        인간포배기 배아의 효과적인 유리화 동결법의 개발을 위한 연구

        Objective: The purpose of this study was to evaluate the survival rate of vitrified blastocyst according to the freezing vessels, equilibration time in cryoprotectant and artificial dehydration method. Methods: Human blastocysts were vitrified after loading onto the plastic straw, open-pulled straw (OPS), electron microscopy grid (EM grid) for 1.5 min or 3 min. They also were directly plunged into LN2 within 30sec. For artificial shrinkage of blastocysts, 36 gauge fine needle was pushed at the cellular junction of the trophectoderm into the blstocoele cavity until it shrank without damage of inner cell mass. Results: The survival rate of vitrified blastocysts on plastic straw, OPS, EM grid as freezing vessels were 26.7, 13.0 and 60.5%, respectively. The survival rate of EM grid was significantly higher than that of plastic straw and OPS (p<0.05). For 1.5 min equilibrium, the survival rates of early blastocyst (EB), middle blastocyst (MB) and late blastocyst (LB) were 64.4, 81.0, and 20.0% respectively. For 3 min equilibrium, the survival rates of EB, MB, and LB were 69.9, 50.0 and 57.5% respectively. The survival rates of EB and MB were significantly higher than that of LB in 1.5 min equilibrium group (p<0.05), however, the significance was not observed in 3 min equilibrium groups. In cytoplasmic shrinkage before vitrification, the survival rates of EB, MB and LB were 92.9, 100 and 75.9% respectively. The survival rate of MB was significantly higher than that of LB (p<0.05). The survival rates of vitrified blastocysts by artificial dehydration and slow-frozen blastocysts were not significantly different as 88.9 and 66.7%, respectively. Conclusion: This study showed that the vitrification of human blastocysts using EM grid and artificial dehydration is an effective method. Therefore, these methods would be an useful techniques for blastocyst cryopreservation.

        Effect of spark plasma sintering conditions on the thermoelectric properties of (Bi<sub>0.25</sub>Sb<sub>0.75</sub>)<sub>2</sub>Te<sub>3</sub> alloys

        <P><B>Abstract</B></P> <P>As a field-assisted technique, spark plasma sintering (SPS) enables densification of specimens in a very short period of time compared to other sintering techniques. For high performance thermoelectric material synthesis, SPS is widely used to fabricate nanograin-structured thermoelectric materials by rapidly densifying the nanopowders suppressing grain growth. However, the microstructural evolution behavior of thermoelectric materials by SPS, another important process during sintering, has been rarely studied. Here, we explore SPS as a tool to control the microstructure by long-time SPS. Using p-type (Bi<SUB>0.25</SUB>Sb<SUB>0.75</SUB>)<SUB>2</SUB>Te<SUB>3</SUB> thermoelectric materials as a model system, we systematically vary SPS temperature and time to understand the correlations between SPS conditions, microstructural evolution, and the thermoelectric properties. Our results show that the relatively low eutectic temperature (∼420 °C) and the existence of volatile tellurium (Te) are critical factors to determine both microstructure and thermoelectric property. In the liquid-phase sintering regime, rapid evaporation of Te leads to a strong dependence of thermoelectric property on SPS time. On the other hand, in the solid-phase sintering regime, there is a weak dependence on SPS time. The optimum thermoelectric figure-of-merit (Z) of 2.93 × 10<SUP>−3</SUP>/K is achieved by SPS at 500 °C for 30 min.         고압멸균 처리시간에 따른 닭 체내 조류인플루엔자와 뉴캐슬병 바이러스의 멸균효과

        Avian influenza (AI) and Newcastle disease (ND) distress a variety of avian species, especially domestic poultry. Severe syndromes are caused by highly virulent specific virus strains termed highly pathogenic AI and velogenic ND viruses, which are potential agrobioterrorism agents. This outbreak emphasizes the need for continuing cooperation between public health and veterinary medical communities in controlling AI and ND when it has a zoonotic potential. Up to date, the stamping out and burying system were applied for controlling methods against these highly infectious diseases in the ordinary way, however these methods had many environmental problems, including leachat and effluvium. Thus, we assessed that sterilization effect of AI and ND virus dependent on several treatment conditions, such as autoclaving time and cutting types of chicken. As a result, we found that the cutting type of chicken meat revealed a reduced HA titer (20) against both of AI and ND virus after 10 min of autoclaving, while whole chicken showed same titer after 30 and 60 min. Therefore, we propose that the conditions of treatment on infected chicken should be developed for convenient, affordable, and effective prevention methods against for AI and ND.

