      • Choline dehydrogenase interacts with SQSTM1/p62 to recruit LC3 and stimulate mitophagy.

        <P>CHDH (choline dehydrogenase) is an enzyme catalyzing the dehydrogenation of choline to betaine aldehyde in mitochondria. Apart from this well-known activity, we report here a pivotal role of CHDH in mitophagy. Knockdown of CHDH expression impairs CCCP-induced mitophagy and PARK2/parkin-mediated clearance of mitochondria in mammalian cells, including HeLa cells and SN4741 dopaminergic neuronal cells. Conversely, overexpression of CHDH accelerates PARK2-mediated mitophagy. CHDH is found on both the outer and inner membranes of mitochondria in resting cells. Interestingly, upon induction of mitophagy, CHDH accumulates on the outer membrane in a mitochondrial potential-dependent manner. We found that CHDH is not a substrate of PARK2 but interacts with SQSTM1 independently of PARK2 to recruit SQSTM1 into depolarized mitochondria. The FB1 domain of CHDH is exposed to the cytosol and is required for the interaction with SQSTM1, and overexpression of the FB1 domain only in cytosol reduces CCCP-induced mitochondrial degradation via competitive interaction with SQSTM1. In addition, CHDH, but not the CHDH FB1 deletion mutant, forms a ternary protein complex with SQSTM1 and MAP1LC3 (LC3), leading to loading of LC3 onto the damaged mitochondria via SQSTM1. Further, CHDH is crucial to the mitophagy induced by MPP+ in SN4741 cells. Overall, our results suggest that CHDH is required for PARK2-mediated mitophagy for the recruitment of SQSTM1 and LC3 onto the mitochondria for cargo recognition.</P>

        당뇨병환자에서 뇨증 Fibronectin농도의 증가에 관한 연구

        Plasma fibronectin is an α₂-glyoprotein, which is produced by vascular endothelial cells. Raised level of plasma fibronectin has been observed in diabetic patients particularly in the presence of microvascular complications. However, no available data exist about urine level of this glycoprotein in diabetic patients. We measured urine fibronectin level by ELISA methods in 54 diabetic patients who have microvascular comlications or not. The following results were obtained. 1) Urine fibronectin level(㎍/g creatinine) in diabetic group(1740.0±678.0) is increased compaired with those in normal control group(471.0±59.0). but this results were not significantly different among two groups(P>0.05). 2) Urine fibronectin level(㎍/g creatinine) was significantly increased in patients with nephropathy (6188.0±3144.0) compared with those in normal control group(471.0±59.0) and patients without nephropathy(645.0±251.0)(P<0.001). 3) There were a significant correlation between BUN, creatinine, creatinine clearance, 24hr urine total protein and urine fibronectin level in diabetic patients. Our data suggest that urine fibronectin excretion level might be used as a sensitive guide for diabectic nephropathy.

