        Kartogenin Promotes the BMSCs Chondrogenic Differentiation in Osteoarthritis by Down-Regulation of miR-145-5p Targeting Smad4 Pathway

        Background: Transplantation of mesenchymal stem cells (MSCs) is a potential therapeutic strategy for cartilage degeneration of osteoarthritis (OA). But controlling chondrogenic differentiation of the implanted MSCs in the joints remains a challenge. The role of kartogenin (KGN) for chondrogenesis of MSCs has been widely reported, however, the mechanism of chondrogenesis has not been elucidated in OA. Methods: In this study, we investigated the miR-145-5p, TGF-β, Samd4, and p-stat3/stat3 expression in cartilage of OA patients and bone marrow mesenchymal stem cells (BMSCs) treated with KGN or miR-145-5p inhibitor. In addition, the cell proliferation and chondrogenic differentiation in vitro and in vivo of BMSCs treated with KGN was also detected. Results: In OA patients, the expression of miR-145-5p was up-regulated, and the expression of TGF-β, Samd4, and p-stat3/stat3 was inhibited. When the BMSCs treated with miR-145-5p inhibitor, the expression of TGF-β, Samd4, and p-stat3/stat3 was also significantly up-regulated. KGN-treated BMSCs had better proliferation and chondrogenic differentiation by up-regulating the expression of Sox 9, Col-2a1, aggrecan in vitro and in OA by down-regulation of miR-145-5p targeting Smad4 pathway. Moreover, intra-articular injection of KGN-treated BMSCs had a better pain relief effect in OA. Conclusion: The double effect on cartilage protection and pain relief indicates a great potential of intra-articular injection of KGN-treated BMSCs for the treatment of OA.         Functional analysis of prv-miR-LLT11a encoded by pseudorabies virus

        Viral-encoded microRNAs (miRNAs) play have vital roles in the regulations of virus replications and host immune responses. The results of previous studies have indicated that miRNA clusters are involved in the replication and virulence of the pseudorabies virus (PRV), which may potentially lead to the immune escape or facilitation of PRV replications. This study's previous research revealed that the prv-mirmiR-LLT11a was differentially expressed during PRV infections. The present study's results have demonstrated that the prv-miR-LLT11a could significantly inhibit PRV replications. It was further determined that SLA-1 was the target gene of the prv-miR-LLT11a, and simultaneously, thate overexpression of prv-miR-LLT11a could down-regulate the mRNA and protein levels of SLA-1 in a dose-independent manner. Furthermore, the present study also found observed that the prv-miR-LLT11a canhad also down-regulated the TAP1 expressions. Our findings provide a better understanding of the molecular mechanism involved in on the effects of prv-miR-LLT11a on SLA-1 and TAP1, as well as and its involvement in a potential immune system evasion of PRV.

        Automatic Optical Inspection of Bump Offsets in Flex-on-glass Bonding Using Differential Interference Contrast Imaging

        Bump offsets have harmful effects on the quality of flex-on-glass (FOG) bonding in liquid crystal display (LCD) module industry. Here we report a novel automatic optical inspection (AOI) method to achieve fast inspecting FOG bonding bump offsets. Overcoming difficulties to image targets at different depths in light absorption medium when using ultra-short-exposure and high-speed imaging, the proposed method innovatively achieves inspecting FOG bonding bump offsets using differential interference contrast imaging based on spatial Poisson point process approximation. Theoretical analysis proves that the proposed method has high reliability in FOG bonding bump offset AOI inspection. Experimental results show missed detection rate of the inspection algorithm is 0.01%, and false rate is 0.1%, and verify the accuracy and validity of the proposed method, all of which are significant for guiding practice in LCD module industry.

