      • GO에 의해 유도된 培養 脊髓感覺神經細胞 독성에 대한 鎭肝熄風湯의 방어효과

        To evaluate the effect of Jingansikpungtang water extract on cultured mouse spinal sensory neuron which was inhibited by glucose oxidase(GO)-induced cytotoxicity, NR assay and TBARS assay for lipid peroxidation were carried out after the cultured mouse spinal sensory neuron were pre-incubated with various concentrations of Jingansikpungtang water extract for 3 hours prior to exposure of GO. GO, a oxygen radical, decreased the survival rate of the cultured mouse spinal sensory neuron cells on NR assay. Jingansikpungtang water extract have efficacy of decreasing lipid peroxidation increasing by GO in cultured mouse spinal sensory neuron. From above the results, it is concluded that Jingansikpungtang has marked efficacy as a treatment for the damages caused in the GO-mediated oxidative process.

        Guinea Pig에서 16, 16-Dimethyl Prostaglandin E2와 콜레스테롤이 담즙의 점액성 당단백질과 담석 형성에 미치는 영향

        Background/Aims: Prostaglandins is proposed to be responsible for gallstone formation in the animal models by hypersecretion of mucus in the gallbladder. This study was performed to determine the effect of 16,16-dimethyl prostaglandin E2 (dmPGE2) and cholesterol on mucous glycoprotein secretion of gallbladder in guinea pig. Methods: The prostaglandin was administered intraperitoneally to 30 guinea pigs fed with normal chow for 5 weeks at daily doses of 0 (controls), 15, 30 ㎍/kg (n=10 at each dose). There was another group (10 animals) fed with chow containing 1% cholesterol and 0.5% cholic acid and administered no dmPGE2. Results: The concentration of biliary mucous glycoprotein was significantly higher in the animals fed with chow containing 1% cholesterol and the animals treated with dmPGE2 than in the control animals (p$lt;0.05). The animals treated with dmPGE2 formed pigment gallstones, and the incidence was dose-dependent (p$lt;0.001). All animals fed with a cholesterol-rich diet also formed pigment gallstons. Infrared spectometry showed that the pigment gallstones contained calcium phosphate and calcium bilirubinate as major components. Conclusions: These results suggest that exogenous prostaglandin E2 increases concentration of mucous glycoprotein in the gallbladder bile and results in pigment gallstone formation through the mechanism similar to that of cholesterol-promoted gallstone formation.

        Highly photoluminescent N-isopropylacrylamide (NIPAAM) passivated carbon dots for multicolor bioimaging applications

        <P><B>Abstract</B></P> <P>We report a highly luminescent Carbon Dots passivated by nitrogen rich molecules, i.e. N-isopropylacrylamide (NIPAAM), with a QY as high as ca. 94%, which is the highest yield until now. NIPAMM@C-Dots are synthesized directly from the precursors composed of CA, EDA and NIPAAM by hydrothermal reaction. The optical properties of in situ synthesized NIPAAM@C-Dots showed EX-dependent EM at high concentration and EX-independent EM at low concentration. In most cations and anions except Fe<SUP>3+</SUP> and Bi<SUP>3+</SUP> added to the aqueous solution, NIPAAM@C-Dots exhibit an extremely stable inert behavior. In addition, in order to verify its usefulness as a fluorescent marker, C6 glioblastoma cells and NIPAAM@C-Dots were cultured together and then applied by both confocal bioimaging and flow cytometry analysis.         Glutamate and GABA concentrations in the cerebellum of novel ataxic mutant Pogo mice

        The Pogo mouse is an autosomal recessive ataxic mutant that arose spontaneously in the inbred KJR/MsKist strain derived originally from Korean wild mice. The ataxic phenotype is characterized by difficulty in maintaining posture and side to side stability, faulty coordination between limbs and trunk, and the consequent inability to walk straight. In the present study, the cerebellar concentrations of glutamate and GABA were analyzed, since glutamate is a most prevalent excitatory neurotransmitter whereas γ-aminobutyric acid (GABA) is one of the most abundant inhibitory neurotransmitters, which may be the main neurotransmitters related with the ataxia and epilepsy. The concentration of glutamate of cerebellum decreased significantly in ataxic mutant Pogo mouse compared to those of control mouse. However, GABA concentration was not decrease. These results suggested that the decrease in glutamate concentration may contribute to ataxia in mutant Pogo mouse.