        지리산국립공원 심원계곡 생태경관 복원공사지역 식생 변화

        본 연구는 지리산국립공원 심원계곡 생태경관 복원공사지역의 모니터링 결과를 분석하여 체계적인 복원관리를 도모할 수 있는 기초자료를 확보하는데 그 목적이 있다. 복원사업이 완료된 2017년도에 식생 모니터링을 위한 조사구 12개소와 대조구 4개소를 설치하였고, 2017년과 2020년의 상대우점치, 종다양도지수, 유사도 등의 변화를 분석하였다. 복원사업 시 수목이 식재되었던 조사구는 종다양도지수가 0∼1.4552로 나타났고, 대조구와의 유사도지수에서는 1개 조사구가 1.32%로 분석된 것을 제외하고 모두 0%로 나타났다. 조사구의 종다양도지수 및 유사도지수가 매우 낮은 것은 복원사업 시 식재된 수목이 고사하거나 멧돼지 피해, 유수에 의한 침식으로 유실되는 등의 영향으로 판단되었다. 반면 복원사업 시 수목이 식재되지 않은 미식재 조사구는 종다양도지수가 0.9538∼2.3222로 나타났고, 유사도지수는 최대 8.33%까지 분석되었다. 국립공원 생태경관 복원공사지역의 복원 공법 개발을 위한 장기 모니터링과, 미식재 조사구의 천이과정을 분석하는 지속적인 연구가 필요하다. This study aims to obtain basic data for systematic restoration by analyzing the monitoring results of the Shimwon Valley Ecological Landscape Restoration Project area in Jirisan National Park. In 2017, when the restoration project was completed, 12 monitoring plots and 4 control plots were installed for vegetation monitoring, and changes in the relative dominance, species diversity index and similarity between 2017 and 2020 were analyzed. The species diversity index of the surveyed areas where trees were planted during the restoration project was 0-1.4552, and the similarity index with the control group was 0% except for one survey area at 1.32%. The very low species diversity index and similarity index in the survey areas were attributed to the loss of trees planted during the restoration project due to death, damage by wild boars, or erosion by running water. On the other hand, the species diversity index was 0.9538-2.3222 in the monitoring plot where no tree was planted, and the similarity index was analyzed to be as high as 8.33%. It is necessary to continue the long-term monitoring for the development of ecological landscape restoration methods in the national park and analysis of the succession in monitoring plots where no trees were planted.

        하악골 전돌증 환자에서 양악 수술과 하악 단독 수술의술 후 안정성에 관한 연구

        Le-Fort I Osteotomy is considered by many surgeons to be a safe and predictable method for correcting various dentofacial deformities. However, there are controversial options concerning the stability of two-jaw surgery (Le-Fort I osteotomy and sagittal split ramus osteotomy) compared to single mandibular surgery (sagittal split ramus osteotomy). The purpose of this study was to evaluate the post-operative stability of mandible in two-jaw surgery and single mandibular surgery, to help the establishment of accurate treatment planning in patients with a skeletal class Ⅲ malocclusion. There were 15 patients who had been undergone two-jaw surgery and 20 patients who had been undergone single mandibular surgery. Lateral cephalograms of these 2groups ; Preoperation and immediate post-operation, postoperatively 1month, postoperatively 3months, at the latest follow up(>6months) ; were analyzed by linear measurement to evaluate changes in position (hard tissue B, Pogonion point) and compare repapse both group. The results obtained were as follows ; In view of B-point, the horizontal repapse rate of single mandibular surgery was 10.17% at post-op 1month, 12.52% at post-op 3months, 13.01% at post-op 6months and two-jaw surgery was 10.23% at post-op 1month, 11.84% at post-op 3months, 12.97% at post-op 1month, 11.33% at post-op 3months, 12.01% at post-op 6months in two-jaw surgery. In terms of vertical relapse rate, rates of B point were 22.96% at post-op 1month, 28.30% at post-op 3months, 29.56% at post-op 6months in single mandibular surgery and 21.14% at post-op 1month, 25.45% at post-op 3months, 28.86% at post-op 6months in two-jaw surgery. Those of Pogonion point were 26.63% at post-op 1month, 28.40% at post-op 3months, 29.29% at post-op 6months in single mandibular surgery and 20.20% at post-op 1month, 25.44% at post-op 3months, 27.68% at post-op 6months in two-jaw surgery. There were no statistical difference between single mandibular surgery and two-jaw surgery in mandibular stability.

