        Preparation of [bis(amido)-phosphine] and [amido-phosphine sulfide or oxide] hafnium and zirconium complexes for olefin polymerization

        New phosphine-based bidentate ligands, 2-Me-8-Ph<SUB>2</SUB>P(X)C<SUB>9</SUB>H<SUB>8</SUB>NH (3, X = O; 4, X = S) and N-R-2-Ph<SUB>2</SUB>P(X)C<SUB>6</SUB>H<SUB>4</SUB>NH (5, R = Et, X = O; 6, R = Me, X = S; 7, R = Et, X = S) were prepared via ortho-lithiation of 1,2,3,4-tetrahydroquinaldine (2-Me-C<SUB>9</SUB>H<SUB>9</SUB>NH) and aniline derivatives (N-R-C<SUB>6</SUB>H<SUB>5</SUB>NH). Reaction of the ortho-lithiated compounds with 0.5 equiv of PhP(OPh)<SUB>2</SUB> afforded the bis(amido)-phosphine ligands (2-Me-C<SUB>9</SUB>H<SUB>8</SUB>NH-8-yl)<SUB>2</SUB>PPh (8) and (N-R-C<SUB>6</SUB>H<SUB>4</SUB>NH-2-yl)<SUB>2</SUB>PPh (9, R = Me; 10, R = Et). Using these ligands, [amido-phosphine oxide]Hf(CH<SUB>2</SUB>Ph)<SUB>3</SUB>, [amido-phosphine sulfide]Hf(CH<SUB>2</SUB>Ph)<SUB>3</SUB>, [bis(amido)-phosphine]MX<SUB>2</SUB> (M = Hf, Zr; X = CH<SUB>2</SUB>Ph, Cl, Me), and [amido-phosphine-amine]MCl<SUB>3</SUB> complexes were prepared. The molecular structures of [amido-phosphine sulfide]Hf(CH<SUB>2</SUB>Ph)<SUB>3</SUB> (13) (prepared using 6), [bis(amido)-phosphine]ZrMe<SUB>2</SUB> (22) (prepared using 8), and [amido-phosphine-amine]MCl<SUB>3</SUB> (23, M = Hf; 24, M = Zr; prepared using 9) were confirmed by X-ray crystallography. Most of the prepared complexes exhibited negligible or low activity for ethylene/1-octene copolymerization. The [amido-phosphine sulfide]Hf(CH<SUB>2</SUB>Ph)<SUB>3</SUB> complex (13) exhibited relatively high copolymerization activity (19 x 10<SUP>6</SUP> g/mol-Hf h); however, this activity was unsatisfactory compared to that of the related [amido-phosphine]Hf(CH<SUB>2</SUB>Ph)<SUB>3</SUB> complexes (up to 48 x 10<SUP>6</SUP> g/mol-Hf h).

        Identification and evaluation of potent Middle East respiratory syndrome coronavirus (MERS-CoV) 3CL<sup>Pro</sup> inhibitors

