The ammonium ion-selective electrodes ($NH^+_4$-ISEs) based on the tetrahydrofuran(THF)-containing-16-crown-4 derivatives,1,4,6,9,11,14,16,19-tetraoxocycloeicosane ($L^1$) and 5,10,15,20,-tetramethyl-1,4,6,9,11,14,16,19-tetraoxocycloeicosane ($L^2$), were prepared and the electrode characteristics were tested. The conditioned $NH_4^+$-ISEs (E1) based on $L^1$ with TEHP as a plasticising solvent mediator gave best results with near-Nernstian slope of 53.9 mV/decade of activity, detection limit of $10^{-4.9}$ M, and enhanced selectivity coefficients for the $NH^+_4$ ion with respect to an interfering $K^+$ ion (log $K^{pot}_{NH_4^+,K^+}$ = -1.84). This result was compared to other ammonium ionophores reported previously, for example, that of nonactin (log $K^{pot}_{NH_4^+,K^+}$ = -0.92). The proposed electrode showed no significant potential changes in the range of 3.0 < pH < 9.0.

단백질 2DE 이미지 분석의 주요작업은 스팟 매칭에 의한 동일한 종류의 단백질 그룹인 패어링 클래스를 구성하는 것으로서 단백질간의 상호 작용, 질병에 관련한 단백질의 변화 등을 관찰할 수 있다. 하지만 2DE 실험의 여러 가지 문제점으로 인하여 패어링 클래스는 먼지, 공기방울 등 에러를 포함하게 되며 이런 에러들은 왜곡된 분석결과를 초래한다. 따라서 본 논문에서는 동일한 조직에서 같은 종류의 단백질은 발현량이 비슷하다는 특성을 이용하여 패어링 클래스의 개개의 스팟을 참조스팟 속성으로 나눈 값을 유사도로 정의하고, 스팟의 유사도가 사용자에 의하여 선택되는 필터링 배수에 의한 범위를 벗어날 때 에러 스팟으로 간주하여 제거되는 에러 필터링 기법을 제안한다. 실험에서는 정확도(Precision), 재현율(Recall) 및 조화평균(Harmonic-mean) 값을 사용하여 제안된 필터링 기법의 타당성을 보여준다.
Impact of a Glyphosate-Tolerant Soybean Line on the Rhizobacteria, Revealed by Illumina MiSeq

The global commercial cultivation of transgenic crops, including glyphosate-tolerant soybean, has increased widely in recent decades with potential impact on the environment. The bulk of previous studies showed different results on the effects of the release of transgenic plants on the soil microbial community, especially rhizosphere bacteria. In this study, comparative analyses of the bacterial communities in the rhizosphere soils and surrounding soils were performed between the glyphosate-tolerant soybean line NZL06-698 (or simply N698), containing a glyphosate-insensitive EPSPS gene, and its control cultivar Mengdou12 (or simply MD12), by a 16S ribosomal RNA gene (16S rDNA) amplicon sequencing-based Illumina MiSeq platform. No statistically significant difference was found in the overall alpha diversity of the rhizosphere bacterial communities, although the species richness and evenness of the bacteria increased in the rhizosphere of N698 compared with that of MD12. Some influence on phylogenetic diversity of the rhizosphere bacterial communities was found between N698 and MD12 by beta diversity analysis based on weighted UniFrac distance. Furthermore, the relative abundances of part rhizosphere bacterial phyla and genera, which included some nitrogen-fixing bacteria, were significantly different between N698 and MD12. Our present results indicate some impact of the glyphosate-tolerant soybean line N698 on the phylogenetic diversity of rhizosphere bacterial communities together with a significant difference in the relative abundances of part rhizosphere bacteria at different classification levels as compared with its control cultivar MD12, when a comparative analysis of surrounding soils between N698 and MD12 was used as a systematic contrast study.

To investigate the air temperature difference between windward and leeward side at high-rise building area, the air temperature and relative humidity data were observed for 10 minute interval from July 9, 2011 to November 30, 2011. The observed data were compared, analyzed and examined to illustrate air temperature between windward side (H Apartment) and Leeward side (W Apartment). The diurnal and seasonal variation of air temperature difference between windward and leeward site were also investigated. After the analysis, the overheat of windward side by 0.4℃ irrespective short distance of two observation positions. It was also lower than those of surrounding air temperature observing stations. It is mainly due to the air temperature decreasing effects of leeward side of high rise buildings.

