        Study of upfront surgery versus neoadjuvant chemotherapy followed by interval debulking surgery for patients with stage IIIC and IV ovarian cancer, SGOG SUNNY (SOC-2) trial concept

        Background: Two randomized phase III trials (EORTC55971 and CHORUS) showed similarprogression-free and overall survival in primary or interval debulking surgery in ovariancancer, however both studies had limitations with lower rate of complete resection and lack ofsurgical qualifications for participating centers. There is no consensus on whether neoadjuvantchemotherapy followed by interval debulking surgery (NACT-IDS) could be a preferred approachin the management of advanced epithelial ovarian cancer (EOC) in the clinical practice. Methods: The Asian SUNNY study is an open-label, multicenter, randomized controlled,phase III trial to compare the effect of primary debulking surgery (PDS) to NACT-IDS instages IIIC and IV EOC, fallopian tube cancer (FTC) or primary peritoneal carcinoma (PPC). The hypothesis is that PDS enhances the survivorship when compared with NACT-IDS inadvanced ovarian cancer. The primary objective is to clarify the role of PDS and NACT-IDS inthe treatment of advanced ovarian cancer. Surgical quality assures include at least 50% of nogross residual (NGR) in PDS group in all centers and participating centers should be nationalcancer centers or designed ovarian cancer section or those with the experience participatingsurgical trials of ovarian cancer. Any participating center should be monitored evaluatingthe proportions of NGR by a training set. The aim of the surgery in both arms is maximalcytoreduction. Tumor burden of the disease is evaluated by diagnostic laparoscopy orpositron emission tomography/computed tomography scan. Patients assigned to PDS groupwill undergo upfront maximal cytoreductive surgery within 3 weeks after biopsy, followed by6 cycles of standard adjuvant chemotherapy. Patients assigned to NACT group will undergo 3cycles of NACT-IDS, and subsequently 3 cycles of adjuvant chemotherapy. The maximal timeinterval between IDS and the initiation of adjuvant chemotherapy is 8 weeks. Major inclusioncriteria are pathologic confirmed stage IIIC and IV EOC, FTC or PPC; ECOG performancestatus of 0 to 2; ASA score of 1 to 2. Major exclusion criteria are non-epithelial tumors as wellas borderline tumors; low-grade carcinoma; mucinous ovarian cancer. The sample size is 456subjects. Primary endpoint is overall survival.       • Lobaplatin-TACE Combined with Radioactive <sup>125</sup>I Seed Implantation for Treatment of Primary Hepatocellular Carcinoma

        Aim: To investigate the efficacy and safety of lobaplatin-transcatheter arterial chemoembolization (TACE) combined with radioactive $^{125}I$ seed implantation in treatment of primary hepatocellular carcinoma (HCC). Methods: 75 patients with primary HCC were enrolled in the study, among them 43 receiving lobaplatin-TACE (TACE group) and 32 lobaplatin-TACE combined with $^{125}I$ seed implantation (TACE+$^{125}I$ group). After treatment, the local remission rates and postoperative complications of two groups were compared using the Pearson Chi-square test. Overall survival in the two groups was calculated using Kaplan-Meier survival curves and the differences were tested using Log-rank test. Results: There were 7 cases of complete response (CR), 13 of partial response (PR), 6 of stable disease (SD) and 17 of progressive disease (PD) in the TACE group, with 13 cases of CR, 9 of PR, 5 of SD and 5 of PD in the TACE+$^{125}I$ group. The disease control rates of TACE and TACE+$^{125}I$ group were 60.5% (26/43) and 84.4% (27/32), respectively, with a significant difference between them (P < 0.05). The survival rates at 6, 12 and 18 months in the TACE group were 100.0%, 81.8% and 50.0%, respectively, and those in TACE+$^{125}I$ group were 100.0%, 93.8% and 65.6%. The mean survival times in the TACE and TACE+$^{125}I$ groups were 19.5 and 22.9 months, respectively. There was a significant difference in the overall survival rate between two groups (P < 0.05). No serious complications were encountered in either group. Conclusion: Lobaplatin-TACE combined with $^{125}I$ seed implantation is favorable and safe for treatment of primary HCC.

