        Reversibility and safety of KISS1 metastasis suppressor gene vaccine in immunocastration of ram lambs

        Objective: The aim of this study was to investigate the reversibility and safety of KISS1 metastasis suppressor (KISS1) gene vaccine in immunocastration. Methods: Six eight-week old ram lambs were randomly divided into vaccinated and control groups. The vaccine (1 mg/ram lamb) was injected at weeks 0, 3, and 6 of the study. Blood samples were collected from the jugular vein before primary immunization and at weeks 2, 4, 6, 10, 14, 22, and 30 after primary immunization. All ram lambs were slaughtered at 38 weeks of age, and samples were collected. Results: The specific anti-KISS1 antibody titers in vaccinated animals were significantly higher and the serum testosterone level was significantly lower than those in the control groups from week 4 to 14 after primary immunization (p<0.05). No significant difference was observed at weeks 22 and 30 after the primary immunization. Similar results were also found for scrotal circumference, testicular weight, length, breadth, and spermatogenesis in seminiferous tubules in week 30 after primary immunization. KS (KISS1-hepatitis B surface antigen S) fusion fragment of KISS1 gene vaccine was not detected in host cell genomic DNA of 9 tissues of the vaccinated ram lambs by polymerase chain reaction. Conclusion: The effects of KISS1 gene vaccine in immunocastration were reversible and no integration events were recorded.

      • Prognostic Significance of Beclin-1 Expression in Colorectal Cancer: a Meta-analysis

        Objective: Beclin-1 has recently been observed as an essential marker of autophagy in several cancers. However, the prognostic role of Beclin-1 in colorectal neoplasia remains controversial. Our study aimed to evaluate the potential association between Beclin-1 expression and the outcome of colorectal cancer patients. Materials and Methods: All related studies were systematically searched in Pubmed, Embase, Springer and Chinese National Knowledge Infrastructure databases (CNKI), and then a meta-analysis was performed to determine the association of Beclin-1 expression with clinical outcomes. Finally, a total of 6 articles were included in our analysis. Results: Our data showed that high Beclin-1 expression in patients with CRC was associated with poor prognosis in terms of tumor distant metastasis (OR=2.090, 95%CI=1.061-4.119, p=0.033) and overall survival (RR=1.422, 95%CI=1.032-1.959, p=0.031). However, we did not found any correlation between Beclin-1 over-expression and tumor differentiation (OR=1.711, 95%CI=0.920-3.183, p=0.090). In addition, there was no evidence of publication bias as suggested by Egger's tests for tumor distant metastasis (p=1.000), differentiation (p=1.000) and OS (p=0.308). Conclusions: Our present meta-analysis indicated that elevated Beclin-1 expression iss associated with tumor metastasis and a poor prognosis in patients with CRC. Beclin-1 might serve as an efficient prognostic indicator in CRC, and could be a new molecular target in CRC therapy.

        Effect of Prior Microstructures on Cementite Dissolution Behavior During Subcritical Annealing of High Carbon Steels

        The variation of the morphology and distribution of cementite particles in different prior structures with spheroidizingannealing process has been proceeded in this paper. It is found that the dissolution and coarsening progresses of variousinitial structures in spheroidizing annealing process are quite asynchronous due to the different interlamellar spacing. Thedissolution rate of degenerated pearlite (D-P) with finer interlamellar spacing is faster. The granular cementite in the spheroidizedmicrostructure is fine, uniform and dense. The mean diameter of spherical cementite is refined to 233 nm. However,the initial structure of degenerated pearlite (D-P) is highly sensitive to the austenitization temperature. The cementite iseasy to be coarsened under high austenitizing temperature. The coarsening is accompanied by the gradual increase of theCr content in the cementite, which increases the stability of the cementite. Therefore, the optimal austenitizing temperaturefor degenerated pearlite (D-P) is suggested to be 770 °C.

