Elevational Distribution of Breeding Bird Communities in Seoraksan National Park, Korea

        ( Hyun-su Hwang ),( Jae-kang Lee ),( Tae-kyung Eom ),( Ho-kyoung Bae ),( Dong-ho Lee ),( Jong-hwan Lim ),( Sung-cheol Jung ),( Chan-ryul Park ),( Shin-jae Rhim ) 한국산림과학회(구 한국임학회) 2020 한국산림과학회지 Vol.109 No.1

In this study, the elevational distribution of breeding birds in Seoraksan National Park, Korea was investigated. Forty-six species of birds were documented from line transect surveys taken from Seorakdong at 230 m above sea level (a.s.l.) to the Daechungbong summit at 1708 m asl. Birdspecies richness and diversity were highest in Seorakdong and lowest at the Daechungbong summit. As elevation increased, bird species richness and diversity decreased, with a humped-shape trend being observed between 700 m and 1200 m a.s.l. Stepwise analyses revealed that breeding bird species diversity indices were significantly negatively correlated with elevation (r2 = 0.327, P < 0.001) and positively correlated with vegetational coverage (r2 = 0.324, P = 0.046). Higher elevations supported fewer birds than low and intermediate elevations. Projections at the local scale, including data on behavior and habitat use by birds, will be necessary for optimal conservation and management of the bird communities in Seoraksan National Park.

We reported a case of pancreatic pseudoaneurysm, involving pancreaticoduodenal artery, developed after endoscopic retrograde cholangiopancreatography(ERCP) with endoscopic sphinterotomy. A 54 year old woman was transferred to our hospital due to sustained hematochezia and hypovolemia. A week before admission, she underwent ERCP with endoscopic sphinterotomy for acute cholangitis in local clinics. She developed abdominal pain and hematochezia at 12 hours after the procedures. A post procedure contrast enhanced computed tomography of abdomen demonstrated a pseudoaneurysm in inferior portion of pancreatic head. This was not present on the pre procedure contrast enhanced computed tomography of abdomen and was thought to arise from the pancreaticoduodenal artery as a complication of the pre sphincterotomy. She was brought to our emergency room in a state of hypovolemia. A upper endoscopy showed active bleeding from ampula of vater. An celiac angiography demonstrated pancreatic pseudoaneurysm involving inferior pancreaticoduodenal artery and selective coil embolization of was performed.

      • Protein kinase A-induced phosphorylation at the Thr154 affects stability of DJ-1

        Ko, Yeon Uk,Kim, Su-Jeong,Lee, Juhyung,Song, Min-Young,Park, Kang-Sik,Park, Jun Bae,Cho, Hyun-Soo,Oh, Young J. Elsevier 2019 Parkinsonism & related disorders Vol.66 No.-

Abstract

Introduction

Most cases of Parkinson's disease (PD) are sporadic, but genetic variations have been discovered in PD patients. PARK7/DJ-1 is a known cause of early-onset autosomal-recessive PD and is implicated in neuroprotection against oxidative stress. Although several post-translational modifications of DJ-1 have been proposed, phospho-modification of DJ-1 and its functional consequences have been less studied.

Methods

Putative phosphorylation sites of DJ-1 were determined by liquid chromatography-tandem mass spectrometry (LC-MS/MS analysis). Subsequently, phosphorylation site of DJ-1 was confirmed by in vitro kinase assay and cell-based pull-down assay. Impaired dimer formation of phospho-null mutant was measured using DSS crosslinking assay and immunoprecipitation assay. To evaluate physiological consequences of this event, protein stability of DJ-1 WT and DJ-1 phospho-null mutant were compared using cycloheximide chase assay and ubiquitination assay.

Results

Here, we showed that DJ-1 directly bound to the catalytic subunit of protein kinase A (PKAcα). We found that PKAcα is responsible for phosphorylation of DJ-1 at the T154 residue. Interestingly, dimerization of DJ-1 was not detected in a DJ-1 T154A mutant. Furthermore, stability of the DJ-1 T154A mutant was dramatically reduced compared with that of wild-type DJ-1. We found that DJ-1 T154A was prone to degradation by the ubiquitin proteasome system (UPS).

Conclusion

We identified a novel phosphorylation site of DJ-1. Furthermore, we determined protein kinase A that is responsible for this posttranslational modification. Finally, we demonstrated physiological consequences of this event focusing on dimerization and protein stability of DJ-1.

Highlights

PKAcα phosphorylates PARK7/DJ1 at the Thr154 residue.

Disruption of Thr154 phosphorylation is linked to a lower stability.

