        Heme oxygenase-1 induced by desoxo-narchinol-A attenuated the severity of acute pancreatitis via blockade of neutrophil infiltration

        <P><B>Abstract</B></P> <P>Heme oxygenase-1 (HO-1) has an anti-inflammatory action in acute pancreatitis (AP). However, its mechanism of action and natural compounds/drugs to induce HO-1 in pancreas are not well understood. In this study, we investigated the regulatory mechanisms of HO-1 during AP using desoxo-narchinol-A (DN), the natural compound inducing HO-1 in the pancreas. Female C57/BL6 Mice were intraperitoneally injected with supramaximal concentrations of cerulein (50 μg/kg) hourly for 6 h to induce AP. DMSO or DN was administered intraperitoneally, then mice were sacrificed 6 h after the final cerulein injection. Administration of DN increased pancreatic HO-1 expression through activation of activating protein-1, mediated by mitogen-activated protein kinases. Furthermore, DN treatment reduced the pancreatic weight-to-body weight ratio as well as production of digestive enzymes and pro-inflammatory cytokines. Inhibition of HO-1 by tin protoporphyrin IX abolished the protective effects of DN on pancreatic damage. Additionally, DN treatment inhibited neutrophil infiltration into the pancreas via regulation of chemokine (C-X-C motif) ligand 2 (CXCL2) by HO-1. Our results suggest that DN is an effective inducer of HO-1 in the pancreas, and that HO-1 regulates neutrophil infiltration in AP via CXCL2 inhibition.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Desoxo-narchinol-A (DN) is a natural compound of HO-1 inducer in pancreas. </LI> <LI> Mechanism of DN-induced HO-1 is mediated by MAPK/Activator Protein-1/HO-1 signaling. </LI> <LI> DN-induced HO-1 blocks neutrophil infiltration into pancreas via inhibition of CXCL2. </LI> <LI> DN inhibits cerulein-induced acute pancreatitis (AP) and AP-associated lung injury. </LI> </UL> </P> <P><B>Graphical abstract</B></P> <P>[DISPLAY OMISSION]</P>

        The diagnostic requirements were suggested and explained regarding the systems of differentiation of symptoms and signs in the third year study of standardization and unification of the terms and conditions used for diagnosis in oriental medicine. The systems were as follows : - analyzing and differentiating of epidemic febrile disease - analyzing and differentiating in accordance with the Sasang constitution medicine based on four-type recognition - differentiation of disease according to pathological changes of Chong and Ren channels - standards for diagnosis of women's disease - standards for diagnosis of children's disease - standards for diagnosis of motor and sensor disturbance (-muscle. born, joint, etc.) - standards for diagnosis of neuropsychiatric disease - standards for diagnosis of five sense organ disease - standards for diagnosis of external disease The indivisual diagnosis pattern was arranged by the diagnostic requirements in the following order : another name, notion of diagnosis pattern, index of differentiation of symptoms and signs, the main point of diagnosis, analysis of diagnosis pattern, discrimination of diagnosis pattern, prognosis, a way of curing a disease, prescription, herbs in common use, disease appearing the diagnosis pattern, documents.         현재 초· 중 · 고등학교(이하 ' 학교기관' )에서 산이고 있는 기존의 수작업 방식의 회계관리와 DOS 방식의 행정전산관리 프로그램의 한계점을 벗어나 새로운 GUI 원도우 체계의 프로그램의 개발 필요성이 대두되어 본 시스템을 개발 하게 되었다.또한 학교기관에 납입해야 할 모든 납부금, 급식비, 장부관리에서부터 수입, 지출 등 학교에서 쓰이는 회계부분의 수작업을 모듈화 및 집약화 하고 누구나 쉽게 운영할 수 있는 행정관리 시스템, 일괄처리로 구성하도록 하였으며, DB를 이중모드(사용자 모드와 관리자모드)로 분리, 보안문제를 보충하고, 문서의 표준화로 정보교류의 용이하다.전국 학교기관의 회계운영방식을 표준화하여 구축 설계하고, 초보자 입장을 지향한 순차적 처리방식과 회계관리의 중복처리를 집약화 하여 구축하였다.