        <P>Middle East respiratory syndrome coronavirus (MERS-CoV) causes severe acute respiratory illness with fever, cough and shortness of breath. Up to date, it has resulted in 1826 human infections, including 649 deaths. Analogous to picornavirus 3C protease (3C(PrO)), 3C-like protease (3CL(PrO)) is critical for initiation of the MERS-CoV replication cycle and is thus regarded as a validated drug target. As presented here, our peptidomimetic inhibitors of enterovirus 3C(PrO) (6b. 6c and 6d) inhibited 3CLPr0 of MERS-CoV and severe acute respiratory syndrome coronavirus (SARS-CoV) with IC50 values ranging from 1.7 to 4.7 M and from 0.2 to 0.7 M, respectively. In MERS-CoV-infected cells, the inhibitors showed antiviral activity with EC50 values ranging from 0.6 to 1.4 1.1M, by downregulating the viral protein production in cells as well as reducing secretion of infectious viral particles into culture supernatants. They also suppressed other cc and 8-CoVs from human and feline origin. These compounds exhibited good selectivity index (over 70 against MERS-CoV) and could lead to the development of broad-spectrum antiviral drugs against emerging CoVs and picornaviruses.         Purpose: This study aims to identify the impact that cultural intelligence and learning engagement of Korean overseas students in Korea have in depression, also, to propose direction for further research, and to suggest a nursing intervention program that aims to the reduction of depression in overseas students. Methods:. The selected participants of the present study were Korean Overseas University Students enrolled in a special admission for overseas students. Data were obtained through the use of SPSS Window 25 and analyzed using descriptive statistics, t-test, ANOVA, Scheffe test, Pearsons correlation coefficient, and multiple linear regression analysis. Results: Depression in Korean overseas university students is influenced by the education system(ß=-.193, p=.046) and continent of residency(ß=0.197, p=.03). It is necessary to take into account the differences of education systems as well as the cultural differences from where they come from for the purpose of reducing depression rates in overseas Koreans. Conclusion: The results of this study show that depression in Korean overseas students is related to the differences in continents of residency and the education systems. Consequently, the development and implementation of a nursing intervention program considering both factors, the continent and education system, has become a necessity that relates to the reduction of depression in Korean overseas students.

        Antimicrobial peptide, hdMolluscidin, purified from the gill of the abalone, Haliotis discus

        <P>A 4.7 kDa antimicrobial peptide was purified from the acidified gill extract of the Abalone, Haliotis discus, by cation-exchange and C-18 reversed-phase high performance liquid chromatography (HPLC). Comparison of the amino acid sequences and molecular weight of this peptide with those of other known antimicrobial peptides revealed that this antimicrobial peptide have high sequence homology with that of cgMolluscidin and was designated hdMolluscidin. hdMolluscidin is composed of 46 amino acid residues containing several dibasic residue repeats like K-K or K-R. hdMolluscidin showed potent antimicrobial activity against both Gram-positive bacteria including Bacillus subtilis and Staphylococcus aureus (minimal effective concentrations [MECs]; 0.8-19.0 mu g/mL) and Gram-negative bacteria including Aeromonas hydrophila, Escherichia coli, Pseudomonas aeruginosa, Salmonella enterica, Shigella flexneri, and Vibrio parahemolyticus ([MECs]; 1.0-4.0 mu g/mL) without hemolytic activity. However, hdMolluscidin did not show any significant activity against Candida albicans. The secondary structural prediction suggested that hdMolluscidin might not form an ordered or an amphipathic structure. hdMolluscidin did not show membrane permeabilization or leakage ability. The full-length hdMolluscidin cDNA contained 566-bp, including a 5'-untranslated region (UTR) of 63-bp, a 3'-UTR of 359-bp, and an open reading frame of 144-bp encoding 47 amino acids (containing Met). cDNA study of hdMolluscidin suggests that it is expressed as a mature peptide. Our results indicate that hdMolluscidin could relate to the innate immune defenses in abalone and it may not act directly on bacterial membrane. (C) 2016 Elsevier Ltd. All rights reserved.</P>

        Antiviral activity of KR-23502 targeting nuclear export of influenza B virus ribonucleoproteins

