        사상체질별 약물의 항당뇨효능에 관한 실험적 연구

        The purpose of this research was to investigate inhibitory activity of Sasang medicines on diabetes. Reduce 36 kind of medicines to powder, abstract add 100 times methanol. Examine inhibitory activity on α-amylase and α-glucosidase in vitro. Thirty six oriental herbs were screened for inhibitory activities against carbohydrate digesting enzymes in vitro. Methanol extract of Pleuropterus mulitiflorus Turez, Ephedra sinica, Moutan cortex Radicis, Magnolia obovata, Alpinia officinarum, Amomi Semen, Rubus coreanus, and Cinnamomi cortex inhibited yeast α-glucosidase activity and extract of Moutan cortex Radicis and Rubus coreanus inhibited porcine α-amylase activity by more 50%. Rubus coreanus showed the strongest inhibition against both α-glucosidase and α-amylase. Therefore, Rubus coreanus was the most effective medicice of 36 kind of medicines.

        風의 韓醫哲學的 意味

        In the ancient time wind is considered as the life·soul and the human’s breath which represents essence of universal creation and the absolute power and also the messenger of the god or gods state. Greek’s ”pneuma”, India’s ”Brahman” and the Old Testament’s ”ruach” are used to signify the wind. Also Wind(風) in traditional culture, it is related to 玄妙之道 of 花郞, the flower of youth in Shilla dynasty and sexual intercourse in shaman ’ s dream which can be thought as Freud’s libido. In this aspect we can see the connection between the wind and the libido which can be meaning of sexual desire. Ancient Chinese wrote word ’風’ as phoenix, the gods bird, the phoenix in inscriptions on bones and tortoise carapace(甲骨文) because one can feel the wind but can’t see it. The word Ki(氣) origins from 風 therefore 風 is 氣’s fundamental notion. The wind can be understood by ki which travels around the world to create all nature. And the Wind is associated with 風化, 玄鳥, 八僧舞, which are related with reproduction. In the book of change (周易) the 震 巽卦; 雷風 come under wind which means the function of ki and also menas the 精(essence of life) of 恒久(eternity) means the reprodution ; that performs succession of life. In the Oriental Medicine 風 is a phenomenon that appears by movement of Ki by 相火(Ministerial fire). 相火 is core of the succession of life which means preservation of descendants; therefore 風 has very similar concept with sexual desire. 風 is the beginning ki of universe and in human body aspect 風 belong to the Liver Meridian. If 風 makes movement then the Pericardium Meridian of Hand Kwolum responds and the genital organs which belongs to Liver Meridian of Leg Kwolum reproductive function by contraction and expansion. Generally 風 understood as movement and origination and this is recognized as meaning of 氣. Therefore as studied above the present writer believe 風 participates closely to reproductive function.

        滋陰健脾湯加枳殼·天麻가 腦血流力學 變動에 미치는 작용기전

        Jaeumgenby-tang(JGT) have been used in oriental medicine for many centuries as a therapeutic agent of vertigo cause by deficiency of qi(氣) and blood(血). Effect of Aurantii Fructus(AF) take off the phlegm by promoting the circulation of qi, Gastrodae Rhizoma(GR) has effects treating for headache, vertigo by calming the liver and suppressing hyperactivity of the liver-yang(陽). And, We reported that JGT adding AF · GR extract(JGTAG) was significantly increased regional cerebral blood flow(rCBF) by dilating pial arterial diameter(PAD). Therefor we designed to investigate whether JGTAG is mediated by adrenergic β-receptor, cyclooxygenase or guanylate cyclase in normal rats. The changes of rCBF and mean arterial blood pressure(MABP) were determinated by laser-doppler flowmetry(LDF), and the change of PAD was determinated by video microscope and width analyzer. The results were as follows in normal rats ; Pretreatment with propranolol(1mg/kg, i.v.) was significantly inhibited JGTAG induced increase of rCBF, PAD and MABP, and pretreatment with indomethacin(1mg/kg, i.v.) was significantly inhibited too. But pretreatment with methylene blue(10㎍/kg, i.v.) were accelerated JGTAG induced increase of rCBF and MABP, but pretreatment with methylene blue was inhibited JGTAG induced increase of PAD. This results suggest that the mechanism of JGTAG is mediated by adrenergic β-receptor and cyclooxygenase.

