        Phosphorylation in Novel Mitochondrial Creatine Kinase Tyrosine Residues Render Cardioprotection against Hypoxia/Reoxygenation Injury

        Objective: Ischemic cardiomyopathy (ICM) is the leading cause of heart failure. Proteomic and genomic studies have demonstrated ischemic preconditioning (IPC) can assert cardioprotection against ICM through mitochondrial function regulation. Considering IPC is conducted in a relatively brief period, regulation of protein expression also occurs very rapidly, highlighting the importance of protein function modulation by post-translational modifications. This study aimed to identify and analyze novel phosphorylated mitochondrial proteins that can be harnessed for therapeutic strategies for preventing ischemia/reperfusion (I/R) injury. Methods: Sprague-Dawley rat hearts were used in an ex vivo Langendorff system to simulate normal perfusion, I/R, and IPC condition, after which the samples were prepared for phosphoproteomic analysis. Employing human cardiomyocyte AC16 cells, we investigated the cardioprotective role of CKMT2 through overexpression and how site-directed mutagenesis of putative CKMT2 phosphorylation sites (Y159A, Y255A, and Y368A) can affect cardioprotection by measuring CKMT2 protein activity, mitochondrial function and protein expression changes. Results: The phosphoproteomic analysis revealed dephosphorylation of mitochondrial creatine kinase (CKMT2) during ischemia and I/R, while preserving its phosphorylated state during IPC. CKMT2 overexpression conferred cardioprotection against hypoxia/reoxygenation (H/R) by increasing cell viability and mitochondrial adenosine triphosphate level, preserving mitochondrial membrane potential, and reduced reactive oxygen species (ROS) generation, while phosphomutations, especially in Y368, nullified cardioprotection by significantly reducing cell viability and increasing ROS production during H/R. CKMT2 overexpression increased mitochondrial function by mediating the proliferator-activated receptor γ coactivator-1α/estrogen-related receptor-α pathway, and these effects were mostly inhibited by Y368A mutation. Conclusion: These results suggest that regulation of quantitative expression and phosphorylation site Y368 of CKMT2 offers a unique mechanism in future ICM therapeutics.

      • 여자 체조경기 난도분석에 관한 연구 : 마루운동을 중심으로

        The purpose of this paper is to present the required difficulty related to Korean Women's gymnastics training to our national woman gymnasts and the concerned instructors. With comparing the degree of the difficulty above B class performed by our national 24 leading woman gymnasts with that shown by international 3 top leading medalists in All Around Competition as Women's gymnastics in 1979 World Champion Ship game and making a study of the difficulty of each performer's exercise, a new possibility to improve the weakness of our national gymnasts is also provided to our national leaders. For the purpose of obtaining necessary data, 24 national leading performers range from compulsorty school to senior high school were chosen and 3 world champion ship medalists, Nadia Comaneci (Rom), Nelli Kim (URS) and Ludmilla Tourischeva (URS) were sellected. The table of the degree of the difficulty of national gymnasts was obtained by analizing their performance in the national official gymnastic competitions, but data of international performers were collected from the recent articles of medilist's techniques. In case of the floor exercise, performance skill and numbers of the difficulty were no different between national gymnasts and international ones. However, the harmony between music and performance seemed somewhat insufficient in our national gymnasts. Even though gymnasts are throwing the harmoneous performance matched with music, they cannot gain high credit without outstanding tumbling and dance elements.