        Voltage-dependent Calcium Channel (VDCC) α1A Subunit Expression in the Ataxic Mutant, Pogo Mice Cerebellum

        본 연구는 한국산 야생마우스에서 유래된 선천성 행동이상 Pogo마우스 소뇌 voltage-dependent calcium channel (VDCC) α1A의 발현을 면역조직화학법으로 조사하여 행동이상의 발생과 소뇌 P/Q calcium channel 발현의 상관관계를 밝히고자 계획되어 다음과 같은 결과를 얻었다. 첫째, VDCC α1A 면역반응은 행동이상 Pogo마우스와 대조군 마우스 소뇌겉질 조롱박세포, 과립세포 및 깊은소뇌핵에서 관찰되었다. 소뇌겉질 조롱박세포와 과립세포의 면역반응성은 행동이상 Pogo마우스와 대조군에서 특별한 차이가 관찰되지 않았다. 둘째, 깊은소뇌핵의 VDCC α1A 면역반응은 대조군에 비하여 행동이상 Pogo마우스 소뇌에서 증가됨이 관찰되었다. 깊은소뇌핵의 사이핵과 가쪽핵에서는 VDCC α1A 면역반응성이 증가됨이 뚜렷하였으나 정중핵에서는 대조군과 유사한 면역반응성을 보여 깊은소뇌핵 내에서도 차이가 있음을 관찰하였다. 이상의 결과로 행동이상 Pogo마우스에서 관찰되는 깊은소뇌핵 VDCC α1A 발현의 증가는 소뇌 칼슘 항상성의 변화를 유발하여 행동이상의 발생과 밀접한 연관관계가 있음을 확인할 수 있었다. The pogo mouse is a new ataxic mutant derived from a Korean wild mouse. The pogo mutation is inherited as an autosomal recessive trait on chromosome 8. Mutations in gene coding for the α1A subunit of voltagegated P/Q-type Ca2+ channel have been shown to cause phenotypes in humans and mice, i.e., tottering, leaner, rolling mouse mouse Nagoya. Using immunohistochemistry, the expression of the α1A subunit of voltage-gated P/Q-type Ca2+ channel was examined in pogo mice cerebellum including deep cerebellar nuclei (DCN). We observed α1A immunoreactivity in the cerebellar cortex (Purkinje cell and granule cell) and DCN of ataxic pogo mice and heterozygote control mice. There was no difference in cerebellar cortical α1A immunoreactivity between ataxic pogo mice and heterozygous littermate controls (pogo/+). However, we observed α1A immunoreactivity in the Purkinje cells of control and ataxic pogo mice cerebellum and DCN. We found a significant difference between pogo and heterozygous controls in terms of α1A immunoreactivities in the DCN. α1A immunoreactivity in this nucleus in pogo was much higher than in heterozygous littermate controls. No significant differences were observed in the interposed nucleus between pogo and heterozygous controls, but we found that the α1A subunits were clearer and more abundant in the lateral and medial regions of pogo than in control mice in these regions, where only weak immunoreactivity was observed. This elevated expression of the α1A subunit in deep cerebellar neurons of pogo might be a compensation for the altered function of P/Q type calcium channel and be related with the induction of the ataxic phenotype in pogo mice.

        In this study, localized development of a submerged cryogenic pump for use in LNG containers is conducted with large-sized commercial vehicles as the target. The submerged pump installed in an LNG storage container is the key module that supplies fuel to the engine through the reciprocating motion of a piston. Research and development on 660-L fuel containers is performed herein. The target is to achieve a mileage of 600 ㎞ or more by applying it to major NGV vehicles worldwide. In this manner, the present study aims to verify the operating mechanisms of the major parts of currently advanced products through reverse engineering in the early development process and draw basic design data.

        Differences of Zebrin II Expression Pattern Between Normal Balb/C and Ataxic Pogo Mouse Cerebellum

