        MicroRNA 452 regulates ASB8, NOL8, and CDR2 expression in colorectal cancer cells

        Background MicroRNAs play important roles in the pathogenesis of human diseases by regulating target gene expression in specifc cells or tissues. Previously, we identifed microRNA 452 (MIR452), which was specifcally up-regulated in early stage human colorectal cancer (CRC) tissue. Objective The current study aims to identify and verify the target genes of MIR452 associated with CRC. Methods A luciferase reporter system was used to confrm the efect of MIR452 on ASB8, NOL8, and CDR2 expression. The expression levels of MIR452 and the target genes were evaluated by quantitative RT-PCR (qRT-PCR) and western blotting. Results We verifed the association between MIR452 and three genes, ASB8, NOL8, and CDR2, and showed that their transcripts were down-regulated by MIR452. Up-regulated MIR452 also down-regulated ASB8, NOL8, and CDR2 mRNA and protein levels in CRC cells. CDR2 protein expression was decreased in CRC tissues compared to adjacent non-tumor tissues. Conclusions These results suggest that ASB8, NOL8, and CDR2 were target genes of MIR452 in CRC cells and that upregulated MIR452 in CRC tissue regulated ASB8, NOL8, and CDR2 expression during colorectal carcinogenesis.

      • Newcastle Disease Virus의 Fusion 단백 유전자의 제한효소분절형태(RELP)의 특이성

        To evaluate the specificity of restriction fragment length polymorphism (RFLP) of Newcastle disease virus (NDV) fusion (F) protein gene for various NDV strains. F protein genes of NDV CBP-1 isolate, LaSota, B1 and Kyojungwon (KJW) were amplified by reverse transcriptase-polymerase chain reaction (RT-PCR) and RFLP were examined. In RT-PCR using F primer sets for 1.7 kb F gene, the amplified products were observed in CBP-1, LaSota and B1 except KJW. However, F2 primer set for 521 bp yielded the amplified bands with all strains. The amplified F genes (1.7 kbp) were treatd with 15 different restriction enzymes to analyze RFLP and the cleavage patterns. CBP-1 showed various cleavage patterns with 8 enzymes, LaSota, with 11 enzymes and B1, with 11 enzymes. According to the cleavage patterns, the physical maps were constructed, and it was found that CBP-1, LaSota and B1 strains had specific cleavage sites with HinfI, XbaI and XhoI. These findings suggest that RFLP are worth applying in the diagnosis and molecular epidemiological studies of Newcastle disease.

Proceeding of International Symposium on East-West Medicine Seoul. 187-200, 1999.-Twirling the needle (TN) is one of several needing methods frequently used for acupuncture in the oriental medicine. The present study was conducted to see if TN enhanced the antinociceptive effect produced by plain-or electroacupuncture in the rat tail flick test. Rats (Sprague-Dawley, 200-300g)lightly anesthetized with thiopental sodium(40mg/kg, i.p.) were used in the tail-flick test. The basal reaction time for tail-flick was 2 sec. For "plain" acupuncture(PA), a needle was inserted for 20 min into a Chock-samni (ST36) acupuncture point which (is located at the anterior tibial muscle and about 10mm below the knee joint). For electroacupuncture(EA), train-pulses(3 Hz,0.3-ms pulse width, 0.2-0.3mA) were applied to the inserted needle for the period of insertion. For combining TN with either PA or EA, TN was performed during the each acupuncture period. Two methods for TN were used; twirling 3 times for 1 second persisted for 1 min in every 5 mins(long-duration and long-interval;LDLI) and twirling 3 times for 1 second persisted for 10 sec in every 1 min(short-duration and short-interval;SDSI). PA and EA produced an increase in tail flick latency (TFL); peak increases were 21.3±5.1% and 42.1±17.3% of the pre-acupunture control, respectively, and occurred immediately after cessation of 20min-acupuncture. Performing TN combined with PA increased TFL more than PA itself by showing a greater peak increase in TFL when performed with a SDSI-TN than with a LDLI-TN(60.6±12.5% and 42.7±22.7% of the pre-acupuncture control, respectively). Performing TN combined with EA also increased TFL more than EA itself and a greater peak increase in TFL was observed with a SDSI-TN as compared to a LDLI-TN(77.5±13.8% and 67.3±14.0% of the pre-acupuncture control, respectively). These results indicate that TN enhances both PA-and EA-produced antinociception where an greater enhanced antinociception than a LDLI-TN. It is suggested that performing a SDSI-TN combined with EA seems to be one of the most effective ways to produce analgesia.

      • 대전광역시 대전천 물로부터 분리한 고온성, 호염성 야생효모, Pichia spartinae WJSL0087의 생육 특성

        대전광역시 대전천 주변의 물에서 분리한 고온성이며 호염성인 Pichia spartinae WJSL0087의 생육특성을 조사하였다. Pichia spartinae WJSL0087는 yeast extract-peptone-dextrose 배지에서 생육이 양호하였고 37℃와 15% NaCl 함유 YPD 배지에서 생육하는 고온성, 호염성 효모이었다. 또한, Pichia spartinae WJSL0087는 포도당 30%, 10% 에탄올 함유 배지에서도 생육하였고 2가의 Co 와 Cu, 1가의 리튬 양이온들에 대하여 500ppm까지, NaCl, KCl등은 15%까지 내성을 보였다. The goal of this study was to investigate growth characteristics of the thermpphilic and halotolerant wild yeast, Pichia spartinae WJSL0087 from freshwater of riverside in Daejeoncheon, Daejeon Metropolitan city, Korea. Pichia spartinae WJSL0087 grew yeast extract-peptone-dextrose (YPD) medium and also grew in 15% NaCl-YPD medium at 37℃. The strain were resistrant to 30% glucose, 10% ethanol, !5% KCl and CaCl2 and further, also grew in the 500 ppm Co, Cu and Li metal ions-containing YPD medium, respectively.