In Vitro Antiviral Activity of Ribavirin Against Severe Fever with Thrombocytopenia Syndrome Virus Myung Jin LEE1, Kye-Hyung KIM1, Jongyoun YI2, SuJin CHOI1, Chung-Jong KIM1, Nak- Hyun KIM1, Kyoung-Ho SONG1, Pyoeng Gyun CHOI1, Ji-Hwan BANG1, Wan Beom PARK1, Eu Suk KIM1, Sang-Won PARK1, Hong Bin KIM1, Nam Joong KIM1, Myoung- Don OH1 Seoul National University College of Medicine, Korea1, Pusan National University School of Medicine, Korea2 Background: Severe fever with thrombocytopenia syndrome (SFTS) is an emerging infectious disease caused by a novel Bunyavirus, severe fever with thrombocytopenia syndrome virus (SFTSV). No effective antiviral therapy is proven yet, but clinical use of ribavirin (RBV) has been tried. We investigated the antiviral effect of RBV against SFTSV in vitro. Methods: To test for cytotoxicity of RBV, Vero cells were treated with different concentrations of RBV (3.90 to 500 μg/mL, two-fold dilution) and analyzed by cell viability MTS assay 48h post-infection. To determine antiviral activity of RBV against SFTSV, Vero cells were infected with SFTSV strain Gangwon/Korea/2012 at 100 TCID50 (50% tissue culture infective dose) per well in a 96-well plate, and RBV was added at the concentrations showing no or minimal cytotoxicity. Viral RNAs were extracted from the culture supernatants and quantifi ed using one-step real-time reverse transcription- PCR to amplify the partial large segment of SFTSV. Statistical analysis was done by one-way ANOVA with Tukey`s post hoc test. Results: Cytotoxicity due to RBV was not observed at RBV concentration =31.3 μg/ mL. Viral RNAs at 24h post-RBV treatment were reduced with increasing RBV concentrations (1-32 μg/mL), compared with those of mock-treated cells (P <0.01, Figure). Half maximal inhibitory concentration (IC50) of RBV was 3.69 μg/mL at 24h post-RBV treatment. Conclusions: Our study shows that RBV has antiviral effect against SFTSV in a dose-dependent manner. Further studies are required to evaluate the effi cacy of RBV in SFTS.

Monitoring ecosystem changes is an important process for collecting basic information for ecosystem management. Since many ecological phenomena and their processes take place over a long period of time, it is necessary to conduct long-term monitoring of ecosystems for the continuous conservation and efficient management of national parks. This study suggested an optimized monitoring design, standardized operation plan, human resource and data management system, stable financial preparation and follow-up support as methods for successful long-term monitoring through case analysis of domestic and foreign long-term monitoring programs. Based on this, it proposes specific measures such as establishing a standardized protocol with adaptive monitoring methods, continuous data management with QA/QC, and strengthening the linkage of investigation items in the development direction of the “HS oil spill accident long-term monitoring program” that has been conducted for more than a decade.

The morphological analysis could be used as a pre-processing of the syntactic and the semantic analysis in one hand, as well as the automatic indexing, the post-processing of the OCR system and the spell checking/correcting in another hand. The linguistic rules improved the accuracy of the morphological analysis and the disambiguation while the corpus provided the better analysis speed and the better efficiency of the application systems. As for the disambiguation which is one of the most important problems in morphological analysis, our approach depends on rules in the first stage, such as specific disambiguation rules for 63 morphemes which are related 1/3 of Korea morphological ambiguities, the syntactic constraint rules, collocation rules, and so on. After applying all the rules filtering incompatible strings, the system chooses the remaining one using statistical data from the corpus. When more than two strings still remain, their priority in the context is decided by statistics in the second stage, such as the frequency value of the morpheme strings or that of the phrases containing them. The accuracy of the system is about 98.27%

Ligand-based QSAR Studies on the Indolinones Derivatives as Inhibitors of the Protein Tyrosine Kinase of Fibroblast Growth Factor Receptor by CoMFA and CoMSIA

        Hyun, Kwan-Hoon,Kwack, In-Young,Lee, Do-Young,Park, Hyung-Yeon,Lee, Bon-Su,Kim, Chan-Kyung Korean Chemical Society 2004 Bulletin of the Korean Chemical Society Vol.25 No.12

Ligand-based quantitative structure-activity relationship (QSAR) studies were performed on indolinones derivatives as a potential inhibitor of the protein tyrosine kinase of fibroblast growth factor receptor (FGFR) by comparative molecular field analysis (CoMFA) and comparative molecular similarity indices analysis (CoMSIA) implemented in the SYBYL packages. The initial X-ray structure of docked ligand (Su5402) to FGFR was used to minimize the 27 training set molecules using TRIPOS force field. Seven models were generated using CoMFA and CoMSIA with grid spacing 2 ${\AA}$. After the PLS analysis the best predicted CoMSIA model with hydrophobicity, hydrogen bond donor and acceptor property showed that a leave-one out(LOO) cross validated value $({r^2}_{cv})^$ and non-cross validated conventional value $({r^2}_{ncv})^$ are 0.543 and 0.938, respectively.