        민법상 친양자제도가형법상 존속살해죄의 행위객체에 미치는 영향

        The Consideration of Full Adoption in the Civil Law is a system in which the children who are adopted as volladoptions are legally separated from the family before adopted, resulting in the extinguishment of the family relationship that was formed earlier and the acquisition of same right as children that were born during marriage in the adopted households. This system was introduced to provide a stable adoption environment for adopted person and to give a harmonization and stabilization for the adoptive family, respectively. But, the introduction of such a system provides some difficulties in interpreting some articles of the Criminal Law. In the parricide crime, which is stipulated in the Criminal Law, the object of action is a lineal ascendant. In such cases, the concept prescribed by the Civil Law must be borrowed. In the general adoption system, it is acceptable that the parricide crime is established, when the adopted person kills his / her biological parents because the kinship before adoption is maintained among them. However, because the family relationship are terminated between the volladoption and a former biological parent, there is controversy as to whether it is reasonable for a murderer not to applied to parricide crime, when the volladoption kills his or her biological parents. In other words, some provisions of the Civil Act affect in interpretation of the criminal law. In this regard, this study suggests the interpretation that the biological parent must be included in action object of parricide crime. Also, this paper derives the possibility that volladoption should be applied to a parricide crime when an adoptee kills a biological parent by the following three methods: First, interpretation of the relationship between civil and criminal law; second, expanding the scope of victim's damages; and third, interpretation with natural science. The parricide crime as a kind of murder is a serious crime that eventually deprives life and makes the its recovery impossible forever. Therefore, even if the murder of a lineal ascendant extends the coverage of punishment power, it can not be said to be an abuse of the penal code. In conclusion, it should be said that the action object of parricide crime includes not only new parents who are newly established by law but also former parents who are biological parents. 민법의 친양자제도는 친양자로 입양되는 자녀가 입양 전의 가정에서 법적으로완전히 분리되는 것으로 기존의 친족관계가 소멸되며 입양 가정의 혼인 중의 출생자와 같은 효력을 발생시킨다. 이러한 친양자제도는 친양자로 입양된 자에게는안정된 입양환경을 제공하고 재혼가정에는 가족의 화합과 안정에도 도움이 되고자 도입되었다. 이러한 친양자제도의 도입은 형법에서 해석상의 어려움을 동반하였다. 형법상 존속살해죄의 행위객체는 법률상 직계존속으로 민법의 개념을 가져오기 때문이다. 일반양자제도에서 입양자는 입양 전의 친족관계가 유지되기 때문에 입양자가친생부모를 살해한 경우에 존속살해죄가 성립한다고 봄에 무리가 없었다. 그러나친양자는 기존의 친족관계가 종료되기 때문에, 이와 같은 민법상의 효과가 형법에도 영향을 줄 것인지 그렇다면 친양자가 친생부모를 살해한 경우 존속살해죄가성립하지 않는 것인지와 관련해 논란이 있다. 이와 관련해서 본 논문에서는 존속살해죄의 행위객체에 친생부모가 포함되어야 한다는 해석을 제시하였다. 검토방법으로 첫째, 민법과 형법의 관계에 따른 해석방법, 둘째, 피해자 보호측면에서의 범위 확대에 대한 검토, 셋째, 자연과학적 해석을 통해서 친양자의 친생부모 살해에대한 존속살해죄 성립 가능성을 제시하였다. 존속살해도 결국은 살인의 한 종류로서 생명을 박탈하여 영원히 그 회복을 불가능하게 하는 심각한 범죄라 할 수 있다. 특히 직계존속에 대한 살인은 그 범위를 확장시킨다 하여 형벌권의 남용이라 할 수는 없을 것이다. 따라서 존속살해죄의 행위객체에는 법률로써 새롭게 맺은 양부모뿐만 아니라 생물학적 부모인 친생부모도 포함된다고 해야 할 것이다.

        먹물버섯 Coprinellus congregatus에서 분열자를 사용한 형질전환

        먹물버섯의 하나인 Coprinellus congregatus를 대상으로 유전자의 도입을 위한 형질전환실험에서 원형질체를 생성하지 않고 분열자 (oidium)를 사용하는 방법을 확립하였다. 분열자는 20일 이상된 CKMM 한천배지에서 생성되며, 이를 밤샘배양으로 발아를 촉진시킨 상태에서 전기천공방법으로 항생물질 basta에 대한 형질전환을 수행한 결과 10-20 형질전환체/${\mu}g$ DNA의 수율로 형질전환체를 확보할 수 있었다. 이 형질전환체들은 도입된 벡터가 염색체 상에 삽입되어 유전적으로 안정된 상태를 유지하였다. Genetic transformation of a mushroom-forming fungus Coprinellus congregatus to antibiotic resistance gene had been successfully carried out by electroporation to oidia instead of protoplasts. Since there was no protoplast generation step which required not only cell wall degrading enzymes but many skillful procedures, commercial herbicide (basta) could be used without any difficulty with simple procedure. The transformation yield was 10-20 $transformants/{\mu}g$ DNA, and the transformants were very stable even after 10 consecutive transfers through the non-selective medium.

        Copine1 Enhances Neuronal Differentiation of the Hippocampal Progenitor HiB5 Cells

        Copine1 is a ubiquitously expressed protein found in various tissues including the brain, but little is known about the physiological function of this protein. Here, we showed that copine1 is involved in neuronal differentiation. Over-expression of copine1 clearly increased neurite outgrowth and expression of Tuj1, a neuronal marker protein, in HiB5 cells. In addition, endogenous copine1 was transiently increased at the early time during neuronal differentiation of HiB5 cells. When the expression of endogenous copine1 was knocked-down by its specific shRNA, PDGF-mediated neurite outgrowth was clearly decreased in HiB5 cells. Furthermore, over-expression of copine1 increased phosphorylation of Akt and copine1-specific shRNA de-creased phosphorylation of Akt during neuronal differen-tiation of HiB5 cells. Interestingly, the phosphorylation level of PI3K, generally known as an upstream protein of Akt, was not changed by copine1 expression. These re-sults suggest that copine1 enhances neuronal differentiation of HiB5 cells not through the PI3K-Akt pathway, but by using another Akt activated signal pathway.