        Research on a fault diagnosis method of rolling bearings using variation mode decomposition and deep belief network

        The working conditions of rolling bearings during the running change in real time. Aiming at the problem of fault diagnosis of rolling bearing under complex working conditions, a new fault diagnosis (VHDBN) method based on variation mode decomposition (VMD), Hilbert transform (HT) and deep belief network (DBN) is proposed in this paper. Firstly, the proposed fault diagnosis method performs the VMD decomposition for the time domain signal in order to obtain a series of intrinsic mode functions (IMFs), and Hilbert envelope spectrum is obtained by Hilbert transform. The Hilbert envelope spectrum is used to construct the feature matrix, which is used as an input of the DBN network in order to obtain a fault diagnosis model. In order to test and verify the effectiveness of the proposed fault diagnosis method, the experimental data of rolling bearings under variable load is used in here. The experimental results show that the VMD-Hilbert envelope spectrum can better reflect the fault characteristics than the time domain spectrum, and the proposed fault diagnosis method under variable load has higher recognition accuracy than other comparison methods.

      • Forage Production and Water Use Efficiency of Three Lucerne Varieties at Different Soil Water Availabilities

        Forage production and water use efficiency (WUE) of Lucerne were investigated in three varieties at different water availabilities. Forage production decreased with the severity of soil water availability. At 50% field water capacity (FWC), forage production dropped but in two varieties, Algonquin and Longdong, was still high and from 75%FWC to 50%FWC, forage production in Longdong decreased at the least rate. The greatest leaf WUE was observed in Longdong at all soil water availabilities. From 75%FWC to 50%FWC, it increased in Longdong and Xinjiangdaye, but decreased in Algonquin. With the severity of water deficit, δ¹³C value increased in all three varieties. At the same water availability, the greatest value was observed in Longdong. It suggested that moderate water stress can improve WUE in Lucerne. Longdong is to some extent more efficient in water use and may be more drought-tolerant with more steady production at moderate water deficit.

      • Application of “Social Work Method” in Social Adaptation of Difficult Teenagers

        Purpose - The purpose of this paper is to introduce the perspective of social work, utilizing professional means of helping others, integrating various social resources, and deeply exploring relevant strategies to improve the social adaptation of disadvantaged adolescents. Design/Methodology/Approach - Based on the particularity, complexity, diversity, and the increasing number of teenagers in distress in recent years have aroused widespread concern from all walks of life. Through the investigation and interview of the distressed adolescents in some areas of Shandong Province, this paper discusses the present situation of their social maladjustment by using the mixed research method. Findings - This paper analyzes the main influencing factors of social maladjustment from three dimensions: selfadaptation, behavior adaptation, and interpersonal adaptation. Self-adaptation is examined from two dimensions: identity and cultural value Behavioral adaptation is mainly the prosocial behavior and anti-social behavior of troubled adolescents in the process of social adaptation. And puts forward the strategies to solve the social maladjustment of the distressed adolescents by using three major methods of social work. Research Implications - In the management of this paper, as a highly practical course, ‘Social Work Methods’ cannot achieve practical results solely relying on classroom teaching methods. Therefore, it is necessary to apply it to specific social life practices. This study focuses on the use of social work methods in the social adaptation of troubled adolescents to expand the use of social work methods in groups.

        Experimental Research on Impact Resistance of Partially Precast Concrete Beams

        A building may be subjected to multiple accidental loads during its service-life. Partially precast concrete (PC) beam is a primary force-bearing member. When subjected to an impact load, its impact resistance will significantly affect the overall safety of the structure. In this study, five PC beams were designed and manufactured, and drop-hammer impact tests were performed. The influences of impact mass and impact height on the failure mode and the dynamic response of the component under different loading conditions were examined. The typical failure mode of the test beam under a dynamic load was derived from the failure mode and crack distribution. The dynamic responses such as impact force, supporting reaction force, and displacement–time-history curve were obtained by the test. With increasing impact energy, the peak impact force increased from 3,353 to 3,708 kN, the peak mid-span displacement increased from 33.6 to 47.3 mm, and the residual mid-span displacement increased from 11.9 to 15.1 mm. Subsequently, the law of energy dissipation in the impact process was analyzed using the combined curves of the impact force-mid-span displacement and support reaction force-mid-span displacement. Furthermore, the result obtained by the integration of the reaction force-displacement curve in the middle of the span is more suitable for evaluating the overall energy consumption of the beam. The vibration energy and internal energy of the beam itself account for the primary part of the remaining energy.

