analysis revealed that porcine liver miR-20a and LDLR levels were significantly negatively correlated (r = –0.656, p<0.05). Conclusion: LDLR is a potential target of miR-20a, which might directly bind the LDLR 3′-UTR to post-transcriptionally inhibit expression. These results have implications in understanding the pathogenesis and progression of porcine cardiovascular diseases.

        Local Field Enhancement Due to Multiple Scattering in Random Sb-SiN Thin Films

        Random Sb-SiN films consisting of random small Sb particles embedded in a SiN thin film were deposited by radio-frequency magnetron sputtering. Specimens comprising in order a random Sb- SiN film and an optical-phase-change recording layer were exposed to a focused laser beam. It was shown that, with a random Sb-SiN layer deposited above the recording layer, the ablation of the recording layer occurred much faster and under much lower power than that of a single recording layer. Scanning Near-field Optical Microscope (SNOM) images of the transmitted light after passing through the random Sb-SiN film were produced as well as a Finite Difference Time Domain (FDTD) calculation of the field distribution. They both showed a local optical near-field enhancement due to multiple scattering effects of the Sb nanoparticles. This is promising for application in superresolution optical storage to get a higher Carrier to Noise Ratio (CNR).

        Study of upfront surgery versus neoadjuvant chemotherapy followed by interval debulking surgery for patients with stage IIIC and IV ovarian cancer, SGOG SUNNY (SOC-2) trial concept

        Background: Two randomized phase III trials (EORTC55971 and CHORUS) showed similarprogression-free and overall survival in primary or interval debulking surgery in ovariancancer, however both studies had limitations with lower rate of complete resection and lack ofsurgical qualifications for participating centers. There is no consensus on whether neoadjuvantchemotherapy followed by interval debulking surgery (NACT-IDS) could be a preferred approachin the management of advanced epithelial ovarian cancer (EOC) in the clinical practice. Methods: The Asian SUNNY study is an open-label, multicenter, randomized controlled,phase III trial to compare the effect of primary debulking surgery (PDS) to NACT-IDS instages IIIC and IV EOC, fallopian tube cancer (FTC) or primary peritoneal carcinoma (PPC). The hypothesis is that PDS enhances the survivorship when compared with NACT-IDS inadvanced ovarian cancer. The primary objective is to clarify the role of PDS and NACT-IDS inthe treatment of advanced ovarian cancer. Surgical quality assures include at least 50% of nogross residual (NGR) in PDS group in all centers and participating centers should be nationalcancer centers or designed ovarian cancer section or those with the experience participatingsurgical trials of ovarian cancer. Any participating center should be monitored evaluatingthe proportions of NGR by a training set. The aim of the surgery in both arms is maximalcytoreduction. Tumor burden of the disease is evaluated by diagnostic laparoscopy orpositron emission tomography/computed tomography scan. Patients assigned to PDS groupwill undergo upfront maximal cytoreductive surgery within 3 weeks after biopsy, followed by6 cycles of standard adjuvant chemotherapy. Patients assigned to NACT group will undergo 3cycles of NACT-IDS, and subsequently 3 cycles of adjuvant chemotherapy. The maximal timeinterval between IDS and the initiation of adjuvant chemotherapy is 8 weeks. Major inclusioncriteria are pathologic confirmed stage IIIC and IV EOC, FTC or PPC; ECOG performancestatus of 0 to 2; ASA score of 1 to 2. Major exclusion criteria are non-epithelial tumors as wellas borderline tumors; low-grade carcinoma; mucinous ovarian cancer. The sample size is 456subjects. Primary endpoint is overall survival.         Estimation of Genetic Parameters for Four Reproduction Component Traits in Two Chinese Indigenous Pig Breeds

        The reproduction component traits are important components of sow efficiency. The objective of this study was to evaluate the phenotypic and genetic parameters of four reproduction component traits (age at puberty (AP), preweaning number dead (PND), weaning to service interval (WSI), and intra-individual SD in litter size (IISDLS)) of sows in two Chinese indigenous pig breeds. Available reproductive records including 22,591 piglets born from 2,054 litters by 574 Jiangquhai sows and 464 Meishan sows were used in this investigation. A set of mixed models and restricted maximum likelihood methodology were used for the multiple trait analyses of these traits. The results showed that the estimates of heritabilities (${\pm}$standard error) for AP, PND, WSI and IISDLS were $0.40{\pm}0.05$, $0.06{\pm}0.03$, $0.20{\pm}0.02$ and 0.09{\pm}0.03 in Jiangquhai sows, and $0.35{\pm}0.06$, $0.05{\pm}0.03$, $0.18{\pm}0.03$ and $0.10{\pm}0.04$ in Meishan sows, respectively. There was moderate genetic correlation between AP and WSI, while there were low genetic correlations between the other pairwise traits. The genetic correlations were positive for most of the pairwise traits, except for the one between AP and IISDLS. The results indicated that all traits except for AP were difficult to make genetic improvement by traditional selection methods due to low heritabilities and the favorable improvement of AP might result in unfavorable changes of IISDLS due to the trend of genetic antagonism.

      • Kernel Reference Set and Its Computation Algorithm

        This paper proposes a new concept of graphs: kernel reference set. A kernel reference set is a fraction of vertices in a graph such that if their positions are known, positions of all other vertices can be derived from information of the distance matrix. We present an algorithm to find kernel reference set of graphs. Because the problem of kernel reference set is firstly proposed in this paper, and the theoretical limit of computation complexity is unknown currently. So we implement our algorithm with C to do the simulation. From simulation results, we verified the validity of our algorithm and evaluate its performance. Inspired by anchor nodes in wireless sensor networks, kernel reference set could be used to analyze anchorbased localization schemes of WSNs applications such as coalmine security monitoring, etc.
