        The essential oils from the aerial parts of three varieties of Chrysanthemum zawadskii cultivated in the herbal garden of Duksung Women' s University were extracted by steam distillation method and their compositions were compared by GC-MS analysis. The composition of essential oil fractions from the three varieties of C. zawadskii significant differences in their compostion. While the predominant components of C. zawadskii were camphor and borneol, C. zawadskii var. tenuisectum contained myrtenol and germacrene D as representing compound of its essential oil. C. zawadskii var. nakdongense oil showed higher contents of germacrene D than other two varieties of C. zawadskii. As the results of the determination of the effects of this oil and its main components on antibiotic-susceptible and -resistant strains of Salmonella enteritidis and S. typhimurium, the main components of C. zawadskii oil fraction, borneol and camphor, significantly inhibited all strains of the two Salmonella species examined, while its oil fraction showed relatively weak activity. In evaluation tests of the combination effects of the oils with kanamycin or streptomycin using a checkerboard microtiter assay, C. zawadskii oil fraction combined with streptomycin resulted the most significant synergism in this study with the fractional inhibiting concentration index (FICI) of 0.19.

        polystyrene beads를 사용한 경량 시멘트 모르타르의 성질을 개선하기 위해 EVA혼화용 폴리머를 첨가한 모르타르를 제조하여, 물리ㆍ역학적 성질을 살펴보았던 바 polystyrene beads 골재 재부의 공극 때문에 높은 공기량을 보였고, 플로우 값은 폴리머-시멘트비 10%에서 가장 높은 값을 나타냈다. 또한 폴리머-시멘트비가 증가할수록 압축강도와 휨강도는 증가하였고, 시멘트-경량골재비가 낮을수록 압축강도는 증가하였으나, 휨감도에는 크게 영향을 주지 않았다. The objective of this study was to improve performance of lightweight polymer cement mortars by treating with redispersible polymer powders. Lightweight polymer-cement mortars were prepared with respect to different polymer-cement ratios, and tested for water absorption, specific gravity, flexural and compressive strengths, workability and freeze-thaw resistance. Manufactured lightweight polymer-cement mortars in this study showed lower specific gravity and lower thermal diffuseness than ordinary cement mortars. Water absorption, specific gravity, flexural, compressive strengths and workability were measured according to appropriate KS test methods. As a results, the optimum mix ratios for cement-aggregate, polymer-cement, and water-cement were found to be 1:3, 10% and 30%, respectively.

        The effects of medium pH and culture temperature on the expression and secretion and secretion of inulinase and invertase were investigated with recombinant Saccharomyces cerevisiae cells. These cells were obtained by transformation of 2μ-based plasmids pYI10 and pYS10 which contain Kluyveromyces marxianus inulinase gene (INU1A) and S. cerevisiae invertase gene (SUC2), respectively, in the downstream of GAL1 promoter. The expression level and localization of inulinase and invertase were not affected significantly by the initial medium pH: secretion efficiencies of inulinase and invertase into the medium were about 90% and 60%, respectively, in the pH ranges of 4.0 to 6.5. However, the expression and secretion of both enzymes were strongly dependent on the culture temperature. The highest expression (7.7 units/mL) and secretion (6.7 units/mL) of inulinase were observed at 28℃ and 30℃. As a consequence of decreased localization of inulinase in the periplasmic space, the secretion efficiency increased from 68% at 20℃ to 95% at 35℃. The total expression level and secretion efficiency of invertase increased from 19 units/mL and 55% at 20℃ to 25 units/mL and 68% at 35℃, respectively. Irrespective of the culture temperature, the invertase activity in the cellular fraction (periplasmic space and cytoplasmic fractions) was kept constant at around 33∼45%.

        Targeting Super-Enhancers for Disease Treatment and Diagnosis

        The transcriptional regulation of genes determines the fate of animal cell differentiation and subsequent organ development. With the recent progress in genome-wide technologies, the genomic landscapes of enhancers have been broadly explored in mammalian genomes, which led to the discovery of novel specific subsets of enhancers, termed super-enhancers. Super-enhancers are large clusters of enhancers covering the long region of regulatory DNA and are densely occupied by transcription factors, active histone marks, and co-activators. Accumulating evidence points to the critical role that super-enhancers play in cell type-specific development and differentiation, as well as in the development of various diseases. Here, I provide a comprehensive description of the optimal approach for identifying functional units of super-enhancers and their unique chromatin features in normal development and in diseases, including cancers. I also review the recent updated knowledge on novel approaches of targeting super-enhancers for the treatment of specific diseases, such as small-molecule inhibitors and potential gene therapy. This review will provide perspectives on using super-enhancers as biomarkers to develop novel disease diagnostic tools and establish new directions in clinical therapeutic strategies.

