      • Distance Estimation Methods in Vehicular Application: An Experimental Study

        Passive Entry and Passive Start (PEPS) system allows drivers to enter the vehicle and start the engine without activating key fob. With growing demand for location-based service and rapid development of Bluetooth Low Energy (BLE), BLE PEPS with localization service becomes a promising choice for next generation of PEPS. This paper studies the critical problem of localizing the vehicle for user based on BLE received signal strength (RSS) provided by preinstalled BLE Access Points (APs) inside vehicle. Multiple APs are employed to reduce the error caused by RSS fluctuation on distance estimation. Fusion algorithms including Extended Kalman Filter, Monte Carlo Localization and Iterative Trilateration are compared based on experiments in vehicle. Experimental results show that distance estimation based on Extended Kalman Filter gets high accuracy in both long distance and short distance scenario.

        3D-quantitative structure–activity relationship and antiviral effects of curcumin derivatives as potent inhibitors of infl uenza H1N1 neuraminidase

        Curcumin derivatives have been shown to inhibitreplication of human infl uenza A viruses (IAVs). However, itis not clear whether curcumin and its derivatives can inhibitneuraminidase (NA) of influenza virus. In this study, ameaningful 3D quantitative structure–activity relationshipmodel (comparative molecular fi eld analysis R 2 = 0.997,q 2 = 0.527, s = 0.064, F = 282.663) was built to understandthe chemical–biological interactions between their activitiesand neuraminidase. Molecular docking was used to predictbinding models between curcumin derivatives and neuraminidase. Real-time polymerase chain reactions showed thatthe fi ve active curcumin derivatives might have direct eff ectson viral particle infectivity in H1N1-infected lung epithelial(MDCK) cells. Neuraminidase activation assay showed thatfi ve active curcumin derivatives decreased H1N1-inducedneuraminidase activation in MDCK cells. Indirect immunofluorescence assay indicated that two active curcuminderivatives (tetramethylcurcumin and curcumin) down-regulatedthe nucleoprotein expression. Curcumin inhibited IAVin vivo. The therapeutic mechanism of curcumin in the treatmentof infl uenza viral pneumonia is related to improvingthe immune function of infected mice and regulating secretionof tumor necrosis-α, interleukin-6, and interferon-γ. These results indicate that curcumin derivatives inhibit IAVby blocking neuraminidase in the cellular model and curcuminalso has anti-IAV activity in the animal model.

      • Enhancement Effect of Confined Pressure and Formation Mechanism in Jointed Rock Tunnel with High Geostress

        Large squeezing deformation is currently becoming one of the most remarkable problems of tunnel construction in Western China. During the tunnel excavation, some geotechnical problems were encountered in the surrounding rock, such as excessive overbreak and intense deformation by asymmetric squeezing, mainly because of intense anisotropic of structural strength. The large deformation led in severe damage of the primary support, even splitting of secondary support, which have a significant influence on construction and security of tunnels. The complex failure phenomena and a series of experimental results in tunnels are summarized in this paper. According to this, the paper proposed a synthetic interpretation, namely enhancement effect of confined pressure (EECP), which resides in different types of jointed rockmass under high in-situ stress. Considering different mechanic parameters of rockmass, sensitivity analysis on EECP has been carried out through numerical simulation (FLAC<sup>3D</sup>). It is indicated that EECP is remarkably influenced by Poisson's ratio of rockmass. Combined statistic mechanics analysis of rockmass with geological record of Zhifang Tunnel and Liangshui Tunnel of Lanzhou-Chongqing railway in China. The calculated pressure between surrounding rock and lining according to present standard exhibits relatively smaller than that exists in tunnels. It is revealed that the formation of EECP can be interpreted by large Poisson's ratio effect, which resides in layered rockmass under compress condition.

        Graded-Three-Dimensional Cell-Encapsulating Hydrogel as a Potential Biologic Scaffold for Disc Tissue Engineering

        BACKGROUND: Intervertebral disk (IVD) degeneration, which can cause lower back pain, is a major predisposing factor for disability and can be managed through multiple approaches. However, there is no satisfactory strategy currently available to reconstruct and recover the natural properties of IVDs after degeneration. As tissue engineering develops, scaffolds with embedded cell cultures have proved critical for the successful regeneration of IVDs. METHODS: In this study, an integrated scaffold for IVD replacement was developed. Through scanning electron microscopy and other mechanical measurements, we characterized the physical properties of different hydrogels. In addition, we simulated the physiological structure of natural IVDs. Nucleus pulposus (NP) cells and annulus fibrosusderived stem cells (AFSCs) were seeded in gelatin methacrylate (GelMA) hydrogel at different concentrations to evaluate cell viability and matrix expression. RESULTS: It was found that different concentrations of GelMA hydrogel can provide a suitable environment for cell survival. However, hydrogels with different mechanical properties influence cell adhesion and extracellular matrix component type I collagen, type II collagen, and aggrecan expression. CONCLUSION: This tissue-engineered IVD implant had a similar structure and function as the native IVD, with the inner area mimicking the NP tissue and the outer area mimicking the stratified annulus fibrosus tissue. The new integrated scaffold demonstrated a good simulation of disc structure. The preparation of efficient and regeneration-promoting tissueengineered scaffolds is an important issue that needs to be explored in the future. It is hoped that this work will provide new ideas and methods for the further construction of functional tissue replacement discs.

