      • 상처치유시 Antisense TGF-β1 Oligodeoxynucleotides가 Inducible Nitric Oxide Synthase발현 조절에 미치는 영향

        Wound healing in the skin may be complicated by both microbial invasion and inflammation. Following injury, platelet degranulation releases a number of chemotactic factors including transforming growth factor (TGF-fl) and platelet derived growth factor (PDGF). TGF-19 is a cytokines that modulates many cellular function and gene expression. However, it is not known whether TGF-P could regulate inducible nitric oxide synthase (iNOS) gene expression during wound healing. Here we report that down regulation of TGF-Q gene expression by antisense oligodeoxynucleotides increase iNOS during wound healing. Our reverse transcription- polymerase chain reaction (RT-PCR) result has shown that antisense TGF-fil oligodeoxynucleotides (ODN) targeting the TGF-J11 translation initiation region markedly reduced TGF-(il mRNA levels in wounded skin. Also, marked reduction in TGF-fl1 mRNA after antisense treatment invited that the increment of iNOS mRNA in wounded skin. Therefore, our irnmunohistochemical studies revealed a "pattern of iNOS product in wounded skin treated antisense TGF-fl1 ODN that was similar to the pattern of RNA synthesis detected by RT-PCR. In conclusion, our results indicated that the regulatory actions of TGF fl1 on iNOS. might be involved in the initiation and faciliates the resolution of inflammation following wounding.

      • 경포호의 어류상 및 어류군집 특성

        The characteristics of ichthyofauna and fish community in the Lagoon Gyeongpo, Korea were investigated from May to October 2005. The total number of fish caught during the period was representing 26 specis and 18 families. Most dominant species was T. hakonensis(50.65%) and subdominant specis was A. flavimaus(25.41%). K. punctatatus(11.07%), M. cephalus(6.53%), E. japonicus(1.46%), C. castaneus(1.01%) were also numerous. The total biomass of collected fish showed 147.00kg, 135.83kg, 85.60kg,23.76kg, repectively. A pattern of the fish community according to seasonal veriations appear to be related tp spawning periods of species. Comparison of the ichthyofauna in present study with the references from early years suggests that there has been a gradual decrease, in composition, of primary freshwater specis accompanying with the increase of peripheral freshwater and seawater specis.

        蓄血證과 蓄水證의 病理에 關한 文獻的 考察

        연구배경 : 축혈증과 축수증은 태양표증이 풀리지 않아 邪熱이 안으로 들어가 형성된 양대병증으로 이들의 부위 및 병리에 대한 설명이 명확치 못한 점이 많다. 이에 축혈증과 축수증의 본래 의미를 검토하고 그것의 병리에 대해서 부위, 증상, 개체별 차이 및 측수ㆍ축혈증의 실질에 대해서 연구하고자 함. 연구방법 : 축혈증과 축수증에 관련된 조문 및 유사증에 관한 조문들을 가려서 역대 주가들의 주를 참고로 연구하였다. 축수ㆍ축혈의 실질에 관해서는 처방을 중심으로 사용된 약재들의 현대적 약리작용을 통해 연구하였다. 결과 : 축혈증은 하초소복부의 혈액순환계통의 질환과 관련된 것으로 나타났으며, 축수증은 방광자체의 문제이기 보다는 기화분리로 인한 수액대사 장애와 관련된 병증으로 볼 수 있다. Through the philological study on syndrome of stagnated blood & syndrome of stagnated water, the results are obtained as following; 1. It is resonable to understand that the location of Syndrome of Stagnated Blood(蓄血證) is the lower abdomen and Syndrome of Stagnated Waste (蓄水證) is not exactly the disorder of Gall Bladder itself, but the disturbance of waterbolism derived from the disturbance of Qi (氣) transformation. 2. Syndrome of Stagnated Blood is that, being uncured Taiyang Channel Syndrom (太陽病), pathogen enters inside and pathogenic heat combines with blood at the lower abdomen; its main signs are spasmatic distention or muscular rigidity of the lower abdomen, mental derangement and normal urination. 3. Syndrome of Stagnated Water is that, being uncured Taiyang Channel Syndrom, pathogenic heat enters inside and combines with water and leads to disturbance of waterbolism; its main signs are thirsty, dysphoria, and difficulty in urination. 4. In the view of Western Medicine, Syndrome of Stagnated Blood is related with the disease of the blood circulation-especially in microcirculation- of the lower abdomen; Syndrome of Stagnated Water is similar to the disease which is related with the abnomal state of osmotic density derived from the unblance of electrolytes. 5. Lee Je-Ma regared the Syndrome of Stagnated Blood as SinSooYeolPyoYeolByung(腎受熱表熱病) of ShaoYin-Ren (少陰人), so explained it as the disease derived from the lower abdominal accumulation of Yang-Qi (陽氣) which coudn't have ascended; and regarded the Syndrome of Stagnated Water as PiSooHanPyoHanByung (脾受寒表寒病) of ShaoYang-Ren (少陽人), so explained it as the disease derived from the accumulation of internal heat because of discending disturbance of Yin-Qi (陰氣).

