Photonic-crystal (PC) structures are used as biosensors that detect the changes in the surrounding refractive index due to biomolecular interactions. In this study, a method of modeling a PC structure that considers the sidewall deposition effect during high-index layer deposition was proposed to precisely predict the performance of a label-free PC biosensor. A PC composed of nanoreplicated grating and a TiO2 high-index layer was fabricated. To replicate nanograting, a silicon mold with a 450 nm pitch and a 100 nm grating height was fabricated via photolithography and reactive ion etching. A mold cavity was coated with a self-assembled monolayer, and UV replication was performed. To realize the PC structure, a TiO2 high-index layer was deposited using the E-beam evaporation system. To design the simulation model considering the sidewall deposition effect, the cross-sectional surface profile of each PC layer was measured. Finally, the changes in the transmission spectrum of the fabricated PC structure with different buffer solutions were measured and compared with the simulated results obtained from rigorous coupled wave analysis to examine the prediction accuracy of the proposed simulation model.

Han, Hyoung-Yun,Huh, Jung-Im,Han, So-Ri,Kang, Myung-Gyun,Yoon, Seokjoo,Han, Ji-Seok,Lee, Byoung-Seok,Kim, Jeong Ah,Min, Byung-Sun Elsevier 2018 Regulatory toxicology and pharmacology Vol.94 No.-

Abstract

Ephedrae Herba (EH) has been used in Asian traditional herbal medicine to cure bronchial asthma, cold, flu, chills, fever, headache, nasal congestion, and cough. In this study, we evaluated the subchronic toxicity of an Ephedrae Herba aqueous extract (EHAE) in male and female F344 rats. The EHAE was administered orally daily at doses of 0, 125, 250, 500, and 1000 mg/kg bw/day for 13 weeks. Toxicological assessment was performed to determine mortality, clinical signs, and changes in body weight, food consumption, ophthalmological, urinary, hematological, and serum biochemical parameters, macroscopic and microscopic evaluations, and organ weights. We found that oral administration of EHAE to F344 rats for 13 weeks resulted in histopathological changes in the kidneys and salivary glands. In the kidneys, increased incidence and severity of tubular basophilia were observed in females administered 1000 mg/kg bw/day of the extract. In the salivary glands, acinar cell hypertrophy was observed in males administered 500 mg/kg bw/day and in both sexes administered 1000 mg/kg bw/day of the extract. All test article-treated groups of males and females administered ≥250 mg/kg bw/day showed increased absolute and relative salivary gland weights. Therefore, the NOAEL (No Observed Adverse Effect Level) was determined as 125 mg/kg bw/day for both sexes of rats under the present experimental conditions.

Highlights

There is little data in the literature on the toxic effects of Ephedrae Herba.

Three males and two females in the highest dose group (1000 mg/kg bw/day) were found dead at weeks 1 and 9.

In macroscopic observation, test article-related changes were observed in the kidneys and salivary glands.

The no-observed-adverse-effect level was determined as 125 mg/kg bw bw/day for both sexes.

An Arabidopsis 관-glucosidase, AtBG1 is known to hydrolyze glucose-conjugated, biologically inactive abscisic acid (ABA) to produce active ABA, which increases the level of ABA in plants. Since an increase of ABA in plants confers tolerance against abiotic stress such as drought, we introduced the pCAMBIA3301 vector harboring the AtBG1 gene into creeping bentgrass through Agrobacterium-mediated transformation. After transformation, putative transgenic plants were selected using the BASTA resistance assay at a concentration of 0.8%. Genomic integration of the AtBG1 gene was confirmed by genomic PCR and Southern blot analysis, and gene expression was validated by Northern blot and Western blot analyses. Interestingly, the transgenic bentgrass plants overexpressing AtBG1 had a dwarf phenotype with reduced growth rates when compared to wild-type creeping bentgrass. In addition, the transgenic plants accumulated higher ABA levels and displayed enhanced drought tolerance. These results suggest that the expression of AtBG1 in plants induces the accumulation of higher ABA levels, which results in the formation of dwarf creeping bentgrass and enhances the survival in water-limiting environments.

X-linked recessive myotubular myopathy with MTM1 mutations
Han, Young-Mi,Kwon, Kyoung-Ah,Lee, Yun-Jin,Nam, Sang-Ook,Park, Kyung-Hee,Byun, Shin-Yun,Kim, Gu-Hwan,Yoo, Han-Wook The Korean Pediatric Society 2013 Clinical and Experimental Pediatrics (CEP) Vol.56 No.3
X-linked recessive myotubular myopathy (XLMTM) is a severe congenital muscle disorder caused by mutations in the MTM1 gene and characterized by severe hypotonia and generalized muscle weakness in affected males. It is generally a fatal disorder during the neonatal period and early infancy. The diagnosis is based on typical histopathological findings on muscle biopsy, combined with suggestive clinical features. We experienced a case of a newborn who required intubation and ventilator care because of profound hypotonia and respiratory difficulty. The preliminary diagnosis at the time of request for retrieval was hypoxic ischemic encephalopathy, but the infant was clinically reevaluated for generalized weakness and muscle atrophy. Muscle biopsies showed variability in fiber size and centrally located nuclei in nearly all the fibers. We detected an MTM1 gene mutation of c.1261-1C>A in the intron 10 region, and diagnosed the neonate with myotubular myopathy. The same mutation was detected in his mother.