        zebrin II는 소뇌 Purkinje 세포에서 발현되는 36-kDa의 호흡효소(respiratory isoenzyme aldolase C)로 지금까지 알려진 가장 대표적인 소뇌구획표지자(cerebellar compartmentation marker)로 알려져 있다. 본 연구는 국내 최초로 발견된 선천성 운동실조 마우스인 pogo 마우스와 Balb/C 마우스 소뇌에서 zebrin II 발현양상의 차이를 확인하기 위하여 시행되었다. zebrin II 면역반응은 pogo 마우스와 Balb/C 마우스 소뇌 Purkinje 세포의 세포질과 수상돌기에서 관찰되었다. pogo 마우스와 Balb/C 마우스 소뇌 zebrin II 면역반응 Purkinje 세포들은 zebrin II에 면역반응을 보이지 않는 Purkinje 세포들과 서로 교대로 위치하여 소뇌의 앞-뒤를 연결하는 parasagittal band 형태로 관찰되었다. 소뇌 전체에 걸쳐 pogo 마우스와 Balb/C 마우스 소뇌 zebrin II 면역반응 Purkinje 세포들의 분포양상은 거의 유사하였다. 그러나, 소뇌 앞부분(anterior zone, AZ)의 lobule III에서는 pogo 마우스와 Balb/C 마우스 소뇌 zebrin II 면역반응 Purkinje 세포들의 분포양상에 차이가 관찰되었다. Balb/C 마우스 소뇌의 lobule III에서는 10~15개 정도의 zebrin II 면역반응 Purkinje 세포들이 모여 parasagittal band를 형성하지만, pogo 마우스의 경우 lobule III의 정중에 위치한 P1 band는 유사하나 P1 band의 양쪽에 위치한 P2 band의 경우 50~60개의 zebrin II 면역반응 Purkinje 세포들이 모여 parasagittal band를 형성한다. 이와 같은 lobule III의 zebrin II 면역반응 Purkinje 세포들의 분포차이는 pogo 마우스와 Balb/C 마우스의 유전적 차이에서 기원하는 것으로 생각되며 본 연구의 결과는 운동실조 pogo 마우스의 소뇌 형태적 연구의 기초적 자료로 이용될 수 있다고 생각된다. The purpose of this study is to identify the differences of zebrin II expression between ataxic pogo and normal Balb/C mouse cerebellum. Zebrin II is expressed by subsets of Purkinje cells that form an array of parasagittal bands that extend rostrocaudally throughout the cerebellar cortex, separated by similar bands of Purkinje cells that do not express zebrin II. Zebrin II immunoreactivity was localized in the perikarya of Purkinje cells, and the dendrites. Distribution of zebrin II-immunoreactive Purkinje cells were very similar pattern in pogo and Balb/C mouse cerebellum. But, in the lobule III, distribution of zebrin II expression was different between pogo and Balb/C mouse cerebellum. In lobule III of Balb/c mouse cerebellum, 10~15 zebrin II-immunoreactive Purkinje cells were observed and clustered to form a parasagittal bands. On the other hand, zebrin II expressions of lobule III in pogo mouse cerebellum showed a little different patterns. In lobule III of pogo mouse cerebellum, three bilateral zebrin IIimmunoreactive parasagittal band were observed. P1 band was almost same with lobule III of Balb/C mouse cerebellum. But, P2 bands were composed of 50~60 Purkinje cells which were immunoreactive with zebrin II. These kind of thickening in zebrin II expression of pogo mouse cerebellum may be due to the genetical difference. Furthermore, these results may provide useful information with further ataxic pogo mice cerebellum studies.

        Elevated Serotonin Transporter (5-HTT) Expression in the Ventral Horn of Lumbar Spinal Cord of Ataxic Pogo Mice

        Serotonin transporter (5-HTT) has been considered to exist in the presynaptic membrane of the serotonin (5-HT) neurons and play an important role in the terminating the 5-HT transmission. After all, maintaining 5-HT homeostasis by returning high synaptic concentrations to normal level. This study aimed clarify whether serotonergic (5-HTergic) innervation of the spinal motoneurons was altered in ataxic pogo mice, that characterized by a failures of interlimb coordination and frequent hyperextension of hindlimb. Using by immunohistochemistry and western blotting, we observed that the density of 5-HTT immunoreactive fibers and boutons in the ventral hon of lumbar region of spinal cord was significantly increased in pogo mice than in controls (P<0.01). Also, the 5-HTT protein was significantly increased in the only lumbar spinal cord of the ataxic pogo mice (P<0.05). However, the ventral horn of cervical and thoracic spinal cord of the pogo mice has no difference, compared with control mice. Furthermore, 5-HTT immunoreactivity levels were no difference in dorsal horn of cervical, thoracic and lumbar division of each group spinal cords. The results suggest that an increased 5-HTergic innervation of the lumbar motoneurons might be correlated with the abnormal hindlimb extension in the ataxic mice, pogo.

        The purpose of this study was to survey the actual condition about field learning materials in Gwangju and Jeonnam area. The questionnaire was developed to examine teachers` perceptions and actual condition of a field learning related science subject at 145 elementary schools in Gwangju city and Jeonnam province. The results of this study were as follows. First, the rate of school in performing field learning related to `strata and fossils` in science class was 6.8%. Second, the place of dianosaur track fossils in Jeonnam province, which was suitable for field learning `strata and fossils`, were well known by 75% of teachers. Also 68% of teachers have not visited those places. On the basis of this result, the followings are proposed. First, field learning materials and place should be developed in each school district. Second, field learning strategies should be developed in various level. Third, some programs to study how to teach in the field learning and experience the various field places should be suggested to preservice teachers as well as inservice teachers.

        간경변환자에서 복부외상후 일시적으로 관찰된 99mTc - MDP Bone Scan의 비장 섭취

        We report a case of 33-year-old man with a transient splenic uptake who had traumatic rnultiple rib fractures and hemoperitoneum combined with underlying liver cirrhosis, liver cirrhosis with liver bed laceration and splenomegaly without any other demonstrable splenic lesion due to traffic accident was found by abdominal ultrasound and surgery. Incidentally, 99mTc-MDP Bone scan showed whole prominent splenic uptake, spleen was also visible on 99mTc Tin coIloid liver scan. We suggest the splenic uptake of the radionuclide temporary splenic infarct to the transient total splenic infarction or unknown traumatic effect.
