        애기장대 Nit유전자 발현 오이 형질전환체 개발

        To produce transgenic cucumber expressing Nit gene coffering abiotic resistance, the cotyledonary-node explants of cucumber (cv. Eunsung) were inoculated with A. tumefaciens transformed with pPZP211 or pCAMBIA2300 carrying Nit gene, that has cis-acting element involved in resistance to various abiotic environmental stresses. After co-cultivation, the procedures of selection, shoot initiation, shoot elongation, and plant regeneration were followed by cotyledonary-node transformation method (CTM, Jang et al. 2011). The putative transgenic plants were selected when shoots were grown to a length greater than 3 cm from the cotyledonary-node explants on selection medium supplemented with 100 mg/L paromomycin as a selectable agent. The confirmation of transgenic cucumber was based on the genomic PCR, Southern blot analysis, RT-PCR, and Northern blot analysis. A 105 shoots (4.12%) selected from the selection mediums were obtained from 2,547 explants inoculated. Of them, putative transgenic plants were only confirmed with 45 plants (1.77%) by genomic PCR analysis. Transgenic plants showed that the Nit genes integrated into each genome of 39 plants (1.53%) by Southern blot analysis, and the expression of gene integrated into cucumber genome was only confirmed at 6 plants (0.24%) by RT-PCR and Northern blot analysis. These results lead us to speculate that the genes were successfully integrated and expressed in each genome of transgenic cucumber.

        복합 비애(Complicated Grief)의 개념과 진단

        Complicated grief is defined as a deviation from the normal symptoms of grief. CG has been subsumed under major depressive disorder or posttraumatic stress disorder. However, there has been a broad consensus that existing diagnoses are not sufficient for CG. A number of research groups have shown that how complicated grief is distinguished from those disorders and demonstrated that CG constitutes a distinct psychopathological diagnostic entity. The purpose of this study is to review current concepts of CG and empirical evidences of CG, and its related issues. 복합 비애(Complicated Grief, 이하 CG)란 사별 후 나타나는 정상적이고 문화적으로 받아들여지는 애도 과정을 벗어나 지속적인 심리적․신체적 부적응을 야기하는 비애 반응을 말한다. 지금까지는 비애 반응이 주로 우울장애나 외상후 스트레스 장애와 같은 기존의 진단체계 내에서 이해되곤 하였으나, 여러 연구결과들을 통해 기존의 진단으로는 CG에서 나타나는 독특한 증상을 반영하지 못한다는 지적이 계속되어 왔다. 이에, 본 연구에서는 CG라는 새로운 개념, CG를 기존의 진단 체계로 분류하려고 할 때의 제한점, CG가 PTSD 진단만으로 부족한 이유, 독립적인 CG 진단의 필요성 및 경험적 근거들, 잠정적으로 합의된 진단 기준과 CG 진단이 치료적으로 어떤 중요한 의미를 갖는지를 개관하여 소개함으로써 아직까지 우리나라에서는 본격적으로 논의된 바가 없는 CG에 대한 관심을 고취하고자 한다.

        게슈탈트치료와 수용전념치료에서의 정신병리 이론과 변화 과정 비교 고찰

        The focus of this study is to review two psychotherapies, Gestalt Therapy and Acceptance and Commitment Therapy (ACT), with regards to mental illness, psychological health and therapeutic change process. After brief presentation of two approaches focused on theories of psychological health and therapeutic change, some similarities and differences of these two approaches were discussed. Despite the orientation of each therapy is differ from, both approaches are experiential and process-oriented intervention based on awareness and acceptance. The purpose of these approaches is not to directly eliminate the symptoms, but rather to come up with more holistic and contextualistic understanding in dealing mental distress. However, each has a different emphasis on the central principle of the change mechanism and also differs in how it is applied. The results of this study presented a theoretical basis for how each of these approaches can be more effective and developed in their field. 본 연구에서는 정신병리 및 심리적 문제에 대한 관점과 변화 과정을 중심으로 게슈탈트치료와 수용전념치료 접근의 특징을 비교 고찰하고자 하였다. 이를 위해 각 접근에 대한 주요 문헌을 고찰하였고 정신병리에 대한 관점과 변화 과정에 대한 각 접근의 이론을 소개한 후, 두 접근의 공통점과 차이점을 논의하였다. 두 접근은 이론의 뿌리는 다르지만 경험에 대한 알아차림과 수용을 지향하는 체험 중심 접근으로서 직접적인 증상의 감소를 일차적 목표로 삼지 않으며 인간의 심리적 고통을 맥락적이고 전체적인 관점에서 바라본다는 공통점을 갖는다. 그러나 변화 기제의 중심 원리에 있어 각각 다른 강조점을 가지며 적용 방법에 있어서도 차이를 보이는 것으로 나타났다. 본 연구의 결과는 두 접근의 효과적이고 통합적인 발전을 위한 이론적 기초를 제시했다는 점에서 의의가 있다.