        쪽파 에탄올 추출물이 果糖 誘導 고혈압 백서의 腎臟 기능에 미치는 영향

        The present study was aimed to investigate whether ethanol-extract of Allium wageki has an ameliorative effect on the renal function in high fructose-diet induced hypertensive rats. The urine osmolality (Uosmol) was decreased in rats with high fructose-diet (60%) during the whole experiment period without change of the urine volume (UV). The urinary excretion of sodium (UNaV) and chloride (UCIV) were decrease significantly in rats with fructose-induced hypertensive rats, whereas urinary excretion of potassium (UKV) was increased. The creatinine clearance (Ccr) and solute-free water reabsorption were also decreased by treatment of fructose-rich diet. Among these renal functional parameters, Ccr was partially restored by the administration of ethanol-extract of Allium wageki. The Uosmol was also partially restored by the administration ethanol-extract of Allium wageki at the end of the experimental period. Taken together, ethanol-extract of Allium wageki has the ameliorative effect on glomerular filtration rate in rats with high fructose-diet induced hypertension.

        加味生料四物湯의 抗炎症효과와 止痒膏의 아토피피부염 손상 및 止痒 효과에 미치는 영향

        This research was performed to examine an anti-inflammatory effects of Gamisangryosamul-Tang(GSS) and anti-pruritus effects of Ziyang-Go(Salve). This study was processed by three experiments; Experiment 1: Inhibitory activity of GSS extract on the degranulation of mast cell and histamine release in plasma induced by compound 48/80 i.p. injection after the pretreatment of GSS extract i.p. injection in Sprague-Dawley rats. Experiment 2: Anti-inflammatory effect of GSS extract on macropharge raw 264.7 cells treated by LPS 250 ppm (before 2 hours). Experiment 3: Measurement of passive cutaneous anaphylaxis and atopic dermatitis using NC/Nga mice. GSS extract inhibited histamine release by 70% compared to compound 48/80 treated control group and histologically significantly reduced (P<0.01) the degranulation of mast cell in SD rats. In GSS extract treated group, the expression of TNF- α in macropharge cell showed the remarkable inhibitory effect about 62% (P<0.01) compared to LPS treated control group. The expression of IL-6 appeared more effective by 46% than the LPS treated control group and by 6% compared to hydrocortison treated group. Comparing with steroid (0.05% prednisolon) ointment, Ziyan-Go treated group showed the significant(30%) recovery on skin response index in atopic dermatis like anaphylaxis mice(NC/Nga). Finally, in scratching behavioral tests of NC/Nga mice for three weeks, Ziyang-Go treated group significantly (P<0.05) suppressed the pruritus on the face, neck, ears and dorsal skin than inbred NC/Nga mice. However, the change of IgE and IFN-7 from the spleen cell of NC/Nga mice was not significantly different between the oral intake of GSS extract group and of saline intaked control group. Summary and Conclusion: This study demons trates that Ziyang-go have the equal anti-pruritus effect to steroid ointment and GSS extract have the notable immunologic activity on inflammatory in vivo and in vitro model. Advanced experiment of this study will be required for more reliable information about the correlation between the lymphokine (i.e. IgE) and the anti-allergic effects of GSS.

        六味地黃湯의 항산화작용에 관한 연구

        In order to examine the antioxidant actions of YMJHT, the study was done through measurement of parameters such as LPO(lipidperoxidation), GSH(glutathione), SOD(superoxidation dismutase), catalase, GOT, GPT, ALP, the results were obtained as follows: For the weight changes, in the testis the group given YMJHT showed significant increase compared to the control group. In the left cerebrum, the group given YMJHT showed significant increase compared to the control group on the activities of SOD, catalase. In the right cerebrum, the group given YMJHT showed significant decrease on the content of LPO and showed significant increase on the activity of catalase. In the cerebellum, the group given YMJHT showed significant decrease on the content of LPO and showed significant increase on the activities of SOD, catalase. In the liver, the group given YMJHT showed significant decrease on the content of GSH and showed significant increase on the activity of SOD. In the kidney, the group given YMJHT showed significant decrease on the contents of GSH, GSH and showed significant increase on the activities of SOD, catalase. In the testis, the group given YMJHT showed significant decrease on the contents of LPO, GSH and showed significant increase on the activity of SOD. From above results, the antioxidant action of YMJHT is effective. And it is expected to be necessary to the study of the mechanism in the antioxidant of YMJHT.