        친환경 세척제의 처리 방법을 달리하여 착즙한 당근 주스의 미생물 안전성 및 품질

        본 연구에서는 당근의 세척에 사용되는 친환경 세척제의 초기 미생물 제어 효과를 향상시키기 위해 탄산수소나트륨과 구연산의 단독 및 병합 처리 조건을 설정하였으며 저장 중 당근 주스의 미생물 수 및 품질 특성 변화를 분석하였다. 친환경 세척제의 단독 처리 방법으로는 0.5, 1, 2% 탄산나트륨수와 0.2, 0.5, 1% 구연산수를 적용하였고, 살균 효과를 비교하기 위해 무처리구, 수돗물 처리구, 50 ppm 차아염소산나트륨 처리구를 사용하였다. 당근의 세척을 위한 병합처리 방법으로는 단독 처리에서 우수한 살균 효과를 나타낸 1% 탄산수소나트륨수와 1% 구연산수를 이용하였고, 1% 탄산수소나트륨수 처리 후 1% 구연산수로 처리하는 단계적 병합 처리구가 가장 우수한 살균 효과를 나타내었다. 1% 탄산수소나트륨수와 1% 구연산수의 단계적 병합 처리구는 무처리구에 비해 일반 세균수 1.87 log CFU/mL, 대장균군 1.56 log CFU/mL를 감소시켰고, 50 ppm 차아염소산수에 비해서도 일반 세균수 1.07 log CFU/mL, 대장균군 0.22 log CFU/mL의 미생물 감균효과를 나타냈다. 1% 탄산수소나트륨수 처리 후 1% 구연산수의 단계적 병합 처리구를 4°C에서 7일 동안 저장한 결과 수돗물, 50 ppm 차아염소산수나 혼합 병합 처리구, 1% 구연산수 처리 후 1% 탄산수소나트륨 처리구에 비해 미생물 제어 효과가 확실하게 나타났지만, pH, 당도, 산도, 색도에는 유의적인 차이가 나타나지 않았다. 본 연구 결과 1% 탄산수소나트륨과 1% 구연산의 단계적 병합 처리는 당근 세척 시 초기 미생물 제어와 품질 유지에 있어 효과적인 살균처리 방법으로 비가열 당근 주스의 유통기간 동안 미생물 안전기준에 부합하며 주스의 품질을 유지할 수 있는 바람직한 세척방법으로 판단된다. The aim of this study was to evaluate the microbial inhibitory activity and physicochemical quality of fresh carrot juice prepared with different environmentally-friendly washing methods during low temperature storage. Individual and combined treatments with sodium bicarbonate (baking soda, NaHCO₃) and citric acid were applied to carrots for 10 min. Tap water and 50 ppm of sodium hypochlorite (NaOCl) were used as the control. Combined treatment of 1% NaHCO₃ and 1% citric acid significantly reduced total aerobic counts and coliforms. In addition, combined treatment of 1% NaHCO₃ and 1% citric acid inhibited microbial growth for 7 days at 4°C and 10°C in a shelf-life study. There were no significant differences among the sanitizers in terms of °Brix, acidity, pH, and color. Changes in physicochemical quality were not significantly different by sanitizer but were affected by storage temperature. These results indicate that washing with combined treatment of 1% NaHCO₃ and 1% citric acid is an effective method to inhibit the microbial population and maintain physicochemical quality. Therefore, combined treatment of 1% NaHCO₃ and 1% citric acid can be effectively used to sanitize and prepare carrot juice without affecting other properties.

      • 일개 대학병원에 내원한 원발성 폐암 환자의 인구학적 및 병리학적 특성

        Background : Recently, the lung cancer mortality rank first in Korea. The prognosis of lung cancer is not good enough with current treatments and multimodality treatments are required. So we are interested in the characteristics of lung cancer patients for better treatment. Methods : We searched lung cancer patients since 1994. Then we collected the data of primary lung cancer patients about the demographic features, histologic types and diagnotic methods. We divided the total period into 2 haves and specific analysis was tried. Results : 1) Total 850 patients were included. 633 (74.5%) were male and 217 (25.5%) were female. 2) The mean age was 65 ± 11 years old. and the peak age was 7th decades. 3) Squamous cell carcinoma(334/850, 39.3%) was slightly more prevalent than adenocarcinoma(302/850, 35.5%). Conclusion : From these results, we expected that older persons will be diagnosed to have lung cancer and adenocarcinoma will be more dominant histologic type with time.

      • GPS를 이용한 2차로 고속도로에서의 추월시거 산정방법에 관한 연구

        This study applied GPS(global positioning system) into the field of traffic and transportation, measured overtaking mechanism at two lane highway. After the analysis of the collected data from the measurement, it was constructed passing sight model which is based on AASHTO(american association of state highway and transportation officials) model and; as the result, passing sight distance was computed. To accomplish this goal, speed and position information for leading and following vehicles were recorded under actual highway conditions. Each vehicle's distance, speed, and acceleration characteristics were calcuated using the passing algorithm equations proposes. The computed value of acceleration was 1.37m/sec2. This value was twice as large as the value calculated earlier(0.6m/sec2). This study proposed 326m as passing sight distance at maximum speed(80km/hr) in 88 Olympic highway.

