        Effect of Non-animal-Derived Nitrogen Sources on the Production of Hyaluronic Acid by Streptococcus sp. KL0188

        Hyaluronic acid (HA) is a linear high-molecular-weight polysaccharide with useful biomedical applications. Streptococcus zooepidemicus, a typical HA-producing bacterium, requires an animal-derived nitrogen source such as tryptone, peptone or sheep blood as a nutrient. Sixteen non-animal-derived (NAD) nitrogen sources were tested as a replacement for the expensive animal-derived nitrogen sources, which may have safety issues. Among the sixteen tested NAD nitrogen sources, a yeast-derived nitrogen source (YE 0251) showed the highest HA productivity, which was equivalent to the control HA production medium containing tryptone in a 5-L jar and in 3,000-L industrial fermentations. In the 3,000-L fermentation, YE 0251 increased cell mass (dry cell weight) and HA production by 11% and 8%, respectively, compared with the control HA production medium. The fmal specific volumetric productivity (0.41 g/L h) was improved by about 70% after reducing the fermentation time from 20 h to 12 h, compared to the conventional production medium.

        To investigate the method of the simultaneous reduction of Soot and NOx, we developed a new conception for dual fuel stratified injection system in a Diesel Engine. Some parts of the injection system, ie. nozzle holder, delivery vale, were remodeled to inject diesel and methanol in order from one injector. The quantity of diesel and methanol was controled precisely by micrometers that were mounted at control lack of injection pump. The real injection quantity ratio of dual fuel was certificated by volumetric ratio at injection Quantity experiment. We could confirm the capability that soot and NOx simultaneously were reduced by diesel-methanol stratified injection from the results of combustion and exhaust characteristic.

Then this study will consider the relationship between cultural globalization and contemporary architecture, by studying on design processes of contemporary architects who attempt to construct cultural flow as an experience element.

      • Dimecron 중독증 환자 6예

        The authors experienced 6 cases of Dimecron intoxication, an organophosphate, that required mechanical ventilator treatment. Among 6 cases, 4 cases were intoxication due to suicidal attempt and 2 cases were due to accidental exposure. Among 4 cases of intoxication due to suicidal attempt, only one patient recovered even after aggressive treatments using gastric lavage, mechanical ventilation, atropine and PAM(pralidoxime). Two cases of intoxications due to accidental exposure, 1 case needed mechanical ventilation. Both cases were recovered without any sequele, however. Organophosphate, especially Dimecron, showed high rates of resfiratory muscle paralysis and mechanical ventilator support. The mortality rate of organophosphate agents can be decreased by education, wearing protectors, and aggressive mechanical ventilator treatment when respiratory failure develops.

      • 상황버섯(Phellinus linteus)으로부터 Ergosterol의 분리

        상황버섯을 80% MeOH 용액으로 추출하고, 추출물을 EtOAc, n-BuOH 및 물로 분배, 추출하였다. 이 중 EtOAc 분획을 silica gel 및 octadecylsilica gel(ODS) column chromatography로 정제하여 1종의 sterol 화합물을 분리하였다. 화합물의 화학구조는 NMR, MS 및 IR 등의 스펙트럼 데이터를 해석하여, ergosta-7,22E-diene-3β,5α,6β-triol(cerevisterol, 1)로 동정하였다. 이 화합물은 상황버섯에서 처음 분리 보고 되었다. Phellinus linteus was extracted with 80% aqueous MeOH, and the concentrated extract was partitioned with EtOAc, n-BuOH and H_(2)O. The repeated silica gel and ODS column chromatographies of the EtOAc fraction led to the isolation of one sterol. From the result of spectral data including NMR, MS and IR, the chemical structure of the sterol was determined as ergosta-7,22E-diene-3β,5α,6β-triol (cerevisterol, 1). Compound 1 was isolated from the Phellinus linteus for the first time in this study.

