        Successful Management of Hepatic Lipidosis Accompanied by a Feline Skin Fragility Syndrome-like Lesion in a Cat

        A 7-year-old spayed female Somali-cross cat was admitted with a 4-week history of anorexia, vomiting, weight loss, and lethargy. The cat was diagnosed with hepatic lipidosis. With intensive care and nutritional support via a nasogastric feeding tube for 3 weeks, the clinical signs of hepatic lipidosis were improved. However, skin lesions were found in the left and right scapular regions during the treatment that were suspected to be due to feline skin fragility syndrome (FSFS). Intensive wound healing therapy with granulated sugar, laser therapy, and a surgical flap was conducted. Skin lesions improved uneventfully without other clinical signs or recurrence of any skin lesion for a year. To our knowledge, this is the first report of a good prognosis in a hepatic disorder and concurrent FSFS. 7년령의 불임 암컷 소말리 교잡종 고양이가 4주 동안 지속된 식욕부진, 구토, 체중감소, 무기력을 주증으로 내원하였다. 환자는 지방간으로 진단 되었으며, 3주 동안 집중치료와 더불어 비위 영양관을 통해 유동식을 공급하였다. 치료기간 중, 지방간으로 인한 임상증상은 개선 되었지만 고양이 피부유약증으로 인한 것으로 의심되는 피부병변이 왼쪽과 오른쪽 어깨 부위에서 발견되었다. 과립당치료, 레이저치료 및 외과적 피판술을 통한 집중적인 상처 치유를 실시하였으며, 그 결과 환자는 다른 임상 증상이나 피부 병변의 재발 없이 성공적으로 치료되었다. 보고된 바에 따르면, 본 증례는 간질환과 고양이 피부유약증이 동반한 환자에서 좋은 예후를 보인 첫 번째 증례이다.

        인버터 DC 저항점용접 공정에서 CRS 알고리즘을 이용한 정전류 퍼지 제어시스템 설계에 관한 연구

        The purpose of this study is to propose a method to decide near-optimal settings of the constant current fuzzy control parameters using a controlled random search. This method tries to find the near-optimal settings of the constant current fuzzy control parameters through experiments. It has an advantage of being able to carry out searches in the search domain which includes some irregular points. The method suggested in this study was used to determine the fuzzy control parameters by which the desired welding current were formed during inverter DC resistance spot welding. The output variable was the ITAE (integral of time multiplied by the absolute error). This output variable was determined according to the input variables, which are the GE, GDE, and GDU. This study described how to obtained near-optimal welding current condition over a wide search space conducting a relatively small number of experiments.

      • 방사면역측정을 위한 항 Gastrin 혈청의 생산 및 평가

        New Zealand white 계의 토끼에게 EDC를 사용하여 hemocyanin에 접합시킨 human G-17-I을 면역한 결과 gastrin의 방사면역측정에 적합한 항 gastrin 혈청을 얻는데 성공하였다. 본 실험에서 얻은 항 gastrin 혈청의 역가는 1:25,000이며 human G-17-I과 human big gastrin에 매우 친화력이 높은 반면 다른 위장관 peptide 들과는 교차반응성이 아주 낮았다. 본 항 gastrin 혈청의 effective affinity constant는 $1.77{\times}10^{11}/M$이었다. 또한 총 결합부위(total binding sites)는 255 nM, 이질성지수(heterogeneity index)는 0.84 그리고 average affinity constant는 $0.79{\times}10^{11}/M$이었다. 본 실험에서 얻은 항 gastrin 혈청을 사용하여 사람과 흰쥐의 혈장 gastrin 농도를 방사면역측정법으로 측정한 결과 이미 잘 알려진 결과와 동일한 성적을 얻을 수 있었다. 이러한 결과들을 종합하여 볼때 본 실험에서 얻은 항 gastrin 혈청은 생리적농도로 존재하는 gastrin의 방사면역측정에 적합한 것으로 생각된다. In order to produce antibody for use in radioimmunoassay of gastrin in physiological concentration, four rabbits of New Zealand white were immunized with synthetic human gastrin-17-I conjugated to hemocyanin with EDC. Among them, only one rabbit produced antibody that could bind 50% of $^{125}I-gastrin$ at a final dilution of 1:25,000. $^{125}I-gastrin$ was prepared with synthetic human gastrin-17-I and $NaI^{125}$ by lactoperoxidase technique. The product was then purified on a column of Sephadex Gl5/G5O (7:3, w/w) followed by a column of DEAE sephadex A-25. The specific radioactivity of the purified $^{125}I-gastrin$ was in the range of 347-1429 ${\mu}Ci/nmole$ when determined by the self-displacement method. The effective affinity constant $(K_{eff})$, total binding sites (N), heterogeneity index $({\alpha})$ and average affinity constant $(K_{0})$ of the anti-gastrin serum calculated from Scatchard plot as well as Sips plot were $1.77{\times}10^{11}/M$, 255 nM, 0.84 and $0.79{\times}10^{11}/M$, respectively. When radioimmunoassay was performed with the anti-gastrin serum, it was confirmed that the mean concentration of gastrin immunoreactivity in plasma was increased by feeding in humans and rats, and also increased by bombesin administration in rats. The results indicate that the anti-gastrin serum produced in the present investigation is suitable for radioimmunological determination of gastrin in physiological concentration.