HMGN5 is a typical member of the HMGN (high mobility group nucleosome-binding protein) family which may function as a nucleosomal binding and transcriptional activating protein. Overexpression of HMGN5 has been observed in several human tumors but its role in tumorigenesis has not been fully clarified. To investigate its significance for human lung cancer progression, we successfully constructed a shRNA expression lentiviral vector in which sense and antisense sequences targeting the human HMGN5 were linked with a 9-nucleotide loop. Inhibitory effects of siRNA on endogenous HMGN5 gene expression and protein synthesis were demonstrated via real-time RT-PCR and western blotting. We found HMGN5 silencing to significantly inhibit A549 and H1299 cell proliferation assessed by MTT, BrdU incorporation and colony formation assays. Furthermore, flow cytometry analysis showed that specific knockdown of HMGN5 slowed down the cell cycle at the G0/G1 phase and decreased the populations of A549 and H1299 cells at the S and G2/M phases. Taken together, these results suggest that HMGN5 is directly involved in regulation cell proliferation in A549 and H1299 cells by influencing signaling pathways involved in cell cycle progression. Thus, our finding suggests that targeting HMGN5 may be an effective strategy for human lung cancer treatment.
Heavy concrete shielding properties for carbon therapy

As medical facilities are usually built at urban areas, special concrete aggregates and evaluation methods are needed to optimize the design of concrete walls by balancing density, thickness, material composition, cost, and other factors. Carbon treatment rooms require a high radiation shielding requirement, as the neutron yield from carbon therapy is much higher than the neutron yield of protons. In this case study, the maximum carbon energy is 430 MeV/u and the maximum current is 0.27 nA from a hybrid particle therapy system. Hospital or facility construction should consider this requirement to design a special heavy concrete. In this work, magnetite is adopted as the major aggregate. Density is determined mainly by the major aggregate content of magnetite, and a heavy concrete test block was constructed for structural tests. The compressive strength is 35.7 MPa. The density ranges from 3.65 g/cm<sup>3</sup> to 4.14 g/cm<sup>3</sup>, and the iron mass content ranges from 53.78% to 60.38% from the 12 cored sample measurements. It was found that there is a linear relationship between density and iron content, and mixing impurities should be the major reason leading to the nonuniform element and density distribution. The effect of this nonuniformity on radiation shielding properties for a carbon treatment room is investigated by three groups of Monte Carlo simulations. Higher density dominates to reduce shielding thickness. However, a higher content of high-Z elements will weaken the shielding strength, especially at a lower dose rate threshold and vice versa. The weakened side effect of a high iron content on the shielding property is obvious at 2.5 µSv=h. Therefore, we should not blindly pursue high Z content in engineering. If the thickness is constrained to 2 m, then the density can be reduced to 3.3 g/cm<sup>3</sup>, which will save cost by reducing the magnetite composition with 50.44% iron content. If a higher density of 3.9 g/cm<sup>3</sup> with 57.65% iron content is selected for construction, then the thickness of the wall can be reduced to 174.2 cm, which will save space for equipment installation.

A nano-sized double-walled carrier composed of chitosan and β-lactoglobulin (β-Lg) for oral administration of epigallocatechin gallate (EGCG) was developed to achieve a prolonged release of EGCG in the gastrointestinal tract. Carboxymethyl chitosan (CMC) solution was added dropwise to chitosan hydrochloride (CHC) containing EGCG to form a primary coating by ionic complexation. Subsequently, β-Lg was added to create a secondary layer by ionic gelation. The obtained EGCG-loaded chitosan/β-Lg nanoparticles had sizes between 100 and 500 nm and zeta potentials ranging from 10 to 35mV. FT-IR spectroscopy revealed a high number of hydrogen-bonding sites in the nanoparticles, which could incorporate EGCG, resulting in high encapsulation efficiency. EGCG incorporated in the primary coating was released slowly over time by diffusion from the swollen CMC-CHC matrix after the outer layer of β-Lg was degraded in the intestinal fluid. The sustained-release property makes chitosan/ β-Lg nanoparticles an attractive candidate for effective delivery of EGCG.
