      • 초피 추출물의 수율 및 항균활성

        Extraction yields of Zunthoxylum piperiturn by organic solvents and supercritical carbon dioxide(SC-CO_(2)) with and without ethanol as a cosolvent, and antimicrobial activities of the extracts against Hucillus cereus were determined. Extraction yield by distilled water was the highest, 20%, while those by ethanol, ethyl ether and hexane were less than 3%. Extraction yield by SC-CO_(2) Lvith ethanol was 1.42 times greater than that by only SC-CO_(2). Ethyl ether- and ethanol-extracts had potent antimicrobial activities against B. cereus. Extraction yield by SC-CO_(2) with ethanol showed almost the same degree of microbial growth inhibition as that by only SC-CO_(2).

      • 이소골 전위 및 골절에서의 고해상 CT 소견

        목적 : 측두골 외상으로 인한 이소골 전위나 골절의 고해상 CT 소견을 알아보고자 하였다. 대상 및 방법 : 1998년 1월부터 2002년 6월까지 측두골 외상 환자 중 고해상 CT에서 이소골 전위나 골절이 있었던 11명의 환자를 대상으로 하였다. 8명의 환자에서는 필름을, 3명의 환자에서는 PACS를 이용한 고해상 CT를 후향적으로 분석하였고 이소골 전위 및 골절의 분류, 측두골 골절의 분류, 그 외 주위조직의 동반된 이상소견을 알아보았다. 결과 : 11명의 이소골 이상이 있었던 환자 중 침추골 관절 분리가 10예로 가장 많았고 침등골 관절 분리가 4예 있었으며 추골 골절, 침골 골절, 침골의 장각과 두상돌기 전위, 침골돌기가 보이지 않았던 경우가 각각 1예씩 있었다. 측두골 골절은 8예에서 있었으며 종위골절이 7예, 황위골절 1예였고 그 외에도 경동맥관 골절, 경동맥해면루, 접형골과 후두골의 골절이 동반되었다. 필름을 이용한 고해상 CT에서 보다 PACS를 이용한 CT에서 확대 및 창폭(window width) 변화가 가능하여 이소골 평가에 이용하였다. 결론 : 측두골 외상으로 인한 이소골의 전위가 골절은 고해상 CT상 횡단 및 관상면의 정확한 해부학적 지식이 필요하였으며 침추골 관절 분리가 가장 흔하고 측두골의 종위골절이 많이 동반되며 특히 PACS를 이용한 고해상 CT가 이소골 평가에 유용하였다. Purpose : To evaluate the CT findings of ossicular disruption and fracture utilizing high resolution computed tomography(HRCT) of the temporal bone. Materials and Methods : The authors retrospectively reviewed the HRCT scans of 11 patients with ossicular injuries between January 1998 and June 2002 using films in 8 patients, PACS in 3 patients. The type of ossicular injuries, temporal bone fracture, and associated adjacent injuries were evaluated. Results : Among the 11 patients of ossicular injuries, there were 10 cases of incudomalleal disruption, 4 cases of incudostapedial disruption, and 1 case of fracture of malleus, fracture of incus, disruption of long and lenticular process of incus, nonvisualization of processes of incus. The temporal bone fractures were longitudinal in 7 cases, transverse in I case. Other injuries were fracture of the carotid canal, caroticocavemous fistula, fracture of sphenoid and occipital bones. Conclusion : Incudomalleal disruption was the most frequent tupe of ossicular injuries and associated temporal bone fracture was longitudinal type. HRCT using PACS was useful in evaluation of ossicular injuries than films.

      • All-Trans Retinoic Acid에 의한 HL-60 세포 분화유도중 H2B Histone, c-Myc 및 DNA Topoisomerase I 발현에 관한 연구

        Effects of all-trans retinoic acid(retinoic acid) on DNA replication, H2B histone and DNA topoisopmerase I(Topo I) gene expression have been investigated in human promyelocytic leukemia cell line HL-60. DNA synthesis decreased at 24 hours after exposure of retinoic acid in the HL-60 cells. H2B histone mRNA rapidly reduced at 48 hours and Topo I and c-myc mRNA at 24 hours in retinoic acid-exposured HL-60 cells, respectively. The levels of c-myc, H2B histone and Topo I gene expression were reduced in proportion to the concentration of retinoic acid. The H2B histone mRNA in retinoic acid-exposured cells was elevated by cycloheximide treatment, while the level of Topo I mRNA was constant. These results suggest that regulations of H2B histone, c-myc and Topo I gene expression are different from one another, and repression of Topo I gene is closely correlated with c-myc gene during retinoic acid-induced differentiation of HL-60 cells.

