      • Differential Effects of Pioglitazone in the Hippocampal CA1 Region Following Transient Forebrain Ischemia in Low- and High-Fat Diet-Fed Gerbils.

        <P>In the present study, we investigated the effects of pioglitazone (PGZ) in the hippocampal CA1 region of low- or high-fat diet (LFD or HFD) fed gerbils after transient forebrain ischemia. After 8 weeks of LFD or HFD feeding, PGZ (30 mg/kg) was intraperitoneally administered to the gerbils, following which ischemia was induced by occlusion of the bilateral common carotid arteries for 5 min. Administration of PGZ significantly reduced the ischemia-induced hyperactivity 1 day after ischemia/reperfusion in both LFD- and HFD-fed gerbils. At 4 days after ischemia/reperfusion, the neurons were significantly reduced and microglial activation was observed in the hippocampal CA1 region in LFD- and HFD-fed gerbils. The microglial activation was more prominent in the HFD-fed gerbils compared to the LFD-fed gerbils. Administration of PGZ ameliorated ischemia-induced neuronal death and microglial activation in the hippocampal CA1 region 4 days after ischemia/reperfusion in the LFD-fed gerbils, but not in the HFD-gerbils. At 6 h after ischemia/reperfusion, tumor necrosis factor-α (TNF-α) and interlukin-1β (IL-1β) levels were significantly increased in the hippocampal homogenates of LFD-fed group compared to control group, and HFD feeding further increased TNF-α and IL-1β levels. PGZ treatment significantly ameliorated the increase of TNF-α and IL-1β levels in LFD-fed gerbils, not in the HFD-fed gerbils. At 12 h after ischemia/reperfusion, superoxide dismutase (SOD) and malondialdehyde (MDA) levels in hippocampal homogenates were significantly increased in the LFD-fed group compared to the control group, and HFD feeding significantly showed relatively reduction in SOD activity and increase in MDA level. PGZ administration significantly reduced the increase in MDA levels 12 h after ischemia/reperfusion in the LFD-fed gerbils, but not in the HFD-fed gerbils. These results suggest that PGZ ameliorates the neuronal damage induced by ischemia by maintaining the TNF-α, IL-1β, SOD and MDA levels in LFD-fed gerbils. In addition, HFD feeding affects the modulation of these parameters in the hippocampus after transient forebrain ischemia.</P>

        Objectives : There are a number of preceding epidemiological studies reporting gender differences in the genetic etiology of alcohol dependence. The author investigated gender difference in the frequencies of ADH2 and ALDH2 genoypes between the patients with alcohol dependence and normal control. Methods : The subjects were 141 alcohol dependent patients (104 males, 37 females) and 138 normal control (79 males, 59 females). The frequencies of 1/1 and 1/2+2/2 (2+ afterward) genotypes for ADH2 and ALDH2 were investigated in male and female between alcohol dependence and normal control group. DNA was extracted from WBC in peripheral venous blood and PCR-RFLP method was used out for genotyping. Results : First, the frequency of ADH2 1/1 genotype was significantly higher in alcohol dependent patients than normal control in both genders. Second, while there was no gender difference in the frequency of ADH2 1/1 genotype in normal controls, in the patient group however, the frequency was significantly higher in females than males. Third, in male subjects with alcohol dependence, the frequency of ALDH2 1/1 genotype was significantly higher than in male normal control subjects. On the other hand, in female subjects with alcohol dependence, the frequency of ALDH2 2+ genotype was significantly higher than in female normal control subjects. Conclusion : These results suggest that while the risk of alcohol dependence is predominantly affected by ALDH2 1/1 geno-type in male, the female ADH2 1/1 genotype is mainly associated with the risk of alcohol dependence. This means that there are gender differences in the genetic etiology of alcohol dependence.

        Effect of SME and Chaebol Loan Contracts on Debt Covenant Hypothesis: Evidence from Korea

        This study provides an in-depth analysis of the covenant-based hypothesis that focuses on whether and how correlations between debt ratios and earnings management (measured by discretionary accruals) are affected by the size and nature of firms. We performed a pooled ordinary least squares regression using a sample of 10,983 firms listed in Korea from 2002 through 2012. In the Korean financial market, SMEs and firms belonging to chaebols (conglomerates) are markedly different in terms of their bargaining power with financial institutions. The results suggest that SMEs have a larger portion of secured loans and that chaebols can use their credit in the bond market to access alternative sources of financing. Accordingly, neither SMEs nor chaebols are motivated to make income-increasing choices for larger discretionary accruals to relax their debt constraints. In contrast, a large firm with a higher debt ratio is likely to make accounting choices to increase reported earnings, which results in more discretionary accruals. The debt agreements between lenders (financial institutions) and borrowers (firms) have different implications in the Korean financial market depending on a firm`s size and nature.

