        The Fantasy Garden in East Asian Tradition: A Case Study of a Garden Record in Seo Yugu’s Treatises on Rural Living

        This paper presents an annotated translation and hermeneutic interpretation of a garden record in Seo Yugu’s nineteenth-century Korean encyclopedia Treatises on Rural Living (Imwon gyeongje ji 林園經濟志). This particular garden record is about a fantasy garden called To-Become Garden (Jangchwiwon 將就園). The translation, historical annotations, and hermeneutic interpretation presented in this paper help define the concept of a fantasy garden in East Asian architecture and uncover the Taoist hermitage in the humanistic, social, and environmental context of premodern Korea, at a time when Korea was experiencing a philosophical shift from traditional Neo-Confucianism to the emerging social movement of “practical learning” (Sirhak). This paper demonstrates that Seo’s encyclopedic composition of garden theories mixes his cultural perception of Korean mountain landscapes, his life experience of a practical hermitage, and his fantasy of a Taoist paradise. A comparison is drawn between Seo’s philosophy of life and Western critical philosophy at the dawn of modernity.

        Expression and Purification of a Functional Recombinant Aspartate Aminotransferase (AST) from Escherichia coli

        Aspartate aminotransferase (AST; E.C. 2.6.1.1), a vitamin B6-dependent enzyme, preferentially promotes the mutual transformation of aspartate and α-ketoglutarate to oxaloacetate and glutamate. It plays a key role in amino acid metabolism and has been widely recommended as a biomarker of liver and heart damage. Our study aimed to evaluate the extensive preparation of AST and its application in quality control in clinical laboratories. We describe a scheme to express and purify the 6His-AST fusion protein. An optimized sequence coding AST was synthesized and transformed into Escherichia coli BL21 (DE3) strain for protein expression. Ideally, the fusion protein has a volumetric productivity achieving 900 mg/l cultures. After affinity chromatography, the enzyme activity of purified AST reached 150,000 U/L. Commutability assessment between the engineered AST and standard AST from Roche suggested that the engineered AST was the better candidate for the reference material. Moreover, the AST showed high stability during long-term storage at -20ºC. In conclusion, the highly soluble 6His-tagged AST can become a convenient tool for supplying a much better and cheaper standard or reference material for the clinical laboratory.

        Physical Layer Security for Two-Way Relay NOMA Systems with Energy Harvesting

        Due to the wide application of fifth generation communication, wireless sensor networks have become an indispensable part in our daily life. In this paper, we analyze physical layer security for two-way relay with energy harvesting (EH), where power splitter is considered at relay. And two kinds of combined methods, i.e., selection combining (SC) and maximum ratio combining (MRC) schemes, are employed at eavesdropper. What’s more, the closed-form expressions for security performance are derived. For comparison purposes, this security behaviors for orthogonal multiple access (OMA) networks are also investigated. To gain deeper insights, the end-to-end throughput and approximate derivations of secrecy outage probability (SOP) under the high signal-to-noise ratio (SNR) regime are studied. Practical Monte-Carlo simulative results verify the numerical analysis and indicate that: i) The secure performance of SC scheme is superior to MRC scheme because of being applied on eavesdropper; ii) The secure behaviors can be affected by various parameters like power allocation coefficients, transmission rate, etc; iii) In the low and medium SNR region, the security and channel capacity are higher for cooperative non-orthogonal multiple access (NOMA) systems in contrast with OMA systems; iv) The systematic throughput can be improved by changing the energy conversion efficiency and power splitting factor. The purpose of this study is to provide theoretical direction and design of secure communication.

        Low-Temperature Synthesis of Near-Monodisperse Globular MoS2 Nanoparticles with Sulphur Powders

        Nanoparticles (NPs) with high uniformity have been extensively investigated for their excellent chemical stability. Near-monodisperse globular MoS2 NPs were prepared with sulphur powders (SPs) as a sulphur source by a one-pot polyol-mediated process without surfactants, transfer agents and toxic agents at 170–190 ℃. The as-processed SPs greatly affected the formation of the MoS2 NPs after low-activity sulphur (S8)n was reassembled from common SPs (S8). The average size of MoS2 NPs can be reduced remarkably from 100–200 nm to 50 nm by introducing low amounts of MnCl2. A preliminary four-step growth mechanism based on the aggregation-coalescence model was also proposed. This green and simple method may be an alternative to the common hot-injection and heating-up methods for the preparation of monodisperse NPs, particularly transition metal dichalogenides.