      • 초임계 이산화탄소에 의한 멸치어유의 추출

        고품질의 고도불포화지방산 제품을 제조할 목적으로, 멸치어유를 초임계 이산화탄소로 추출분획한 후 각 분획들의 추출수율과 지방산 조성을 측정하였다. 멸치어유의 주요 지방산은 palmitic acid. DHA, EPA, palmitoleic acid. myristic acid. oleic acid 였다. 멸치어유의 추출수율은 동일압력에서 추출시간의 증가에 따라 감소하였으며, 40과 50℃에서는 추출온도의 증가에 따라 감소하였으나, 60℃에서는 50℃ 보다 다소 높았다. 동일온도에서 C14와 C16 지방산들은 추출물 분획에 농축되었고, 나머지 지방산들은 추출 잔류물 분획에 농축되었다. EPA는 분획들간에 거의 일정한 경향을 보였으나, DHA는 추출잔류물 분획에 농축되었다. Silver nitrate를 silica gel에 코팅시킨 후 어유와 혼합하여 추출 2시간 동안은 초임계 이산화탄소로만으로 추출하고, 그 후 보조용매로 ethyl acetate를 첨가하여 2시간 동안 추출하였을 때 추출물 분획에서의 저급지방산인 C14:0은 추출잔류물에 비하여 5.0∼6.4배 농축되었고, C16:0은 1.9∼2.0배 농축된 반면, 추출잔류물 분획에서 EPA와 DHA는 23.9와 31.6%로 원료어유의 13.3과 16.2%에 비하여 1.8배와 1.9배 농축되었다. Anchovy oil was extracted by supercritical CO₂, and extraction yield and fatty acid composition were measured in the extract and residual fraction. EPA and DHA were 7.6∼13.3% and 16.2∼18.3% for the original anchovy oil. The extraction yield of anchovy oil increased as the extraction temperature decreased. Essentially no great differences were noticed in the fatty acid composition of the oil extracted under different extraction temperatures. Anchovy oil mixed with silver nitrate-coated silica gel was extracted by only SC-CO₂ for 2 hr and by ethyl acetate-modified SC-CO₂ for 2 hr at 60℃/276 bar. This produced higher extraction yield and higher concentration of EPA and DHA in the residual fraction. Starting with anchovy oil containing 13.3% EPA and 16.2% DHA, the residue fraction containing 23.9% EPA and 31.1% DHA was obtained.

      • 고추장 숙성 중 품질 변화와 초고압처리에 의한 α-amylase 활성 변화

        Qualily changes in pH, titratable acidity and amino nitrogen of Kochujang were measured during the period of fermentation. α-amylase activity was also determined in Kochujang treated with ultra-high pressure. Initial pH of Kochujang was 5.05. There was a little decrease after 10 days of aging and then drastic change occurred. It reached 4.74 after 80 days. Titratable acidity of Kochujang just after preparation was 11.8 mL per l0g of Kochujang and then increased rapidly. It was 15.2 mL after 80 days. Amino nitrogen was increased significantly from 90.0 to 114.9 mg% after 10 days of aging. It increased as the increase of aging period and then reached 160.7 mg% after 80 days of fermentation. α-amylase activities decreased as the increase of treatment pressure, temperature and time. During storage at 37℃, α-amylase activities of all samples regardless of treatment conditions decreased significantly after 45 days, and then increased gradually.