        <P>The optimal timing of percutaneous coronary intervention (PCI) in patients with non–ST-segment elevation myocardial infarction (NSTEMI), complicated by acute decompensated heart failure (ADHF), is unclear. A total of 1,027 patients with NSTEMI complicated by ADHF who underwent successful PCI were analyzed using a Korean multicenter registry. All patients were divided into 4 groups by the timing of PCI: group 1 (PCI < 2 hour after admission, n = 149), group 2 (2 to 24 hours, n = 577), group 3 (24 to 72 hours, n = 189), and group 4 (≥72 hours, n = 112). We analyzed the incidences of 12-month mortality, nonfatal myocardial infarction (MI), target-vessel revascularization, and rehospitalization because of HF. The prevalence of ADHF in patients with NSTEMI was 15.2% at initial presentation, and in-hospital mortality was higher in group 1 than in the other groups. There were no significant differences in mortality, nonfatal MI, target-vessel revascularization, or rehospitalization for HF during the 12-month follow-up between groups, regardless of initial PCI timing, except for a higher 12-month mortality in patients who received PCI within 24 hours (vs ≥24 hours) (hazard ratio 1.52, 95% confidence interval 1.09 to 2.29, p = 0.046). Early PCI did not reduce adverse clinical outcomes in patients with NSTEMI complicated by ADHF. Delayed PCI after stabilization may be reasonable in such high-risk patients.</P>

        Background : In recent years, gonorrhea has been panedemic and remains one of the most commom STDs in the world, especially in developing countries. Objective & Methods: For the detection of a more effective therapeutic regimen and assessing the prevalence of PPNG, we have been trying to study the patients who have visited the VD Clinic of Choong-Ku Public Health Center in Seoul since 1980 by means of the chromogenic cephalosporin method. Results: In 1996, 139 strains of N. gonorrhoeae were isolated, among which 53(39.0%) were PPNG. Conclusion: Our results suggests that after a peak of 74.3% in 1993, the prevalence of PPNG in Seoul is gradually declining.

        Background : In recent years, gonorrhea has been pandemic and remains one of the most common STDs in the world, especially in developing countries. Objective & Methods : For the detection of a more effective therapeutic regimen and assessing the prevalence of PPNG, we have been trying to study the patients who have visited the Venereal Disease Clinic of Choong-Ku Public Health Center in Seoul since 1980 by means of the chromogenic cephalosporin method. Results : In 1997. 99 strains of N. gonorrhoeae were isolated, among which 45(45.5%) were PPNG. Conclusion : The prevalence of PPNG in Seoul, which had been decreased to 39% in 1996 after a peak of 74.3% in 1993, is increased to 45.5% in 1997.

        This study investigated the effect of thermal treatment (blanching) conditions on the physicochemical and nutritional properties of spinach. For thermal treatments, fresh spinach was treated by three blanching methods including boiled water, steam and pan frying treatments. At the varying time intervals of treatments, pH, color, shear force, proximate composition, organic acid content, peroxidase activity and microbial counts were evaluated. The pH of all treatments tended to increase with increasing blanching time, however, no significant differences among treatments were obtained. The shear force of spinach decreased with blanching time, particularly hot water treatment showed the rapid decrease in the shear force of spinach. The lightness of spinach showed similar pattern with shear force. For proximate compositions, the highest moisture content was found in hot water treatment, while crude protein and ash were highest in steam treatment. Due to the usage oil, pan frying treatment involved in the highest lipid content. The organic acid content treatment was in the order of hot water, pan frying and steam treatment. The thermal treatments attributed to low total plate count of spinach. In addition, thermophilic bacteria, coliform, mold and yeast were not detected in all thermal treatments. Peroxidase activity was lowered by applied thermal treatments. Based on the results, the best blanching condition for spinach was found in steam treatment for 60 s where the spinach showed the minimal changes in physicochemical and nutritional properties of spinach.

        Dust Collection and Nutrient Absorption by Halophyte Communities in Saemanguem Reclaimed Land

        Halophyte community was established for dust prevention in Saemangeum reclaimed land from 2006 to 2008. In the present study, the functions of halophyte community were examined on-site in aspects of dust collection and nutrient absorption. In dust collection experiments, total suspended particulate (TSP) decreased through transplanted halophyte community and the reduction effect continued to 50 m leeward, which was 5.6 times of plant height. TSP reduction behind in-situ halophyte communities amounted to 25.6% on seven-monthly average. TSP collected within four halophyte communities varied among halophytic species in the field. Harvested in the reclaimed land, halophytic samples contained significant amount of nitrogen (0.84 to 1.71% of dry weight), $P_2O_5$ (0.05 to 0.21% of dry weight), and $Na^+$ (0.08 to 3.20% of dry weight). On the basis of halophyte community area in 2006, the amount of total nitrogen, $P_2O_5$ and $Na^+$ absorbed by Suaeda asparagoides was estimated up to 404,000, 47,000, and 498,000 kg, respectively. These results implied that halophyte communities are capable of both collecting significant dust particulates and absorbing of nitrogen, phosphorus, and sodium in the reclaimed land.