        Production of transgenic cucumber expressing phytoene synthase-2A carotene desaturase gene

        The objectives of this study were to 1) evaluate the efficiency of the protocol of Agrobacterium-mediated transformation of cucumber to introduce phytoene synthase-2a carotene desaturase (PAC genes); 2) demonstrate the integration of PAC genes into the genome of putative transgenic cucumber based on growth on selection medium, PCR and Southern analysis; 3) evaluate the expression of PAC genes in transgenic cucumber based on the analysis of RT-PCR and Northern blot hybridization. Out of 5,945 cotyledonary-node explants inoculated with Agrobacterium, 65 (1.1%) explants produced 238 shoots. Integration of PAC genes into the genome of the cucumber was demonstrated based on the analysis of gDNA- PCR, 21 out of the 238 plants regenerated; while 6 plants proved positive for Southern blot hybridization. Transgene expression was demonstrated based on analysis of RT-PCR, 6 plants proved positive out of the 6 plants analyzed; while 4 plants out of 6 proved positive during Northern blot hy bridization. This study successfully demonstrated the production of transgenic cucumber, integration, and expression of the PAC gene in cucumber.

        한국형 유산균 Bifidobacterium 속 균주의 항생물질에 대한 감수성

        Minimal inhibitory concentrations (MICs) of Bifidobacterium spp. strains (Bifidobacterium breve K-110, B. breve K-111 and B. infantis K-525) isolated from healthy Korean against antituberculosis agents and fluoroquinolones were determined. From the MICs it was found that Bifidobacterium breve K-110, B. breve K-111 and B. infantis K-525 were susceptible to rifampicin and fluoroquinolenes and resistant to other antituberculosis agents.

        포도 탄저병균의 엽면접종을 통한 국내 육성 포도나무의 품종 간 저항성 검정 및 유전적 다양성 분석

        본 연구는 품종 간 변이의 폭이 좁은 포도속(Vitis spp.) 유전자원을 대상으로 탄저병 저항성 품종을 판 별하기 위해 국내 주요 재배 품종 및 유전자원 159품종을 공시하고,Colletotrichum acutatum과 C. gloeosporioides를 엽면접종하여 병원성 검정을 수행하였다. 또한, Genotyping-by-sequencing(GBS) 방 법을 이용하여 포도나무 품종 간 유전적 다양성을 비교 분석하였다. 그 결과, C. acutatum을 접종한 잎에서는 58품종은 감수성을, 17품종은 저항성을 나타내었고, C. gloeosporioides를 접종한 경우에는 34품 종이 감수성을, 25품종은 저항성을 나타냈다. 두 균주에 대해 공통적으로 저항성을 보인 품종은 ‘Agawan’, ‘Huangguan’, ‘Xiangfei’ 등 8품종이였으며, 이들 대부분은 유럽종과 미국종의 교잡종으로 분류되었다. 유럽종에 기원한 ‘Emerald Seedless’, ‘Tano Red’, ‘Rem 46-77(Aestivalis GVIT 0970)’ 등의 품종들은 대부분 감수성으로 판별되었다. 따라서 본 연구 결과를 통해 확보된 병 검정 및 유전 분석 결과는 포도나무 병해를 효과적으로 관리하는데 유용하게 활용될 수 있으며, 특히 포도나무 탄저병 저항성 품종들을 개발하고 육성하는데 매우 중요한 육종 소재 및 정보가 될 것으로 사료된다. Here, we reported 159 varieties of major cultivars using grapevine genetic resources to identify the resistant grape ripe rot cultivars. To do this, we performed pathogenicity assays from these grape cultivars by inoculating Colletotrichum acutatum and Colletotrichum gloeosporioides. Genotyping-by-sequencing(GBS) method was also used to compare genetic diversity among grape varieties. As a result, leaves inoculated with C. acutatum showed that 58 cultivars were susceptible, while 17 cultivars were resistant. In the case of C. gloeosporioides, 34 cultivars were found to be susceptible, while 25 cultivars were resistant. The 8 cultivars that showed resistance to both species were ‘Agawan’, ‘Huangguan’, ‘Xiangfei’, and 5 other cultivars from the hybrids of European and American species. Most of the varieties such as ‘Emerald Seedless’, ‘Tano Red’, and 'Rem 46-77(Aestivalis GVIT 0970)' originated in European species were identified as susceptible. These results can be used in the effective management of grape disease. In addition, these findings provide information for the development and cultivation of resistant to grape ripe rot disease cultivars.

        Lactobacillus plantarum MKHA15와 Leuconostoc mesenteroides MKSR을 첨가한 발효 채소 주스의 특성

        The objective of this study was to develop fermented vegetable juices that possess antidiabetic and antioxidant activities. Lactobacillus plantarum MKHA15 (MKHA15) and Leuconostoc mesenteroides MKSR (MKSR) were applied to ferment onion, cabbage, and tomato juices at 37oC and 30oC for 72 h, respectively, and their functionality was tested using the 12 h hour-fermented juice by MKHA15, and 48h hour-fermented juice by MKSR. Inhibition of α-glucosidase activity was observed in all fermented juices. The onion juice fermented by MKHA15 showed significantly higher α-glucosidase inhibition activity compared to other juices. All juices showed more than 70% inhibition of α-amylase activity. The DPPH radical scavenging activity of onion juice fermented by MKSR showed significantly lower activity than cabbage and tomato juices; however, no difference was observed between the types of starter cultures. The SOD-like activity of cabbage juice fermented by MKSR was the highest among the fermented juices. The juices fermented by MKHA15 showed higher reducing power than those by MKSR. Therefore, we believe that cabbage, onion and tomato juice fermented by MKHA15 and MKSR would be useful in probiotic juices, as they possess antidiabetic and antioxidant activities.
