        백선의 형태적 특성, 항균 효과 및 분자 세포학적 분석

        The importance of wild plant resources along with the development of high biotecyhnology is highlighted for exploitation of new materials which can make the added value. The goal of this study is providing fundamental data bases for developing new materials through the analyses of morphological characteristics, antibiotics and molecular cytogenetics in Baegseon (Dictamnus dasycarpus Turcz.). Baegseon has several characteristics; there are two types of flower color, pink and white, the seed germination starts about February 20th, the maximum flowering season is around May 17th in southern Korea, the growth period is about 60 days. The number of chromosomes are 2n=2x=36, the size of chromosomes in metaphase is $4.2{\sim}8.1{\mu}m$. The amount of 2C nuclear DNA is 1.93pg, and there is no variation of genome size amoung varieties. The extraction juice of baegseon young roots has the excellent antibiotic activity against the mold (Botrytis cinerea).

        木花 二面交雜에 의한 量的形質의 遺傳分析

        The experiment was conducted to get basic information for cotton breeding program through seven-variety diallel cross population. Significant variance analysis was observed in days to flowering, days to maturity, plant height, length of stem internode, bolls per plant, boll weight, seeds per boll, lint weight per boll, 100-seed weight, staple length, length of seed, width of seed and lint percentage. Based on the Vr-Wr graphical analysis, the characters, days to flowering, bolls per plant, seeds per boll, lint weight per boll, 100-seed weight, staple length, length of seed and width of seed were found to inherit incomplete dominance, and the characters of days to maturity, plant height, length of stem internode, boll weight and lint percentage were assumed to inherit over dominance. The component of genetic variance analysis for days to flowering, lint weight per boll, 100-seed weight, staple length, length of seed, width of seed and lint percentage showed that additive effects were higher than dominant effects. The narrow-sense heritability(h²ns) for days to flowering, bolls per plant, boll weight, seeds per boll, lint weight per boll, 100-seed weight, staple length and length of seed showed high values as more than 0.41. The estimate of broad-sense heritability(h²bs) value was higher than that of h²ns because of the low importance of dominance effects.

        국내 거베라 육종계통 형질전환 기초 조건 확립

        본 연구에서는 거베라 형질전환 기초기술개발을 위한 연구를 수행하였다. 경남원예화훼시험장에서 육성된 국내 거베라 18품종 중, Agrobacferium 감염에 순응적인 품종을 선발하기 위하여 18개의 국내 거베라 품종을 스크린하여 011, 016, GM023 등 12개의 순응형 계통을 스크린 하였으며 효율적인 형질전환을 위한 제균 항생제로는 cefotaxime이 선택되었다. 더 많은 캘러스 유도를 위한 전처리 배지의 호르몬 농도는 BA 2 ppm, Zeatin 2 ppm, IAA 0.2 ppm (2 x media)이 효율적이었고 재분화 배지의 호르몬 농도도 전처리 배지 호르몬의 농도와 동일하게 BA 2 ppm, Zeatin 2 ppm, IAA 0.2 ppm (2 x media)에서 효율적이었으나 GUS 발현에는 다른 차이가 없었다. 고효율 형질전환 체계 확립을 위하여 네 가지 다른 전처리 기간 처리와 두 가지 다른 Agrobacterium tumerfaciens 사용, 두 가지 다른 공배양 기간을 처리한 결과 전처리 하지 않은 엽병과 엽신을 LBA4404 Agrobacterium tumerfaciens을 사용하여 5일 간 공배양 기간을 가졌을 때와 dipping 처리하여 접종한 엽병과 엽신에서 높은 GUS 발현빈도를 나타냈다. Gerber (Gerbera hybrida) is a valuable ornamental species grown as a potted plant and cut flowers. However, genetic variability within the gerbera genus is very limited. So it is absolutely needed to introduce and widen genetic resources into gerbera lines by genetic transformation. For the purpose, 18 Korean gerbera lines were screened to establish Agrobacterium-mediated genetic transformation procedure. In an experiment to select Korean gerbera lines which are amenable to Agrobacterium-inoculation, 12 lines turned out to be positive in Agrobacterium-inoculation. More callus were produced from BA 2ppm, Zeatin 2ppm, IAA 0.2ppm in pre-culture and regeneration medium (2X media) but there was no difference in the frequency of GUS expression rate. In another experiment to find out optimal condition for highly efficient Agrobacterium-inoculation, petiole and leaf explants have been treated with four different pre-culture periods, two different co-culture periods and two different Agrobacterium strains. As a result, high GUS expression has been showed from petiole and leaf explants treated no pre-culture period with LBA4404 Agrobacterium tumerfaciens, 5 day co-culture period and dipping treatment.