      • 독성물질의 세포사 기전 및 세포사 유발물질의 검색법 개발에 관한 연구(Ⅰ) : 독성물질로 인한 파킨슨병 모델에서의 세포사 기전 연구 Study on the cell-death mechanisms of toxin-induced parkinsonism

        MPTP 독성물질이 도파민성 신경세포에 선택적으로 작용하여 산화성 손상에 의한 신경세포사를 일으키는 것을 이용하여 파킨슨병의 동물모델을 만들고, 이를 통해서 아폼토시스를 비롯한 포사의 기전에 대한 연구 및 너코틴의 신경세포 보호효과 여부를 판정하는 실험을 병행하고자 하였다. 파킨슨꾐의 동물모델을 MPTf 독성 물질을 이용하여 확립하였으며, MPTP(30mgag, i.p.)를 투여한 후 1, 2,3, 4, 5일째 흑질 조직을 채춰하여 tarm로 박걸하여 tyrosine hydroxylase 면역조직화학염색을 수행하여 cell countif우한 결과, control은 57.635ce11s, 1일째 친.OfDells,2일째 57.9±6cells,3일릴 없.3±죠ells, 4일째 49.0츠3cells, 5일째 39.4±Scells료 4, 3일째 뚜렷한 신경세포 수의 감소를 보였다. 신경세포사 기전 규명을 위한 아폼토시스 분걱에서는 벼PTP 투여 후 1, 2, 3, 4, 5일째 조직을 채취하여 Hoechst staining, TUNEL staining을 수곡하였는데 양성 반응을 보인 신경세포는 관찰되지 않아. 아폼토시스로 인한 세포사가 관찰되지 않았다. bIPTP 파킨슨병 동물모델에서 nicotine 보호효과 탐색에 관한 실험은 nicat푸e 0.2mgAg을 5일 퐁안 투여 후 리『fP(30mgag)를 CS7Bt/6 마은스에 복강 내주사로 nicotine과 병용 투여한 후 1, 2, 3, 4, 5일째 뇌를 적출하땄다. 신경세포사가 뚜렷이 관찰되기 시작하는 4, 5일째의 신경세포 수의 감소 정도를 20. 30% 정도 약화시키는 경향을 보였으나, nicotine 보호효과에 대한 추가 실헝이 현재 수행 중에 있다. The cause of Parkinson's disease (PD) is largely unknown. However, free radical toxicit? may plaf a role ip. the degeneration of substantia nigra, which is the Hajorfocus of pathological damages in PD. Recently, a neuroprotective effect of nicotine in PD has been suggested. Therefore, the mechanism of neurodegenerafion and protective potential o( nicotine in PD were investigated in the experimental modeB of Pll using a neurotoxin, C57BL/6mice were administered with 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP, 30 mg/kg,j.p.). The degree of neurodegenerafion was determined by immunohistochemical stainiHB oftyrosine hydroxylase (TH). TH-positive cells on nigral sections were found 56.0 ±4, 57.9 ±6,52.315ce11s, 49.0±3cells, and 39,4±Scells at days 1, 2, 3, 4, 5, respectively (controls : 57.6±Scells). Hoechst and TUNEL staining showed no evidence of apoptosis. The exandnation on themice co-adrunistered with nicotine(0.2mgAg) and MPTP(30mgag) revealed a tendency ofnicotine protective effects. At days 4 and 5, the degree of TH-positive cells was decreased by20-30%, In corclusiffn, the role of apoptosis was not evidenced in this MPTP modeB of PB.The possible proteccon by nicotine should be elucidated with further studies.

