        Generation of Buffalo Mammary Epithelial Cells with Targeted Knockout of 18s rDNA by a CRISPR/Cas9 Adenovirus System

        In recent years, CRISPR/Cas9 has rapidly become one of the most promising genome editing tools because it is simple and easy to use and cost effective. However, the large size of Cas9 sequences limits its application in clinically promising vectors and it also impacts non-viral transfection. In this study, CRISPR/Cas9 adenovirus vectors that target the buffalo 18s rDNA gene were constructed, transfected into 293 cells for adenovirus packaging, and the adenovirus was used to knockout the 18s rDNA gene in buffalo mammary epithelial cells. The results demonstrated that the CRISPR/Cas9 adenovirus vectors for the buffalo 18s rDNA gene could efficiently target the sites as revealed by the fluorescence reporter system. After amplification, the adenovirus titer of Sn458- 18s1 and Sn458-18s2 reached 1.03 × 109PFU/mL and 1.05 × 109 PFU/mL, respectively. For buffalo mammary epithelial cell infection, the efficiency was 100% when the multiplicity of infection (MOI) 100 PFU/mL. There were 9 mutational clones found in the 20 clones, and the gene mutagenesis rate reached 45%. Of these, 2 clones were 35-bp deleted and 7 clones were 12-bp deleted. These results suggested that the adenovirus system overcame the low transfection efficiency of the buffalo mammary epithelial cells associated with using lipid-based methods or electroporation. Moreover, we preliminary developed an efficient technique for multiple-locus gene targeting at repeated sequences of the buffalo genome.

      • Enhancement Effect of Confined Pressure and Formation Mechanism in Jointed Rock Tunnel with High Geostress

        Large squeezing deformation is currently becoming one of the most remarkable problems of tunnel construction in Western China. During the tunnel excavation, some geotechnical problems were encountered in the surrounding rock, such as excessive overbreak and intense deformation by asymmetric squeezing, mainly because of intense anisotropic of structural strength. The large deformation led in severe damage of the primary support, even splitting of secondary support, which have a significant influence on construction and security of tunnels. The complex failure phenomena and a series of experimental results in tunnels are summarized in this paper. According to this, the paper proposed a synthetic interpretation, namely enhancement effect of confined pressure (EECP), which resides in different types of jointed rockmass under high in-situ stress. Considering different mechanic parameters of rockmass, sensitivity analysis on EECP has been carried out through numerical simulation (FLAC<sup>3D</sup>). It is indicated that EECP is remarkably influenced by Poisson's ratio of rockmass. Combined statistic mechanics analysis of rockmass with geological record of Zhifang Tunnel and Liangshui Tunnel of Lanzhou-Chongqing railway in China. The calculated pressure between surrounding rock and lining according to present standard exhibits relatively smaller than that exists in tunnels. It is revealed that the formation of EECP can be interpreted by large Poisson's ratio effect, which resides in layered rockmass under compress condition.

        Fast Video Motion Estimation Algorithm Based on Motion Speed and Multiple Initial Center Points Prediction

        본 논문은 모션 속도와 다수의 초기 중심점에 기반한 빠른 모션 추정 알고리즘을 제안한다. 제안한 방법은 시공간적 이웃 모션 벡터들에 의해 초기 탐색점을 예측한다. 모션벡터를 빠르게 구하기 위하여 모션 속도와 예측된 초기 중심점들에 기반한 동적인 탐색 패턴이 이용된다. 제안한 방법은 시공간 정보와 동적 탐색 패턴을 이용하여 탐색 속도를 매우 빠르게 할 뿐만 아니라 양질의 영상화질을 유지할 수 있다. 실험결과를 통해 제안한 방법이 완전 탐색, 새로운 삼단계 탐색, 사단계 탐색 방법들과 비교하여 검색시간을 줄이면서 PSNR 관점에서 양질의 영상화질을 제공함을 알 수 있다. This paper proposes a fast motion estimation algorithm based on motion speed and multiple initial center points. The proposed method predicts initial search points by means of the spatio-temporal neighboring motion vectors. A dynamic search pattern based on the motion speed and the predicted initial center points is proposed to quickly obtain the motion vector. Due to the usage of the spatio-temporal information and the dynamic search pattern, the proposed method greatly accelerates the search speed while keeping a good predicted image quality. Experimental results show that the proposed method has a good predicted image quality in terms of PSNR with less searching time comparing with the Full Search, New Three-Step Search, and Four-Step Search.

      • An Improved Two-Dimensional Run-Length Encoding Scheme and Its Application

        In this paper, we propose an improved two-dimensional run-length encoding (I2DRLE) scheme for representing grayscale images. Conventional 2D run-length encoding scheme is simple and effective that has been widely used, while it is not suitable to represent non-block images. Our approach is a new data compression algorithm inspired by 2D run-length encoding and quadtree, which apply some predefined patterns to represent various data and can sharply reduce the number of blocks in image representation. Experimental results show that this method is an effective lossless grayscale image encoding scheme.

