        Heme oxygenase-1 mediates nicotine- and lipopolysaccharide-induced expression of cyclooxygenase-2 and inducible nitric oxide synthase in human periodontal ligament cells

J Periodont Res 2010; 45: 177–183. © 2010 John Wiley & Sons A/S</I></P><P>Background and Objective: </P><P>Although heme oxygenase-1 (HO-1) plays a key role in inflammation, its anti-inflammatory effects and mechanism of action in periodontitis are still unknown. This study aimed to identify the effects of HO-1 on the proinflammatory mediators activated by nicotine and lipopolysaccharide (LPS) stimulation in human periodontal ligament (PDL) cells.</P><P>Material and Methods: </P><P>The production of nitric oxide (NO) and prostaglandin E<SUB>2</SUB> (PGE<SUB>2</SUB>) was evaluated using Griess reagent and an enzyme immunoassay, respectively. The expression of inducible nitric oxide synthase (iNOS), cyclooxygenase-2 (COX-2) and HO-1 proteins was evaluated by Western blot analysis.</P><P>Results: </P><P>Lipopolysaccharide and nicotine synergistically induced the production of NO and PGE<SUB>2</SUB> and increased the protein expression of iNOS, COX-2 and HO-1. Treatment with an HO-1 inhibitor and HO-1 small interfering RNAs blocked the LPS- and nicotine-stimulated NO and PGE<SUB>2</SUB> release as well as the expression of iNOS and COX-2.</P><P>Conclusion: </P><P>Our data suggest that the nicotine- and LPS-induced inflammatory effects on PDL cells may act through a novel mechanism involving the action of HO-1. Thus, HO-1 may provide a potential therapeutic target for the treatment of periodontal disease associated with smoking and dental plaque.</P>

      • 부산지역 공무원 및 교직원의 당뇨병에 관한 역학적 조사연구

        조사대상자(공무원 및 교직원) 4,755명에 대한 당뇨병의 유병율은 2.9%(남 3.0%, 여 2.5%)이었으며 연령이 높을수록, 남자에서, 그리고 비만자의 경우, 당뇨병의 유병율이 각각 고율이었다 (P<0.05) Authors surveyed on the prevalence rate of diabetes and several factors related with the occurrence of diabetes on 4,755 numbers of public officials and school personnels in Busan for one year from July, 1982. The obtained results were as follows: 1.The prevalence rate of diabetes among total aubjects was 2.9% (Male 3.0%, Female 2.5%, and it was gradually increased with aging (P < 0.05). 2.The positive rate of lasting urine sugar was 2.7% among total subjects, and the negative rate of diabetic group was 78.3%. 3.The rate of obesity (120% and over of standard body weights in diabetic group wart higher than normal (P<0.05). 4.Among 76 diabetics, 58 (76.3%) numbers were 110mg/dl and over in postprandial 2 hour blood sugar level.

      • Trichloroethylene 취급 작업장의 공기 중 농도와 근로자의 요중 Trichloroacetic Acid 농도의 상관관계에 관한 조사연구

        부산ㆍ경남지역의 Trichloroethylene 취급 사업장 중 4개업종 20개소를 대상으로 기중 Trichloroethylene 농도를 측정하고, 이들 사업장 근로자 중 101명을 대상으로 요중 Trichloroaceticacid 농도를 분석하여 Trichloroethlene 폭로 정도를 평가하였다. Trichloroethylene, which is a chlorinated unsaturated hydrocarbon, is widely used as a solvent in degreasing operations. In the present study, the time-weighted average intensity of trichloroethylene exposure at work environments in 20 manufactures of 4 industries was assessed, at the same time trichloroacetic acid excretion in urine among 101 workers exposed to trichloroethylene was measured, and the exposure-excretion relationship was investigated. The results were summarized as follows; 1.The environmental trichloroethylene level was highest in metal machine industry(121.3ppm), followed by fabricated metal industry (106.0ppm), precision machine industry(65.0 ppm) and electrical machine industry(48.0 ppm). 2.The rate of urinary trichloroacetic acid concentration over 75mg/l among 101 workers was 42.6%(43 workers). 3.There is a linear correlation between the environmental exposure and urinary excretion(Y=1.05584X+35.65591, r=0.52401, p=0.0000).