        Effects of Androgen Receptor Overexpression on Chondrogenic Ability of Rabbit Articular Chondrocytes

        Background: The role of sex hormones and their receptors has drawn much attention in the process of cartilage regeneration. This study aimed to investigate the effect of androgen receptor (AR) on the chondrogenic ability of articular chondrocytes and the related mechanism. Methods: Articular chondrocytes were isolated, cultured, identified by toluidine blue staining and then transduced with lentivirus carrying the AR gene. The cell viability was determined using Cell Counting Kit-8, and cell apoptosis was assessed by flow cytometry analysis. The effects of AR overexpression on the expression of cartilage-specific proteins and some signalling molecules were evaluated by real-time PCR and Western blotting. Using 24 New Zealand rabbits, the regeneration of rabbit articular cartilage defects was further investigated in vivo and evaluated histologically. Results: The overexpression of AR significantly reduced the apoptosis rate of chondrocytes but did not affect their proliferation. The overexpression of AR also promoted the expression of Sry-related HMG box 9, collagen II and aggrecan, decreased the expression of matrix metalloproteinase-13, and downregulated p-S6 and RICTOR. The experimental group with AR-overexpressing chondrocytes exhibited superior regeneration of cartilage defects. Conclusion: AR overexpression can maintain the phenotype of chondrocytes and promote chondrogenesis in vitro and in vivo. mTOR-related signalling was inhibited.       • Association of Leptin Receptor Lys109Arg and Gln223Arg Polymorphisms with Increased Risk of Clear Cell Renal Cell Carcinoma

        Background: Although roles of genetic polymorphisms of leptin receptor (LEPR) gene in several cancers have been documented, the association between polymorphisms of LEPR and clear cell renal cell carcinoma (CC-RCC) remains unknown. The aim of this study was to explore any relation. Materials and Methods: The study population consisted of 77 patients with CC-RCC and 161 healthy control subjects. Polymorphism analyses of Lys109Arg and Gln223Arg were performed by direct DNA sequencing and PCR-restriction fragment length polymorphism approaches respectively. Results: Comparisons of allelic and genotypic frequencies in Lys109Arg and Gln223Arg showed no significant difference between the cases and controls. However, when evaluating the combined genotype of Lys109Arg and Gln223Arg, risk with GG/GG was increased (OR=1.85, 95%CI=1.04-3.30) and with GA/GG or GG/GA was decreased (OR=0.07, 95%CI=0.01-0.54; OR and 95%CI of the latter could not be calculated for a value of zero). Furthermore, the G-G haplotype frequency of Lys109Arg and Gln223Arg in the cases was higher (OR=1.68; 95%CI=1.02-2.76). In contrast, the A-G and G-A haplotype frequencies in the cases were lower than those in the controls (OR=0.06; 95%CI=0.01 to 0.47; OR and 95%CI of the latter could not be calculated for a value of zero). In addition, the Lys109Arg A allele was in LD with the Gln223Arg A allele (d'=0.9399) in the CC-RCC subjects, but not in the controls. Conclusions: Our data suggest that the GG/GG combined genotype and G-G haplotype of Lys109Arg and Gln223Arg can act as evaluating factors for CC-RCC risk.

        Advice on Standardized Diagnosis and Treatment for Spinal Diseases during the Coronavirus Disease 2019 Pandemic

        Coronavirus disease 2019 (COVID-19) outbreak started in December 2019 that caused difficulties for clinical work. Practical work experience in our spinal outpatient and emergency department during the COVID-19 pandemic is summarized in this article, with combined evidence-based medical evidence to explore a standardized process of diagnosis and treatment for spinal diseases. Outpatient reservation, continuous screening, triage, and isolation, first consultation accountability system, pandemic reporting system, and online revisit were strictly followed. We hope that our experience in prevention and control of COVID-19 can help spine surgeons globally in stopping the spread of COVID-19. Spine surgeons should collaborate with infection control specialists to avoid cross-infection in hospitals and optimize treatment.
