        Overexpression of Zoysia ZjCIGR1 gene confers cold stress resistance to zoysiagrass

        Zoysia japonica Steud. is a warm-season lawn grass popular in Korea and elsewhere. They are cultivated in many places such as river banks, roadside, and play grounds. However, there still is a disadvantage of frequent mowing, and the grass grows poorly under the chilly conditions. To develop a grass variety that circumvents these drawbacks, we cloned the chitininducible gibberellins-responsive1 gene (CIGR1) from zoysiagrass. The full length of the ZjCIGR1 (Zj; Zoysia japonica Steud.) gene was obtained by 5′/3′ RACE PCR and the phylogenetic tree showed that it belonged to the CIGR1-subgroup in the PAT1-group of GRAS protein family. Expression of the ZjCIGR1 in wild-type plants was confirmed in roots, meristems, leaves, and flowers, especially high in the flowers. The transgenic zoysiagrass was confirmed by PCR using gene-specific primers, phosphinothricin-acetyl-transferase (PAT) strip test, and Southern blot analysis. ZjCIGR1-overexpressing plants acquired tolerance to cold stress displaying morphological phenotypes characteristic of stress resistance. In addition, in the transformants, expression of the ZjCIGR1 as well as cold-regulated (COR) gene was increased compared to the wild-type plants under cold stress condition. These results suggest that ZjCIGR1 gene is an important candidate for regulating cold stress resistance.

      • 편측 폐환기에서 Prostaglandin F2α가 심혈관계에 미치는 영향

        The lung is usually considered to have one function, namely gas exchange. Another function of lung is pharmacokinetic function, which the cells and enzymes of pulmonary vascular bed act to change biological activities of a variety of substances transported to them through pulmonary circulation. The pulmonary vasoconstrictor response to hypoxia is a part of a self-regulatory mechanism by which capillary blood flow is automatically adjusted to alveolar ventilation. Some vasoactive substances mediate the hypoxic pulmonary vascular response, and prostaglandin F2a, which is used for uterine contraction during obstetric surgery, is considered to potentiate the hypoxic pulmonary vasoconstriction. The author investigated the cardiovascular effects of PF2α during one-long ventilation in Korean mongrel dogs. Dogs were anesthetized with pentobarbital, morphine and pancuronium and intubated with Rush endotracheal tube No. 6.5.. One-lung ventilation was induced by deep right endobronchial intubation, which was confirmed by chest movement and abscence of breathing sound in the left lung. Prostaglandin F2α in dose of 0.5, 1.0, 2.0 ㎍/kg/min was infused into peripheral vein. The results were as follows: 1. One-lung ventilation group was compared with control group: The heart rate was decreased and MPAP, PVR and SVR were increased, In blood gas, pH and PO2 were decreased but PvCO_2 and shunt were increased. 2 Prostaglandin F2α infusion groups were compared with one-lung ventilation group: The cardiovascular function and blood gas changes were not effected by prostaglandin F2α, but MPAP was increased with dose-dependent manner and PVR were increased in prostaglandin F2α in doses of 1.0, 2.0㎍/kg/min infusion group. Thus hypoxic pulmonary vasoconstriction was found to be enhanced b usual dose of prostaglandin F2α (0.5 ~ 2.0 ㎍/kg/min).

      • 사람 위선암에서 cathepsin L의 발현증가

        cathepsin L은 lysosomal cysteine 단백분해효소로서 기저막(basement membrane)과 세포외기질(extracellular matrix)을 파괴하여 암세포의 침윤과 전이에 중요한 역할을 하는 물질로 알려져 있다. 이러한 cathepsin L에 대한 mRNA 발현도를 5개의 사람 위선암(gastric adenocarcinoma) 세포주와 5명의 위선암 환자에서 채취한 조직에서 방사능으로 표지된 cathepsin L특이 cDNA를 사용한 Northern blot법으로 측정하였다. 위암의 전이병소에서 수립한 세포주인 SNU-5, SNU-16, MKN-45와 Kato Ⅲ에서는 cathepsin L mRNA가 발현되었으나 원발병소에서 수립한 AGS 세포주에서는 mRNA의 발현이 관찰되지 않았다. 5명의 위암 환자에서는 원발병소, 전이가 확인된 임파절 및 암 근처 정상 위점막에서 각각 조직을 채취하여 cathepsin L mRNA의 발현을 측정하였다. 원발병소와 전이병소에서는 모두 cathepsin L mRNA가 발현되었으나 정상 위점막조직에서는 전예에서 mRNA 발현이 관찰되지 않았다. 한편 mRNA의 발현도는 1예에서는 전이병소가 원발병소에 비해 높았으나, 2예에서는 전이병소에서 발현도가 낮았으며, 나머지 2예에서는 원발병소와 전이병소 사이에 차이가 없어, 원발병소와 전이병소 사이에 mRNA의 발현도의 일관성 있는 경향은 관찰되지 않았다. 이상의 결과는 cathepsin L은 위암의 발생과 진행에 있어 암세포의 침윤과 전이를 촉진하는 것 이외에 또 다른 역할을 할 가능성을 시사하고 있다고 사료된다. Cathepsin L, a lysosomal cysteine protease, is known to play an important role in cancer invasion and metastasis by degrading the components of basement membrane and extracellular matrix. The mRNA expression of cathepsin L was determined by Northern blot analysis using a radiolabeled cDNA specific for cathepsin L in five human gastric adenocarcinoma cell lines and five surgical specimens of primary gastric adenocarcinomas, their metastatic lymph nodes and matched adjacent normal mucosae. The mRNA of cathepsin L was expressed in all of the four cell lines established from the metastatic sites, SNU-5, SNU-16, MKN-45 and Kato Ⅲ, while not detected in one cell line established from the primary site, AGS. The mRNA was expressed in all of the five primary and five metastatic cancer specimens tested, while it was not detected in all matched normal mucosae. The intensities of the mRNA expressions, however, did not show the consistent pattern between primary sites and metastatic lymph nodes. These results suggest that cathepsin L may have the other function in addition to facilitation of the invasion and metastasis during the development and progression of stomach cancer.