      • 고무 및 화학제품제조업 산업장의 작업환경 실태에 관한 조사연구

        고무 혹은 화학제품제조업 산업장내 각종 유해작업환경 인자 가운데 특히 톨루엔ㆍ키실렌 등과 같은 유기용제가 산업보건학적 견지에서 보아 중요한 유해인자로 고려되었다. 따라서 고무 혹은 화학제품제조업 산업장과 그 근로자들에 대한 보건관리사업은 유기용제 중독의 예방과 조기치료에 중점을 두어야 할 것으로 생각된다. This study was conducted on some rubber and chemical products manufacturing industries of relatively larger sires and 3,356 workers in Busan area over a period of 4 months from June 1 to ,September 30, 1979. The purpose of the study was in investigating the status of working environment in rubber and chemical products manufacturing industries. The summarized results were as follows: 1.The highest and lowest mean values of harmful environmental elements in workroom of rubber products manufacturing industries were noted in cutting process (93.8 dB) and decorative designing(72.3 dB) in case of noise, vulcanizing (6.92 mg/m3) and rolling (5.55 mg/m3) in class II dust, spreading (883.3 ppm and 803.3 ppm, respectively) and decorative designing (155.0 ppm and 93.3 ppm, respectively) in toluene and xylene, and spreading (87.3 ppm) and assembling (61.0 ppm) ill benzene, respectively. Also in chemical products manufacturing industries, they were noted in culling process (96.2 dB) and inspecting (74.3 dB) in case of noise, cutting (9.07 mg/m3) and grinding (2.30 mg/m3) in class II dust, casting (160.0 ppm) and tube (71.7 ppm) in toluene. printing (133.3 ppm and 43.5 ppm respectively) and tube (66.7 ppm and 11.7 ppm, respectively) in xylene and benzene, respectively. 2.The workers of rubber products manufacturing industries who exposed to working environment above threshold limit value of harmful elements were highly noted in pressing process, cutting and compressing in case of noise, mixing, cutting and vulcanizing in class II dust, decorative designing and spreading in toluene and benzene, and spreading in xylene, respectively. Also in chemical products manufacturing, they were highly noted in cutting process, sheet and grinding in case of noise, mixing and cutting in class II dust, and printing and milling in toluene. xylene and benzene, respectively.

      • 소음성 청력장애에 관한 조사연구

        부산지역 일부 섬유, 고무 및 제강 산업장의 소음 작업부서 종사 근로자들중 6분법에 의한 청력장애(21dB이상)자는 조사대상의 6.30%이었다. Authors surveyed on the status of prevalence and several characters related with the occurrence of noise induced hearing impairment on 2,963 numbers of workers engaged in noisy department of twelve industries which were Textile, Rubber and Steel-Wire in Busan from July 1st, 1980 to September 30th, 1981. The obtained results were as follows: 1.About 90 percent of workers in noisy department engaged in the place of noise level less than 100dB(A). 2.The prevalence of occupational healing loss of workers engaged in noisy department by each level of hearing loss was respectively 5.43 percent at 21dB and above. 2.53 percent at 31dB and above, 0.80 percent at 41dB and above. 3.Workers of noise induced hearing impairment showed typical Cs-dip in Audiogram. 4.About 80 percent and above of workers of noise induced hearing impairment determined hearing loss of 40dB and below. 5.Hearing loss of workers of noise induced hearing impairment increased with aging. 6.Hearing loss of workers of noise induced hearing impairment was in accordance with the duration of employment in 7 years and below. 7.The occurred rate of noise induced hearing impairment using ELI(Early Loss Index) was about 92 percent(D-E scale).

