구강 악성 흑색종에서 PCNA 발현에 관한 면역조직화학적 연구

Malignant melanoma arising from the mucosa of head and neck region was rare and showed poor prognosis. Some of malignant melanoma were transformed from benign melanotic lesion. Malignant melanoma had high cellular proliferation and rapid growth. The percentage of PCNA-positive cell (labeling index) is high in many malignant tumor. So we compared the pattern of PCNA expression in the melanotic lesion. We performed the immunohistochemical study in malignant melanoma(19 cases), benign melanotic macule(24 cases) and normal mucosa(20 cases) were diagnosed in Seoul National University Dental Hospital between 1980 and 2000. Positive PCNA staining was found mainly in malignant melanoma. The mean PCNA expression in malignant melanoma, melanotic macule, normal tissue were 29.2%, 1.4%, 0%, respectively. Significant differences in PCNA expression were noted between malignant melanoma and melanotic macule(p<0.01), normal mucosa(p<0.01). These result suggested that the PCNA expression seems to be used as a diagnostic indicator for malignancy in malignant melanoma and melanotic lesion.

More than 7,000 ton of sewage sludge was produced daily in korea, of them more than 80 percent reclaimed in land and dumped into ocean, while less than 7 percent were recycled. In this study, geotechnical and environmental laboratory tests were accomplished to evaluate material characteristics of solidified sewage, which fermented first, as possible civil construction materials, Geotechnical tests included liquid and plastic limit test, sieve analysis, compaction test, and shear strength thest, while environmental tests were pH test, heavy metal content and elution test. The results of tests said that geotechnical characteristics of solidified sewage sludge were satisfied the standard specification of covering materials for waste dumping ground, pH test said that pH of solidified sewage sludge was about 7.7 which is weak alkaline, Each elution concentration of heavy metals, which are Cd, Cu, Pb, Cr, and Zn, were also satisfied with the geotechnical and environmentally safe as civil construction material such as covering materials for waste dumping ground and seaside reclamation materials.

목 적: 저자들은 제3형 개방성 경골 골절로 수술적 치료를 시행한 환자의 결과를 분석하여 이에 대한 치료 지침을 제시하고자 한다. 대상 및 방법: 최근 7년간 본원에서 치료한 환자 42례를 대상으로 하였으며 연령 분포는 15세에서 76세로 남자가 35례, 여자가 7례였으며, 골절의 분류는 Ⅲa형 14례, Ⅲb형 20례, Ⅲc형 8례였다. 최종 기능적 평가는 Tucker의 분류법을 사용하였다. 결 과: 골 유합 기간은 Ⅲa이 9.2개월, Ⅲb형11.0개월, Ⅲc형이 13.8개월이었고 치료 기간 중 회전 피판 이식술과 유리 피판 이식을 시행하였으며 Ⅲc형에서는 골 연장술을 시행하였다. 기능적 평가로는 Ⅲc형로 갈수록 불량하였다 결 론: 비활성 조직의 충분한 제거, 노출 골편의 조기 피복 후 Ⅲa형에서는 골수강내 금속정을 이용한 견고한 내고정으로 좋은 결과를 얻을 수 있고 Ⅲb와, Ⅲc형에서는 외고정 기구를 사용하여 견고한 고정력을 얻을 수 있을 것으로 사료된다. Purpose: The purpose of this study was to analyze the functional outcome and propose the guidelines in the surgical treatment of open type Ⅲ tibial fracture. Materials and Methods: We have analyzed the treatment results of 44 cases; age was ranged from 15 to 76 years, and there were 35 males and 7 females. The type of fracture according to the classification by Gustilo revealed typeⅢa 14 cases, type Ⅲb 20 cases and type Ⅲc in 8 cases. We analyzed functional outcome according to the classification of functional result by Tucker. Results: The average union time of type Ⅲa was 9.2 months, Ⅲb 11.0 months, and Ⅲc was 13.8 months. The rotational flap and free flap were done during treatment and bone lengthening especially in type Ⅲc. Functional results were poor especially in type Ⅲc. Conclusion: Debridgement of devitalized tissue, early soft tissue coverage and sufficient stability using intramedullary nails will be necessary in type Ⅲa fracture. Rigid external fixation, early soft tissue coverage by rotational muscle flap and free flap reduced infection rate with satisfactory functional outcome especially in type Ⅲb and Ⅲc fractures.
12-O-Tetradecanoylphorbol-13-Acetate에 의한 HL-60 세포 분화유도중 Vimentin 유전자 전사조절에 대한 AP-1의 역할

Purpose: To gain insight on the role of AP-1 in transcriptional regulation of vimentin gene during differentiation of HL-60 cells by 12-O-tetradecanoylphorbol-13-acetate (TPA), the levels of vimentin mRNA and AP-1 have been investigated with Northern blot hybridization and DNA mobility shift assay. Materials and Methods: HL-60 cells were grown in RPMI 1640 medium supplemented with 10% heat-inactivated fetal calf serum and antibiotics in a humidified 5% CO_2 at 37℃. Total RNA was prepared by a modification of the method of Karlinsey et al. Northern blot hybridization was performed by the method of Virca et al. EcoRI fragment of pVIM-GEM was used as probe for vimentin mRNA. DNA mobility shift assay was performed by the method of Lim et al. End labeled DNA probe(Upper strand, 5'-CGCTTGATGAGTCAGCCG- 3') for AP-1 binding activity was mixed with nuclear extracts in a 20 μl reaction volume containing 300 mM KCI, 60 mM HEPES, pH 7.9, 25 mM MgCI_2, 1 mM EDTA, 1 mM DTT, 60% glycerol, and 2 ㎍ of poly[dI-dC]. Results: TPA increased vimentin mRNA levels, with maximal stimulation reached at 24 hr. The level of vimentin mRNA was induced in proportion to the concentration of TPA. TPA-induced vimentin mRNA was almost reduced by actinomycin-D pretreatment. TPA-induced stimulation of vimentin gene was completely reduced by staurosporin pretreatment. In DNA mobility shift assay, AP-1 newly appeared at 24 hr during TPA-induced differentiation and was almost not detected after the pretreatment of staurosporin. Conclusions: These results suggest that the induction of vimentin mRNA during TPA-dependent differentiation in HL-60 cells may be mediated by protein kinases C signal transduction and AP-1 is important to transcriptional regulation.