      • 비병원성 야생효모 Rhodosporidium toruloides HGG35-2와 호중구 elastase 저해제 생성효모인 Wickerhamomyces anomalus HO9-2로부터 Killer toxin 생산

        항진균 바이오의약의 소재인 killer toxin을 비병원성 야생효모로부터 생산하기 위하여 25개 비병원성 색소 생성 효모들의 killer toxin 활성을 측정하여 우수균주로 Rhodosporidium toruloides HGG35-2와 Wickerhamomyces anomalus HO9-2을 선발하였다. 이들 killer toxin 생성균주들의 효모 감수성 균주들에 대하여 killer 활성을 측정한 결과 시판 포도주등의 알콜발효로 이용되고 있는 Saccharomyces cerevisiae (La Parisienne®)에 대하여 우수한 항진균활성을 보였다. Killer toxin production of non-pathogenic wild yeasts, Rhodosporidium toruloides HGG35-2 and Wickerhamomyces anomalus HO9-2 were investigated. Rhodosporidium toruloides HGG35-2 showed strong killer activity against pathogenic yeast, Candida guilliermondii AHP28-1 and commercial alcohol fermentation yeast, Saccharomyces cerevisiae(La Parisienne). Wickerhamomyces anomalus HO9-2 also showed killer activity against Rhodotorula nothofagi JSC10-1 and commercial alcohol fermentation yeast, Saccharomyces cerevisiae(La Parisienne®).

      • 비병원성 야생효모들로부터 생산된 항산화물질과 항통풍성 Xanthin 산화효소 저해물질의 부분정제 및 특성

        비병원성 Rhodosporidium paludigenum HHGG35-1와 Rhodosporidium diobovatum NMD18-1 균주들이 생산하는 항산화물질과 항통풍성 물질을 산업적으로 응용하기 위하여 먼저 이들 각각의 YPD 배양 상등액을 한외여과를 실시하여 3kDa 이하의 활성물질을 얻은 후 Sephadex G-50으로 여과하여 부분 정제된 항산화물질과 xanthin 산화효소 저해물질을 얻었다. 부분 정제한 항산화물질의 작용 최적 온도와 pH는 각각 30℃, pH 7.0 이었고, xanthin 산화효소 저해물질의 작용 최적 온도와 pH는 각각 15℃, pH 5.0 이었다. The antioxidants of Rhodosporidium paludigenum HHGG35-1 was partially purified by ultrafiltration and Sephadex G-50 filtration and investigated its optimal reaction condition. Optimal reaction temperature and pH of the partial purified antioxidants were 30℃ and pH 7.0. The xanthin oxidase inhibitor from Rhodosporidium diobovatum NMD18-1 was purified partially by enzyme treatment, ultrafiltration and Sephadex G-50 filtration. Optimal reaction temperature and pH of the partial purified xanthin oxidase inhibitor were 15℃ and pH 5.0. Some heavy metal ions and sugars did not affect inhibitory activity of the parial purified xanthin oxidase inhibitor.

      • 도시 쉼터 노숙자의 정신장애 유병율과 삶의 질

        연구목적 : 외환위기 이후에 급증한 노숙자 문제는 경제적인 상황의 호전에도 불구하고 만성화하는 경향을 보이고 있다. 쉼터의 노숙자를 대상으로 정신질환의 유병율과 삶의 질에 대한 조사를 시행하였다. 방 법 : 2000년 3월에 서울의 대표적인 쉼터에 거주하는 노숙자220명을 대상으로 인구학적 자료에 대한 설문조사를 한 뒤 Structured Clinical for DSM-Ⅳ(SCID-I)을 이용하여 AxisⅠ의 주요 정신질환을 진단하였다. 또 SmithKline Beecham Quality of Life Scale(SBQOL)를 사용하여 삶의 질정도를 평가하였다. 결 과 : DSM-Ⅳ AxisⅠ의 정신질환 중 기분장애, 정신병적 장애, 물질 사용 장애에 대한 진단 평가 결과 상기정신질환의 전체 평생 유병율은 73.6%, 현재 유병율은 59.1%이었다. 알코올 의존과 남용의 평생 유병율은 59.5%로 가장 높았고, 기분장애와 장애가 각각 39.5%, 3.6%이었다. 삶의 질은 노숙의 기간이나 실직 기간과 유의한 관계가 없었고, 자신이 지각한 건강상태, 자살과거력, 우울장애 유무와 통계적으로 유의하게 상관관계가 있었다(p<0.01). 결 론 : 노숙자에서 정신질환 중 알코올 장애의 유병율이 가장 높았으며, 우울장애가 있는 경우에 삶의 질이 저하되는 것으로 나타났다. Objectives : Homeless people surged after financial crisis tend to be chronic despite late economic recovery. So we tried to estimate prevalence of mental illness and quality of life of the homeless population living in shelter. Method : The study subjects were 220 homeless peoples who stayed at a shelter in Seoul around March 2000. Questionnaires on sociodemographic data were administered to the subjects, and then diagnoses of major DSM-Ⅳ Axis I mental disorders were made using Structured Clinical Interview for DSM-Ⅳ Axis I Disorders(SCID). And quality of life(QOL) was evaluated by SmithKline Beecham Quality of Life(SBQOL). Results : The lifetime prevalence of major DSM-Ⅳ mental disorders(mood disorders, psychotic disorder, and substance use disorder) of 220 homeless people was 73.6%, and current prevalence was 59.1%. The lifetime prevalence of alcohol dependence and abuse was highest, 59.5% and those of mood disorder and psychotic disorder were 39.5%, 3.6% respectively. Quality of life was not significantly correlated with duration of homelessness and unemploy-ment but marital status, self perceive health status, suicidal attempt history were significant factors. The quality of life in the people with mood disorders were estimated to be lower than those with any other diseases(p<0.01). Conclusion : Alcoholism was the most prevalent mental disorder in homeless people and mood disorder was negatively related to the quality of life scale.