        An Attention-based Temporal Network for Parkinson's Disease Severity Rating using Gait Signals

        Parkinson's disease (PD) is a typical, chronic neurodegenerative disease involving the concentration of dopamine, which can disrupt motor activity and cause different degrees of gait disturbance relevant to PD severity in patients. As current clinical PD diagnosis is a complex, time-consuming, and challenging task that relays on physicians' subjective evaluation of visual observations, gait disturbance has been extensively explored to make automatic detection of PD diagnosis and severity rating and provides auxiliary information for physicians' decisions using gait data from various acquisition devices. Among them, wearable sensors have the advantage of flexibility since they do not limit the wearers' activity sphere in this application scenario. In this paper, an attention-based temporal network (ATN) is designed for the time series structure of gait data (vertical ground reaction force signals) from foot sensor systems, to learn the discriminative differences related to PD severity levels hidden in sequential data. The structure of the proposed method is illuminated by Transformer Network for its success in excavating temporal information, containing three modules: a preprocessing module to map intra-moment features, a feature extractor computing complicated gait characteristic of the whole signal sequence in the temporal dimension, and a classifier for the final decision-making about PD severity assessment. The experiment is conducted on the public dataset PDgait of VGRF signals to verify the proposed model's validity and show promising classification performance compared with several existing methods.

        Appraisal of Guidelines for the Management of Blood Pressure in Patients with Diabetes Mellitus: The Consensuses, Controversies and Gaps

        Background: Currently available guidelines contain conflicting recommendations on the management of blood pressure (BP) in patients with diabetes mellitus (DM). Therefore, it is necessary to appraise the guidelines and summarize the agreements and differences among recommendations.Methods: Four databases and the websites of guideline organizations were searched for guidelines regarding BP targets and thresholds for pharmacologic therapy in DM patients, and the included guidelines were appraised with the Appraisal of Guidelines for Research and Evaluation (AGREE) II instrument.Results: In 6,498 records identified, 20 guidelines met our inclusion criteria with 64.0% AGREE II scores (interquartile range, 48.5% to 72.0%). The scores of the European and American guidelines were superior to those of the Asian guidelines (both adjusted P<0.001). Most of the guidelines advocated systolic BP targets <130 mm Hg (12 guidelines, 60%) and diastolic BP targets <80 mm Hg (14 guidelines, 70%) in DM patients. Approximately half of the guidelines supported systolic BP thresholds >140 mm Hg (10 guidelines, 50%) and diastolic BP thresholds >90 mm Hg (nine guidelines, 45%). The tiny minority of the guidelines provided the relevant recommendations regarding the lower limit of official BP targets and the ambulatory BP monitoring (ABPM)/home BP monitoring (HBPM) targets and thresholds in DM patients.Conclusion: The lower official BP targets (<130/80 mm Hg) in patients with DM are advocated by most of the guidelines, but they contain conflicting recommendations on the official BP thresholds. Moreover, the gaps regarding the lower limit of official BP targets and the ABPM/HBPM targets and thresholds need to be considered by future study.

        Modeling and Analyzing for Magnetic Field of Segmented Surface-Mounted PM Motors with Skewed Poles

        The skewed PM (permanent magnet) in the axial direction and the segmented PM in the circumferential direction, can reduce the cogging torque of the permanent magnet synchronous motor (PMSM). In this paper, the magnetization expression, which can be applied to diff erent circumferential sectioning modes, is derived. The quasi-3D magnetic fi eld analytical model suitable for diff erent circumferential and axial segmentation modes is proposed. The magnetic fi eld, cogging torque, back-EMF, and electromagnetic torque are obtained by the proposed modeling. The accuracy of the analytical method is verifi ed by 3D fi nite-element analysis. Finally, the modeling is used to analyze the variation of electromagnetic performance with diff erent radial skew angle and axial segments. The proposed modeling can shorten the calculation time, and analyze the variation law of motor performance with PM segmentation and skew comprehensively. It provides a reference for the design and optimization of segmented PMSM with skewed poles and has great signifi cance for improving motor performance.
