      • High-performance liquid chromatography method development for the quality control of Ginkgonis Semen

        <P>Ginkgonis Semen (GS) is the seed of Ginkgo biloba Linne and a valuable material for herbal medicines and functional foods in China, Japan and Korea. The main bio-compounds of GS are GA, GB and GC like the leaves. There are many studies for the analysis of ginkgolides in the leaves or leaves extract of G. biloba because the leaves extract is a valuable material in pharmaceutical industries. However, there is no efficient analytical method for the quality control of GS based on the quantitation of ginkgolides because of matrix effect induced by different chemical composition. So, there are no content criteria of GS in Pharmacopoeia of Korea, Japan and China until now. This study aimed to develop HPLC method using ginkgolides based on the quantitation of GA, GB and GC for the quality control of GS with the optimization of sample preparation to enhance the analytical sensitivity and reproducibility. At first, defatting process using petroleum ether and liquid-liquid extraction were applied for sample preparation to remove matrix effect. The HPLC-ELSD method was developed with the mobile phase of a 0.5% aqueous acetic acid and methanol-acetonitrile solution (1: 1 ratio) under gradient conditions. GA, GB and GC contents in GS were different between Korea and China. The mean quantity of Korean samples was 4.85 +/- 2.33 lg/g GA, 48.38 +/- 5.10 lg/g GB, and 37.83 +/- 7.64 lg/g GC. Those contents of Chinese samples were higher than Korean samples as 9.39 +/- 2.51 lg/g GA, 123.59 +/- 26.24 lg/g GB and 53.39 +/- 4.97 lg/g GC. It indicated that the discrimination of GS between Korea and China could be achieved by marker compound contents. Furthermore, the geographical discrimination of GS between Korea and China was confirmed by PCA using the quantitative data of marker compounds. By statistical analysis, the calculated content criteria of GS by regression method were 2.35 lg/g of GA, 29.20 lg/g of GB, and 27.75 lg/g of GC, based on dry weight. Thus, our HPLC method shows potential toward the development of a universal quality control methodology to quantify GS quality and origin. (C) 2016 The Authors.         고인슐린증이 없었던 인슐린종 1 예

        Insulinoma is a disorder characterized by hypoglycemia due to endogenous hyperinsulinemia. The elevated plasma insulin level and hypoglycemia should be established for the diagnosis of an insulinoma. Other laboratory methods such as immunoreactive insulin to glucose(IRI/G) ratio, C-peptide, proinsulin, and some provocation tests are also employed to detect an insulinoma because hyperinsulinemia is often not found. Especially, IRI/G ratio has been considered as very sensitive and specific test to confirm the insulinoma without peripheral hyperinsulinemia. We report a case of insulinoma without hyperinsulinemia and elevation of IRI/G ratio, which was diagnosed by endoscopic ultrasonography and THPVS(Transhepatic portal venous sampling). It is suggested that more confirmatory tests will be necessary for the diagnosis of insulinoma especially when it is highly suspected although IRI/G ratio is not increased(J Kor Soc Endocrinol 10: 439-444, 1995).

        A brushless DC motor has the high quality of torque output and silence, has been more widely used in industrial area. As the driver and controller of BLDC motor have been more complicated and precise, simulation method has been much used m motor design. And the output characteristics of BLDC motor is determined by the waveform of BACK-EMF in instinct. But because the conventional model of BLDC motor is obtained by approximation of real nonlinear waveform to ideal trapezoidal waveform, the error is occurred in slmulation result. Thus in this paper, for the correction of this error in simulation, the model of real nonlinear waveform considered is proposed, and the simulation result is obtained in case of three-phase, four-poles Y-connected, surface mounted permanent magnet BLDC motor.

      • Rgs19 regulates mouse palatal fusion by modulating cell proliferation and apoptosis in the MEE

        Palatal development is one of the critical events in craniofacial morphogenesis. During fusion of the palatal shelves, removal of the midline epithelial seam (MES) is a fundamental process for achieving proper morphogenesis of the palate. The reported mechanisms for removing the MES are the processes of apoptosis, migration or general epithelial-to-mesenchymal transition (EMT) through modulations of various signaling molecules including Wnt signaling. RGS19, a regulator of the G protein signaling (RGS) family, interacts selectively with the specific α subunits of the G proteins (Gαi, Gαq) and enhances their GTPase activity. Rgs19 was reported to be a modulator of the Wnt signaling pathway. In mouse palatogenesis, the restricted epithelial expression pattern of Rgs19 was examined in the palatal shelves, where expression of Wnt11 was observed. Based on these specific expression patterns of Rgs19 in the palatal shelves, the present study examined the detailed developmental function of Rgs19 using AS-ODN treatments during in vitro palate organ cultivations as a loss-of-function study. After the knockdown of Rgs19, the morphological changes in the palatal shelves was examined carefully using a computer-aided three dimensional reconstruction method and the altered expression patterns of related signaling molecules were evaluated using genome wide screening methods. RT-qPCR and in situ hybridization methods were also used to confirm these array results. These morphological and molecular examinations suggested that Rgs19 plays important roles in palatal fusion through the degradation of MES via activation of the palatal fusion related and apoptotic related genes. Overall, inhibition of the proliferation related and Wnt responsive genes by Rgs19 are required for proper palatal fusion.

