        Medilite 침출수에 의한 녹차 추출물의 항산화 활성연구

        The chemical compositions and antioxidant activity of green tea extracts in medilite-extraction water were compared to that of distilled water(DW). Antioxidant activity was fetermined by the formation of thiobarbituric acid reactive substances (TBARS) in rat liver homogenates and microsomes and the scavenging activity of free radicals by DPPH ($\alpha$, $\alpha$'-diphenyl-$\beta$-picrylhydrazyl). The order of total polyphenolic compounds and extracted yield by extracts was medilite 325 mesh-extraction water, medilite 600 mesh-extraction water and distilled water(DW). The ranges of scavenging activity of green tea extracts in DPPH method were 60.95% - 64.51%. The inhibition ratios of TBARS formation in the rat liver homogenates and microsomal fractions were significantly lower with green tea extracts by DW-extraction than with both medilite 325 mesh and 600 mesh-extraction water. The concentration of iron ion of water containing medilite 325 mesh and green tea extracts and of water containing medilite 600 mesh and green tea extracts were significantly higher compared to DW. Therefore, this result suggested that enhanced concentration of iron ion in green tea extracts by medilite-extraction water containing high iron ion content was associated with enhanced peroxidation of the rat liver microsomal fractions. These results showed that total polyphenolic compounds, the % of yield and mineral compounds of green tea extracts were increased using medilite 325 mesh and 600 mesh-extraction water.

        Bacillus sp. FF-7에 의한 항산화물질 생산조건과 항산화 활성

        발효식품 시료로부터 분리된 세균을 DPPH(α,α¹-diphenyl-β-picrylhydrazyl) 전자공여능으로 항산화 활성을 측정하여 가장 활성이 강한 균주를 선별하여 형태학적, 생화학적, 생리학적 특성 및 16S rRNA 염기서열을 조사한 결과 Bacillus sp.으로 판명되어 FF-7로 명명하였다. DPPH 전자공여능법에 의한 Bacillus sp. FF-7이 생산하는 항산화물질의 최적 생산 배지조건은 탄소원 2% galactose와 질소원 1% tryptone 첨가였다. Bacillus sp. FF-7에 의해 생성된 항산화 물질의 활성을 DPPH 전자공여능, 흰쥐 각 조직 microsomal 실험계 및 linoleic acid 과 산화지질 실험계에서 malondialdehyde를 thiobarbituric acid(TBA)방법으로 측정하였다. 흰쥐 각 조직 microsomal 실험계에서 지질과산화에 대한 항산화 효과는 뇌(97.50%)>심장(79.95%)>신장(77.84%)>비장(77.47%)>고환(69.96%)>간장(62.45%) 순이였다. Linoleic acid의 과산화지질률 TBA법으로 측정한 결과 반응 4일째까지 억제 효과가 강하게 나타났으며, 동시에 대조구로 사용한 0.05% BHT 첨가구에서도 실험종료시까지 항산화 활성이 강하게 나타났다. The antioxidative activity of antioxidative substances produced from several bacterial strains isolated from fermented foods were tested by DPPH (α,α'-diphenyl-β-picrylhydrazyl) free radical scavenging activity. One of the strains showing the highest antioxidative activity was identified as Bacillus sp. based on the morphological, biochemical, physiological characteristics, and 165 rRNA sequence, and named FF-7. The molt optimal medium condition for the production of antioxidative substance from Bacillus sp. FF-7 was 2% galactose as carbon source and 1% tryptone as nitrogen source. The antioxidative substance produced from FF-7 in these cultural medium was also tested by in vitro experimental models, the peroxidation of linoleic acid and the peroxidation of rat tissues microsomes by using thiobarbituric acid (TBA) for assay of free rnalondialdehyde production. The antioxidative avtivity against lipid peroxidation of rat tissues microsomes was shown in the following order; brain 97.50% > heart 79.95% > kidney 77.84% > spleen 77.47% > testis 69.96% > liver 62.45%. The antioxidative substance produced from FF-7 on linoleic acid peroxidation by TBA method was effectively inhibited during four days, and 0.05% BHT(butylated hydroxytoluene) used comparative control was also effectively inhibited. Results showed that the highest antioxidative activity by DPPH method of antioxidative substance produced from Bacillus sp. FF-7 was obtained by supplementing 2% galactose as carbon source and 1% tryptone as nitrogen source in cultured medium, this substance effectively inhibited the formation of TBARS in brain microsome in vitro system and in linoleic acid peroxidation.

        Orotic acid 투여 흰쥐의 조직 과산화지질 농도에 미치는 capsaicin의 영향

        The antioxidative activities of capsaicin (Cap, 0.02 and 0.04%) on the lipid peroxidation of tissues in male Sprague Dawley rats fed diets with or without orotic acid (1.0%, w/w) were studied in vivo system by measuring thiobarbituric acid reactive substances (TBARS) concentrations. Body weight gain, food intakes, food efficiency ratio and the relative tissues weights of brain, kidney, spleen, heart, and testis were not significantly different among dietary groups. Relative weights of liver were higher in the OA group than that in the other groups. TBARS concentrations in liver were significantly higher in the OA or 0.04% Cap groups than in the normal group, while this raise was not observed 0.02% Cap group. A significant increase in TBARS concentrations was found in the liver of the OA +0.04% Cap group compared with the OA or the 0.04% Cap groups. Nonheme iron concentrations were significantly higher in the liver of the OA, 0.04% Cap, OA+0.02% Cap, OA+0.04% Cap groups than that in the normal group. TBARS concentrations in kidney were lower in the 0.02% or 0.04% Cap groups than that in the normal group, but this concentrations were higher in either the OA, OA+0.02% Cap or OA+0.04% Cap groups than that in the normal group. Meanwhile, TBARS concentrations of brain, spleen, heart and testis were not significantly different among groups. The present study suggested that the lipid peroxidation was increased in the rats liver fed diet with erotic acid, and the simultaneous supplementation of capsaicin further enhanced.