      • 스틸벤의 치환기 효과에 대한 분자궤도함수론적 해석

        치환 시틸벤 계열에 대해 HMO법으로 Hammett 치환기 상수를 해석하였다. 이 계열에서 치환기 상수에 대한 유도효과와 공명효과의 기여를 양자화학적 지수로 취하여 계산한 이론값이 Hammett 치환기 상수 σ_p와 병행성이 있음을 알았으며 이 이론값으로 치환 스틸벤의 쌍극자능률에 미치는 치환기 효과를 설명할 수 있었다. 아울러 이 화합물의 전자전이에 대한 최대 흡수파장(λ_max)은 HOMO와 LUMO 에너지의 차에 의존됨이 확인되었다. The Hammett's substituent constants were interpreted for substituted stilbenes by HMO method. The appropriate quantum chemical indices are chosen as independent contribution of the inductive and the resonance effects for substituent constants. It has been found that theoretical values, σ_p^th, defined as sum of the net charge, self atom polarizability and difference in HOMO energy between substituted- and unsubstituted stilbenes, correlated with experimental Hammett's substituent constants. The dipole moments were found to be correlated with differences in σ_p^th between two substituents for disubstituted stilbenes. It has been also found that transition optical spectra, λ_max of the substituted stilbenes depend on difference between the HOMO and the LUMO energy as expected.

        The contents for the chemistry parts in the five textbooks of middle school science 1, 2, 3 based on 5th middle school new curriculum were analyzed and compared with each other. The results showed as follows : (1) Most of textbooks were well organized in accordance with objectives and contents based on the new curriculum. (2) The contents of the textbooks had a little difference according to authors. (3) The quantity of space increased as the grade is moved up. (4) Most of textbooks dealt with observation and measurements, conditional controls, performance of laboratory equipments and discussion of results exceedingly in the content of experiments, but sefety educations were somewhat neglected.

      • 초임계이산화탄소에 의한 유기염소계 잔류농약의 포집효율

        Collection efficiencies of organochlorine pesticides by supercritical fluid extraction(SFE) was investigated to develop a simple, fast and inexpensive extraction method. Standard organochlorine pesticides mixture(SOPM, no water) showed no evidence of metering valve plugging in contrast to the plugging associated with SFE of fish tissues(wet sample). Maximum percent recovery of SOPM was obtained at bar by collecting the extract within 3 min through the metering valve without heating into a capped screw-top tube half filled with solid CO_(2) Recovery of SOPM itself shwes 98.3% while recovery of that spiker in fish tissue 85.6%.

        2003년 5월의 연무 관측시 에어로졸의 기상·물리·화학 특성

        Severe haze, mist, and fog phenomena occurred in the central part of Korea during 15~25 May 2003 resulted in poor visibility and air quality. When these phenomena occurred, Korean peninsula was under the effects of anticyclone. The atmosphere was stable, and wind speed was so weak. Under this meteorological conditions, air quality was worse and worse. The characteristics of aerosol in Seoul, Incheon, and Gosan (Jeju) during this period are investigated from the PM_(10). TSP concentrations and aerosol number concentrations. Concentrations of PM_(10) and TSP measured at KMA increased upto 176 and 230㎍/㎥ on 22 May 2003, respectively. Aerosol number concentrations of size range from 0.82 to 6.06 ㎛ increased in Seoul on 17, 19, and 21~24 May 2003, and the concentrations of NO₂ and SO₂had maximum value of 0.165 ppm at Gwanak Mt. and 0.036 ppm at Guro-dong on 23 May 2003, respectively. Result from analysis on heavy metal elements showed high concentrations of Zn, Pb, Cr, Ni, Cu, and Cd during 20~24 May 2003. This event is examined by comprehensive analyses of synoptic weather conditions, satellite images, concentrations of suspended particles and air pollutants, and heavy metal elements.

      • 운동이 일부 남녀 대학생들의 혈청과 뇨중 칼슘, 마그네슘 및 아연함량에 미치는 영향

        This study investigated the effect of regular exercise on mineral status in college students. Mineral status for calcium, magnesium, and zinc was evaluated prior to beginning the study and again after 35 days of exercise training for 30 minutes per day. The results obtained were summarized as follows: 1) Serum concentrations of calcium and magnesium were decreased in female subjects but were not affected in males. 2) Urinary excretion of calcium and magnesium was significantly increased in male subjects but unchanged in females. 3) Serum and urinary zinc concentrations were not affected in either male or female subjects. These results suggest that regular exercise may have different effect on calcium and magnesium in humans, depending on gender.