        Background: We examined changes in hepatitis B core-related antigen (HBcrAg) during the four sequential phases of chronic hepatitis B virus (HBV) infection: hepatitis B e antigen (HBeAg)-positive chronic infection (EPCI) and hepatitis (EPCH), followed by HBeAg-negative chronic infection (ENCI) and hepatitis (ENCH). We compared the performance of serum HBcrAg, hepatitis B surface antigen (HBsAg), and HBV DNA in predicting EPCH and ENCH.

        Efficient Production of ε-Poly-L-Lysine by Streptomyces ahygroscopicus Using One-Stage pH Control Fed-Batch Fermentation Coupled with Nutrient Feeding

        ε-Poly-L-lysine (ε-PL) is a homopolymer of L-lysine molecules connected between the epsilon amino and alpha carboxyl groups. This polymer is currently used as a natural preservative in food. Insufficient biomass is a major problem in ε-PL fermentation. Here, to improve cell growth and ε-PL productivity, various nitrogen-rich nutrients were supplemented into flask cultures after 16 h cultivation, marking the onset of ε-PL biosynthesis. Yeast extract, soybean powder, corn powder, and beef extract significantly improved cell growth. In terms of ε-PL productivity, yeast extract at 0.5% (w/v) gave the maximum yield (2.24 g/l), 115.4% higher than the control (1.04 g/l), followed by soybean powder (1.86 g/l) at 1% (w/v) and corn powder (1.72 g/l) at 1% (w/v). However, supplementation with beef extract inhibited ε-PL production. The optimal time for supplementation for all nutrients examined was at 16 h cultivation. The kinetics of yeast-extract-supplemented cultures showed enhanced cell growth and production duration. Thus, the most commonly used two-stage pH control fed-batch fermentation method was modified by omitting the pH 5.0-controlled period, and coupling the procedure with nutrient feeding in the pH 3.9-controlled phase. Using this process, by continuously feeding 0.5 g/h of yeast extract, soybean powder, or corn powder into cultures in a 30 L fermenter, the final ε-PL titer reached 28.2 g/l, 23.7 g/l, and 21.4 g/l, respectively, 91.8%, 61.2%, and 45.6% higher than that of the control (14.7 g/l). This describes a promising option for the mass production of ε-PL.

        Half Load-Cycle Worked Dual SEPIC Single-Stage Inverter

        The two-stage converter is widely used in traditional DC/AC inverter. It has several disadvantages such as complex topology, large volume and high loss. In order to overcome these shortcomings, a novel half load-cycle worked dual SEPIC single-stage inverter, which is based on the analysis of the relationship between input and output voltages of SEPIC converters operating in the discontinuous conduction mode (DCM), is presented in this paper. The traditional single-stage inverter has remarkable advantages in small and medium power applications, but it can’t realize boost DC/AC output directly. Besides one pre-boost DC/DC converter is needed between the DC source and the traditional single-stage inverter. A novel DC/AC inverter without pre-boost DC/DC converter, which is comprised of two SEPIC converters, is studied. The output of dual SEPIC converters is connected with anti-parallel and half load-cycle control is used to realize boost and buck DC/AC output directly and work properly, whatever the DC input voltage is higher or lower than the AC output voltage. The working principle, parameter selection and the control strategy of the inverters are analyzed in this paper. Simulation and experiment results verify the feasibility of the new inverter.

        A series of novel chiral tert-amino alcohols 4a-h derived from enantiomerically pure phenylalanine were synthesized efficiently and used as chiral ligands in the catalytic enantioselective ethylation of aldehydes with diethylzinc (diethylzinc-to-aldehyde addition). The use of 10 mol % of the amino alcohols led to the corresponding sec-alcohols with excellent enantioselectivities (up to 100% ee) and high yields.