        버섯 추출물과 이를 함유한 유산균 발효유가 in vivo 및 vitro 과산화지질에 미치는 영향

        The antioxidative effects of fermented milk, mushroom extract and fermented milk containing its extract (Lentinus edodes, Ganoderna lucidum, and Pleurotus ostreatus) on the lipid peroxidation in the tissues of female Sprague-Dawley rats and on the DPPH ($\alpha,\alpha$' -diphenyl-$\beta$-picrylhydrazyl) radical donating ability were studied. The total concentrations of polyphenolic compound in Lentinus edodes, Ganoderma lucidum and Pleurotus ostreatus were 0.34, 0.20 and 0.34%, respectively. The DPPH donating abilities of mushroom extract, fermented milk, fermented milk containing its extract and BHT (butylated hydorxytoluene) as standard were 33.9, 34.9, 51.9 and 95.6%, respectively. Experimental diet groups were divided into five groups: the normal diet (ND), the cholesterol diet (CD), and cholesterol + fermented milk diet (CDFM), cholesterol + mushroom extract diet (CDME) and cholesterol + fermented milk containing mushroom extract diet (CDFMME). The concentrations of lipid peroxide in liver and its microsome were significantly lower in both CDFM and CDFMME groups than in the other groups. The kidney concentration of lipid peroxide was significantly higher in the CD group than in the ND group, but this rise were significantly decreased in the CDFM and CDFMME groups. Meanwhile, the concentrations of heart and spleen and their fractions were not significantly different among dietary groups. This study was suggested that the fermented milk diet containing mushroom extract effectively reduced the lipid peroxidation in liver and kidney of cholesterol-fed female rats.

        콜레스테롤 급여 흰쥐에서 함초 요구르트의 콜레스테롤 저하효과

        The effect of yogurt containing Hamcho (Salicornia herbacea) extract (Hamcho yogurt: HY) on the lipid concentrations in male Sprague-Dawley rats fed a cholesterol diet were studied. Rats were assigned three groups in the experiment; cholesterol diet (the CHOL group), cholesterol diet plus yogurt by lactic acid bacteria (the Y group) or cholesterol diet plus yogurt supplemented Hamcho extract by lactic acid bacteria (the HY group). Fermented milk with or without Hamcho extract in the diet supplemented at the levels 5.0% for 3 weeks. The concentrations of total cholesterol and bile acid in serum was significantly lower in the HY group than the CHOL group. This effect was also observed in rats fed a diet supplemented fermented milk. This cholesterol-lowering effect was more pronounced in the HY group compared to the Y group. The atherogenic index was significantly lower in the HY or Y groups than the CHOL group. The concentration of non esterified fatty acid in serum was significantly lower in the HY group than the CHOL and the Y groups. These results suggested that Hamcho yogurt exert the hypocholesterolemic effect in cholesterol fed rats.

        Effect of Chicory Extract on Triglyceride Metabolism in Rats

        We investigated the effect of chicory (Chicorium intybus) extract on triglyceride concentration and microsomal triglyceride transfer protein (MTP) activity in rats. The effect of water-soluble extract of chicory fed at the $2.0\%\;and\;4.0\%(w/w)$ levels for 2 weeks on the concentration of serum triglyceride and the activity of hepatic microsomal riglyceride transfer protein (MTP) was investigated in male Sprague-Dawley rats. The triglyceride concentrations in serum of the chicory extract fed groups were significantly lower than in the control group. MTP activity, known to be essential for the assembly/secretion of apolipoprotein B-containing lipoproteins, was also significantly lower in the chicory extract groups than in the control group. The concentrations of other lipids in serum and liver and the activity of phosphatidate phosphohydrolase, the rate-limiting enzyme in triglyceride synthesis, showed no significant differences among in the chicory fed groups. These results indicate that dietary chicory extract decrease hepatic MTP activity and serum triglyceride concentration, and therefore reduces hepatic lipoprotein assembly and secretion.

        Physiological Effect of Hamcho Yogurt on Streptozotocin-Induced Diabetic Rats

        함초 추출물 분말을 첨가하여 유산 발효시킨 함초 요구르트를 streptozotocin-유발 당뇨쥐에 투여하여 항당뇨효과 및 콜레스테롤 저하 효과를 규명하기 위하여 3주간 실험하였다. 당뇨 대조군에 비해 함초 요구르트 투여군에서 혈당치 농도 및 식이 섭취량이 현저히 감소하였다. 경구당부하 실험에서도 당뇨 대조군에 비해 함초 요구르트 투여군에서 현저한 혈당상승 억제작용을 나타내었다. 당뇨쥐에 함초 요구르트와 일반 요구르트 섭취에 의해 혈중 총 콜레스테롤 농도 감소와 더불어 동맥경화지수도 감소하였다. 따라서, 함초 요구르트에 의한 혈당치 및 콜레스테롤 농도 감소효과는 고혈당증 및 고 콜레스테롤 혈중에도 유익한 작용을 할 수 있는 기능성식품 개발 가능성이 있음을 시사하였다.