        완전뇌허혈후 재관류시에 국소뇌혈류, 체성감각유발전위 및 신경학적 회복에 대한 Mannitol과 Thiopental의 효과

        The cardiopulmonary bypass or cerebral circulation arrest is often used in the treatment of complex aneurysm or of arteriovenous malformation to decrease the risk of intraoperative aneurysm rupture. Although experimental studies have suggested that some drugs may protect the brain from ischemic injury, there are limitations in maintaining cerebral perfusion arrest without incurring neurologic deficits due to the initiation of detrimental processes including excitotoxic neuronal injury, activation of phospholipases, influx of calcium, and generation of damaging free radicals. The purpose of this study is to determine wheter mannitol or thiopental has any favorable effects on the recovery of neurologic deficits and on the regional cerebral blood flow(rCBF), somatosensory evoked potential(SEP) and electroencephalogram(EEG) in cats which underwent 15 minutes-complete global ishcemia-reperfusion. The complete global ischemia was produced in 38 cats by temporary intrathoracic occlusion of the innominate artery and the subclavian artery following ligation of bilateral mammary arteries and simultaneous induction of hypotension. The cats were allocated randomly to one of 4 treatment groups : (1) control group, 8 cats received equal volume of saline solution : (2) thiopental group, 10 cats received 45㎎/㎏ thiopental intravenously, (3) mannitol group, 10 cats received 2g/㎏ mannitol intravenously, (4) combined mannitol and thiopental group, 10 cats received equal dose of mannitol and thiopental intravenously. The drugs were administrated in a equally divided dosage before and after the ischemic episode. The results were as follos : 1) Eight animals which received saline showed the severe postischemic hypoperfusion and poor recovery of SEP and EEG, and 6 of them died within 6 hours after the ischemia. 2) Ten thiopental-treated animals also showed the severe postischemic hypoperfusion and poor recovery of SEP and EEG, and 6 of them died within 6 hours the ischemia. 3) Both mannitol-and combined treated groups showed early recovery of EEG, good recovery of SEP and EEG without the severe postischemic hypoperfusion, and 7 of 10 mannitol-treated animals and 8 of 10 combined treated animals were significantly recovered in all parameters. There were not significant differences in all parameters between the mannitol-and combined treated groups. 4) Thirty-four of 38 animals involved in this 15 minutes-ischemia resulted in the severe neurologic deficits inspite of treatment with mannitol, thiopental or both of them. These results suggest that, in cats, mannitol treatment is effective but not thiopental in preventing severe neurologic injury following complete global ischemia and the duration of complete ischemia should be far less than 15 minutes.

        The purpose of this study is to investigate growth environment of street trees planted on main roads in Kimhae city. The results of investigation that soil hardness, soil acidity, vitality classified by characteristic of area are as follows ; The results of soil hardness classified by residential area(A area), commercial area (B area), industrial area (C area) are as follows; A section 20.7㎜, B section 17.9㎜, C section 13.4㎜. Soil hardness of investigative areas are satisfactory, but it needs to establish of tree-grate for healthful growth of street trees. The results of soil acidity classified by residential area(A area), commercial area (B area), industrial area (C area) are as follows ; A section pH 5.42, B section pH 4.28, C section pH 3.89, B, C areas are investigated extreme acidity, so it needs to improve of the soil structure.

      • 肉鷄後期飼料의 給與時期가 鷄肉의 理化學的 性質에 미치는 影響 : 貯藏期間別에 따른 脂質含量과 脂肪酸造成의 變化 Changes in lipid contents and fatty acid compositions as the pass age of storage time

        後期飼料를 3주령부터 조기에 급여한 9주령 Hubbad鷄肉의 3부위 근육을 공시육으로 하여 4℃에서 저장하면서 경시적(도살직후 저장후 2일 4일 7일)으로 지질의 함량을 추출하고 지방산의 조성을 GLC로 분석한 결과 다음과 같은 결과를 얻었다. 1. 지질함량은 도살직후 대퇴부위가 2.67%로 가장 높고 흉심부가 2.38%, 근위 1.28% 가장 낮았으며, 저장기간에 따른 지질함량의 변화는 3부위 모두 저장기간의 경과에 따라 지질함량이 감소하였다. 2. GLC로 분석한 지방산은 3부위 모두 myristic acid(14:0)로 부터 arachidonic acid(20:4)까지 10種을 분리 동정하였다. 3. 함량이 제일 높은 지방산은 세부위 모두 불포화 지방산인 Oleic acid로 40.7~45.5%였고 다음 Palmitic acid로 20.5~27.9%였으며 linoleic acid는 6.9~18.3%로 타육류에 비해 필수지방산인 linoleic acid의 함량이 높았다. 4. 3部位 모두 저장시간이 경과함에 따라 포화지방산의 함량이 증가되고 불포화지방산의 함량은 상대적으로 감소되었다. The meat cut 3 parts of leg, breast and gizzard of broiler-Hubbard at 9 weeks fed early finisher diet from 3 weeks were stored at 4℃ for four levels, immediately after slaughtering, 2 days, 4 days and 7 days after storing in order to extract lipid contents and fatty acid compositions was analyzed by the GLC method. The results obtained in this experiment were summarized as follows: 1. Lipid contents were the highest with 2.67% in the leg immediately after slaughtering and the lowest, with 1.28% in the gizzard. Fatty acid compositions were decreased in all the three parts as the storage time passed. 2. Ten fatty acids from myristic acid(14:0) to arachidonic acid(20:4) in all the there parts were identified by GLC. 3. The highest fatty acid was oleic acid with 40.7 to 45.5% and the next, palmitic acid with 20.5 to 27.9%. Linoleic acid was higher with 6.9 to 18.3% in broiler-Hubbard then in the other broiler. 4. As the storage time passed, saturation fatty acid increased and unsaturation fatty acid decreased relatively.

