        Quantitative and Rapid Analysis of Transglutaminase Activity Using Protein Arrays in Mammalian Cells

        We developed a novel on-chip activity assay using protein arrays for quantitative and rapid analysis of transglutaminase activity in mammalian cells. Transglutaminases are a family of Ca2+-dependent enzymes involved in cell regulation as well as human diseases such as neurodegenerative disorders, inflammatory diseases and tumor progression. We fabricated the protein arrays by immobilizing N,N′-dimethylcasein (a substrate) on the amine surface of the arrays. We initiated transamidating reaction on the protein arrays and determined the transglutaminase activity by analyzing the fluorescence intensity of biotinylated casein. The on-chip transglutaminase activity assay was proved to be much more sensitive than the [3H]putrescineincorporation assay. We successfully applied the on-chip assay to a rapid and quantitative analysis of the transglutaminase activity in all-trans retinoic acid-treated NIH 3T3 and SH-SY5Y cells. In addition, the on-chip transglutaminase activity assay was sufficiently sensitive to determine the transglutaminase activity in eleven mammalian cell lines. Thus, this novel on-chip transglutaminase activity assay was confirmed to be a sensitive and high-throughput approach to investigating the roles of transglutaminase in cellular signaling, and, moreover, it is likely to have a strong potential for monitoring human diseases.

        삼차신경통 태음인 환자의 만성 설사 치험례

        Objectives This study was aimed to report a significant improvement of chronic diarrhea patient, diagnosed as 'Taeeum type liver heat-lung dryness pattern' based on Sasang medicine. Methods The patient had treatment with Sasang constitutional medicine. The treatment was executed three times everyday from August 27th to September 3rd and two times everyday from September 3rd to September 16th of 2021. Clinical improvement was evaluated with Bristol stool form scale, the degree of abdominal discomfort and nausea compared to the day of hospitalization and the number of defecations and nocturia per day. Results The symptoms of diarrhea, abdominal discomfort and nausea were improved. The number of night urination was decreased from 3-4 times to once a night. Conclusion After the treatment with Sasang constitutional medicine, diarrhea, abdominal discomfort, nausea and the general condition of the patient also improved.

        Functional and Immunofluorescence Evaluations of Vascular and Neural Integrities in Urinary Bladder of Streptozotocin-Induced Diabetic Mice

        Purpose: To assess functional and structural changes in vascular and neural structures associated with diabetic bladder dysfunction (DBD) in the bladders of streptozotocin (STZ)-induced diabetic mice. Methods: Eight-week-old C57BL/6 mice were injected with STZ at 50 mg/kg daily for 5 consecutive days. Catheters were inserted 12 weeks later, and 5 days after catheter placement bladder functions were assessed by conscious cystometry. Neurovascular and extracellular matrix marker changes in harvested urinary bladders were investigated by immunofluorescent staining. Body weights and fasting and postprandial blood glucose levels were measured 12 weeks after STZ injection. Results: STZ-induced diabetic mice had significantly lower body weights and significantly higher blood glucose levels. Assessment of bladder function in STZ-induced diabetic mice revealed a nearly 3-fold increase in bladder capacity and intercontractile interval compared to controls. However, basal pressure, maximal bladder pressure, and threshold pressure were not significantly different. Morphological and structural analysis showed that STZ-induced diabetic mice had significantly reduced microvascular density in lamina propria (33% of the nondiabetic control values), and severely decreased nerve contents in the detrusor region (42% of the nondiabetic control values). Conclusions: STZ-induced diabetic mice exhibit functional and structural derangements in urinary bladder. The present study provides a foundation and describes a useful means of evaluating the efficacies of therapeutic targets and exploring the detailed mechanism of DBD.