      • 하수슬러지 발효고화에 의한 Biosoil의 매립장 복토재 Lysimeter 평가

        현재 하수슬러지의 발생양은 매년 증가하는 추세로 함수율이 높은 유기성 하수 슬러지는 2차 오염을 유발시키고 처리, 처분 단계에서 여러 가지 방법이 논의되고 있지만, 합리적인 처리가 어려워 세계적으로 중요한 과제로 떠오르고 있다. 이에 하수슬러지의 최종처분 및 재활용 방안 마련을 위하여, 하수슬러지를 발효·고화시킨 Biosoil을 매립지의 복토재로 사용하기 위해 현재 매립장에서 복토재로 일반적올 사용되고 있는 토양을 비교, 검토하고자 하였다. 하수 슬러지를 고화, 발효시킨 Biosoil과 일반 복토재로 사용하고 있는 흙을 Lysimeter를 통해 비교 연구하여 복토재로서의 재활용 가능성을 평가해 보고자 하였다. 이에 본 연구에서는 실험 재료의 물리, 화학적 성질을 알아보기 위해 매립전후 일반 토양과 Biosoil의 유기물량, pH. 등을 측정하였으며, 동일한 Lysimeter 2기에 각각 일반토양과 Biosoil을 복토하여 비교실험을 하였다. 장치 내부에는 매립장과 동일하게 생활쓰레기를 성상 분석하여 충진 하였고, 인공강우를 주기적으로 살수하였다. 연구 결과 매립전후 일반토양의 유기물량은 일반흙과 비슷하였으나 Biosoil은 매립 후 유기물량은 다소 낮아졌으며 pH는 매립후 일반토양, Biosoil 모두 낮아졌다. 또한 토질시험결과 Biosoil의 투수게수는 5.68 x 10^(-6)cm/sec로 매립장 복토재로서의 조건을 충분히 만족하는 것으로 나타났다. 실제로 복토후의 모습을 관찰하기 위해 실외에서 실시한 기상실험에서도 건조한 하수슬러지에 비해 재슬러지화가 일어나지 않았으며, 일반흙과 비슷하게 다져지게 나타났다. 위와 같이 하수슬러지를 재활용한 복토재로의 가능성을 살펴본 결과 복토재로서의 사용가능성을 보이고 있으며, 더 많은 연구를 통해 재활용 복토재로의 사용가능성을 더높여야 할 것으로 판단된다.

        본 논문은 1997년에 발생한 아시아 경제위기에 대해서 고찰하였다. 한국이 IMF의 구제 금융을 받게된 것을 계기로 IMF는 아시아 경제위기의 치유에 주도적인 역할을 맡게 되었다. 그러나 IMF의 구제금융은 아시아의 경제위기를 해결하지 못하였다. 구제금융의 지원에도 불구하고 이들 3개국의 신용등급은 더욱 멀어졌고 통화가치는 빨리 떨어져 외채위기는 더욱 증폭되어 IMF의 목표가 완전히 실패로 돌아갔다.

        Taekwondo became a world-wide sport beyond the symbol of Korean traditional culture. Taekwondo go through its value and put love for humanity and love of peace in practice in order to share love and participation with a citizen of the world. There are three essential prerequisite essentials for Taekwondo to be loved by people from all of the world in a globalizational era. Those are renovation, reinforcement, and solidarity and it can be practiced through revision of match rules, improvement of equipment, continuance of exhibition tour, and dispatch of masters.Also, educational values of Taekwondo is very important to achieve peace and happiness of all mankinds. Those are follows;First, Taekwondo has a basis of education for the whole man. Second, Taekwondo masters have authority as a leader. Third, Taekwondo training places are educational sites.       • 유산소성 운동이 비만 남고생의 전신지구력 향상에 미치는 영향

        While measuring 1000m running record of high school students, I witnessed that corpulent students feel harder than ordinary students do while running and confirmed that the 1000m running record of the corpulent students is poor as compared with that of ordinary students. I felt a sense of mission that it is required to take measures for the corpulent students, and conducted this study under the understanding of the corpulent students themselves, their class teachers and parents. The conclusion of this study is as follows: 1)The corpulent students who intend to do physical exercise but really carry out their intention have successfully participated in this program as the instruction programs that consider their life style, physical conditions and preference of physical exercise have been effectively implemented including relatively small mountain climbing. 2)Amusing games have been offered to the corpulent students so that they are accustomed to do warming-up and finishing exercise as well as reinforcing exercise form time to time. And relatively over-corpulent students could improve their physical endurance by climbing mountain in a systematic manner.