        瀉白散 추출물에 의한 인체 폐암세포의 Apoptosis 유도 기전에 관한 연구

        We investigated the effects of Sabaek-san (SBS) water extract on the growth of human lung carcinoma A549 cells. Upon treatment with SBS extract, a concentration-dependent inhibition of cell viability was observed and cells developed many of the hallmark features of apoptosis, including condensation of chromatin. Flow cytometry analysis confirmed that SBS treatment increased populations of apoptotic-sub Gl phase. In addition, proteolytic cleavages of poly(ADP-ribose) polymerase and β-catenin protein were observed after treatment of SBS extract. These apoptotic effects of SBS in A549 cells were associated with marked inhibition of Bcl-2 and Bcl-xL mRNA in a dose-dependent manner, however the levels of Bax expression were not affected. SBS treatment also induced a proteolytic activation of caspase-3, which is believed to play a central role in the apoptotic signaling pathway. The previous and present results indicated that SBS-induced inhibition of lung cancer cell proliferation is associated with the blockage of G1/S progression and the induction of apoptosis.

        척수운동신경세포에 있어서 Glucose Oxidase의 독성에 대한 동과의 영향

        It has been suggested that oxidative stress of reactive oxygen species(ROS) may play an important role in the pathogenesis of neurological disorder. The aim of this study was to elucidate the oxidative stress of glucose oxidase(GO) in the cultured mouse spinal motor neurons and the preventing effect of Benincasae Semen(BS) on ROS-induced neurotoxicity. Cytotoxic effect of GO and protective effect of BS were performed by MTT assay. 30mU/ml GO decreased cell viability in dose-and time-dependent mannner, and BS diminished GO-induced neurotoxicity in these cultures. From above the results, ROS such as GO has toxic effect, and herb extract of BS is very effective against GO-induced neurotoxicity in cultured spinal motor neurons of neonatal mouse.

        산사 Butanol 분획이 PGF2α-유도 혈관평활근수축의 억제에 미치는 신호전달 연구

        The mary mechanism of smooth muscle contraction is phosphorylation of the 20 kDa myosin light chains(LC20) by a myosin light. chain kinase(MLCK). Relaxation, then, is generally the result of dephosphorylation of LC20 by myosin phosphatase(MP). Changes in MP activity is one of the important mechanisms in the regulation of Ca2^+-sensitivity. Inhibition of MP activity is linked to an increase in phosphorylated myosin light chain(MLC) without an increase in [Ca^(2+)]i-levels. It is now generally accepted that Rho-kinase phosphorylates 130 kDa regulatory and myosin binding subunits(M130, MYPT) of MP, which results in an inhibition of MP activity. In addition Rho-kinase can also directly phosphorylate MLC. In the present study, LC20 phosphorylation and MP subunits translocation to the cell membrane were investigated in freshly isolated ferret portal vein smooth muscle single cells treated with PGF2α. We also examined the effect of Y27632(10-5mol/L), Rho-kinase inhibitor, in the MP subunits localization to compare with butanol fraction of Fructus Crataegi in its effect. Butanol fraction of Fructus Crataegi(BFFC; 1 mg/ml) was more effective in PGF2 α induced contraction than those of phenylephrine in its vasodilation effect. It significantly(P<0.05) dephosphorylated the LC20 at time indicated. In addition, the dissociation of subunits are inhibited by BFCF treatment. The results indicate that, in the smooth muscle cells, the relaxation effect of BFFC is associated with increase of MP activity based on inhibition of dissociation of the catalytic and targeting subunits of the phosphatase, and thus decrease the sensitivity of LC20 phosphorylation for Ca^(2+)

        활성산소로 손상된 대뇌신경세포에 대한 천오두의 영향

        Neurotoxicity of reactive oxygen species(ROS) and neuroprotective effect of Aconiti Radix(AR) against ROS-induced cytotoxicity were determined on cultured mouse cerebral neurons by MTT assay after cerebral neurons were cultured for 5 hours in various concentrations of GO. GO was toxic in a dose-dependent manner on cultured cerebral neurons after cerebral neurons were incubated for 5 hours in media containing 5~40mU/ml GO. While, cultures were pretreated with 180μg/ml AR for 2 hours increased remarkably cell viability. From these results, it is suggested that GO has toxic effect on cultured mouse cerebral neurons by the decrease of cell viability. And also, herb extract such as AKR is very effective in the protection of neurotoxicity induced by GO.