        단기간의 금연과 일회성 유산소 운동이 흡연자의 심혈관 반응에 미치는 영향

        The purpose of this study was to investigate cardiovascular responses to a bout of aerobic exercise and acute smoking cessation in smoker. nine collegiate male smoker who have been smoking(duration of smoking: 7.5 ± 0.9yrs; number of cigarettes per day: 15.5 ± 1.5) participated in this study. All subjects performed PEI to stimulate Group IV metaboreceptor at immediately after smoking, 24hours after smoking cessation and smoking after a bout of aerobic exercise. they were measured SBP, DBP, HR, SV, CO, TVC using by finometer(FMS co, Netherland). As a result, compared to immediately after smoking, SBP, DBP, and MAP were significantly decreased 24-hours after smoking cessation at rest and PEI(p<0.05). In addition, smoking after a bout of aerobic exercise caused by post-exercise hypotension indicated that SBP was significantly decreased compared to immediately after smoking(p<0.05). also, the changes in DBP and MAP at rest and handgrip exercise were significantly smaller compared to immediately after smoking(p<0.05). Thus, the results showed that smoking cessation during 24-hours and a bout of aerobic exercise relieve excess blood pressure response and enhanced arterial baroreflex in collegiate male smoker. It is concluded that caused exercise pressor reflex by stimulated Group IV may be effective to improve excess blood pressure responses.

        고연령 연금계좌 가입자 세액공제 확대에 따른 수익비 효과 분석

        [연구목적] 본 연구에서는 정부의 2019 세법개정안이 가입자들의 연금계좌 수익비에 미치는 영향을 살펴보고, 정책적인 시사점을 제공하였다. [연구방법] 본 연구는 연금 세제개편 흐름 및 이와 관련된 선행연구들을 충분히 검토하였으며 3가지의 제도 시나리오를 설정하여 수익비 분석을 진행하였다. 나아가 본 연구에서는 현실적인 분석을 수행하기 위하여 통계청의 마이크로 데이터를 활용하였으며, 이를 통해 연령 별 소득분위에 따른 보유 자산, 소비수준 등을 고려하여 분석을 진행하였다. [연구결과] 분석결과, 수익비 관점에서 저소득층이 연금계좌 추가납입에 따른 혜택을 가장 많이 받는 것으로 나타났으나 현실적으로 저소득층 고령자의 경우 추가납입을 위한 여유자금을 보유하고 있지 않아 추가납입이 제한되는 것으로 나타났다. 이에 따라 연금계좌 세액공제 확대에 따른 혜택은 고소득 고령자들에게 집중되는 것으로 나타났다. [연구의 시사점] 연금계좌의 추가납입을 제도적으로 유인하기 위하여 저소득층 세액공제 확대와 함께 보조금 매칭이나 고령자의 재고용 지원정책 등과 같이 현실적으로 저소득 고령자의 연금계좌 추가납입을 실현할 수 있는 방안이 모색될 필요가 있다. [Purpose] The purpose of this paper is to analyze the effect of expanding tax credit on contribution to private pension accounts for the elderly. [Methodology] For this purpose, we described prior literature review and summarized the details of the tax reform. Then, we conducted the money’s worth analysis in consideration of asset and consumption level according to the income quintile by age. [Findings] Our results shows that lower income people receive more benefits from additional payments to the private pension accounts from the perspective of money’s worth. In reality, however, additional payments are limited as they do not have any sufficient assets for additional payments. As a result, benefits from expanding tax credit on contribution amounts were concentrated on the elderly with high incomes. [Implications] Therefore, it would be necessary to seek ways to realistically realize additional payments to low-income senior citizens, such as introducing the marginal tax credit, matching subsidies and so on.