        Diallel Cross Analysis of Characters in Barley

        In this experiment, gene actions were analysized for seven barley parents in order to obtain basic information on their genetic improvement by diallel crosses. The results obtained were summarized as follows. In analysis of variance, additive, dominant, maternal and reciprocal effects were observed significantly for culm length, tiller number, number of spiked per plant, culm diameter, awn length, leaf width, number of grains per spike, and 1000 grain weight. Over-dominance was shown by Vr-Wr graphic analysis in five characters such as tiller number, number of spikes per plant, length of flag leaf, number of grains per spike, and 1000 grain weight, and partial dominance in four characters such as culm length, culm diameter, awn length, and leaf width. Component of genetic varience analyzed for four characters such as culm diameter, awn length, length of flag leaf, and leaf width showed that additive effects were higher than dominant effects. Culm length, tiller number, number of spikes per plant, number of grains per spike, and 1000 grain weight showed dominant effects higher than additive effects. The narrow-sense heritability for awn length, leaf width, and number of grains per spike showed high values as more than 0.62, while broad-sense heritability for culm length, number of spikes per plant, culm diameter, awn length, leaf width, number of grains per spike, and 1000 grain weight showed high values as more than 0.65.

        사탕무(Beta vulgaris L.) 의 자엽소 배양에 의한 callus 유기 적색색소 및 단백질 \ulcorner량의 변화

        In order to produce betacyanins from sugar beet(Beta vulagris L.) in vitro, callus induction, shoot formation, root formation, betacyanin contents and protein contents determined from callus which was induced cotyledons of sugar beet seedling under an addition of NAA and BAP on the MS medium. The results were summarized as follows; The combination 3.0mg/l NAA and 1.0mg/l BAP treatment showed the most high callus induction rate, betacyanin and protein contents. The combination NAA and BAP treatments were not shoot formation, but BAP treatments showed high root formation rate. But high concentrations of BAP have not shown root formation.

        우리나라 재래종(在來種)들깨의 품종육성(品種育成)에 관한 연구(硏究)(2)

        1979年度 本大學 農科大學에서 들깨의 品種改良 및 育成을 위하여 全國 109個 地域 在來系統 蒐 集種中에서 比較的 優秀한 20個 系統을 選拔하여 1980年度에 계속 사업으로 實施한 本實驗에서 몇가지 結果가 있었기에 이를 報告하는 바이다. 1. 本實驗에서 收量은 大邱種보다 제천-3070-28이 35.9%, 연기-4120-35가 51.5%, 남제주--7010-67이 53.7%, 문경-8090-71이 69.1%, 하동-9220-97이 44.2% 가량의 增收를 하였다. 2. 各 形質間의 相關關係는 收量과 莖直徑, 有效分枝數間에는 正의 相關關係가 認定되었고 1000粒重에 있어서는 花房數 및 油脂含量間에 否의 경향을 나타내었다. 3. 油脂含量에 있어서는 갈색계통인 금능-80 50-69가 49.6%, 제천-3070-28이 47.4%, 성주-8130-72가 47.7%로 높았고 白色系統은 比較的 油脂含量이 낮았다. 4. 多重不飽和 脂肪酸에 있어서도 갈색계통은 53.4%인데 비하여 白色系統은 46% 정도로 낮았다. 5. 들깨 잎에는 칼슘함량에 있어선 연기-2060-17이 284mg/100g으로 가장 높았고 나주-60 80-49가 241.4mg/100g으로 다음 이었다. 그리고 마그네슘은 성주-1830-72가 77.5mg/100g으로 가장 높았고 蛋白質含量은 남해-932 0-102, 금능-8050-69가 5.5g/100g 以上으로 높은 數値를 나타내었다. The experiment was carried out continualy from 1979 for the Perilla, breeding and variety improvement of excellent lines selected from 109 local species, and the results obtained were summarized as fellow: 1. The increasing ratio of yield per 10a were 35.9% in Jecheon-3070-28 lines, 51.5% in Yeongi-8090-71, 53.7% in Namje ju-70l0-67, 69.1% in Mungyeong-8090-71 and 44.2% in Hadong-9220-97 as compared with the yield amount of Daegu. 2. Statistically positive correlation were convinced between yields and plant length, stem diameter, and number of effective valid branches. 3. In percentage of oil composition, Geumnung-8050-69 was 49.6%, Jecheon-3070-28 was 47.4%, Seong ju-8130-72 was 47.7%, and the oil contents of these brown seed coat lines were considered to be comperatively higher than those white seed coat lines. 4. Contents of ploy-unsaturated fatty acid was 53% in brown seed coat lines, on the other hand, that of white seed coat lines was about 43.9%. 5. In calcium contents of perilla leaf, the amounts of 284.7mg in Yanggu-2060-17 was the highest and Na ju-6080-49 (241.4mg) was the next, Seongju-8130-72(77.5mg) was highest in magnesium composition, and the protein content in Namhae-9320-102, and Geumnung-8050-69 were more than 5.5g.

