        캠벨얼리(Vitis labruscana B.) 잎 에탄올 추출물이 신경세포에서 아밀로이드 전구 단백질의 발현과 아세틸콜린에스테라제 활성에 미치는 영향

        Alzheimer's disease (AD) is the most common form of dementia, and the accumulation of β-amyloid (Aβ) in the brain triggers AD, followed by hyperphosphorylation of tau protein, neurofibrillary tangles, and synapses loss, neuronal cell death, and cognitive decline occur in a chain. In APPswe neuronal cell line, 50 ㎍/ml of Campbell early (Vitis labruscana B.) leaves 50% ethanol extract (VLL) treatment inhibited the secretion of Aβ1-42 by about 63% and the secretion of Aβ1-40 by about 50%. VLL did not target the enzymatic activity of the amyloidogenic pathway and decreased the protein expression of APP. As a result of RT-qPCR (Reverse transcription-quantitative real-time PCR) of the APPswe cell line treated with VLL, it is thought that the protein expression of APP was reduced by inhibiting the transcription process of the APP gene. In addition, VLL inhibited acetylcholinesterase (AChE) enzyme activity in vitro by 27.6% and 54.7%, respectively, at 50 and 100 ㎍/ml concentrations. We found that VLL inhibited the production of Aβ, a dementia-inducing substance, by suppressing the transcription of the APP gene, and that VLL inhibited AChE activity. We suggest that VLL has the potential as a natural drug material that modulates the alleviation of dementia symptoms.

        LMK02의 품질규격화와 $A{\beta}$ 올리고머에 의해 유도된 희주해마 H19-7세포주에 미치는 항치매효과

        For standardization of LMK02 quality, Ginsenoside Rg3 of Red Ginseng and Decursin of Angelica gigas Nakai in the constituents of LMK02 were estimated as indicative components. From LMK02 water extract, has been used in vitro test for its beneficial effects on neuronal survival and neuroprotective functions, particularly in connection with APP-related dementias and Alzheimer's disease (AD). $A{\beta}$ oligomer derived from proteolytic processing of the ${\beta}$-amyloid precursor protein (APP), including the amyloid-${\beta}$ peptide ($A{\beta}$), play a critical role in the pathogenesis of Alzheimer's dementia. We determined that oligomer amyloid-${\beta}$ ($A{\beta}$) have a profound attenuation in the increase in rat hippocampus H19-7 cells from. Experimental evidence indicates that LMK02 protects against neuronal damage from cells, but its cellular and molecular mechanisms remain unknown. Using a hippocampus cell line on $A{\beta}$ oligomer-induced neuronal cytotoxicity, we demonstrated that LMK02 inhibits formation of $A{\beta}$ oligomer, which are the behavior, and possibly causative, feature of AD. In the Red Ginseng, the average amounts of Ginsenoside Rg3 were $47.04{\mu}g/g$ and $42.3{\mu}g/g$, 90 % of its weight were set as a standard value. And, in the Angelica gigas Nakai, the average amounts of Decursin were 2.71 mg/g and 2.44mg/g, 90 % of its weight were also set as a standard value. The attenuated $A{\beta}$ oligomer in the presence of LMK02 was observed in the conditioned medium of this $A{\beta}$ oligomer-induced cells under in vitro. In the cells, LMK02 significantly activated antiapoptosis and decreased the production of ROS. These results suggest that neuronal damage in AD might be due to two factors: a direct $A{\beta}$ oligomer toxicity and multiple cellular and molecular neuroprotective mechanisms, including attenuation of apoptosis and direct inhibition of $A{\beta}$ oligomer, underlie the neuroprotective effects of LMK02 treatment.

        Objective : Recent studies indicate that the deposition of beta-amyloid (Aβ) is related in the pathogenesis of Alzheimer`s disease (AD), but the underlying mechanism is still not clear. Method : To investigate the potential cellular functions of APP and LMK02, we use transgenic drosophila as a model was treated with either LMK02, and the effect in APP expression was determined by climbing assay. LMK02 have been shown to be neuroprotective in fly model systems. We asked whether dietary supplementation with LMK02 would influence behavior and AD-like pathology in a transgenic fly model. Result : LMK02 water extract have attenuated fly death in vivo. LMK02-treated fly increased percentage of flight ability more longly and survival ratio more than controls. APP-GRIM drosophila treated with LMK02 had significantly less accumulation of APP deposition in the eye and brain as compared to control drosophila. Conclusion : These results suggest that LMK02 prevent APP-induced neurotoxicity through attenuating flies death induced by APP, and may be useful as potential therapeutic agents for AD.

        여우주머니의 Phenol성 화합물

        The herbaceous species of phyllanthus in this genus are used as hyperglycemic, diuretic, and malaria agent in the world. For the phytochemical studies and the investigation of medicinal resources in the Phyllanthu species, phyllanthus species, (Euphorbiaceae) were used and the studies of constituents in this plant were carried out. From the aqueous fraction of methanolic extract, one flavonoid (quercetin-3-0-rutinoside), two gallotannins(gallic acid, methyl gallate), and two ellagitannins (corilagin, geraniin)were isolated through fractionation and repeated column chromatography using Amberlite XAD-4, ODS gel, and sephadex LH-20. The structures of these compounds were identified on the basis of spectroscopic evidences.