        Many viruses infect cucurbits. One of the well-known symptoms is mosaic disease. Those that cause mosaic are cucumber mosaic virus (CMV), squash mosaic virus (SqMV), watermelon mosaic virus (WMV), zucchini yellow mosaic virus (ZYMV) and cucumber green mottle mosaic virus (CGMMV). WMV resistant GM squash was developed many years ago in the United States and it was on the market, but no further information was available by now pertinent to commercial aspect. Usually these viruses are not easily controlled by frequent applications of chemicals that target the insect as carriers of viruses. Therefore, it is necessary to develop commercial varieties possessing resistance against viral diseases. We have developed GM watermelon rootstocks called gongdae, using a coat protein gene of CGMMV as transgene. Those GM watermelon rootstocks showed highly resistant to CGMMV, and have been crossed to get the several BC and T generation. In order to obtain the virus resistant watermelon, watermelon lines were crossed to the selected GM watermelon rootstock. Here, we present the successful watermelon cultivars that show resistance to CGMMV. The resistance must have obtained by transferring the transgene from the GM watermelon rootstock to watermelon line.

        Repression of adenosine triphosphate–binding cassette transporter ABCG2 by estrogen increases intracellular glutathione in brain endothelial cells following ischemic reperfusion injury

        <P><B>Abstract</B></P> <P>The adenosine triphosphate–binding cassette efflux transporter ABCG2, which is located in the blood-brain barrier limits the entry of endogenous compounds and xenobiotics into the brain, and its expression and activity are regulated by estrogen. This study was aimed to define the role of ABCG2 in estrogen-mediated neuroprotection against ischemic injury. ABCG2 protein levels before and after ischemic stroke were increased in the brain of female mice by ovariectomy, which were reversed by estrogen replacement. In brain endothelial cell line bEnd.3, estrogen reduced the basal ABCG2 protein level and efflux activity and protected cells from ischemic injury without inducing ABCG2 expression. When bEnd.3 cells were transfected with ABCG2 small interfering RNA, ischemia-induced cell death was reduced, and the intracellular concentration of glutathione, an antioxidant that is transported by ABCG2, was increased. In addition, after ischemic stroke in ovariectomized mice, estrogen prevented the reduction of intracellular glutathione level in brain microvessels. These data suggested that the suppression of ABCG2 by estrogen is involved in neuroprotection against ischemic injury by increasing intracellular glutathione, and that the modulation of ABCG2 activity offers a therapeutic target for brain diseases in estrogen-deficient aged women.</P> <P><B>Graphical abstract</B></P> <P>[DISPLAY OMISSION]</P>

        Dual action of apolipoprotein E-interacting HCCR-1 oncoprotein and its implication for breast cancer and obesity

        <P>Obese women have an increased risk for post-menopausal breast cancer. The physiological mechanism by which obesity contributes to breast tumourigenesis is not understood. We previously showed that <I>HCCR-1</I> oncogene contributes to breast tumourigenesis as a negative regulator of p53 and detection of HCCR-1 serological level was useful for the diagnosis of breast cancer<SUB>.</SUB> In this study, we found that the HCCR-1 level is elevated in breast cancer tissues and cell lines compared to normal breast tissues. We identified apolipoprotein E (ApoE) interacting with HCCR-1. Our data show that HCCR-1 inhibits anti-proliferative effect of ApoE, which was mediated by diminishing ApoE secretion of breast cancer cells. Finally, HCCR-1 induced the severe obesity in transgenic mice. Those obese mice showed severe hyperlipidaemia. In conclusion, our results suggest that HCCR-1 might play a role in the breast tumourigenesis while the overexpression of HCCR-1 induces the obesity probably by inhibiting the cholesterol-lowering effect of ApoE. Therefore, HCCR-1 seems to provide the molecular link between the obesity and the breast cancer risk.</P>