      • 탄소성 변형을 받는 냉간 압연강의 X선 잔류응력 특성에 관한 연구

        In textured material, diffraction angle 2θ usually shows a nonlinear relation against sin^2Ψ due to elastic anisotropy of crystals. SPHD and SPCD steel is cold-rolled carbon steel for automobile. The characteristics X-ray for stress measurement is Cr K_α and Mo K_α1characteristic X-ray. The sin^2Ψ-2θ diagram under elastic strain seems to have a linear behavior using regression line of data but has a nonlinear behavior in distribution of data by Cr K_αcharacteristic X-ray. As the plastic strain of specimen increases, the nonlinearity of 2θ with respect to sin^2Ψ increases remarkably. On the other hand, the diffraction angle 2 θ by Mo K_α1 characteristic X-ray shows a good linearity on sin^2Ψ-2θ diagram under plastic strain as well as elastic strain. Therefore, This paper presents the measurement of residual stress in cold-rolled carbon steel for automobile using penetration depth of Mo K_α1 charateristic X-ray and multiplicity factor of crystal diffraction plane.

        In this study, the potential energy and limitation of the facility of Chungju national university are analyzed in the respect of human knowledge, nature, and visual environment. In addition, the effctiveness and problem of the existing master plan are evaluated through the investigation on the main hart and development procedure of the previous master plan of CJU. These results are compared and reflected to the plan of Vision 2003, e-STRR2010 and effective campus development plans of CJU are presented.

        3차 의료기관 응급실로 내원한 안면부 열상 환자에 대한 임상적 고찰

        Background: Facial laceration is a common clinical problem in the emergency room that is frequently associated with other craniofacial injuries. It elicits an emotional response from the patient because of the underlying concern of permanent scar and secondary facial disfigurement, so they demand prompt and adequate treatment. The aim of this study is to analyze the incidence, etiology, depth, and site of facial lacerations to provide basic data for further understanding. Method: This study reviewed 1,043 facial-laceration patients treated in the emergency room at Pusan Paik Hospital between March 1999 and February 2000. This retrospective study was done by reviewing and analyzing the sex and age distributions, the monthly and daily distributions, the causes of injury, the types of injury, the sites of injury, the lengths of the lacerations, and associated injuries. Results: The sex ratio of men to wemen was 2.4:1, and the 0~10 age group was at the top of the age distribution. The number of patients was the highest on sundays and during March. About 9.2% of the patients visited the emergency room between 10:00 P.M. and 11:00 P.M. Falls(38.1%) were the most common cause of facial laceration, and deep laceration(51.8%) was the most common type of facial laceration. The most common laceration site was the forehead, followed by the chin and the nose. The most common associated craniofacial injury was facial bone fracture(39%), Facial-lacerations were sutured by plastic surgeons(80%), emergency physicians(10%), and oromaxillofacial surgeons(10%). We found statistical significance in the types and lengths of lacerations between men and women, among age groups(p<0.05). Conclusion: Facial laceration is one of the most common craniofacial injuries in the emergency room. By now, most lacerations have been repaired by plastic surgeons, But, the causes of lacerations are diverse and many patients are admitted due to the associated injuries. Therefore, emergency physicians should participate more aggressively in the care of facial-laceration patients to improve the outcome of the patients.

      • 고어텍스를 이용한 비성형 5례

        비성형수술에 삽입하는 사용물로는 자가연골이 가장 이상적이지만 필요한 만큼의 충분한 양을 얻기가 힘들고, 수술시간이 길어지며, 흡수가 일어날 수 있다는 단점이 있다. 이러한 단점을 극복할 수 있는 이종이식물 중에서 고어텍스는 최근에 개발되어 사용이 점차 증가하고 있는 추세이지만 아직까지 고어텍스의 임상적 효용성에 대한 보고는 드물다. 저자들은 고어텍스를 5명의 융비술 환자에서 사용하였는데 3명은 연골과 같이, 2명은 고어텍스 단독으로 사용하였다. 이 중 1명에서 감염이 발생하여 고어텍스를 제거하고 늑연골을 이용하여 융비술을 다시 시행하였고 나머지 4명은 모두 만족스러운 결과를 얻었다. 평균 20개월의 추적관찰에서 이식물의 이동이나 탈출, 피부의 손상 등의 큰 부작용이 없어 고어텍스가 비성형, 특히 융비술에 유용하다고 판단되어 관련문헌 고찰과 함께 보고하는 바이다. Though autogenous cartilage remains the ideal graft material in nasal surgery, situations exist where an alternative or an additional grafting material is needed. Gore-Tex is one of most recently developed alloplastic material and has been widely used for rhinoplasty. But there are few reports about the clinical efficacy of this material in rhinoplasty. The authors used Gore-Tex as a nasal implant in 5 cases of augmentation rhinoplasty. In three patients, the Gore-Tex was used with septal or conchal cartilages because the cartilages were not enough for the needed augmentation. In one patient, the implant was removed due to postoperative infection and subsequent augmentation was done with autogenous rib catilage after infection control. During average 20 months of follow-up, complications such as absorption, mobilization, or extrusion of the implant and thinning of the nasal skin were not found. All patients were satisfied with the surgical results. From these results, Gore-Tex can be considered as an effective material in augmentation rhinoplasty when autogenous material is not available or is not sufficient for the proper augmentation.
