Safety Evaluation of Lactobacillus paracasei subsp. paracasei LC-01,a Probiotic Bacterium

        Hao Zhang,Yu Wang,Jing Sun,Zirui Guo,Huiyuan Guo,Fazheng Ren 한국미생물학회 2013 The journal of microbiology Vol.51 No.5

The safety of Lactobacillus paracasei subsp. paracasei LC-01was evaluated for its use as a potential probiotic. In our in vitro study, the antibiotic resistance and the ability to produce biogenic amine were determined. The results showed that the strain was sensitive to all tested antibiotics and did not produce biogenic amine except for tyramine. The oral toxicity of this strain was evaluated in Balb/C mice. One hundred mice were divided into 10 groups. Four groups were administered 0, 108, 109, or 1010 CFU/mouse per day dissolved in saline solution respectively, for 28 days. Three groups were injected intraperitoneally with 109 CFU/mouse dissolved in saline solution, and were killed 2, 5, and 10 days after injection. The last 3 groups were injected with the vehicle as controls respectively. The results showed that oral administration of the strain had no adverse effects on mouse body weight and that there was no treatment-associated bacterial translocation. Intraperitoneal administration caused a significant translocation to liver, spleen and kidney. However, this translocation did not cause illness or death throughout the experiment. The results suggest that L. paracasei subsp. paracasei LC-01 is likely to be safe for human consumption.

Lactobacillus paracasei subsp. paracasei LC01 Positively Modulates Intestinal Microflora in Healthy Young Adults

        Hao Zhang,Jing Sun,Xianting Liu,Chuan Hong,Yuanbo Zhu,Aiping Liu,Siqi Li,Huiyuan Guo,Fazheng Ren 한국미생물학회 2013 The journal of microbiology Vol.51 No.6

Lactobacillus paracasei subsp. paracasei LC01 (LC01) can tolerate intestinal stresses and has antioxidant activity. To evaluate the effect of the bacterium on human intestinal microflora,a randomized, double-blind, placebo-controlled human trial was carried out. Fifty-two healthy adult volunteers were randomized equally to two groups. One group consumed 12% (wt/vol) skimmed milk supplemented with 1010 CFU of LC01 each day for the 4-week treatment period, and then consumed placebo in the next treatment period, separated by a 2-week washout. The other group followed the reverse order. Group-specific real-time PCR and biochemical analyses was used to determine the intestinal bacterial composition of fecal samples collected at the end of every period, and the concentration of short-chain fatty acids and ammonia. A significant inhibition in fecal Escherichia coli and increase in Lactobacillus, Bifidobacterium, and Roseburia intestinalis were observed after consumption of LC01. Acetic acid and butyric acid were significantly higher in the probiotic stage and fecal ammonia was significantly lower. The results indicated a modulation effect of LC01 on the intestinal microflora of young adults, suggesting a beneficial effect on bowel health. LC01 may have potential value as a probiotic.

Multi-stage Nonlinear Model Predictive Control with Online Scenario Update for Semi-batch Polymerization Processes

        Jing-Gao Sun,Xian-Feng Chen,Guang-Hao Su,Meng Wang,Hong-Guang Pan 제어·로봇·시스템학회 2022 International Journal of Control, Automation, and Vol.20 No.10

In this paper, the problem of multi-stage nonlinear model predictive control with scenario update is investigated for semi-batch polymerization processes. The objective is to propose novel online scenario update schemes such that the more reasonable scenario tree can be generated. Firstly, based on the Orthogonal Configuration of Finite Elements (OCFE) method of direct radau configuration, the dynamic optimization problems are converted to Nonlinear Programing (NLP) problems such that the speed and accuracy of real-time optimization problem solving are effectively improved. Then, the scenario deviation is calculated based on model prediction information of each scenario and process measurement information. After that, calculate the bayesian probability weight of corresponding scenario is obtained. The online scenario reduction scheme uses the weight information update scenarios gradually reduce the scope of scenario tree representation. The online scenario weight update scheme uses the weight information as the basis for weight assignment of each scenario in the optimization problem. They use different methods to make the scenario tree modeling approach the real realization of uncertainty, and reduce the conservativeness compared with the traditional MSNMPC fixed scenario tree method. Through multiple batches numerical simulations of a semi-batch polymerization process, the advantages and effectiveness of the two proposed schemes are verified.