      • 흰쥐의 위궤양 발생에 미치는 대상회전 제거 및 미주신경 절단의 영향

        This study was undertaken to investigate the effect of cingulate cortical ablation upon gastric ulceration, and the pathway through which cingulate cortex exerts the effect. 56 female rats were divided equally into cingulate (cingulate cortical ablation), cingulate-vagal (cingulate cortical ablation and vagotomy), normal control and vagal (vagotomy) groups. Cingulate cortex was ablated through a slit-shaped opening (1 mm in width, 13 mm in length) which was made symmetrically on both sides of, and parallel to, the sagittal suture by removing a bone flap from parietal and frontal bones on each side. Vagus nerves on both sides were transected around the distal end of the esophagus. In the normal control animals, surgical intervention ended with scalp incision. All rats were kept without restraint or food deprivation for 3 weeks after surgery. The stomach of each rat was inflated with 7 ml of physiological saline and then removed under deep anesthesia. The mucosal surface was examined under dissecting microscope for the location, shape and number of ulcers, and then enlarged photograph $(4{\times})$was taken. The incidence of ulcer in each group was counted and the number of ulceration as well as the total area of glandular mucosa were measured on the photograph. Results obtained were as follows: 1. The mean number of ulcer per stomach and the total area of ulcer exprssed as permillage of the total area of glandular mucosa were significantly higher in the cingulate group than the cingulate-vagal, the normal control and the vagal groups. There was no difference among the latter three groups. 2. The incidence of ulcer in the cingulate group was significantly higher than that in the .normal control group and was also higher, though not significantly, than those in the cingulate-vagal and the vagal groups. There was no difference among the normal control, the cingulate-vagal and the vagal groups. It is inferred from the above results that the cingulate cortex exerts an inhibitory influence upon gastric ulceration and that this influence is mediated by controlling the vagal activity.

      • Effect of Reserpine on Pancreatic Exocrine Secretion Induced by Mesencephalic Reticular Stimulation in Rats