      • Rubber Forming에 의한 알루미늄 판재성형의 치수오차에 관한 연구

        The aluminum alloy plate is used for appurtenance of a airplane. The aluminum plane appurtenance is made by the processes : cutting, thermal treatments, rubber forming, dimension modification. The dimensional modification is needed because the aluminum plate appurtenance made with some dimensional error during the rubber forming. The research of the prediction of the deformation has been required in order to reduce the dimension error and extra process. In this study, rubber forming simulation was performed by PAM-STAMP. Input variables, the properties of material, and contact condition, have used general values. The displacement of any node was predicted by the output file, i.e. node-time history. The results from PAM-STAMP with changing the input variables were compared to the actual deformation.

      • 單크론 抗體를 利用한 結核菌 베타抗原의 精製와 結核性 髓膜炎의 診斷的 應用에 關한 硏究

        5F3F2 hyridoma cells produce monoclonal antibody which is reactive with a single epitope on the molecule of beta antigen and shows a specificity restricted to Mycobacterium tuberculosis complex. Ascitic fluids were produced by an intraperitoneal injection of 5.0 x 10^6 hybridoma cells into Balb/c mice previously sensitized with pristane. The reactive pattern of ascitic fluid with reference beta antigen was analyzed by Western blot. Purification of 5F3F2 monoclonal antibody from ascitic fluid was performed by a single step method using hydroxylapatite chromatography. Beta antigen from the cell extract of Mycobacterium tuberculosis was purified by affinity chromatography with 5F3F2 monoclonal antibody and checked its molecular characterization by SDSPAGE. Finally, affinity purified beta antigen was applied for its potential diagnostic utility in dection of IgG antibody in the cerebrospinal fluids of 12 patients with tuberculous meningitis and 24 non-tuberculous patients by ELISA at the CSF dilution of 1 : 5. The results were summarized as follows: 1. Ascitic 5F3F2 monoclonal antibody was reacted with a major band of 83-kDa and a minor band of l l0-kDa molecule of reference beta antigen on Western blot. 2. 5F3F2 monoclonal antibody was purified directly from ascitic fluid by a single step purification of hydroxylapatite chromatography. 3. Affinity purified beta antigen showed two distinct bands of 83-kDa and 52-kDa molecules on SDS-PAGE. 4. The mean ELISA values of IgG antibody activities to affinity purified beta antigen in CSF of 12 patients with tuberculous meningitis and 24 controls were 0.866±0.607 and 0.058±0.040, respectively. The specificity and sensitivity were 96.5% and 93.1%. These results suggest that the major molecule of beta antigen is 83-kDa and the affinity purified beta antigen is useful for the diagnosis of tuberculous meningitis.

        This paper discusses System Identification using observer in time domain. It is carried out for a experimental Three story structure With Active Mass Damper(AMD). Applying gradually stable method. Observer Kalman Filter Identification(OKID) determines Maskov paramaters. System eigen value is presumed utilizing this variables. Simulation system is composed of this fluents's compounding. It is verticified to compare numerical simulation results with experimental measurements.