        낙태죄의 허용 한계 사유와 허용 한계 시기에 대한 고찰

        낙태죄의 존재에 대한 긍정적인 작용을 부인할 수 없으므로, 낙태죄를 폐지하는 것에 찬성하지 않는다. 이에 따라 형법상 낙태죄의 목적 및 보호법익으로서 태아의 생명권과 산모의 자기결정권 사이를 어느 정도로, 어떤 방식으로 조화시킬 것인지를 검토하였다. 그 방법적인 부분으로, 첫째, 먼저 현행 형법에서 금지하는 낙태를 예외적으로 허용하는 모자보건법상 규정을 수정 또는 개정하여, 낙태 행위에 대한 처벌 예외 범위를 합리적으로 조정하여 규정해야 한다. 모자보건법에 규정되어 있는 예외적 낙태 허용사유의 범위를 현 시점의 사회에 맞도록 수정 및 보완해야 한다. 이 작업은 태아의 생명권과 비교해 임부의 자기결정권 보장에 보다 유리하게 작용할 수 있다. 둘째, 모자보건법 시행령에 규정된 낙태 허용 한계 시기에 대한 부분을 재검토해야 한다. 이로 인해 태아의 생명권 보장에 좀 더 유리하게 작용할 수 있다. 모자보건법 시행령 제15조 제1항에 규정된 인공임신중절수술의 허용한계에 따르면 모자보건법 제14조에 따른 인공임신중절수술은 임신 24주일 이내인 사람만 할 수 있도록 하고 있다. 이에 대하여 임신 초기 즉, 10주-12주에 한하여 여성의 자기결정 또는 낙태 선택이 가능하도록 하자는 주장이 있다. 이러한 허용 한계 시기에 대한 논의에 앞서 검토할 것은 용어적인 부분이다. 우리는 임신주령 혹은 임신주수, 태아나이(수정 후), 또는 착상 이후 주수와 혼재해서 사용하고 있다. 몇 주까지를 허용 주수로 할 것인가에 있어서 그 기준이 되는 시기는 매우 중요하므로, 기준에 대한 용어의 통일이 우선적으로 작업되어야 할 것이다. 다음으로 낙태 허용 한계 시기에 대해서 검토하기 위해서는 임신 주령에 따른 태아의 발달 사항을 알아야 한다. 이미 임신 10주령이 끝나면 배아는 기본적으로 사람에게 요구되는 모든 형태를 갖추게 된다. 따라서 지금 가장 논의가 집중되는 임신 12주(수정 후 10주 혹은 착상 후 8주)미만 낙태 허용 입장에 대해서는 재고가 필요한 부분이다. 생각건대 태아의 발생이 끝나서 외부의 침해가 없는 이상 정상적인 출생이 예상되는 임신 10주 즉, 수정 후 8주 까지만 허용해야 한다고 생각한다. By considering both the purpose of an abortion law and the right to life of a fetus, it can not be denied the positive effect of an abortion law. Therefore, the exceptional exemption regulation from the penalty for abortion prohibited by the current criminal law should be revised reasonably. In revising the exceptionally permitted reasons for the abortion as stipulated in Article 14 of the Maternal and Child Health Act, minors and unmarried-mothers should be the first consideration. Mental patients should also be considered because the likelihood of child offenses is very high when a person with a mental illness is a parent. Some argue that social and economic circumstances should be taken into consideration for allowing abortion. But. As such, there is concern that expanding the scope of abortion according to social and economic circumstances may eventually lead to an unlimited expansion. And the expansion of abortion tolerance causes more demand for the national welfare budget. However, considering the current welfare budget of the country, it seems difficult to expand the scope of abortion for social and economic reasons. Next, we examined the time limit for abortion. At present, according to the time limit of artificial abortion operation prescribed in Article 15 (1) of the Enforcement Decree of the Maternal and Child Health Act, the artificial abortion performed according to Article 14 of the Maternal and Child Health Law is allowed only to those who are within 24 weeks of pregnancy. In this regard, it is argued that the selection of abortion should be made possible only in the early pregnancy ages, 10 to 12 weeks by respecting women"s self-determination. First, before reviewing the time limit for abortion, it is necessary to orientate the terms of pregnancy time in order to unify the meanings of the contents to be discussed. We mark the time of pregnancy as the gestational age calcurated from the last menstrual period, the fetal age counted since fertilization, or the fetal age reckoned after implantation. Therefore, the unification of the terms for pregnancy time should be solved first. When a person is pregnant, the fertilized egg formed by fertilization develops continuously until birth, through the embryo in which the major vital organs develop and through the fetus in which most of the organs grow. In other words, at the end of the 8th week after fertilization (10th week of pregnancy), the embryo will have all the basically necessary organs for human beings. Thus, as mainly discussed in this paper, allowing abortion at 12 weeks (10 weeks after fertilization or 8 weeks after fertilization) is problematic in that it apparently kills life. Therefore, it is necessary to reconsider the current position of abortion.