        <P>The spiro compound 5,6-dimethyl-3H,3'H-spiro(benzofuran-2,1'-isobenzofuran)-3,3'-dione (KR-23502) has antiviral activity against influenza A and more potently B viruses. The aim of this study is to elucidate its mechanism of action. Subcellular localization and time-course expression of influenza B viral proteins, nucleoprotein (NP) and matrix protein 1 (M1), showed that KR-23502 reduced their amounts within 5 h post-infection. Early steps of virus life cycle, including virus entry, nuclear localization of NP and viral RNA-dependent RNA replication, were not affected by KR-23502. Instead it interrupted a later event corresponding to nuclear export of NP and M1 proteins. Delivery of viral ribonucleoprotein (vRNP)-M1 complex has been known to be mediated by the viral nuclear export protein (NEP) through interaction with cellular chromosomal maintenance 1 (CRM1) protein. In this study, we experimentally demonstrated that the compound targets the nuclear export of vRNP. Moreover, a single mutation (aspartate to glycine) at amino acid position 54 in M1 [M1(D54G)] was detected after 18 passages in the presence of KR-23502 with a 2-fold increase in 50% effective concentration indicating that this compound has a relatively high genetic barrier to resistance. Interestingly, it was observed that proteasome-mediated degradation of M1 (D54G) was attenuated by KR-23502. In conclusion, we suggest that KR-23502 shows its anti-influenza activity by downregulating NEP/CRM1-mediated nuclear export of influenza vRNP and M1. KR-23502 provides a core chemical skeleton for further structure-based design of novel antivirals against influenza viruses. (C) 2016 Elsevier B.V. All rights reserved.</P>

      • Traditional Brazilian fermented foods: cultural and technological aspects

        Fermented foods production started thousands of years ago and comprised a wide variety of products from diferent cultures and countries. The discovery of fermented foods is considered an empirical process based on human observation and experimentation of food types susceptible to natural biochemical and microbiological efects. Given the historical miscegenation of Brazilian people, the country has rich cultural diversity and a complex mix of ethnicities, religions and culinary traditions, among others. Thus, the current review aims at presenting the main cultural, microbiological and technological aspects of diferent types of fermented foods and beverages produced and consumed in Brazil, such as traditional artisanal cheeses, fermented meat (socol and charqui), non-alcoholic or low-alcohol beverages (aluá, calugi, tarubá and yakupá), alcoholic beverages (cachaça, tiquira, caiçuma, cauim and caxiri) and fermented foods based on cassava (puba, farinha d’água, polvilho azedo and tucupi).

      • Epac2a-null mice exhibit obesity-prone nature more susceptible to leptin resistance

        <P><B>Background:</B></P><P>The exchange protein directly activated by cAMP (Epac), which is primarily involved in cAMP signaling, has been known to be essential for controlling body energy metabolism. Epac has two isoforms: Epac1 and Epac2. The function of Epac1 on obesity was unveiled using Epac1 knockout (KO) mice. However, the role of Epac2 in obesity remains unclear.</P><P><B>Methods:</B></P><P>To evaluate the role of Epac2 in obesity, we used Epac2a KO mice, which is dominantly expressed in neurons and endocrine tissues. Physiological factors related to obesity were analyzed: body weight, fat mass, food intake, plasma leptin and adiponectin levels, energy expenditure, glucose tolerance, and insulin and leptin resistance. To determine the mechanism of Epac2a, mice received exogenous leptin and then hypothalamic leptin signaling was analyzed.</P><P><B>Results:</B></P><P>Epac2a KO mice appeared to have normal glucose tolerance and insulin sensitivity until 12 weeks of age, but an early onset increase of plasma leptin levels and decrease of plasma adiponectin levels compared with wild-type mice. Acute leptin injection revealed impaired hypothalamic leptin signaling in KO mice. Consistently, KO mice fed a high-fat diet (HFD) were significantly obese, presenting greater food intake and lower energy expenditure. HFD-fed KO mice were also characterized by greater impairment of hypothalamic leptin signaling and by weaker leptin-induced decrease in food consumption compared with HFD-fed wild-type mice. In wild-type mice, acute exogenous leptin injection or chronic HFD feeding tended to induce hypothalamic Epac2a expression.</P><P><B>Conclusions:</B></P><P>Considering that HFD is an inducer of hypothalamic leptin resistance and that Epac2a functions in pancreatic beta cells during demands of greater work load, hypothalamic Epac2a may have a role in facilitating leptin signaling, at least in response to higher metabolic demands. Thus, our data indicate that Epac2a is critical for preventing obesity and thus Epac2a activators may be used to manage obesity and obesity-mediated metabolic disorders.</P>