      • 패류중 중금속함량에 관한 연구

        본 연구는 패류중 중금속함량 측정을 위한 분석효율이 높고 간편한 분해법을 찾아내어 그 방법에 의끓 유종중인 패류중 중금속함량을 파악하고자 수행하였다. 시료를 분해시키는 Microwave와 습식, 건식회법을 비교한 결과 변형습식회화르가 가장 적합하였다. 변형습식회차딘는 려린0,3+C104+H2SOf의 혼찰산을 쪼 · j 1 (v/v)의 비율로 혼합하여 사용하였 을 때 회수을 Pb린틴. Cd 97.8, Cu 94.2, Zn 92.8%를 나타내었근 분해시간은 2~2.5시간 소요되었다. 유통중인 패류중 중금속함량 」최소~최대(평균), mgAgl 은 Pb 0.028~o.528(0.153), Cd 0.OO7~1,832(0.587), Cu 0.360~37.375(4.6691, Zn 6.317~233.2iO(38.881), Hg 0.001 ~O.151(0.012)mg/ltg으로 나타났다. 또한 유통중인 5종의 패류중 중금속평균항량이 생산지 하재지역에서 구입한 패류에 비해 모든 중금속언써 약간 높은 함량을 보였으나 오염된 수준이 아닌 자연함유량수준이었다. This study was conducted to improve the digestion method for the determination of heavy metals in shellfishes and to estimate the contents of heavy metal in commercial shellfishes by their digestion method. This digestion method was compared microwave with wet and dry ash digestion for shellfish samples, obtained results showed that the modified wet ash Ⅱ digestion was convenient and easy to use. For the modified wet ash Ⅱ digestion, The mixed acid of HNO₃+HClO₄+H₂SO₄(25:5:1, v/v) was used. Under this condition, Recoveries were Pb 94.3, Cd 97.8, Cu 94.2 and Zn 92.8% and digestion time was ∼2.5 hours. The contents of heavy metal [minimum∼maximum (mean), mg/kg in commercial shellfishes were as follows ; Pb:0.028∼0.528(0.153), Cd:0.007∼1.832(0.587), Cu:0.360∼37.375(4.669), Zn:6.317∼233.250(38.881), Hg:0.001∼0.151(0.013). The contents of heavy metal in commercial shellfishes of five species were slightly greater than those of shellfishes purchased at Hajae. There was no great difference between the contents of heavy metal of Hajae and those of shellfishes purchased at Whole market.

        Clinicopathologic features of early gastric carcinoma with lymphoid stroma and feasibility of endoscopic submucosal dissection

        <P>Gastric carcinoma with lymphoid stroma (GCLS) is a rare disease known to have unique features and a favorable prognosis. This study aimed to determine the clinicopathologic features of early GCLS (EGCLS) and define the feasibility of endoscopic submucosal dissection (ESD) for EGCLS. We performed a retrospective analysis of 70 EGCLS patients treated via ESD or surgery and 1626 patients who underwent surgical resection for early non-GCLS (ENGCLS) between January 2007 and December 2014 at Pusan National University Hospital, Busan, Republic of Korea. The mean age of EGCLS patients was 58 years (range 36-77 years); a male predominance (3.7:1) was observed, and 81.4% showed Epstein-Barr virus positivity. Compared with ENGCLS, EGCLS was macroscopically more elevated (34.3 vs. 18.0%, P = 0.003) and located more proximally (upper third: 37.1 vs. 9.7%, P < 0.001). Tumor size was smaller (2.1 +/- 1.1 vs. 3.1 +/- 2.0 cm, P < 0.001), but submucosal invasion was more frequent (77.1 vs. 44.4%, P < 0.001) and deeper in the EGCLS group. Among the 59 EGCLS patients who were treated surgically, only two (3.4%) showed lymph node metastasis (LNM). Despite submucosal invasion, EGCLS showed a lower LNM rate (4.0 vs. 19.4%, P = 0.007) than ENGCLS, even in patients with SM3 EGCLS (5.3 vs. 24.5%, P = 0.007). There were no recurrences in the available ten patients who underwent ESD alone during a mean follow-up of 37.2 months. In this study, we observed unique clinicopathologic features with a very low LNM rate in EGCLS. We consider ESD a potentially curative treatment strategy for EGCLS despite deep submucosal invasion, especially in patients with poor performance status and significant comorbidities.</P>