        Tailoring ZSM-5 zeolite through metal incorporation: Toward enhanced light olefins production via catalytic cracking: A minireview

        The steam cracking of hydrocarbons has been the major process for light olefins production in recent decades. Recent studies have shown that a considerable amount of propylene and ethylene can be producedby hydrocarbon cracking on modified ZSM-5 zeolite. Various modified elements and post-treatmentmethods have been employed to fulfil the improvement of the catalytic performance over ZSM-5 zeolite. This article focuses on the research progress about the catalytic cracking of hydrocarbons over metalincorporatedZSM-5 zeolite. The mechanism of propylene and ethylene production via catalytic crackingof various hydrocarbons over ZSM-5 zeolite is summarized and the influences of different metal species,such as alkaline earth and alkali metal elements, rare earth metal elements, transition metals, etc. on bothactivity and stability are also discussed. Moreover, the synergistic influence of bimetallic components ismentioned as well. In particular, the effects of different synthetic methods and structures of metal speciesare discussed. We hope that this review will kindle the future rational design of metal-incorporated zeolitecatalysts to realize efficient catalytic cracking of hydrocarbons for light olefins production.

        Study on flexible two-axis roll-bending process for component with non-circular section

        Two-axis roll-bending technology, with the advantages of small curvature radius, short straight section and high efficiency, is widely used in fields of aerospace and automobile. By adjusting indentation depth in two-axis roll-bending process, different curvature radius of circular and elliptic component can be formed. Roll-bending experiments and numerical simulations for component with circular and elliptic section are conducted. The results show that the forming curvature radius of circular component and its deviation both decrease with ascending indentation depth. The relationship between forming curvature radius and indentation depth is polynomial fitted. On this basis, ellipse parameter equation is established and different curvature radius at different position of the ellipse is achieved by adjustment of indentation depth. The relationship between indentation depth and forming time is obtained for elliptic component. Elliptic components with different sizes are formed. The stress distribution is uniform after roll-bending process and it increases with ascending indentation depth.

        Morphology Characterization and Mechanism Analysis of the Microstructure of LiNbO3 Crystals Fabricated by Using Focused Ion Beam Milling

        This paper reports the basic results obtained from an investigation of the microstructure of lithium niobate (LiNbO3) crystals fabricated by using the focused ion beam (FIB) sputtering method. The morphology characterization of the microstructure is explained in terms of scanning electron microscopy observations based on the geometrical integrity, surface smoothness, cross-sectional area, aspect ratio, etc. The optimized micro-fabrication conditions and the sample morphology characterization are presented, and the sputtering rate of Ga ions for a 30-keV energy milling of the LiNbO3 crystal is calculated, which is lower than the theoretical one gained from “The Stopping and Range of Ions in Matter” (SRIM) sputtering simulation due to the redeposition effect being enhanced with increasing milling depth

        Adsorption of hexavalent chromium onto Bamboo Charcoal grafted by Cu2+-N-aminopropylsilane complexes: Optimization, kinetic, and isotherm studies

        The adsorption mechanism of Cr(VI) uptake onto Bamboo Charcoal grafted by Cu2+-N-aminopropylsilanecomplexes (BC/Cu-N) was investigated. The properties of BC/Cu-N were characterized using XRD, FTIR,SEM, EDS, potentiometric acid–base titration and electrochemical analysis. Results indicated that theframework integrity of BC was kept aftermodification and the quantity of functional groups on BC/Cu-Nsurface has changed. There existed functional groups on the BC/Cu-N which could consume OH and thepHpzc was found to be 6.20. Moreover, electrochemical analysis showed that new electron transferpathway was imported. Parameters such as pH, initial Cr(VI) concentration, adsorbent dosage andtemperature were optimized using RSM. Analysis of variance of the quadratic model for Cr(VI) wassuitable to predict the adsorption of Cr(VI) (F value = 134.23 and P value < 0.001) with a high correlation(R2 = 0.9921). The results showed that the initial Cr(VI) concentration and the adsorption capacity ofCr(VI) were positively related. Adsorption data of Cr(VI) were better fitted by Sips, Temkin and D-Rmodels with the maximum adsorption capacity of 17.9383 mg/g. In addition, the pseudo second-orderkinetic model was found to be more suitable for the adsorption of Cr(VI).

        Correlations Between Fasciology and Yin Yang Doctrine

        The aim of this study is to explore the correlations between fasciology and yin yang doctrine. Professor Yuan developed fasciology by three-dimensional reconstruction of connective tissue (fascia) in the trunk and limbs of the human body and tracing back to tissue origins in light of biological evolution and developmental biology. Fasciology states that the human body can be divided into two systems: the supporting-storing system and the functional system. This article elaborates on the roles of the two systems and their mutual relationship. The two systems are used to analyze the yin,the yang, and their relationship. The two systems are promoted but also restricted in different contexts. The supporting-storing system is formed by undifferentiated connective tissue and provides undifferentiated cells and nutrients for differentiated cells of the functional system. Thus, the supporting-storing system could be classified as quiet, similar to yin. The functional system continuously maintains the various functional activities of the human body. Thus, the functional system could be classified as active, similar to yang. In interpreting the yin yang doctrine from the point of view of fasciology, yin can be compared with the supporting-storing system and yang can be compared with the functional system.