        최근에 마취한 흰쥐에서 중뇌망상체를 전기적으로 자극하면 췌장의 외분비 기능이 증가하며 이러한 결과는 망상체의 자극으로 인하여 교감신경계의 활성도가 상승하기 때문이라는 보고가 있다. 한편 교감신경계의 활성도가 상승할 경우 교감신경계의 전달 물질인 catecholamine이 교감신경 종말 뿐만 아니라 부신수질에서도 유리된다고 알려져 있다. 그러므로 본 연구에서는 중뇌망상체의 자극으로 인하여 췌장의 외분비 기능이 증가함에 있어 교감신경계가 중요한 역할을 담당하는지를 확인하고, 이때 부신수질이 관여하는가를 알아보고자 하였다. 마취한 흰쥐에게 atropine (1mg/kg) 또는 reserpine (5mg/kg)을 투여하거나 또는 부신을 적출한 다음 중뇌망상체를 전기 자극하면서 췌장액을 채취하였다. 사용한 전기자극의 매개변수는 1.3V, 40Hz, 2msec이었다. atropine과 reserpine을 투여하면 마취한 흰쥐의 자발적 췌장액 분비량과 단백질 분비량은 모두 유의하게 감소하였으나 부신을 제거하면 췌장액 분비량에는 이렇다할 변동이 없는 반면에 단백질 분비량은 유의하게 감소하였다. 중뇌망상체를 전기자극하면 췌장액 분비량과 단백질 분비량 모두가 유의하게 증가하였다. 이러한 망상체의 자극효과는 atropine 전처치에 의하여 이렇다할 영향을 받지 않았으나 reserpine 전처치에 의하여 소실되었다. 그러나 부신을 적출하면 망상체 자극에 의한 췌장액 분비량의 증가는 유지되는 반면에 단백질 분비량의 증가는 소실되었다. 한편 미주신경을 절단한 흰쥐에서 중뇌망상체를 자극하는 동안에 경동맥의 수축기 및 이완기 혈압이 상승하였는데 이러한 망상체의 자극효과도 reserpine의 투여에 의하여 유의하게 감소되었다. 본 실험의 결과를 종합하여 보면 마취한 흰쥐에서 중뇌망상체의 자극은 교감신경계를 활성화시켜 췌장액 분비량과 단백질 분비량에 촉진적인 영향을 미치며, 이때 활성화된 교감신경계는 부분적으로 부신을 경유하게 췌장의 단백질 분비에 촉진적인 영향을 미치는 것으로 생각된다.

      • Isolation of Bombesin-Like Substances from the Skin of the Frog, Bombina orientalis: Its Molecular Heterogeneity and Biological Activity

        Bombesin (BBS)은 유럽에 서식하는 무당개구리(Bombina bombina)의 피부로 부터 발견되었으며 이것은 14개의 amino acid로 구성되어 있음이 잘 알려져 있다. 한국에도 유럽의 무당개구리와 종(species)이 다른 무당개구리인 Bombina orientalis가 서식함이 알려졌으나 이 무당개구리의 피부에 BBS가 존재하는 지는 확실치 않다. 그러므로 본 실험에서는 한국에 서식하는 무당개구리의 피부로부터 BBS 유사물질(BBS-LS)을 추출하여 분자적 이질성과 생물학적 활성을 측정하고자 하였다. 박리한 무당개구리 피부를 100% methanol로 처리하여 얻은 추출물의 일부를 Sephadex G-50, superfine column에 부하하고 여기에서 얻은 chromatogram을 분석한 결과 BBS-LS의 3.5%는 합성 gastrin releasing peptide (GRP-27) 보다 앞에 용출되었으며 나머지 96.5%는 GRP-27과 합성 BBS-14의 사이에 용출되었다. alkaline alumina column과 Sephadex G-10, fine column 그리고 Sephadex G-50, superfine column을 연속적으로 이용하여 methanol 추출물로 부터 BBS-LS를 정제 하였다. 정제된 BBS-LS를 Sephadex G-50, superfine column으로 분석한 결과 합성 GRP-27과 합성 BBS-14 사이에 용출되어 BBS-LS가 이미 알려진 BBS-14와 분자의 크기에 있어서 다른 물질임을 알 수 있었다. 정제된 BBS-LS를 reversed phase HPLC로 분석한 실험은 이러한 결과를 더욱 확실히 뒷받침하였다. 정제된 BBS-LS를 합성 BBS-14와 비교하여 생물학적 활성을 검토한 결과 흰쥐에서 gastrin의 분비, 췌장액의 분비, 회장의 수축을 촉진하고 guinea pig 담낭의 수축을 증가시키는 등 이들 두물질이 매우 유사한 생물학적 활성을 지니고 있음을 발견하였다. 이러한 결과로 보아 한국에 서식하는 무당개구리의 피부는 서로 다른 두가지 형태의 BBS-LS를 함유하고 있으며 그 중에서 96.5%를 차지하는 BBS-LS는 합성 BBS-14와 분자적 이질성을 지니나 유사한 생물학적 활성을 지니고 있음을 알 수 있었다. Molecular property as well as biological activities of bombesin-like substance (BBS-LS) isolated from the skin of B. orientalis in Korea was compared with those of synthetic BBS-14. BBS-LS in the crude extract was applied on a Sephadex G-50, superfine column $(1.6{\times}112\;cm)$. On chromatogram, the first peak (3.5% of BBS-LS) was found ahead of synthetic GRP-27, and the second peak (rest) between synthetic GRP-27 and synthetic BBS-14. The main form of BBS-LS was successfully purified by using a column of alkaline alumina followed by sequential gel-filtrations on a column of Sephadex G-10, fine and a column of Sephadex G-50, superfine. Chromatographic analysis of the purified BBS-LS using a column of Sephadex G-50, superfine and reversed phase HPLC revealed that the main form of BBS-LS in the skin of B. orientalis could be distinctly different from either BBS-14 or GRP-27 in molecular size. The purified BBS-LS exerted biological activities quite identical to those of synthetic BBS-14. The results of the present investigation indicate that the skin of B. orientalis contains BBS-LS composed of two distinct forms. The main form of BBS-LS purified in the present study is heterogenous to both synthetic BBS-14 and GRP-27 in molecular size but identical to BBS in biological activities.

