        제 2형 콜라겐에 의해 경구관용 유도된 DBA/1 mice에서의 세포면역반응

        Objective: To investigate the dosage of bovine type II collagen (BnCII) for the induction of oral tolerance in CIA animals, and to verify the changes of immune response and TGF-β production of mesenteric lymph node cells in tolerized CIA animals. Methods: Oral tolerance was induced by feeding of variable doses (5㎍, 10㎍, 20㎍ and 40㎍) of BnCII to DBA/1 mice 4 times per week during 2 weeks, and control mice were given ovalbumin (1000㎍), before immunization. We examed clinical assessment; incidence of arthritis, severity of arthritis, arthritic limb by visual analysis. IgG antibodies to BnCII were measured by ELISA, T cell responses to BnCII and PHA were quantified by antigen (CII)-induced 3H-thymidine incorporation into lymphocytes of mesenteric lymph node, draining lymph node, and spleen. TGF-β in supernatants obtained from lymph node culture medium was measured by ELISA. Results: Arthritis limbs were observed in 100% of control at 5 weeks after subcutaneous BnCII injection. The incidences of CIA in all tolerized group were significantly lower than that in control 5 weeks after immunization (control 100% vs. 5㎍ feeding group: 50%, 10㎍ feeding group: 50%, 20㎍ feeding group: 50%, 40㎍ feeding group: 55.5%, P<0.01). In comparison to control, mean articular indices were lower in all tolerized groups (control 5.13: 5㎍ feeding group 3.50, 10㎍ feeding group 2.75, 20㎍ feeding group 2.87, 40㎍ feeding group 2.63, P<0.05). Arthritic limbs were also significantly lower in tolerized groups (control 58.3: 5㎍ feeding group 20.8, 10㎍ feeding group 16.7, 20㎍ feeding group 20.8, 40㎍ feeding group 20.8, P<0.05). The titers of IgG antibody to CII were lower in tolerized group than that in control [tolerized group; median 10 (min. 0, max. 48), control; median 33 (min. 8.6, max. 101), P<0.05]. The proliferative responses to BnCII were significantly suppressed in tolerized (control 8010±2319cpm, tolerized group 4500±2060cpm, P<0.01). High TGF-β production was noted in tolerized group (control; 28pg/ml, BnCII feeding group; 73pg/ml). Conclusion: Oral tolerance in DBA/1 mice was successfully induced from low doses of BnCII (5㎍) and suppressed T and B cell function in conjunction with increased TGF-β production may play an important role for the induction of CII induced oral tolerance in DBA/1 mice.

      • Precise determination of the lithium isotope ratio in geological samples using MC-ICP-MS with cool plasma

        <P>Lithium has two naturally occurring isotopes, <SUP>6</SUP>Li and <SUP>7</SUP>Li, with approximate relative abundances of 7.5% and 92.5%, respectively. Due to large Li isotope variations in nature, lithium isotopes have the potential to reveal important information relevant to nuclear technology, biomedicine, astrophysics, and geochemistry. With the advent of multi-collector inductively coupled mass spectrometry (MC-ICP-MS), studies of Li isotopes have largely focused on the analysis of geological materials, with varying degrees of accuracy. However, this technique has often been affected by either baseline interferences or isobaric interferences on mass 6 and 7 during ionization in Ar plasma, which is mainly due to the Li compound with hydrogen gas, and double-charged nitrogen and carbon ions at higher levels of RF power. In this study, we reduced baseline interferences in Ar plasma using a cool plasma (∼800 W) technique with a X-type cone. Lithium was separated using a cation exchange column (BioRad AG50W-X8, 200–400 mesh) with a mixture of 6 N HNO<SUB>3</SUB> and 80% methanol at <0.2 mL min<SUP>−1</SUP> elution speed. The short-term reproducibility of <I>δ</I><SUP>7</SUP>Li values of the NASS-5 seawater standard was 30.55 ± 0.45‰ (2<I>σ</I>, <I>n</I> = 15). Measured <I>δ</I><SUP>7</SUP>Li values of rock and seawater standards ranged from 2.48 to 30.55‰, in good agreements with reported values.</P> <P>Graphic Abstract</P><P>Average <I>δ</I><SUP>7</SUP>Li values of a pure L-SVEC Li solution and one passed through the column were 0.00 ± 0.23‰ (2<I>σ</I>, <I>n</I> = 26) and ¬0.01 ± 0.20 ‰ (2<I>σ</I> , <I>n</I> = 9), respectively, indicating that our chemical separation procedure for Li induces no mass fractionation. <IMG SRC='http://pubs.rsc.org/services/images/RSCpubs.ePlatform.Service.FreeContent.ImageService.svc/ImageService/image/GA?id=c2ja30293d'> </P>

        Purification and Characterization of Helicobacter pylori ${\gamma}$-Glutamyltranspeptidase