      • 국내 해조류 자원의 항염증 및 세포독성 스크리닝 평가

        본 연구에서는 해조류 자원의 산업적 활용 소재로서의 가치 및 기능성 식품 및 사료 첨가 소재로의 개발을 위한 기초 자료를 제시하고자 국내에 서식하는 10종의 해조류 열수추출물을 이용하여 항염증 활성 및 마우스 대식세포주와 어류 세포주의 세포생존율에 미치는 영향을 확인하였다. 그 결과 S. japonica 100, 1000 ㎍/mL과 G. tenax 1000 ㎍/mL 농도에서 마우스 대식세포주를 LPS로 자극하였을 때 생성되는 NO를 유의적으로 감소시키는 것을 관찰하였으며, 나머지 8종의 해조류 추출물은 실험 농도에서 LPS에 의한 NO 생성에 영향을 미치지 않았다. 고농도에서 항염증 활성을 보인 S. japonica 및 G. tenax 열수추출물은 염증 인자를 조절할 수 있는 기능성 후보 소재로서 분획별 생리활성과 유효성분 규명의 연구가 요구된다. 본 연구 대상 해조류 추출물에 대한 안전성 기초 자료로서 마우스 대식세포 및 넙치 배아 유래 세포 세포생존율에 미치는 영향을 평가한 결과, P. tenera 고농도 (1000 ㎍/mL) 처리군을 제외하고 모든 해조류 추출물은 마우스 대식세포의 생존율에 영향을 주지 않았으며, 모든 해조류 추출물에서 5000 ㎍/mL까지 넙치 배아유래 세포에 대한 세포독성이 나타나지 않았다. 본 연구 결과는 후속 연구를 통해 해조류의 다양하고 복잡한 구조의 생물 활성 화합물의 생체 내 메커니즘을 규명함으로써 기능성 소재 개발을 위한 기초자로료 활용될 수 있을 것으로 기대하며, 본 연구의 대상 10종의 국내 해조류 자원 중 S. japonica과 G. tenax은 잠재적인 천연 항염증제로서 안전한 식품 및 사료 첨가 소재로서의 가능성을 제시한다. In this study, the anti-inflammatory and cytotoxic effects of hot-water extracts from 10 kinds of macroalgae in Korea were investigated. It was selected materials in consideration of biological activity and industrial potential as follows: Caulerpa okamurae; Codium fragile; Ulva australis; Ishige foliacea; Saccharina japonica; Sargassum horneri; Undaria pinnatifida; Gloiopeltis tenax; Gracilaria verrucosa; Porphyra tenera. Results showed that S. japonica and G. tenax significantly decreased NO productionn in LPS-stimulated Raw 264.7 cells at concentrations of 100, 1000 ㎍/mL and 1000 ㎍/mL, respectively. However, most of the other macroalgae used in the experiment did not affect NO production. It was observed that all macroalgae extracts except for the highest concentration (1000 ㎍/mL) treatment group of P. tenera did not affect the viability in Raw 264.7 cells. In addition, there was not significant decrease in cell viability by macroalgae extracts treatment in HINAE cells. These results suggest that S. japonica and G. tenax could be used as potential safe natural anti-inflammatory agents for food and feed additives. Also, the results of this study are expected to be used as basic data for the development of functional materials for 10 kinds of macroalgae resources in Korea.

      • Electroglottography를 사용한 한국어 폐쇄자음의 특성 및 임상적 적용

        An electroglottography (EGG) was used to investigate the function of the vacal folds during their vibration. In this study, four Korean native speakers and 10 vocal polyp patients were selected. To investigate the dynamic change of EGG waveforms for the three-way distinction of Korean stops, a DSP-Sona graph model 5500, a Rino-Laryngeal stroboscope, 3 CSL model 4300B and a Laryngograph were used. An EGG Model 4338 was used to exam the vocal polyp of patients' voices during high, low, comfortable pitch production. The purpose of this study is to investigate the characteristics of Korean stop consonants in relation to pitch and to observe laryngeal movement during vocal fold vibration and speech production. The basic data accumulated during this research can be applied in clinical treatment. The results are as follow: on the Korean stop consonants, the aspirated stop is the highest in the GOT and PC1. On the angle of vowel contour, the angle of lenis is smaller than the angle of heavily aspirated and glottalized stops. The fundamental frequency is lowest at the jenis stop. In vocal polyp patients', the low pitch range is smaller than in normal speakers'. The pitch break and the vocal fry were observed. The jitter and OQ value are higher in vocal polyp patients than in those of normal speakers'. Keywords: electroglottography, stop consonants, vocal polyp