      • A Molecular Cascade Showing NO-HO-1-VEGF-IL-8 Sequence in Human Endothelial Cells

        Heme oxygenase(HO)-1 has been shown to be an important biological target of nitric oxide(NO). NO can induce HO-1 expression and interleukin(IL)-8 production, particularly, in endothelial cells. Interestingly, HO-1 tends to induce the production of vascular endothelial growth factor(VEGF) that is involved in endothelial IL-8 syntheses. Whether HO-1 expression by NO may provide a link with IL-8 or VEGF synthesis was investigated in human umbilical vein endothelial cells(HUVECs). The NO donor S-nitroso-N-acetyl-penicillamine(SNAP) dose-dependently increased IL-8 and VEGF productions and HO-1 expression in HUVECs. Transfection with either HO-1 small interfering RNA or HO-1 antisense oligodeoxynucleotide abrogated the ability of SNAP to induce HO-1 expression and IL-8 and VEGF productions. Both pharmacological induction and gene transfer of HO-1 directly induced IL-8 and VEGF productions. Anti-VEGF neutralizing antibody blocked SNAP-mediated IL-8 production and VEGF itself induced IL-8 production, whereas anti-IL-8 neutralizing antibody had no effect on VEGF production in SNAP-treated HUVECs. Neither anti-VEGF nor anti-IL-8 antibodies influenced SNAP-induced HO-1 expression. Moreover, neither VEGF nor IL-8 showed an additive effect on SNAP-induced HO-1 expression. HO-1 transfection had no significant effect on productions of other CXC chemokines, such as growth-related oncogen-α and epithelial neutrophil activation peptide-78. Taken together, these results provide a molecular cascade showing NO-HO-1-VEGF-IL-8 sequence in human endothelial cells. This molecular cascade may be important in maintaining the angiogenic phenotype of the endothelium, and may provide opportunities to consider molecular targets to attenuate this biology.

        인진슬기의 간섬유화 억제 효과

        The inhibitory effects of Injinseulki including extracts of melanian snail plus phytomedicinal plants on $CCl_4$-induced fibrosis, $IFN-{\gamma}$ plus LPS-induced production of NO, PMA-stimulated production $O_2$, and tacrine-induced hepatic injury were investigated in rats, RAW 264.7 or HepG2 cells. The ethanol extracts of melanian snail significantly inhibited tarcrine-induced inury of HepG2 cells. Injinseulki inhibited the production of NO and $O_2$ in a dose-dependent manner in activated RAW 264.7 macrophages. Injinseulki significantly inhibited chemical-induced fibrosis in Rats. These results show that Injinseulki including extracts of melanian snail plus phytomedicinal plants may explain some known biological activities of Injinseulki including their anti-inflammatory and anti-fibrotic effect, and is of considerable benefit in the treatment for live diseases.

        Caspase-3을 경유한 동충하초 자실체 유래 4-Acetyl-12, 13-epoxyl-9-trichothecene-3, 15-diol의 방광암 세포주(NBT-II) Apoptosis

        The fruiting bodies of Isaria japonica have been traditionally used in Korea to treat cancer. An apoptosis-inducing compound, 4-Acetyl-12, 13-epoxyl -9-trichothecene-3, I5-diol (AETD), was isolated from the methanol extract of fruiting bodies of Isaria japonica Yasuda by bioassay -guided fractionation. The apoptosis of murine bladder cancer cell line (NBT-Ⅱ) by the compound was accessed by propidium iodide staining flow cytometric analysis, and apoptosis-inducing activity at $IC_{50}$ concentration (5 nmol/L) was further confirmed by a nuclear morphological change, a ladder pattern of DNA fragmentation, and an activation of caspase-3. These results indicate that AETD induces apoptosis of NBT-Ⅱ cells via expression of caspase-3.