        Gamma-glutamyltranspeptidase (GGT) was purified to electrophoretic homogeneity from the cell extract of H. pylori. The purified enzyme consisted of heavy and light subunits with molecular weights of 38 kDa and 21 kDa, respectively. N-terminal amino acid sequence of heavy and light subunits revealed that H. pylori GGT was processed into 3 parts for a signal peptide of 27 amino acid residues, a heavy subunit of 352 residues, and a light subunit of 188 residues during translation. The reaction rate for hydrolysis of ${\gamma}$-GpNA was 84.4 ${\mu}mol/min$ per milligram of protein, and that for the ${\gamma}$-glutamyl transfer from ${\gamma}$-GpNA to gly-gly was 23.8 ${\mu}mol/min$ per milligram of protein. The apparent Km values of H. pylori GGT for ${\gamma}$-glutamyl compounds were on the order of $10^{-3}$ to $10^{-4}$ M and those for acceptor peptides and amino acids were on the order of $10^{-1}$ to $10^{-2}$ M. The GGT protein kept approximately 80% of the initial enzymatic activity on incubation at $60^{\circ}C$ for 15 min. The optimum temperature and pH for reactions of both hydrolysis and transpeptidation were $40^{\circ}C$ and 9.0, respectively. The transpeptidation and hydrolysis reactions catalyzed by H. pylori GGT were strongly inhibited by L-Gln and moderately inhibited by L-Ala, L-Ser, ${\beta}$-chloro-L-Ala, and L-Glu. These results demonstrated that the biochemical properties of H. pylori GGT are different from those of other bacterial GGTs. Further, H. pylori GGT might degrade glutathione in the gastric mucous layer of humans if the enzyme could be secreted in the bacterial niches.

      • 대퇴골 경부 골절에 대한 임상적 고찰

        27 cases of femoral neck fractures which were treated in department of orthopaedic surgery of CNUH from 1979 Jan. to 1985 Jan, were analysed clinically and radiologically. Very sensitive, noninvasive, simple and easily reproducible bone scanning with 99m Tc was introduced fro early diagnosis of avascular necrosis as a complication of femoral neck fracture. The results were as follows: 1. The most common cause of fracture was slipping and falling down. 2. In classification, transcervical and subcapital fractures were most common in anatomical classification, Pauwels' type I in Pauwels' classification and Garden stage 3 in displacement. 3. Singh index was decreased according to increase of physiological age. 4. Avascular necrosis develops commonly in Pawels' type 3, Garden's stage 4 and subcapital fracture. 5. Satisfactory results were obtained 75% in internal fixation and 85% in head replacement. 6. Bone scan is considered to be significant technique for early detection of vascular impairment to the femoral head after femur neck fracture, and is important in selecting the method of treatment according to results of bone scan.

        In this study, to control secondary particles of silica, SiO_(2) seed growth experiments have been carried out variable concentrations of tetraethylorthosilicate(TEOS), agitation speed, feeding time, concentration of [NH_(3)], [H_(2)O]. The formation of secondary particles was reduced with increasing feeding time(t_(F)), agitation speed. Increasing TEOS concentration promote formation of the secondary particles. Increasing NH_(3) concentration reduce formation of the secondary particles.

      • Prospidin이 Mouse의 간장에 미치는 영향

        Prospidin, a new alkylating agent, has antimitotic and anticancer effect. Incubation with prospidin increases the stability of DNA and suppresses the incorporation of uridine into RNA by 15-20 percent. Prospidin is cytostatic for suspension culture and cells are blocked in G₂ phase of the cell cycle. The authors have demonstrated the effect of prospidin on the liver of mice histologically and histochemically and observed the cellular changes, loss of pyroninophilic granules and accumulation of fat in the hepatic parenchymal cells. Sixty-five healthy mice of ICR strain, average weight being 20g, were divided into two groups; control and prospidin-treated groups. All experimental animals were administered foo and water ad libitum. Animals of the prospidin-treated groups were administered prospidine 500mg per kg of body weight intraperitoneally fter dilution in the water for injection. Animals of the control group were administered only water for injection. Prospidin treated animals were sacrificed at intervals of 6, 12, 24, 36 and 48 hours after administration of prospidin. Histological preparations were made by way of hematoxylin and eosin stain for the cellular changes, methyl green-pyronin stain for pyroninophilic granules and oil red O stain for fat accumulation in the mouse liver. The results obtained are as follows. 1. Cellular swelling associated with vesicular formation were observed in the hepatic parenchymal cells of entire hepatic lobule at 24 and 36 hours after administration of prospidin. 2. A marked reduction of the pyroninophilic granules were observed in the hepatocyte of he whole hepatic lobule at 24, 36 and 48 hours following drug administration. 3. Fat deposits in the hepatic lobule were seen at 36 hours after administration of prospidin. It is suggested that prospidin acts as a cytotoxic agent and causes cellular swelling with vesicular formation, loss of pyroninophilic granules, and fat accumulation in the parenchymal cells of the liver.