        Encapsulation of Various Guests by an Anionic In-Metal–Organic Framework Containing Tritopic BTB Ligand: Crystal Structure of Reichardt’s Dye Captured in an In-Metal–Organic Framework

        <P>The reaction between 1,3,5-benzenetribenzoic acid (H<SUB>3</SUB>BTB) and In(NO<SUB>3</SUB>)<SUB>3</SUB> hydrate in diethylformamide yielded a new In<SUP>III</SUP>-metal-organic framework, [(CH<SUB>3</SUB>CH<SUB>2</SUB>)<SUB>2</SUB>NH<SUB>2</SUB>]<SUB>3</SUB>[In<SUB>3</SUB>(BTB)<SUB>4</SUB>]·10DEF·14H<SUB>2</SUB>O (<B>I</B>). The countercation and solvent-free doubly interpenetrated <B>I</B> potentially contains 71.0% of solvent accessible void. Although the framework of <B>I</B> was not stable enough to maintain its original structure when the solvent molecules were removed, the as-prepared <B>I</B> was found to be a very good sorbent for acridine orange hydrochloride, a large Reichardt’s dye, and hydrophobic iodine molecule in solution. The as-prepared <B>I</B> exhibited increased uptake amount in the order of Reichardt’s dye > acridine orange hydrochloride > iodine. The largest uptake of the bulky Reichardt’s dye by <B>I</B> could be attributed to the optimized structural fitting of Reichardt’s dye into the large three-dimensional void space of <B>I</B>. The structure of Reichardt’s dye-encapsulated <B>I_RD</B> was unambiguously revealed by X-ray crystallography for the first time.</P><P>Thermal reaction between the tritopic ligand, 1,3,5-benzenetribenzoic acid (H<SUB>3</SUB>BTB), and In(NO<SUB>3</SUB>)<SUB>3</SUB> hydrate in diethylformamide yielded colorless crystals formulated as [(CH<SUB>3</SUB>CH<SUB>2</SUB>)<SUB>2</SUB>NH<SUB>2</SUB>]<SUB>3</SUB>[In<SUB>3</SUB>(BTB)<SUB>4</SUB>]·10DEF·14H<SUB>2</SUB>O (<B>I</B>).         Phenyl 2‐pyridyl ketoxime induces cellular senescence‐like alterations via nitric oxide production in human diploid fibroblasts

        <P><B>Summary</B></P><P>Phenyl‐2‐pyridyl ketoxime (PPKO) was found to be one of the small molecules enriched in the extracellular matrix of near‐senescent human diploid fibroblasts (HDFs). Treatment of young HDFs with PPKO reduced the viability of young HDFs in a dose‐ and time‐dependent manner and resulted in senescence‐associated β‐galactosidase (SA‐β‐gal) staining and G2/M cell cycle arrest. In addition, the levels of some senescence‐associated proteins, such as phosphorylated ERK1/2, caveolin‐1, p53, p16<SUP>ink4a</SUP>, and p21<SUP>waf1</SUP>, were elevated in PPKO‐treated cells. To monitor the effect of PPKO on cell stress responses, reactive oxygen species (ROS) production was examined by flow cytometry. After PPKO treatment, ROS levels transiently increased at 30 min but then returned to baseline at 60 min. The levels of some antioxidant enzymes, such as catalase, peroxiredoxin II and glutathione peroxidase I, were transiently induced by PPKO treatment. SOD II levels increased gradually, whereas the SOD I and III levels were biphasic during the experimental periods after PPKO treatment. Cellular senescence induced by PPKO was suppressed by chemical antioxidants, such as N‐acetylcysteine, 2,2,6,6‐tetramethylpiperidinyloxy, and L‐buthionine‐(<I>S</I>,<I>R</I>)‐sulfoximine. Furthermore, PPKO increased nitric oxide (NO) production via inducible NO synthase (iNOS) in HDFs. In the presence of NOS inhibitors, such as L‐NG‐nitroarginine methyl ester and L‐NG‐monomethylarginine, PPKO‐induced transient NO production and SA‐β‐gal staining were abrogated. Taken together, these results suggest that PPKO induces cellular senescence in association with transient ROS and NO production and the subsequent induction of senescence‐associated proteins<B>.</B></P>