        Fast Depth Video Coding with Intra Prediction on VVC

        In the stereoscopic or multiview display, the depth video illustrates visual distances between objects and camera. To promote the computational efficiency of depth video encoder, we exploit the intra prediction of depth videos under Versatile Video Coding (VVC) and observe a diverse distribution of intra prediction modes with different coding unit sizes. We propose a hybrid scheme to further boost fast depth video coding. In the first stage, we adaptively predict the HADamard (HAD) costs of intra prediction modes and initialize a candidate list according to the HAD costs. Then, the candidate list is further improved by considering the probability distribution of candidate modes with different CU sizes. Finally, early termination of CU splitting is performed at each CU depth level based on the Bayesian theorem. Our proposed method is incorporated into VVC intra prediction for fast coding of depth videos. Experiments with 7 standard sequences and 4 Quantization parameters (Qps) validate the efficiency of our method.

        Video Quality Assessment Based on Short-Term Memory

        With the fast development of information and communication technologies, video streaming services and applications are increasing rapidly. However, the network condition is volatile. In order to provide users with better quality of service, it is necessary to develop an accurate and low-complexity model for Quality of Experience (QoE) prediction of time-varying video. Memory effects refer to the psychological influence factor of historical experience, which can be taken into account to improve the accuracy of QoE evaluation. In this paper, we design subjective experiments to explore the impact of Short-Term Memory (STM) on QoE. The experimental results show that the user’s real-time QoE is influenced by the duration of previous viewing experience and the expectations generated by STM. Furthermore, we propose analytical models to determine the relationship between intrinsic video quality, expectation and real-time QoE. The proposed models have better performance for real-time QoE prediction when the video is transmitted in a fluctuate network. The models are capable of providing more accurate guidance for improving the quality of video streaming services.

        Extracellular products-mediated interspecific interaction between Pseudomonas aeruginosa and Escherichia coli

        The Gram-negative pathogen Pseudomonas aeruginosa adopts several elaborate strategies to colonize a wide range of natural or clinical niches and to overcome the neighboring bacterial competitors in polymicrobial communities. However, the relationship and interaction mechanism of P. aeruginosa with other bacterial pathogens remains largely unexplored. Here we explore the interaction dynamics of P. aeruginosa and Escherichia coli, which frequently coinfect the lungs of immunocompromised hosts, by using a series of on-plate proximity assays and RNA-sequencing. We show that the extracellular products of P. aeruginosa can inhibit the growth of neighboring E. coli and induce a large-scale of transcriptional reprogramming of E. coli, especially in terms of cellular respiration- related primary metabolisms and membrane components. In contrast, the presence of E. coli has no significant effect on the growth of P. aeruginosa in short-term culture, but causes a dysregulated expression of genes positively controlled by the quorum-sensing (QS) system of P. aeruginosa during subsequent pairwise culture. We further demonstrate that the divergent QS-regulation of P. aeruginosa may be related to the function of the transcriptional regulator PqsR, which can be enhanced by E. coli culture supernatant to increase the pyocyanin production by P. aeruginosa in the absence of the central las-QS system. Moreover, the extracellular products of E. coli promote the proliferation and lethality of P. aeruginosa in infecting the Caenorhabditis elegans model. The current study provides a general characterization of the extracellular products-mediated interactions between P. aeruginosa and E. coli, and may facilitate the understanding of polymicrobial infections.

        Molecular Cloning, mRNA Expression, and Localization of the G-protein Subunit Galphaq in Sheep Testis and Epididymis

        The reproductive function of G-protein subunit Galphaq (GNAQ), a member of the G protein alpha subunit family, has been extensively studied in humans and rats. However, no data is available on its status in ruminants. The objectives of this study were to evaluate the expression pattern of the GNAQ in the testis and epididymis of sheep by polymerase chain reaction (PCR). The mRNA expression levels were detected by real-time fluorescent quantitative PCR, and cellular localization of GNAQ in the testis and epididymis was examined by immunohistochemistry. Additionally, GNAQ protein was qualitatively evaluated via western blot, with the results indicating that similarities between GNAQ mRNA levels from sheep was highly conserved with those observed in Bos taurus and Sus scrofa. Our results also indicated that GNAQ exists in the caput and cauda epididymis of sheep, while GNAQ in the testis and epididymis was localized to Leydig cells, spermatogonial stem cells, spermatocytes, Sertoli cells, spermatid, principal cells, and epididymis interstitial cells. The concentrations of GNAQ mRNA and protein in the caput and cauda epididymis were significantly greater than those observed in the corpus epididymis (p<0.01) and testis (p<0.05). Our results indicated that GNAQ exists at high concentrations in the caput and cauda epididymis of sheep, suggesting that GNAQ may play an important role in gonad development and sperm maturation.