        방사선 피부염 치료법에 대한 문헌고찰

        Objective : Radiodermatitis is a common sequelae in 95% of patients receiving radiation therapy, which is important to be well managed as it can affect the patient's quality of life as well as the cancer treatment schedule. The aim of this study is to review and summarize the interventions available for the treatment of acute radiodermatitis, including traditional Korean medicine, and to propose treatment algorithms for clinicians. Methods : To collect studies about managements for radiodermatitis, domestic and foreign database were used such as Korean journal of traditional knowledge portal (KTKP), Korean studies information service system (KISS), national discovery or science leaders (NDSL), and oriental medicine advance searching integrated system (OASIS), Pubmed, Google scholar and Scopus. Results : Thirty-two studies were selected. There were nine studies on usual care and dressing, eleven studies on chemical agents, two studies on biological preparations, and ten studies on herbal medicines. Conclusion : Hygienic options and dressings have proved to be useful in the management of radiodermatitis. Chemical agents such as corticosteroid, statin, and topical antibiotic agent have proved to alleviate symptoms and severity, regenerate damaged skin, and prevent secondary infection. In biological preparations, EGF (epidermal growth factor) and GM-CSF (granulocyte-macrophage colony-stimulating factor) could be used to protect skin and prevent radiodermatitis. For herbal medicines, Calendula, catechin, β-sitosterol, and Jaungo (紫雲膏) may be effective for symptoms including pain, itching, and burning sensation induced by radiation therapy. Because of some research with conflicting results, further studies are needed to propose an algorithm for more optimal treatments.

        소양인 족번열에 대한 숙지황고삼탕 치험 1례

        Introduction This is the first case report of a soyangin suffering from burning feet syndrome who reported a significant improvement after taking Sukjiwhanggosam-tang. Case presentation The patient complained of hot feet, pain and numbness that occurred during the night. The patient was diagnosed with 'Yin deficiency diurnal fever disease' and treated with Sukjiwhanggosam-tang as well as acupuncture and cupping therapy. The degree of improvement in heat sensation, pain and numbness was expressed in numeric rating scale compared to the first visit. Conclusion After treatment, the heat sensation, pain and numbness of feet was decreased significantly, and the condition improved much at night.

        비대면 비실시간 대학 피아노 전공실기 수업의 만족도 분석과 개선방안 연구

        After conducting non-face-to-face non-real-time college applied music piano lessons for two semesters in 2020, 74 students responded to the survey. In this survey, piano playing video recording locations, lesson video learning locations, the satisfaction of the non-face-to-face non-real-time lessons, and the pros and cons of the non-real time non-face-to-face class were investigated. It is confirmed that there were many students who take the lesson video without a piano and the satisfaction score was 6.06. As an positive opinion by students, being able to check their skills voluntarily and objectively through their performance records, being able to freely choose the learning location and time, and repeated learning through repeated playback of lesson videos are confirmed. As an negative opinion, the limitation of musical expression, lack of tension and concentration due to free choice of the learning location and time, and difficulties in immediate communication with the instructor are confirmed. By the thought of instructor, there is the additional advantage that of increasing the time of lessons per person and there is the disadvantage of not being able to observe the performance and development of other students. As a way to improve non-face-to-face non-real-time piano lessons, conducting non-face-to-face non-real-time lessons with non-face-to-face real-time lessons, changing the format of assignment videos for smoother communication, and learning from each other through Havruta teaching method is suggested. 2020년도 두 학기 동안 비대면 비실시간 방식으로 피아노 전공실기 수업을 진행한 후, 74명의 수강생을 대상으로 과제 영상의 촬영 장소, 레슨 영상의 학습 장소, 비대면 비실시간 수업의 만족도, 비대면 비실시간 수업의 장단점 등을 조사하였다. 피아노 없이 레슨 영상을 수강하는 학생이 다수 있는 것으로 조사되었으며, 만족도는 6.06점으로 조사되었다. 학생이 체감하는 비대면 비실시간 수업의 장점 의견으로, 자신의 연주 기록을 통해 자발적이고 객관적인 실력 검증, 자유로운 레슨 시간과 장소, 레슨 영상 반복 재생을 통한 반복 학습이 확인되었으며, 단점 의견으로 음악적 표현 전달의 한계, 자유로운 레슨 시간과 장소에 의한 긴장감과 집중력 부족, 교수자와의 즉각적 소통 어려움이 확인되었다. 교수자는 학생 1인당 레슨시간이 증가한다는 장점과 학생들 서로의 연주 및 발전 과정을 관찰하지 못한다는 단점이 더 있다고 판단하였다. 비대면 비실시간 피아노 전공실기 수업의 개선방안으로, 비대면 비실시간 수업을 중심으로 수행하되 비대면 실시간 수업을 접목하는 방안, 보다 원활한 소통을 위한 과제 영상 형식을 변경하는 방안, 하브루타 교습을 통한 서로 배우기 방안을 제안한다.