        슬관절 인공관절 치환술 후 배액관의 위치에 따른 출혈량 및 임상결과 비교

        목적: 슬관절 인공관절 전치환술 후 배액관의 위치에 따른 외부 배액량, 혈액 손실량 및 임상 결과를 비교하고자 하였다. 대상 및 방법: 양측 슬관절 인공관절 전치환술을 같은 날 동시에 시행받은 환자 100명을 대상으로, 실험 1에서는 50명에서 편측은 관절강 내에, 반대측은 피하지방층에 배액관을 삽입하여 수술 후 외부 배액량과 통증 정도, 관절 운동 범위, 합병증의 빈도를 비교하였다. 실험 2에서는 또 다른 50명에서 양측 모두 관절강 내에 배액관을 삽입하여 총 혈액 손실량을 산출하여 실험 1의 환자 50명과 비교하였다. 결과: 실험 1에서 외부 배액량은 피하지방층에 배액관을 삽입한 군이 관절강 내에 삽입한 군보다 유의하게 적었으나(p<0.001), 통증의 정도는 수술 후 2일경에만 두 군 간 유의한 차이를 보였다. 실험 2에서 총 혈액 손실량을 비교했을 때 두 군 간 유의한 차이를 보이지 않았다. 결론: 피하지방층에 배액관을 삽입한 군의 외부 배액량은 관절강 내에 삽입한 군보다는 적었으나 배액관의 삽입 위치에 따른 총 혈액 손실량은 유의한 차이를 보이지 않았으며 임상 결과의 큰 차이가 없어 피하지방층에 배액관 삽입의 큰 이점은 관찰되지 않았다. Purpose: The aim of this study was to compare the drainage amount, total blood loss, and clinical results between two different positions of suction drainage after total knee arthroplasty. Materials and Methods: A total of 100 patients who underwent one stage bilateral total knee arthroplasty were enrolled. In experiment 1 with 50 patients, we compared the drainage amount, pain, range of motion, and complications of the leg whose suction drain was inserted into the joint cavity with those of the contralateral leg whose suction drain was inserted in subcutaneous tissue. Another 50 patients of experiment 2 had suction drainage in the joint cavity of both legs and the total blood loss (sum of drainage output, exudates, and hematoma of subcutaneous tissue and joint) was calculated and compared with that of experiment 1. Results: In experiment 1, the drainage amount was less in the leg with suction drainage in subcutaneous tissue compared with the contralateral leg with suction drainage in the joint cavity (p<0.001). However, the postoperative joint pain was significantly different only on post-operative day 2 between two legs. In experiment 2, there was no significant difference in the total blood loss between the two groups. Conclusion: Although the drainage amount was less in the leg whose suction drain was kept in subcutaneous tissue compared with the contralateral leg whose suction drain was in the joint cavity, the total blood loss and the clinical results were not significantly different according to the position of the suction drain. Therefore, we can conclude that the subcutaneous position of the suction drain did not yield superior results.

        Clinical Article : 자가 골-슬개건-골을 이용한 전방십자인대 재건술 시수술 전 슬개건 길이의 예측

        Preoperative prediction of patellar tendon length is important during anterior cruciate ligament (ACL) reconstruction using bone-patellar tendon-bone (BPTB) autograft. Three methods of imaging analysis to predict patellar tendon length were compared in this study. One hundred and twenty-three patients who underwent ACL reconstruction using BPTB autograft by single surgeon during October 2002 through April 2011 were included. We measured the patellar tendon length from true and oblique lateral simple radiographs (classified according to degree of rotation) and magnetic resonance image (MRI). These values were compared with actual length measured during operation and assessed accuracy by calculating the coefficient of determination. The mean length of patellar tendon measured during operation and by true lateral and oblique lateral radiographs and MRI were 42.4±0.45 mm (range, 32.0.54.0 mm), 41.7±0.61 mm (range, 24.2.55.3 mm), 40.7±0.57 mm (range, 24.8.51.5 mm), and 41.7±0.52 mm (range, 28.7. 56.0 mm), respectively. The correlation of patellar tendon length was the most strong between actual length and value from true lateral radiograph (coefficient of determination, r2=0.660) according to simple linear regression analysis. R2 values were 0.361 and 0.332 for oblique lateral radiograph and MRI compared to actual value, respectively. In conclusion, Patellar tendon length measured on true lateral radiograph was the best method to coincide with actual patellar tendon length among various preoperative prediction methods.

        Recently, hydrolyzed silk peptide on acid and alkaline were developed and studied on anti bacterial, cryoprotective, antioxidant, anticancer and antidiabetic activities. For the investigation of antidiabetic drug, we were elucidated the effects of enzyme hydrolyzed silk peptide on streptozotocin induced hyperglycemia, silk peptide of two different molecular weight were administrated after injection of streptozotocin (50mg/kg, ip), and blood glucose, serum triglyceride and serum cholesterol levels were determined at 4 days after streptozotocin injection. And the results were as following: 1. The blood glucose levels of streptozotocin induced hyperglycemic rats were significantly decreased by the administrations by various dose of silk peptide of Japan Products and glibenclamide and especially enzyme hydrolyzed silk peptide were showed to decrease glucose levels than another silk peptide. 2. The serum cholesterol levels of streptozotocin induced hyperglycemic rats were significantly decreased by the administrations by various dose of silk peptide of Japan Products and glibenclamide and especially enzyme hydrolyzed silk peptide were showed to decrease cholesterol levels than another silk peptide. 3. The serum triglyceride levels of streptozotocin induced hyperglycemic rats were significantly decreased by the administrations by various dose of silk peptide of Japan Products and glibenclamide and especially enzyme hydrolyzed silk peptide were showed to decrease triglyceride levels than another silk peptide.