Effect of Sand Particle Size on Hydraulic-Mechanical Behavior of Bentonite-Sand Mixtures

        Tian-Hao Lu,Wen-Jing Sun,Ke Liu,Yun-Zhi Tan 대한토목학회 2022 KSCE JOURNAL OF CIVIL ENGINEERING Vol.26 No.8

Bentonite-sand mixtures can be used as the base material of engineered barriers in deep high-level radioactive waste geological repositories. Swelling, compression, and permeability tests were conducted out on bentonite-sand mixtures with 30%, 50%, 70% and 85% sand content. Influence of sand content and particle size on the deformation and permeability characteristics of bentonite-sand mixtures were analyzed. The sand particle size affects the deformation characteristics by influencing the vertical stress required to form a sand skeleton, and the skeleton’s stability. When the sand content is less than the critical sand content, bentonite-sand mixtures cannot form a sand skeleton, and particle size has no effect on their characteristics. When the sand content is greater than the critical sand content and the vertical stress exceeds the initial deviation stress, a sand skeleton forms. In this case, smaller the sand particle size, lesser will be the vertical stress required to form a sand skeleton, and more the swelling of bentonite-sand mixtures. Further, more uniform particle size, more stable will be its structure and lesser the compressibility of bentonite-sand mixtures. The hydraulic conductivity is related to the flow area and the seepage path length, and particle size has no obvious effect on either with the same sand content. Therefore, the sand particle size has no obvious effect on mixtures’ permeability.

      • Computed Tomography Manifestations of Histologic Subtypes of Retroperitoneal Liposarcoma

        Lu, Jing,Qin, Qin,Zhan, Liang-Liang,Yang, Xi,Xu, Qing,Yu, Jing,Dou, Li-Na,Zhang, Hao,Yang, Yan,Chen, Xiao-Chen,Yang, Yue-Hua,Cheng, Hong-Yan,Sun, Xin-Chen Asian Pacific Journal of Cancer Prevention 2014 Asian Pacific journal of cancer prevention Vol.15 No.15

Objective: Liposarcoma (LPS) is the most common soft tissue sarcoma and accounts for approximately 20% of all mesenchymal malignancies, often occurring in deep soft tissue of retroperitoneal space. Accurate preoperative diagnosis is therefore necessary. We explored whether computed tomography (CT) could be used to differentiate between the various types of retroperitoneal liposarcoma (RPLS). Method: Forty-seven cases of RPLS, diagnosed surgically and histologically, were analyzed retrospectively. CT features were correlated with postoperative pathological appearance. Results: The study radiologist identified 29, 11, 2, 2 and 3 RPLS as atypical lipomatous tumor/well-differentiated liposarcoma (ALT/WDL), dedifferentiated liposarcoma (DDL), myxoid/round cell liposarcoma (ML/RCL), pleomorphic liposarcoma (PL) and mixed-type liposarcoma. Analysis of CT scans revealed the following typical findings of the different subtypes of RPLS: ALT/WDL was mainly visible as a well-delineated fatty hypodense tumor with uniform density and integrity margin; DDL was marked by the combination of focal nodular density and hypervascularity. ML/RCL, PL and mixed liposarcoma showed malignant biological behaviour and CT findings need further studies. Conclusions: CT scanning can reveal important details including internal components, margins and surrounding tissues. Based on CT findings, tumor type can be roughly evaluated and biopsy location and therapeutic scheme guided.

China Consensus Document on Allergy Diagnostics

        Chen Hao,Li Jing,Cheng Lei,Gao Zhongshan,Lin Xiaoping,Zhu Rongfei,Yang Lin,Tao Ailin,Hong Haiyu,Tang Wei,Guo Yinshi,Huang Huaiqiu,Sun Jinlyu,Lai He,Lei Cheng,Liu Guanghui,Xiang Li,Chen Zhuanggui,Ma Ha 대한천식알레르기학회 2021 Allergy, Asthma & Immunology Research Vol.13 No.2

The prevalence of allergic diseases has increased dramatically in recent years in China, affecting the quality of life in 40% of the population. The identification of allergens is the key to the diagnosis of allergic diseases. Presently, several methods of allergy diagnostics are available in China, but they have not been standardized. Additionally, cross-sensitization and co-sensitization make allergy diagnostics even more complicated. Based on 4 aspects of allergic disease (mechanism, diagnosis procedures, allergen detection in vivo and in vitro as well as the distribution map of the most important airborne allergens in China) and by referring to the consensus of the European Society of Allergy and Clinical Immunology, the World Allergy Organization, and the important literature on allergy diagnostics in China in recent years, we drafted this consensus of allergy diagnostics with Chinese characteristics. It aims to standardize the diagnostic methods of allergens and provides a reference for health care givers. The current document was prepared by a panel of experts from the main stream of professional allergy associations in China.