        만성 전립선염에 대한 한의학적 치료의 임상적 효과

        Objectives: The prostatitis is characterized as relatively uncurable chronic recurrent illness. Because it is so difficult to treat, this study was designed to evaluate the clinical effects of herb medicine, acupuncture, and herbal acupuncture therapy on chronic prostatitis. Methods: A clinical observation was made on thirty patients with chronic prostatitis. They were treated with herb medicine, acupuncture, and herbal acupuncture therapy. Before and after treatment we measured NIH-CPSI(National Institutes of Health Chronic Prostatitis Symptom Index). Results: After treatment NIH-CPSI was decreased considerably. Conclusions: This suggests a role for oriental medical therapy in treating prostatic diseases. Further study into approach and application of these treatments for prostatitis is needed.

      • 강원도 지역에 서식하는 무당개구리에서 생물학적 활성을 지닌 Bombesin 유사 Peptide에 관한 연구

        본 실험에서는 강원도 지역에 집단으로 서식하고 있는 한국산 무당개구리(Bombina orientalis) 피부에서 bombesin 유사 peptide를 순수정제하여 생리활성과 amino acid 서열을 측정하고자 하였다. 무당개구리의 피부를 methanol에 담가 bombesin 유사반응물질(BLI)을 추출한 다음 alkaline alumina에 이은 Sephadex G-10 겔 크로마토그래피와 RP C18 preparative HPLC로 부분정제하였다. 부분정제시 여러 종류의 BLI가 출현하므로 본 연구에서는 전체 BLI의 65%를 점유하는 peak(BLI-K1)를 RP C18 및 SP-이온교환 HPLC를 이용하여 순수정제하였다. 피부조직 420 g에서 $890\;{\mu}g$의 BLI-K1를 순수정제하였으며, 이것은 RP C18, 겔 투과 그리고 SP-이온교환 HPLC 모두에서 합성 bombesin(BBS)과 동일한 정체시간을 나타내었다. 생리활성에서도 BLI-K1은 마취한 흰쥐에서 혈장 gastrin 농도와 혜장 단백질, 중탄산이온, amylase 및 chymotrypsin 분비량을 모두 유의하게 증가시켜 합성 BBS와 동일한 활성을 나타내었다. 또한 BLI-K1의 분자량 및 C-terminal의 11개 amino acid 서열이 합성 BBS과 동일하였기 때문에 한국산 무당개구리의 피부에도 유럽산 무당개구리의 피부에 존재하는 것과 동일한 bombesin이 존재하는 것으로 생각된다. The present study was performed to purify a BLI from the skin of frogs, B. orientalis inhabiting Kangwon-Do, Korea, and then determine its physiological activities and amino acid sequence. A substance that shows bombesin-like immunoreactivity (BLI) was partially purified from crude methanol extract of the frog skin by using a column of alkaline alumina and Sephadex G-10. BLI was further purified by using RP C18 preparative HPLC in which BLI was separated into 5 peaks. The major peak (BLI-K1) containing 65% of total BLI was subjected to purify as a single component by using sequential HPLC of SP-ion exchange and RP C18. Content of BLI in each fraction was monitored by RIA for which bombesin antiserum with a high titer and affinity to BBS was raised in a guinea pig. Eventually, $890\;{\mu}g$ of BLI-K1 was homogeneously purified from 420g of the skin of B. orientalis and it was not differentiated from synthetic BBS in RP C18, gel permeation and SP-ion exchange HPLC. In the physiological activities, BLI-K1 was identical to synthetic BBS in stimulation of gastrin release and exocrine pancreatic secretion including volume, protein, bicarbonate, amylase and chymotrypsin output in anesthetized rats. Furthermore, molecular weight and partial amino acid sequence of BLI-K1 was revealed to be identical to synthetic BBS. Therefore, it is concluded from the above results that the skin of B. orientalis contains the same BBS that has been isolated from the skin of European Bombina.