        Half Load-Cycle Worked Dual Input Single Output DC/AC Inverter

        A novel half load-cycle worked dual input single output (DISO) DC/AC inverter is presented. The basic circuit consists of a dual buck regulator, which works in continuous current mode. The working principle of DISO DC/AC inverter has been used. The control method applied for half load-cycle worked DISO DC/AC inverter has been studied. The control effects of the open-loop proportional control and closed-loop proportional-integral control are compared by using PSIM software. The parameters are adopted in the realistic simulation and experiment test. Moreover, the waveforms, such as voltage of modulation reference signal and output voltage, were given. The simulation and experiment results proved that the half load-cycle worked DISO DC/AC inverter could achieve good performance, gain a line frequency of 50 Hz, and verify the correctness of theoretical analysis.

      • Soluble Expression of Recombinant Human Smp30 for Detecting Serum Smp30 Antibody Levels in Hepatocellular Carcinoma Patients

        Senescence marker protein 30 (SMP30), a hepatocellular carcinoma (HCC) associated antigen, was earlier shown by our research group to be highly expressed in HCC paracancerous tissues, but have low levels in HCC tissues. In order to detect anti-SMP30 antibody in serum of HCC patients, we established pET30a-SMP30 and pColdIII-SMP30 expression systems in Escherichia coli. However, the expression product was mainly in the form of inclusion bodies. In this research, we used several combinations of chaperones, four molecular chaperone plasmids with pET30a-SMP30 and five molecular chaperone plasmids with pColdIII-SMP30 to increase the amount of soluble protein. Results showed that co-expression of HIS-SMP30 with pTf16, combined with the addition of osmosis-regulator, and a two-step expression resulted in the highest enhancement of solubility. A total of 175 cases of HCC serum were studied by ELISA to detect anti-SMP30 antibody with recombinant SMP30 protein. Some 22 were positive and x2 two-sided tests all showed P>0.05, although it remained unclear whether there was a relationship between positive cases and clinical diagnostic data.

        Application of wavelet transform for the impulse response of pile

        The purpose of this paper is to study the capabilities of the impulse response method in length and flaw detecting for concrete piles and provide a suggested method to find small-size flaws in piles. In this work, wavelet transform is used to decompose the recorded time domain signal into a series of levels. These levels are narrowband, so the mix of different dominant bandwidths can be avoided. In this study, the impulse response method is used to analyze the signal obtained from the wavelet transform to improve the judgment of the flaw signal so as to detect the flaw location. This study provides a new way of thinking in non-destructive testing detection. The results show that the length of a pile is easy to be detected in the traditional reflection time or frequency domain method. However, the small flaws within pile are difficult to be found using these methods. The proposed approach in this paper is able to greatly improve the results of small-size flaw detection within piles by reducing the effects of any noise and clarifying the signal in the frequency domains.

        Optimization of submerged culture conditions involving a developed fine powder solid seed for exopolysaccharide production by the medicinal mushroom Ganoderma lucidum

        To facilitate Ganoderma lucidum submergedcultivation and achieve high productivity, four fine powdersolid substrates incorporated with different nitrogen-richsupplements were utilized to grow the fungus and as solidseed for its submerged culture. Of the four solid seeds, thesoybean meal solid seed gave the highest biomass (10.73 g/L) and exopolysaccharide (EPS) (1.22 g/L), higher thanthose (8.36 g/L biomass and 0.44 g/L EPS) obtained withmycelial liquid seed. The optimal level of soybean mealsupplementation was 20% (w/w) for production of the solidseed. Following single factor experiments, levels of threeselected process variables were optimized as: the moisturecontent of solid seed, 70%; inoculum size, 0.8 g/flask; androtary speed, 160 rpm. These conditions were validatedexperimentally with improved EPS yield of 1.33 g/L. Thedeveloped solid seed can be conveniently used for G. lucidumsubmerged culture with improved EPS productivity.