        Relapsing polychondritis is an uncommon multisystemic disease which is characterized by recurrent episodes of inflammation and progressive destruction of cartilaginous tissue. The cause of relapsing polychondritis is unclear and suspected to be autoimmune pathophysiology. It involves cartilage of nose, vertebra, trachea, peripheral joint as well as structures of eye, heart, blood vessels or inner ear. Auricular chondritis is present in almost all patients, and multiple arthritis, nasal chondritis, saddle nose, eye involvement, laryngotracheal involvement and skin manifestations are also present. We report a case of relapsing polychondritis presented a arthritis with review of literatures.

      • 급성 복통을 주소로 진단된 자발성 양측 부신 출혈에 의한 부신피질부전

        Thomas Addison은 1855년 발표된 그의 논문에 일차성 부신피질부전에 의하여 발생한 근력 약화, 피로, 식욕부진, 복통, 체중감소, 색소침착 등에 대하여 처음으로 기술하였다[1,2]. 부신피질부전은 부신피질의 파괴에 의해 발생하는 당질 코르티코이드 결핍과 관련된 일차성과 뇌하수체에서 혈청 부신피질자극 호르몬(adrenocorticotropic hormone, ACTH)의 분비장애에 의해 발생하는 이차성으로 구분할 수 있다. 일차성과 이차성의 중요한 차이는 무기질 코르티코이드의 결핍이 일차성에는 있고 이차성에 는 없다는 점이다. 일차성 부신피질부전의 원인으로 이전에는 결핵에 의한 양측 부신 파괴가 가장 흔한 원인이었으나, 결핵 진단율 및 치료약제 보급률이 좋아진 20세기 후반부터는 감소 추세를 보이고 있으며, 선진국에서는 자가면역성 부신염에 의한 일차성 부신피질 부전이 80~90%로 가장 흔한 것으로 보고되었다[3]. 이외에 다른 감염성 질환, 전이성 암, 림프종, 부신출혈 또는 경색, 약물 등의 원인을 들 수 있다. 일차성 부신피질부전의 임상 증상과 징후로는 전신무력감, 식욕부진, 피로감, 체중감소, 과다한 색소침착, 오심, 복통, 기립성 저혈압, 저혈당, 전해질 장애 등이 올 수 있고, 심하면 탈수, 저혈압, 의식장애 및 쇼크가 동반될 수 있다. 일차성 부신피질부전은 본 증례와 같이 드물게 양측 부신 출혈성 경색으로 인해 발생되는 경우가 보고되었는데, 위험인자로는 항응고제 또는 헤파린 투여, 혈전성 질환, 항인지질 증후군과 같은 과응고 상태, 물리적 외상, 감염, 심한 스트레스와 관련되어 발생할 수 있으며 드물게 임신, 부신 종양, 쇼크 등이 원인이 되고 있다[4,5]. 그러나 본 증례와 같이 원인 없이 발생한 자발성 출혈에 의한 부신피질부전의 보고는 매우 드물다. 양측 부신 출혈의 임상 증상과 징후로는 저혈압, 쇼크, 복부, 등, 옆구리, 흉부의 통증, 발열, 의식저하, 복부 강직, 반발 압통 등이 있다[6]. 적절한 치료를 시행하지 않으면 쇼크가 진행하여 혼수와 사망에 이를 수 있어 정확하고 빠른 진단과 치료가 필요하다. 환자가 생존한다면 부신 기능은 드물게 수개월 후 정상으로 회복될 수 있다[7]. 저자들은 상복부 통증을 주소로 내원한 환자에서 매우 드물게 원인 없이 발생한 자발성 양측 부신 출혈에 의한 급성 부신피질부전 1예를 경험하였기에 보고하는 바이다.

        Forced suction thrombectomy in patients with acute ischemic stroke using the SOFIA Plus device

        Objective: Stent retrieval thrombectomy has recently been the standard treatment for acute ischemic stroke with large artery occlusion. However, the development of catheters for suction thrombectomy has recently led to results comparable to that of stent retrieval thrombectomy (SRT). This study aimed to analyze the safety and efficacy of forced suction thrombectomy (FST) using the SOFIA Plus (MicroVention Terumo, Tustin, CA, USA) device.Methods: We included patients with acute ischemic stroke who underwent FST using the SOFIA Plus device at our institution. Medical records and angiographic data were reviewed, and the results of this study were compared with those of other FST studies.Results: A total of 35 patients were included in this study. The occlusion sites were the internal carotid artery terminal (4), M1 segment (20), and posterior circulation (11). Of the 35 patients, FST was performed in only 21 (60%) patients, and the remaining 14 (40%) patients underwent SRT and FST. In all cases, the recanalization rate was 100%, and the average time from groin puncture to recanalization was 21±4.94 min. In particular, the average time required to reach the SOFIA Plus lesions from the groin puncture was 10.44±5.06 min and about 67% of the FST patients were recanalized at the first attempt. Three-months modified Rankin Scale (mRS) score of ≤2 was observed in 52% of the patients.Conclusions: Forced suction thrombectomy using the SOFIA Plus yielded a high recanalization rate within a shorter time. In particular, the recanalization rate was higher than that reported in previous studies using other types of suction devices.