      • TMPFS/Polysiloxane 혼합물의 표면특성

        We report on the surface proputies of the films composed of polysiloxane and trimethoxyperfluorosilane(TMPFS). In this study, the PFS was introduced in order to enhance soil-proof property of siloxane polymer. Surface properties of the mixture were investigated. Contact and sliding angle measurements were done in order to investigate the anti-soil, and soil release properties, respectively. The surface free energy of siloxane polymer was decreased remarkably by incorporation of small quantity of PFS with the increment of contact angle and decrease of sliding angle. The TMPFS/Polysiloxane mixture film may provide a great opportunity for the antisoil applications.

      • 야생효모, Wickerhamomyces anomalus HO9-2로부터 급성 폐손상 억제물질인 호중구 elastase 저해물질 생산 및 생산균주의 균학적 특성

        This study was performed to produce acute lung injury-preventing neutrophil elastase inhibitor from non-pathogenic wild yeast and further determine enzyme activity and physiological functionality of the potent wild yeast. Culture supernatants and cell-free extracts of one hundred twenty five non-pathogenic wild yeast strains were prepared and their neutrophil elastase inhibitory activities were measured. The neutrophil elastase inhibitory activity of the cell-free extract from Wickerhamomyces anomalus HO9-2 showed the highest inhibitory activity of 38.4% and its elastase inhibitor was maximally produced(IC50,3.2㎎) when Wickerhamomyces anomalus HO 9-2 was cultured in pH 7.0 of malt extract medium at 20℃ for 24h. Wickerhamomyces anomalus HO9-2 was oval shape and did not form ascospore and pseudomycelium. The HO9-2 strain grew well in YPD and YM media and was halophilic yeast which grew well in YPD medium containing 15% NaCl. Cell-free extract of the HO9-2 strain were showed high 17.2U of glucoamylase activity and anti-diabetic α-glucosidase inhibitory activity of 87.5%. 본 연구는 급성 폐손상 예방(또는 치료)의약 소재를 개발하기 위하여 비병원성 야생효모들의 elastase 저해활성을 측정하여 우수 균주를 선발하였다. 또한 선발균주의 elastase 저해제 생산 조건을 최적화 하였으며 선발균주의 미생물학적 특성 등을 조사하였다. 비병원성 야생효모인 Wickerhamomyces anomalus HO9-2의 무세포추출물이 38.4%의 호중구 elastase 저해활성을 보여 우수균주로 선발하였고 이 균을 pH 7.0 의 malt extract 배지에서 20℃로 24시간 배양하였을 때 최고의 elastase 저해활성(IC50, 3.2mg)을 보였다. 또한 이 균은 난형으로 포자와 의균사를 형성하지 않았고 YPD와 YM배지에서 생육이 양호하였고 NaCl을 15% 함유한 배지에서도 생육하는 호염성 효모이었다.