        피아노 교습을 위한 융합 기술의 발전 동향 연구 -특허 분석을 중심으로-

        특허 분석을 통하여 최근 20년 동안의 피아노 교습 융합 기술 발전 동향을 검토하고 미래의 기술 발전 방향을 전망하였다. 2016년 내지 2017년 이후부터 스마트기기를 함께 사용하는 어쿠스틱 피아노 교습 기술에 대한 특허 출원의 증가 추세가 확인되었으며 전자피아노 교습 기술에 대한 특허 출원 감소가 확인되었다. 2015년까지는 다양한 추가 기기를 활용하는 피아노 교습 기술이 개발되었으나, 2016년 이후부터 스마트기기를 활용하는 피아노 교습 기술이 점진적으로 증가하여 2020년에는 전체 특허의 약 60%에 이르렀다. 또한, 2016년부터 자가 테스트 방식으로, 소프트웨어를 이용한 자동 평가 목적의 피아노 교습 기술 특허의 증가 추세가 확인되었다. 미래에는 스마트기기의 기술 발전과 더불어 어쿠스틱 피아노를 활용하는 교습 기술이 계속 발전할 것이며, 전자피아노를 활용한 교습 기술은 크게 증가하지 않을 것으로 전망된다. 또한, 스마트기기의 정보처리능력 발전과 더불어 피아노 연주 능력을 자가 테스트하는 소프트웨어 기술 발전도 계속될 것으로 전망된다.

        인공지능 피아노 교습 기술의 분석과 전망

        본 연구에서는 인공지능 피아노 교습 기술을 검토함으로써 인공지능이 예술과 교육 분야에 줄 수 있는 영향을 알아보고자 한다. 특허와 상용화된 소프트웨어를 통하여 현재의 기술 수준을 검토하였으며, 3단계의 기술 수준으로 분석되었다. 1단계는 악보 데이터화를 통한 악보 추천 기술이다. 2단계는 소리를 비교하여 정오를 판단하여 오류를 알려주는 기술이다. 3단계는 지속적 오류를 확인하여 연주자의 습관 여부를 판단하여 알려주는 기술이다. 이들을 통하여, 인공지능 피아노 교습 기술은 반복 훈련에 의한 연주 스킬 향상에 도움을 준다는 것을 알 수 있다. 하지만 인공지능 피아노 교습 기술의 한계는 인간의 신체 움직임을 판단하지 못하고 감정을 확인할 수 없다는 점이다. 이러한 한계는, 인공지능 피아노 교습 기술이 음악 본질보다는 표현물인 소리 분석에 집중되어 있음에 기인한 것으로 판단된다. 표현물 이면에 존재하는 인간의 감정을 이해하고 분류하여 인공지능이 학습 가능한 데이터로 가공하고 그 구분 기준을 설정할 수 있어야 한다. 이러한 발전 방향이 인공지능과 예술을 융합하는 정도일 것으로 판단된다. In this study, the effects of artificial intelligence on the arts and education were researched by studying artificial intelligence piano teaching skills. The current level of technology was reviewed through patents and commercially available software. They have three steps of technology. The first step is sheet music recommendation technology. The second step is to compare the sounds to determine and inform the error. The third step is a technology that determines and informs the habits of the player by checking the continuous error. Through these, the AI piano teaching technique helps to improve the playing skills by repetitive training. but cannot determine human body movements and identify emotions yet. This limitation is believed to be due to the fact that AI piano teaching technology is focused on sound analysis rather than music essence. In order to be the stage where artificial intelligence and art are truly converged, it is necessary to understand and classify human emotions that exist behind expressions.