        FK506이 T 림프구 사멸에서 활성산소 생성에 미치는 영향

        Purpose: Tacrolimus (FK506) has been widely used as an immunosuppressant in organ transplanted recipients to suppress organ rejection phenomenon. We investigated the role of oxidative stress and heme oxygense-1 by FK506 on human Jurkat T cells. Methods: The cells viability was examined by DAPI stain, enzyme activity of caspase family proteins, and western blotting for Baks, PUMA, iNOS, HO-1. Cells were cultured in the absence or presence of CoPPIX or ZnPPIX and the fluorescence intensity was analyzed using a flow cytometry. Results: Treatment with FK506 increased the generation of reactive oxygen species (ROS), including hydrogen peroxide and superoxide anion, and NO in Jurkat cells in a dose-dependent manner. Immunohistochemistry and Western blot analysis data revealed the hemoxygenase-1 (HO-1) was induced by the addition of FK506 in Jurkat cells. Induction of CoPP, HO-1 inducer, resulted in decreased intracellular H₂O₂ and NO concentrations. Instead ZnPP, an HO-1 competitive inhibitor did it reversely. In addition, ZnPP regulates iNOS protein synthesis by inhibition of HO-1. Conclusion: Increase of HO-1 expression would induce to decrease the intracellular H₂O₂ and NO concentrations. Also, HO-1 would regulate iNOS protein synthesis. Consequently, we can expect the regulation of HO-1 expression with concomitants use of FK506 to suppress organ rejection phenomenon by enhancing apoptosis.

      • 전통발효식품으로부터 Chitin 분해 미생물의 분리 및 특성 규명

        A bacterial strain CJ-3 which produced chitinase was isolated from Korean traditional soy sauce. Using 16S rDNA analysis, the strain CJ-3 was identified as Bacillus atrophaeus. The approximate molecular weight of the putative chitinase enzyme was 31.0 kDa and the enzyme activity was remarkably induced by addition of colloidal chitin (0.5, 1.0, 2.0%). The antioxidant activity was increased 53% by the browning reaction products of B. atrophaeus CJ-3. Escherichia. coli lipopolysaccharides (LPS)-induced production of nitric oxide (NO) was reduced up to 45% by the browning reaction product in RAW264.7 macrophage. Inhibition of cell viability in the presence of LPS was recovered to normal level by the browning reaction product. These results suggest that browning reaction of B. atrophaeus CJ-3 plays an important role for activation of immune system. B. atrophaeus CJ-3 exhibited optimum temperature and pH of 37℃ and pH 7.8~8.0, respectively. The major intracelluar free amino acid was determined to be glutamate.

        N-acetyl cysteine inhibits H2O2-mediated reduction in the mineralization of MC3T3-E1 cells by down-regulating Nrf2/HO-1 pathway

        There are controversial findings regarding the roles of nuclear factor (erythroid-derived 2)-like 2 (Nrf2)/heme oxygenase-1 (HO-1) pathway on bone metabolism under oxidative stress. We investigated how Nrf2/HO-1 pathway affects osteoblast differentiation of MC3T3-E1 cells in response to hydrogen peroxide (H2O2), N-acetyl cysteine (NAC), or both. Exposing the cells to H2O2 decreased the alkaline phosphatase activity, calcium accumulation, and expression of osteoblast markers, such as osteocalcin and runt-related transcription factor-2. In contrast, H2O2 treatment increased the expression of Nrf2 and HO-1 in the cells. Treatment with hemin, a chemical HO-1 inducer, mimicked the inhibitory effect of H2O2 on osteoblast differentiation by increasing the HO-1 expression and decreasing the osteogenic marker genes. Pretreatment with NAC restored all changes induced by H2O2 to near normal levels in the cells. Collectively, our findings suggest that H2O2-mediated activation of Nrf2/HO-1 pathway negatively regulates the osteoblast differentiation, which is inhibited by NAC.