        Fractional-order control of active suspension actuator based on parallel adaptive clonal selection algorithm

        In current study, a new approach for vehicle active suspension actuator control is reported. For this approach, a nonlinear model for an electro-hydraulic servo actuator and quarter suspension system is established. An Oustaloup recursive approximation (ORA) Fractionalorder proportional–integral–derivative (FOPID) controller is designed to control the displacement of the actuator, and controller parameters are optimized based on error integral criteria using the Parallel adaptive clonal selection algorithm (PACSA). The research indicates that PACSA can tune parameters of the FOPID controller accurately and efficiently, and the tuned FOPID controller performs better in terms of response speed and tracking accuracy than a regular Proportional–integral–derivative (PID) controller. The performance of the active suspension system that uses the FOPID actuator is obviously superior to that of the active suspension system that uses a PID actuator and a passive suspension system.

        EFFECTS OF PREPARATION PARAMETERS ON ONE-DIMENSIONAL CdS NANOSTRUCTURES BY DIPHENYLTHIOCARBAZONE-ASSISTED SOLVOTHERMAL METHOD

        One-dimensional (1D) single crystalline CdS nanostructures have been successfully synthesized via a diphenylthiocarbazone-assisted solvothermal route. The results revealed that the microstructure and optical absorption properties of 1D CdS nanostructures were temperature and time dependent owing to thermodynamically and kinetically controlled growth. Single crystalline CdS nanowires with a diameter of 80 nm and length of 20 μm were synthesized at 180°C for 96 h. At moderate temperature (180°C), the morphology of the products transformed from irregular short rods to uniform long rods as the reaction time prolonging in a kinetically controlled growth regime. Only nanorods were obtained at a temperature as low as 120°C even extending reaction time to 260 h due to thermodynamic limited growth. At high temperature (250°C in this system), the products were nanowires with larger diameter but lower aspect ratio since the growth rates on both lateral and axial directions were accelerated. Moreover, the optical absorption spectra revealed that the CdS nanowires showed a blue shift compared with bulk CdS. Two optical absorption peaks appeared due to the nanometer effect in the radial and lengthwise directions of CdS nanowires. The growth mechanism of 1D CdS nanostructures was discussed.

        Immobilization of lipase onto aminopropyl-functionalized MSU-H type mesoporous silica and esterification

        Candida rugosa lipase (CRL) was immobilized on an aminopropyl-functionalized MSU-H type mesoporous silica (AFMS) through physical adsorption and a covalent cross-linking. It was evaluated as a class of biocatalysts in the esterification of conjugated linoleic acid (CLA) isomers with ethanol. AFMS materials with varied content of aminopropyl were prepared by a simple co-condensation at near neutral pH condition. Introduction of aminopropyl chains and CRL molecules onto the AFMS supports was confirmed by Fourier transform infrared (FT-IR) spectra. CRL was immobilized on the AFMS through electrostatic and covalent interactions. The covalently cross-linked CRL gave a loading amount of 34.3mg CRL/g-support and a hydrolytic activity of 2471.5U/g-catalyst. It exhibited high operational stability and remained 23.9-27.5% of total esterification in 32 h consecutive four runs in the esterification of CLA with ethanol. Moreover, the immobilized CRLs catalyzed 2.8-3.8 times of esterification of cis-(c)9, trans-(t)11- CLA faster than that of t10, c12-CLA.

        Two new tirucallane triterpenoids from the leaves of Aquilaria sinensis

        Two new tirucallane triterpenoids, aquilacallanesA–B (1–2), together with 15 known compounds(3–17) were isolated from the leaves of Aquilaria sinensis. The structures of these new compounds were elucidated onthe basis of extensive spectroscopic analyses. All compoundswere evaluated for their cytotoxic activity againstfive human cancer cell lines. The known compounds,ursolic acid (7) and 5,7,40-trimethoxyflavone (14), exhibitedweak cytotoxic activity against some cells.