        METRx™ 기구를 이용한 후방 경추부 추궁추간공 제거술의 예비 결과

        Objectives: Posterior cervical microscopic laminoforaminotomy has been a preferred procedure for a posterolateral cervical disc or foraminal stenosis. However, neckpain from wide skin incision and muscle injury are some of disadvantages. The authors performed cervical microscopic laminoforaminotomy with METRx™ tubular dilator system and were compared the results with classical cervical microscopic foraminotomy. Methods: Six patients underwent posterior cervical microscopic laminoforaminotomy using the METRx™ during 6 months. We have selected 10 patients from our institution as a comparison group. Preoperative and postoperative pain score composed of the radicular and neck component was examined and disability score also recorded. Diameters and lengths of laminoforaminotomy on the postoperative CT also were compared with two methods Results: Postoperative pain scores to the radiculopathies were not different but postoperative pain scores to the neckpain on the follow up periods were significantly different. The average vertical and transverse diameters of the laminoforaminotomy defect on the postoperative CT scans were not different between two groups. Average hospital stay of the classical methods was 8.8±7.5 days comparison with 4.5±0.54days of new procedure. Conclusions: Posterior cervical microscopic laminoforaminotomy with METRx™ systems seems to provide similar decompression and clinical results as classical methods compared to classical methods and shows superior results for postoperative neckpain and hospital stays.

        유도초음파를 이용한 2.25Cr-1Mo재의 열화도 평가

        고온에서 장시간 사용으로 인하여 발생되는 경년열화 평가에는 파괴적인 방법이 신뢰성이 높지만 시험편 채취의 어려움이 있다. 그러므로 비파괴적인 방법에 의한 열화도 평가가 바람직하다. 본 연구에서는 2.25Cr-1Mo강에 대하여 장시간 등온열처리로 모의 열화시험편을 제작하여 고온설비부재에서 발생되는 탄화물의 석출 및 조대화, 불순물의 입계편석 등 금속학적 미세조직을 관찰하고, 미세조직의 변화와 유도 초음파의 전파특성과의 상관관계를 규명하고, 파괴시험치와 그 결과를 비교하여 초음파 비파괴평가에 의한 고온부재의 열화도 평가의 가능성을 실험적으로 검증하고자 한다. The destructive method is reliable and widely used for the estimation of material degradation but, it have time-consuming and a great difficulty in preparing specimens from in-service industrial facilities. Therefore, the etimation of degraded structural materials by nondestructive evaluation is strongly desired. In this paper, the use of guided wave was suggested for the evaluation of thermally damaged 2.25Cr-1Mo steel as an alternative way to compensate for limitations of fracture tests. The observation of microstructure variations of the material including carbide precipitation increase and spheroidization near gain boundary was conducted and the correlation with the guided wave features such as energy loss ratio and group velocity changes was investigated. Through this study the feasibility of ultrasonic guided wave evaluation for thermally damaged materials was explored.