      • Prostaglandin $E_1$과 Acetazolamide가 흰쥐 전혈(全血)의 Carbonic Anhydrase 활성에 미치는 영향

        This study was undertaken to investigate the influence of prostaglandin $E_1(PGE_1)$ upon the activity of carbonic anhydrase and upon the inhibitory action of acetazolamide on carbonic anhydrase. The heparinized blood was sampled by cardiac puncture from Sprague-Dawley strain rats under ether anesthesia and was hemolysed by adding distilled water 1,000 times the amount of the blood. The activity of carbonic anhydrase of 0.1 ml of the hemolysate was measured by Maren's simplified micro-method. In the first experiment, the 7 rats were used, and the activity was measured by adding 0.1 ml of various concentrations of $PGE_1$(0.5, 1.25, 2.5, 5.0, 10 and $20\;{\mu}g/ml$). In the second experiment, the 6 rats were used and the activity was measured by adding 0.1 ml of $PGE_1(5\;{\mu}g/ml)$ and 0.1 ml of acetazolamide$(6{\times}10^{-7}M/l)$ respectively or simultaneously. Obtained results were as follows: 1) The activity of carbonic anhydrase was significantly inhibited by $PGE_1$ at doses of $0.5{\sim}10\;{\mu}g/ml$, maximally at a dose of $2.5\;{\mu}g/ml$, but inhibition was no more observed at a dose of $20\;{\mu}g/ml$. 2) The activity of the acetazolamide group was significantly less than that of the control group. 3) The activity of the $PGE_1+acetazolamide$ group was significantly less than those of the $PGE_1$ group and the acetazolamide group. It is inferred from the above results that the $PGE_1$ inhibits the activity of carbonic anhydrase dose-dependently and strengthens the inhibitory effect of acetazolamide on carbonic anhydrase.

        극박 다이아프램의 펄스 GTAW 공정 최적화에 관한 연구

        This paper has aimed to prevent excessive heat input by controlling arc distribution and heat input capacity with pulse GTAW in order to improve weld quality in 0.08mm pressure gauge diaphragm and flange welding parts. A design of experiment was designed using Box-Behnken method to optimize a welding process. The pulse GTAW parameters such as pulse current, base current, pulse duty, frequency and welding speed were set to input variables while hydraulic pressure that represents welding characteristics in diaphragm and flange joint were set to output variables. Based on the test result, a second regression equation was obtained between input and output variables and turned out significant. Besides, an influence of parameters has been confirmed through response surface analysis using the second-order regression equation and optimum welding condition was obtained through a grid-search method. The optimum welding condition was set to pulse current 84.4(A), base current 29.6(A), pulse duty 58.8(%), frequency 10(%), and welding speed 596(mm/min). Then, decent bead shape was acquired with no excessive heat input under the $2.3kgf/cm^2$ of hydrostatic pressure.