        Boeravinone B, a natural rotenoid, inhibits osteoclast differentiation through modulating NF-κB, MAPK and PI3K/Akt signaling pathways

        Osteoporosis is a major public health concern, which requiresnovel therapeutic strategies to prevent or mitigate bone loss. Natural compounds have attracted attention as potential therapeuticagents due to their safety and efficacy. In this study, weinvestigated the regulatory activities of boeravinone B (BOB), anatural rotenoid isolated from the medicinal plant Boerhaviadiffusa, on the differentiation of osteoclasts and mesenchymalstem cells (MSCs), the two main cell components responsiblefor bone remodeling. We found that BOB inhibited osteoclastdifferentiation and function, as determined by TRAP stainingand pit formation assay, with no significant cytotoxicity. Furthermore,our results showing that BOB ameliorates ovariectomyinducedbone loss demonstrated that BOB is also effective invivo. BOB exerted its inhibitory effects on osteoclastogenesis bydownregulating the RANKL/RANK signaling pathways, includingNF-κB, MAPK, and PI3K/Akt, resulting in the suppression ofosteoclast-specific gene expression. Further experiments revealedthat, at least phenomenologically, BOB promotes osteoblast differentiationof bone marrow-derived MSCs but inhibits their differentiationinto adipocytes. In conclusion, our study demonstratesthat BOB inhibits osteoclastogenesis and promotes osteoblastogenesisin vitro by regulating various signaling pathways. Thesefindings suggest that BOB has potential value as a novel therapeuticagent for the prevention and treatment of osteoporosis.

        CD64 Expression Is Increased in Patients with Severe Acute Pancreatitis: Clinical Significance

        Background/Aims: Upregulated CD64 expression on neutrophils is the most useful marker for acute bacterial infections and systemic inflammation. However, it is unknown whether CD64 is involved in the pathogenesis of acute pancreatitis (AP). This study was designed to determine whether CD64 is implicated in severe acute pancreatitis (SAP), and thus, is a suitable marker for SAP. Methods: SAP was induced in rats with an intraperitoneal injection of L-arginine. CD64 expression in the rat pancreas was determined by quantitative real-time polymerase chain reaction (qRT-PCR) and immunohistochemistry. Additionally, the CD64 mRNA expression in peripheral blood leukocytes from 21 patients with mild acute pancreatitis (MAP) and 10 patients with SAP was investigated at the time of admission and during remission by qRT-PCR. Results: CD64 mRNA and protein expression in the pancreas was significantly higher in rats with SAP, compared to the controls. The CD64 expression was higher in the patients with SAP than in the patients with MAP. During remission, CD64 mRNA decreased in both the MAP and SAP patients. The area under the curve of CD64 expression for the detection of SAP was superior to both the Ranson and the Acute Physiology and Chronic Health Evaluation II scores. Conclusions: The CD64 level was significantly increased in correlation with the disease severity in SAP and may act as a useful marker for predicting the development of SAP.         Study on host plants for reproduction of Chilo suppressalis

        The rice stem borer, Chilo suppressalis, is generally considered to be a polyphagous pest. The current study challenges this view by investigating its oviposition preference, larval survival and development on different host plants under both laboratory and field conditions. Rice and water-oat populations of the borer inhabiting on corresponding plants respectively have partial reproductive isolation based on previous studies. In a laboratory multiple-choice test, C. suppressalis adults from rice population laid most of eggs on water-oat (46.5% of total eggs) and rice (43.8%), with very fewlaid onwheat (3.6%), sugarcane (4.0%) andmaize (2.0%). Field surveys supported the laboratory study and found no egg on the plants other than rice and water-oat. Neonate inoculation experiments performed in field and laboratory showed that larval survival rate was much higher on rice (49.1%–51.2%) and wheat (36.5%–44.1%) than that on water-oat (10.7%–10.8%), maize (1.2%–7.2%), sugarcane (0–1.5%) and weeds (2.4%). These results were discussed with the data reported from water-oat population and it was concluded that C. suppressalis is not a typical polyphagous pest. Rice population mainly reproduces on rice and use water-oat only as minor host, and water-oat population breeds better on water-oat than on rice as reported. Neither population could thrive on the other recorded host plants, which are used for the supplementary nutrition sources of larvae. These findings provide useful information for the development of control strategies to prevent C. suppressalis laying eggs on rice seedlings in early spring, hence effectively reducing population density of this pest in rice fields.
