PC12 세포와 A123.7 세포에서 차별적으로 발현되는 유전자의 검색

The cAMP-dependent protein kinase(PKA) is an intracellular enzyme with serine-threonine kinase activity that plays a key role in cell growth, differentiation, and apoptosis in eukaryotes. In order to understand the PKA signal transduction pathway regulating cell life cycle and identify its role, we focused on the characterization of up-/down-regulated genes by PKA using the differential display polymerase chain reaction. Seven differentially expressed sequence tags(DEST) have been obtained. Among these DESTs, 2 DESTs were homologous to the sequence of genes from BLAST search result. KC1-5 DEST that was up-regulated in A123.7 cells was highly corresponded to mouse apoptosis-related gene(MA-3) or mouse mRNA for topoisomerase inhibitor suppressed(TIS). MA-3 was induced in various types of apoptosis, specially in NGF-deprived apoptotic PC12 cells. TIS was down-regulated in the RVC lymphoma cells incubated with topoisomerase inhibitor that induces DNA strand breakages. PG1-1 DEST that was highly expressed in PC12 cells was corresponded to transposon Tn10 3'-end. Tnansposon Tn10 was up-regulated in differentiated myeloblastic ML-1 cells by 12-O-tetradecanoylphorbol-13-acetate. This study illuminates that MA-3/TIS was down-regulated by PKA activity, and transposon Tn10 was up-regulated by it.

Drago $Gali{\acute{c}}$ (1907-1992) has been acknowledged as one of most important modern Croatia architects of the 20th century and noted for his controversial apartment buildings at 35-35a and 43-43a blocks on Vukovar Street in Zagreb, Croatia. Although the two housings were highly regarded as the best examples of the post-war housing design in Croatia, a plagiarism controversy arose due to its similar exterior looks to Le Corbusier's $Unit{\acute{e}}$ d'Habitation in Marseille in 1952. This research intends to comparatively analyze architectural features implemented on the works of apartment of Drago $Gali{\acute{c}}$ and Le Corbusier's $Unit{\acute{e}}$ d'Habitation. The analysis focuses on architectural characteristics categorized in three parts: unit plan, community space, and unit combinations. The site survey was carried out to yield more useful information for the analysis. During this process, written and photographic documentations are collected for the further interpretation. In addition, scale drawings are reconstructed for the in-depth analysis of the project.

This study deals with a comparative analysis of wooden assembly methods between Japanese traditional architecture and Kuma, Kengo`s works. Kuma, Kengo has know as an architect who pursues Japanese architectural traditions with integrated solutions for built and natural environments, and expresses a deep commitment to unexpected use of materials such as stone and wood with the clarity of structural solutions. Accordingly, his architecture focuses on the appreciation of the spatial relationship with the surrounding, its local characters, and the selection of materials. This article first examines two of wooden assembly methods that have been shown in Japanese traditional architecture such as `two-way system` and `one-way system`. And then, Kuma, Kengo`s wooden assembly methods in his works are analyzed and compared in relation to Japanese wooden assembly methods to find out similarities and differences between them. Then, it concludes that Kuma, Kengo`s approach is not only relying on the methods of Japanese traditional architecture, but also exploring creative ways beyond traditional assembly methods.
장기간 플루세틴 처리에 의한 흰쥐 해마에서의 NCAM140 유전자 발현의 증가

Objectives : Most of the mechanisms reported for antidepressant drugs are the enhancement of neurite outgrowth and neuronal survival in the rat hippocampus. Neural cell adhesion molecule 140(NCAM140) has been implicated as having a role in cell-cell adhesion, neurite outgrowth, and synaptic plasticity. In this report, we have performed to elucidate a correlation among chronic antidepressant treatments, NCAM140 expression, and activation of phosphorylated cyclicAMP responsive element binding protein(pCREB) which is a downstream molecule of NCAM140-mediated intracellular signaling pathway in the rat hippocampus. Methods : Fluoxetine(10mg/kg) was injected acutely(daily injection for 5days) or chronically(daily injection for 14days) in adult rats. RNA and protein were extracted from the rat hippocampus, respectively. Real-time RT-PCR was performed to analyze the expression pattern of NCAM140 gene and western blot analyses for the activation of the phosphorylation ratio of CREB. Results : Chronic fluoxetine treatments increased NCAM140 expression 1.3 times higher than control in rat hippocampus. pCREB immunoreactivity in the rat hippocampus with chronic fluoxetine treatment was increased 4.0 times higher than that of control. Conclusion : Chronic fluoxetine treatment increased NCAM140 expression and pCREB activity in the rat hippocampus. Our data suggest that NCAM140 and pCREB may play a role in the clinical efficacy of antidepressants promoting the neurite outgrowth and neuronal survival.
한국인의 2중합효소연쇄반응-증폭 좌위에 대한 동위 유전자 빈도 분포

Tetranucleotide STR인 HUMTH01과 HUMF13A1에 대한 대립형질과 유전형질 빈도를 한국인을 대상으로 전기영동 및 중합효소연쇄반응을 이용하여 분석하였다. HUMTH01과 HUMF13A1 유전형질 분포 양상은 Hardy-Weinberg 공식을 따랐다. 부가적으로 한국인 집단의 대립형질 빈도를 중국인과 유럽코커시안 집단의 데이터와 비교하였다. 코커시안집단에서 HUMTH01의 분포양상은 유사하였다. 하지만 한국인과 중국인 집단의 결과와 유사하였다. 본 연구에서 HUMTH01과 HUMF13A1 STR 좌위에 대한 한국인 집단 데이터베이스를 확립하였다. 이들 집단에서 STR의 대립형질빈도 분포 데이터는 인간집단유전학을 위한 연구에 유용하게 사용될 수 있을 것이다. Allele and genotype frequencies for the tetranucleotide STRs (short tandem repeats) of HUMTH01 and HUMF13A1 were investigated in Korean population using electrophoresis and PCR amplification. The HUMTH01 and the HUMF13A1 genotype distributions were in Hardy-Weinberg equilibrium. Additionary, the allele frequencies of Korean population were compared with data for the Chinese population and European Caucasian populations. There were similar distribution patterns of HUMTH01 allete frequencies among the Caucasian populations. However, Korean and Chinese populations were significantly different with them. Allele distribution pattern of Korean was more similar with Chinese population than them of European. Conclusively, the Korean population database has been established for HUMTH01 and HUMF13A1 STR loci. These data of allele frequencies distribution of STRs in population will be useful for studies on human population genetics.