Subdivision of Nasopharyngeal Carcinoma Patients with Bone-Only Metastasis at Diagnosis for Prediction of Survival and Treatment Guidance

        Xue-Song Sun,Yu-Jing Liang,Sai-Lan Liu,Qiu-Yan Chen,Shan-Shan Guo,Yue-Feng Wen,Li-Ting Liu,Hao-Jun Xie,Qing-Nan Tang,Xiao-Yun Li,Jin-Jie Yan,Lin-Quan Tang,Hai-Qiang Mai 대한암학회 2019 Cancer Research and Treatment Vol.51 No.4

Purpose The purpose of this study was to subdivide M1 stage nasopharyngeal carcinoma (NPC) patients with bone-only metastases for prognosis prediction while identifying the treatment effect of locoregional radiotherapy (LRRT) and metastasis radiotherapy (MRT) among patients with different risk. Materials and Methods From November 2006 to October 2016, a total of 226 patients with bone-only metastasic NPC were retrospectively enrolled. All patients developed distant lesions before receiving treatment. All potential prognostic factors were considered and the correlation of the M1 subdivisions with overall survival (OS) was determined by Cox regression hazards model. Kaplan-Meier curves were used to appraise survival condition and log-rank testing was used to compare the differences. Results The median follow-up time was 33.9 months (range, 3 to 126 months). According to multivariate Cox proportional hazard analysis, the number of metastatic lesions and Epstein-Barr virus (EBV) DNA status after palliative chemotherapy (PCT) were independent prognostic factors for OS. Thus, we subdivided patients into three risk groups according to these two factors. Systemic chemotherapy combined with LRRT may benefit patients in low- and intermediate-risk groups but not in the high-risk group. Further aggressive MRT based on systemic chemotherapy showed no survival benefit in any risk group. Conclusion The stratification of NPC patients with bone-only metastasis based on EBV DNA after PCT and the number of metastatic lesions provided promising prognostic value and could aid clinicians in person-specific treatment.

Anti-inflammatory effect of hispidin on LPS induced macrophage inflammation through MAPK and JAK1/STAT3 signaling pathways

        Han Ying-Hao,Chen Dong-Qin,Jin Mei-Hua,Jin Ying-Hua,Li Jing,Shen Gui-Nan,Li Wei-Long,Gong Yi-Xi,Mao Ying-Ying,Xie Dan-Ping,Lee Dong-Seok,Yu Li-Yun,Kim Sun-Uk,김지수,권태호,Cui Yu-Dong,Sun Hu-Nan 한국응용생명화학회 2020 Applied Biological Chemistry (Appl Biol Chem) Vol.63 No.3

Severe inflammatory reactions caused by macrophage activation can trigger a systemic immune response. In the present study, we observed the anti-inflammatory properties of hispidin on LPS induced RAW264.7 macrophage cells. Our results showed that hispidin treatment significantly reduced the production of cellular NO, IL-6 and reactive oxygen species (ROS) while has not inhibitory effect on TNF-α productions. Excitingly, hispidin treatment retains the phagocytosis ability of macrophages which enabling them to perform the function of removing foreign invaders. Signaling studies showed, hispidin treatment dramatic suppressed the LPS induced mitogen activated protein kinases (MAPK) and JAK/STAT activations. In conclusion, our findings suggest that hispidin may be a new therapeutic target for clinical treatment of macrophages-mediated inflammatory responses.

Patterns of Failure and Survival Trends in 3,808 Patients with Stage II Nasopharyngeal Carcinoma Diagnosed from 1990 to 2012: A Large-Scale Retrospective Cohort Study

        Xue-Song Sun,Di-Han Liu,Sai-Lan Liu,Qiu-Yan Chen,Shan-Shan Guo,Yue-Feng Wen,Li-Ting Liu,Hao-Jun Xie,Qing-Nan Tang,Yu-Jing Liang,Xiao-Yun Li,Jin-Jie Yan,Ming-Huang Hong,Jun Ma,Lin-Quan Tang,Hai-Qiang M 대한암학회 2019 Cancer Research and Treatment Vol.51 No.4

Purpose The purpose of this study was to investigate the survival trends and patterns of failure in patients with stage II nasopharyngeal carcinoma (NPC) treated with radiotherapy (RT) and chemotherapy over the last 20 years. Materials and Methods Thirty-eight hundred and eight patients diagnosed with stage II NPC between January 1990 and December 2012 were involved in this retrospective cohort study. All patients were treated with RT. According to the main imaging techniques and RT technology, we categorized these patients into four calendar periods: 1990-1996, 1997-2002, 2003-2007, and 2008-2012. Overall survival (OS), progression-free survival (PFS), locoregional relapse-free survival (LRFS), and distant metastasis–free survival (DMFS) were served as the clinical outcome. Results After a median follow-up period of 84.7 months, we observed increasing trends in survival and disease control. The 3- and 5-year OS rates increased from 87.1% and 78.7% in the first calendar period to 97.4% and 94.5% in the last calendar period, respectively (p < 0.001). Additionally, significant increasing trends could be seen in the PFS and LRFS during the four calendar periods. In the subgroup analysis, the LRFS in patients older than 50 years at diagnosis showed greater improvement than younger patients. However, the rate of distant metastasis was stable and relatively low, as the 5-year DMFS ranged from 90.5% to 94.7% among the four calendar periods. Conclusion The survival rates in patients with stage II NPC showed increasing trends from 1990 to 2012. The advance of RT provided excellent locoregional control and enhanced OS.