      • 급성 A형 간염에 병발한 급성신부전 2 례

        Hepatitis A is a mild, self-limiting disease of the liver, critical complications of which, such as acute kidney injury, are rare. Two cases of patients with acute hepatitis A who had an acute renal failure complication are reported herein. The first case is that of a 30-year-old man who showed up at the author’s hospital complaining of fever and myalgia. His laboratory tests upon his admission showed 9,520 lU/L of aspartate transaminase, 5,600 lU/L of alanine transaminase, 31.9 mg/dL of blood urea nitrogen, and 5.5 mg/dL of creatinine, and he was positive for the Anti-HAV IgM antibody. Upon his admission, supportive treatment was started, which included fluid therapy. The laboratory tests on the next day showed 53 mg/dL of blood urea nitrogen, 9.9 mg/dL of creatinine, and lower urine output per day. Renal replacement therapy was performed only once. A renal biopsy was performed and the result was Acute Tubular Necrosis (ATN). The patient recovered from acute renal failure without sequela. In the second case, a 35-year-old man complained of fever and abdominal pain. His laboratory tests upon his admission showed 8,570 lU/L of aspartate transaminase, 4,510 lU/L of alanine transaminase, 16 mg/dL of blood urea nitrogen, and 1.19 mg/dL of creatinine, and he was positive for the Anti-HAV IgM antibody. He experienced acute renal failure and recovered after two-time renal replacement therapy. The result of the renal biopsy was Acute Tubular Necrosis and C1q nephropathy.

      • 전립선 결절성 증식증의 미세구조의 변화

        전립성 결절성 증식증의 발병기전을 형태학전으로 규명하고자 10례의 환자에서 적출된 전립선을 광학 및 전자현미경적으로 검색하였으며 전립선의 분비선과 간질조직의 증식정도를 알기 위하여 형태계측법을 시행하였다. 아울러 대조군으로 2례의 부검시 적출된 정상인 전립선을 같은 방법으로 검색하였다. 그 결과를 요약하면 다음과 같다. 대조군에서 전립선의 분비선 상피세포는 분비과립, lysosome,mitochondria,ER 및 Golgi장치가 풍부하였으며 간질조직의 평활근세포 및 섬유아세포는 세포소기관의 양이 빈약하였다. 결절성증식증에서는 분비선 상피세포는 분비과립과 lysosome이 약간 있었을뿐 세포소기관의 양이 적었다. 간질조직에서는 평활근세포 및 섬유아세포내의 세포소기관의 양이 증가되었다. 형태계측법에 의한 전립선의 분비선 대 간질조직의 비는 대조군에서 0.28±0.04이었으며 결절성 증식증에서는 0.10±0.03으로 간질조직이 증가되었다. 이상의 성적으로 보아 전립선의 결절성 증식증은 일차적으로 간질조직의 증식에 의해 일어나는 질환으로 인정된다. Normal and prostatic nodular hyperplasia were studied by quantitative light microscopic measurements and electron microscopy. Two cases of normal prostate obtained from the autopsy and ten cases of nodular hyperplasia were examined. Electron microscopically, glandular lining cells of the normal prostate showed abundant secretory granules, lysosomes, endoplasmic reticulums, mitochondria, and Golgi apparatus. Smooth muscle cells and fibroblasts had a few cell organelles. the epithelial cells of the hyperplastic glands demonstrated less cell organelles. However, smooth mucle cells and fibroblasts revealed abundant cell organelles. On the morphometry, the ratio of gland to stromal tissue were 0.28±0.04 in control group and 0.10±0.03 in nodular hyperplasia. According to the above results, it is suspected that the nodular hyperplasia is primarily a stromal disease.

      • 망막아세포종 세포주(Y-79)의 Arylalkylamine N-acetyltransferase 활성에 미치는 Dopamine의 영향

        The arylalkylamine N-acetyltransferase(AA-NAT, EC 2.3.1.87) in vertebral pineal gland and retina is rate-limiting enzyme in the pathway of biosynthesis of melatonin. AA-NAT activities are remarkably increased during night with 10-150 folds. Neurohormone melatonin is thought to play roles in a broad range of physiological functions in human, primarily through effects on the biological clock in the suprachiasmatic nucleus of the hypothalamus, This rhythm reflects large changes in the activities of AA-NAT. So the important regulatory role of AA-NAT has made it central interest in research on melatonin biochemistry and neurochemical signal transduction. The neurotransmitter that regulates the AA-NAT activities is norepinephrine, which acts through a cyclic AMP mechanism. The effect of dopamine on AA-NAT in human retinoblastoma Y-79 cells was studied. the obtained results are as follows; AA-NAT activities in Y-79 cells were increased by cyclic AMP, and dopamine inhibited the AA-NAT activities stimulated by forskolin. The pattern of inhibition of dopamine on the AA-NAT activities in Y-79 cells was time and concentration dependent. The results suggest that dopamine participates in the regulation of the AA-NAT activities through the inhibitory mechanism in the human pineal and retinal tissues.