        수도작에서 왕우렁이 및 오리 방사가 수량구성요소에 미치는 영향

        This experiment was carried out to find the effect of different organic farming types, the Duck - pasture plot, the Pond-snail pasture plot and control of the conventional culture plot farming on the yield and its components of paddy rice. The results obtained are summarized as follows. Plant height and Tillering were significantly increase in the Duck - pasture plot. Panicle number per hill, No. of grain per spike and 1,000grain weight were significantly increase in the conventional culture plot. Yield per 10a were 574.13^ at pond-snail pasture plot, 576.81kg at Duck-pasture plot and 579.04kg at conventional culture plot. From these results, it seems that the organic fanning are usefel and pasture of the Duck, Pond-snail for depression of the weed occurrence or increase of yield per 10a is effective as the environmental perservation farming.

        RAPD를 이용한 짚신나물(Agrimonia pilosa Ledeb.) 수집종 유연관계 분석

        우리 나라 자생 짚신나물(Agrimonia pilosa Ledeb.) 수집종의 분포, 자생지의 환경 및 자생지에서의 형태적 특성조사를 실시하고, 자생종 짚신나물와 도입 짚신나물 간의 형태적 특성 및 유연관계를 연구한 결과는 다음과 같다. 1. 수집된 종 중에서 하동지역의 화개면 쌍계사와 청암면 청학동에서 수집된 2종은 다른 수집종에 비하여 유전적 거리지수가 가장 가까운 수치에서 나타나 근연관계로 나타났으며, 충북 영동읍 비탄재와 경기도 강화읍 수집종간에는 유전적 거리지수는 최대값이 같은 것으로 보아 유연관계가 있는 것으로 판단된다. 2. 경남거제에서 수집(No. 7)된 것과 충북 보은에서 수집(No. 21)된 2종이 유전적으로 가장 가깝게 나타나 지역적 차이에 의한 진화에도 다양함을 보이고 있어 수집지역의 확대와 이에 따른 체계적인 선발을 통해서 우량종의 선발도 가능할 것으로 판단된다 3.증폭된 DNA band pattern에 있어서 절편크기는 300∼21000p로 PCR로 증폭된 종 상호간의 1-F값은 평균 0.624였고, 최소값 0.365, 최대값 0.827이었으며, 2개 군으로 분리되어 유전적으로 상당한 차이가 있는 것으로 나타났다. Agromonia pilosa Ledeb. has been used as a medicinal plant in traditional folk remedy. There are few reports on classification, physiology, ecology and morphological studies of Agromonia pilosa L. in Korea. Therefore, advanced approaches on study and development with this plant would be done urgently. Present stndy was carried out to gain basic information on genetic resources and variation with collected domestic landraces through RAPD analysis in Agromonia pilosa L. Forty two collections of Agromonia pilosa L. from nation-wide area including USA one were analyzed by RAPD test. Molecular marker size by amplified DNA band pattern ranged from 300 to 2,100bp. Among the collection, two landraces of Hadong and Cheonghak-dong showed close relation in genetic similarity. Minimum and maximum value by matrix of 1-F among 26 collected landraces were figured out as 0.365 and 0.827 showing mean value for 0.624, respectively. Those landraces were classified into two groups with cluster analysis by Nei and Li s formula from RAPD-analyzed values, and considerable genetic differences were recognized between two groups.
